Air pollution is a significant public health issue that impacts lung health, particularly in vulnerable populations such as children, the elderly, and individuals with pre-existing respiratory conditions. Both natural and anthropogenic sources of air pollution give rise to a variety of toxic compounds, including particulate matter (PM), ozone (O₃), sulfur dioxide (SO₂), nitrogen dioxide (NO₂), carbon monoxide (CO), and polycyclic aromatic hydrocarbons (PAHs). Exposure to these pollutants is strongly associated with the development and exacerbation of respiratory diseases, including asthma, chronic obstructive pulmonary disease (COPD), lung cancer, and idiopathic pulmonary fibrosis (IPF). Notably, early life exposure to pollutants such as …

The Development Of The Assessment Of Self-Care (Asc), Ashley R. Righetti
Clinical Psychology Dissertations
Self-care is a topic that has piqued the interest of researchers in recent years with much of the focus being on specific subsets of the population such as helping professions and those with chronic conditions. This trend has been consistent when it comes to the development of measures intended to evaluate people’s self-care practices. Designed with Butler and colleagues’ (2019) self-care model in mind, the Assessment of Self-Care (ASC) was developed to broadly assess self-care practices in the general U.S. adult population. This study discusses the development and initial validation of the ASC. After the item generation phase, exploratory factor …

Human Adipose Stem Cell-Derived Small Extracellular Vesicles Modulate Glial Cell Response In Aged And Young Tbi Mouse Models, Salma S. Abdelmaboud
USF Tampa Graduate Theses and Dissertations
Traumatic brain injury (TBI) is a leading cause of long-term neurological impairment, with aging significantly increasing vulnerability and worsening outcomes. Older individuals experience greater cognitive and motor deficits after TBI and often show diminished responses to therapeutic interventions. Both aging and TBI independently promote heightened neuroinflammation and cognitive dysfunction, increasing susceptibility to poor recovery. This study investigated the therapeutic potential of small extracellular vesicles (sEVs) derived from human adipose-derived stem cells (hASCs) to promote neurological recovery and modulate neuroinflammation in a mouse model of TBI. Male C57BL/6 mice aged 3, 15, and 20 months underwent controlled cortical impact (CCI) and …

Evidence links air pollution to dementia, yet its role in Lewy body dementia (LBD) remains unclear. Here we showed in a cohort of 56.5 million individuals across the U.S. that PM2.5 exposure raises LBD risk. Mechanistically, we found PM2.5 exposure led to brain atrophy in wild-type mice, an effect not seen in α-synuclein (αSyn)-deficient mice. PM2.5 exposure generated a highly pathogenic αSyn strain, PM-PFF, with enhanced proteinase K-resistance and neurotoxicity, resembling αSyn LBD strains. PM2.5 samples from China, the U.S., and Europe consistently induced proteinase-resistant αSyn strains and in vivo pathology. Transcriptomic analyses revealed shared responses between PM2.5-exposed mice and …

HPV integration disrupts host genomic structure and expression, but whether these alterations promote cancer development remains unclear. Multiple genomic analyses of oropharyngeal cancers identified several host fusion genes, including recurrent FGFR3-TACC3 fusions, expressed from rearranged genomic loci adjacent to HPV integration sites. Evolutionary modeling implicated integration of virus concatemers into the host genome as a common initiating event in fusion formation. Co-expression of HPV16 E6/E7 and FGFR3-TACC3, but neither alone, was sufficient for tumor development in both xenograft and syngeneic mouse models and led to unique transcriptional programs implicated in carcinogenesis. FGFR3-TACC3 expression decreased the ubiquitination and degradation of …

To explore how early can cancers be detected prior to clinical signs or symptoms, we assessed prospectively collected serial plasma samples from the Atherosclerosis Risk in Communities (ARIC) study, including 26 participants diagnosed with cancer and 26 matched controls. At the index time point, eight of these 52 participants scored positively with a multicancer early detection (MCED) test. All eight participants were diagnosed with cancer within 4 months after blood collection. In six of these 8 participants, we were able to assess an earlier plasma sample collected 3.1 to 3.5 years prior to clinical diagnosis. In four of these six …

An Investigation Of Sex-Specific Effects Of Nucleus Accumbens Dopamine Receptor Signaling In Persistent Reward Seeking Under Punishment, Anna Caroline Toburen
An Investigation Of Sex-Specific Effects Of Nucleus Accumbens Dopamine Receptor Signaling In Persistent Reward Seeking Under Punishment, Anna Caroline Toburen
Theses and Dissertations
Addiction involves persistent reward-seeking behavior despite adverse outcomes, with the nucleus accumbens (NAc) critically mediating motivation and cost-benefit decision-making. Dopamine (DA)-responsive neurons in the NAc, specifically D1 and D2 receptors within the NAc are crucial in encoding motivated behavior. However, the specific role of D1/D2 receptors in driving reward-seeking under punishment remains unclear. Preliminary findings suggest a sex-specific effect, with females exhibiting greater sensitivity to the suppressive effects of footshock on natural reward-seeking compared to males. We investigated the differential roles of NAc D1/D2 receptors in modulating reward-seeking behavior under punishment, with a focus on potential sex-specific effects. Using a …

We present the Causal Pivot (CP) as a structural causal model (SCM) for analyzing genetic heterogeneity in complex diseases. The CP leverages an established causal factor or factors to detect the contribution of additional suspected causes. Specifically, polygenic risk scores (PRSs) serve as known causes, while rare variants (RVs) or RV ensembles are evaluated as candidate causes. The CP incorporates outcome-induced association by conditioning on disease status. We derive a conditional maximum-likelihood procedure for binary and quantitative traits and develop the Causal Pivot likelihood ratio test (CP-LRT) to detect causal signals. Through simulations, we demonstrate the CP-LRT’s robust power and …

Climate Change-Related Threats To Railroads: Implications For Threat, Hazard And Risk Assessment, Frances L. Edwards, Daniel C. Goodrich
Faculty Research, Scholarly, and Creative Activity
Climate change has created new challenges for railroads around the world. The rail systems operate in open environments, subjecting them to increased heat, changing locations of heavy rains, developing sea level rise and storm surge, and a longer and stronger wildfire season. This research investigates how an updated threat and hazard identification and risk assessment (THIRA) can lead to the development of climate adaptation strategies that limit damage and disruption to the railroad enterprise. The THIRA analyzes which threats are of most concern in a specific rail segment, and which climate change adaptation strategies give a beneficial return on investment.
