Direct electrical stimulation of the human brain has been used for numerous clinical and scientific applications. At present, however, little is known about how intracranial stimulation affects activity at the microscale. In this study, we recorded intracranial EEG data from a cohort of patients with medically refractory epilepsy as they completed a visual recognition memory task. During the memory task, brief trains of intracranial theta burst stimulation (TBS) were delivered to the basolateral amygdala (BLA). Using simultaneous microelectrode recordings, we isolated neurons in the hippocampus, amygdala, orbitofrontal cortex, and anterior cingulate cortex and tested whether stimulation enhanced or suppressed firing …

BACKGROUND: Properly characterizing biomechanics in populations with known hip deformities, such as developmental dysplasia of the hip (DDH), requires accurately identifying the hip joint center (HJC). Many studies have assessed the accuracy or repeatability of predictive and functional methods to locate the HJC in normally shaped hips, but not for hips with DDH.
RESEARCH QUESTION: For patients with DDH, which of three common functional and four common predictive HJC methods is most accurate compared to a magnetic-resonance-imaging-based anatomic method?

Addressing employee turnover and boosting retention are major challenges for companies looking to achieve long-term success. Project managers are crucial in tackling this issue, as high turnover can undermine team performance and increase operational costs. Grounded in Herzberg’s two factor theory, the purpose of this qualitative case study was to explore strategies that some project managers use to reduce high employee turnover. The participants included six project managers in Mississippi who had an active role in using strategies to increase employee retention. The current study sought to explain the heterogeneity of social impairment among youth with symptoms of ADHD with the goal of potentially improving a clinician's ability to identify and support those children. A novel model of the relationship between the disorder and social impairment was proposed with serial mediators of parental/family functioning and emotion regulation. It was anticipated that a higher level of ADHD symptoms would lead to more impairment among aspects of parental/family functioning, which would predict more emotion dysregulation which would subsequently predict poorer social functioning. Protein homeostasis, or proteostasis, is responsible for a folded and functional proteome and is essential for cell survival. Proteins can misfold during stress or because of mutation. The accumulation of misfolded proteins is observed in many diseases, including Huntington’s disease. Misfolded proteins will clump together into inclusions, which can be sequestered to membrane-less compartments by proteostasis mechanisms. Several proteins, including chaperones, co-chaperones, and other proteins, have been identified as sequestrases that target inclusions and misfolded proteins to different protein quality control (PQC) compartments. The juxtanuclear quality control (JUNQ) compartment forms in the nucleus-vacuole junction in yeast cells, and several soluble …

Introduction: Trainees experiencing burnout, depression, or career dissatisfaction challenge educators and may have patient care consequences. Grit, defined as the “perseverance and passion for long-term goals,” buffers against hopelessness and quantifies an aspect of personality shown to predict fewer career changes. Higher grit has been associated with positive outcomes in physicians, including lower burnout rates. Our objective was to determine the correlation between emergency medicine (EM) resident Short Grit Scale (Grit-S) scores and components of well-being: burnout, a positive depression screen, and career satisfaction.

RATIONALE: Alkyl nitrites ("poppers") are recreational substances known for their vasodilatory and euphoric effects. Increasing evidence links them to a vision-threatening condition termed poppers maculopathy, characterized by foveal yellow spots and outer retinal changes on optical coherence tomography. While most reports describe acute exposure, little is known about long-term consequences. This case describes one of the longest documented durations of chronic poppers use, combined with a literature review to contextualize the clinical spectrum.

Excess turnover of personnel threatens organizational viability and long-term success. Human resource leaders need to develop strategies to increase employee retention. Grounded in Herzberg’s two-factor and Maslow’s hierarchy of needs theories, the purpose of this qualitative pragmatic inquiry was to explore and identify successful employee retention strategies employed by human resource leaders to mitigate employee turnover. Data were collected from human resources managers using semistructured interviews and public data from companies’ websites. Alzheimer's Disease Dementia (ADD) prognosis is an unmet medical need. Mitochondrial dysfunction is an early AD etiopathogenic factor. The present study analyzed mitochondrial DNA (mtDNA) methylation patterns in blood samples from patients with mild cognitive impairment (MCI) who progressed to ADD (P), MCI remained stable (NP), and Cognitively Normal (CN) individuals. Differentially methylated sites were identified in the D-loop region in both CN vs. NP and NP vs. P comparisons, even before β-amyloid positivity. BACKGROUND: Diagnosis of autism spectrum disorder (ASD) significantly improves quality of life, yet current diagnostic practices rely on subjective behavioural assessments. While machine learning models using functional Magnetic Resonance Imaging (fMRI) show promise for objective diagnosis, existing approaches have critical limitations. High-accuracy models often neglect interpretability, increasing clinical distrust, while interpretable approaches frequently suffer from low accuracy and lack neuroscientific validation of identified biomarkers. Furthermore, no studies have systematically benchmarked which interpretability methods are most effective for fMRI data, hindering the translation of Artificial intelligence (AI) findings into clinical practice.

Financial inducements such as free bets are frequently-used gambling marketing offers which temporarily improve a gambler’s usual pattern of risk and potential return. Previous research has shown that there are up to 15 distinct types of financial inducements in common use, and that gamblers frequently misunderstand inducements’ play-through requirements and other complex terms and conditions. The Australian government has therefore recently banned play-through requirements for inducements shown to new customers, and the Great British regulator the Gambling Commission has recently announced a maximum play-through requirement of 10 times. Deep-space missions, such as those planned for the cis-lunar space and Mars, pose unique challenges to astronaut health due to prolonged microgravity (MG) exposure and low-dose chronic irradiation (CIR). Understanding the impact of these stressors on secondary lymphoid tissues, such as the spleen and intestine, is crucial for ensuring crewmembers' well-being, given the impracticality of conducting this research with human samples. Background. – This study aims to evaluate the impact of obesity on post–liver transplant outcomes stratified by low- and high-performance status. Methods. – Patients undergoing liver transplantation between 2006 and 2023 were identified in the United Network for Organ Sharing data set. Patients with severe obesity body mass index ([BMI] ≥40 kg/m2) and super obesity (BMI >50 kg/m2) were compared with those with lower BMI on perioperative and long-term outcomes stratified by preoperative performance status. Introduction: Ischemia-induced retinopathies, such as retinopathy of prematurity and proliferative diabetic retinopathy, are major causes of vision loss. Both pathological neovascularization and neuronal damage contribute to vision impairment in these conditions, yet current therapies primarily target neovascularization and may further compromise neuronal health.
Objectives: Cyclic AMP (cAMP) is a widely recognized second messenger regulating diverse physiological and pathological processes. Exchange protein directly activated by cAMP (Epac) is a recently identified effector of cAMP signaling. Purpose: Persistent ulnar-sided wrist pain, often because of triangular fibrocartilage complex (TFCC) tears, can lead to instability of the distal radioulnar joint (DRUJ) and the ulnocarpal joint (UCJ). Although the TFCC's role in stabilizing the DRUJ is well established, its contribution to UCJ stability has gained increasing recognition. This study evaluates the clinical outcomes of combined Herbert Sling and suture repair for (UCJ) instability secondary to TFCC tears.

This thesis consists of two empirical studies on elderly mental health and on health behaviors. The first study investigates the long-term impact of childhood hunger on mental health in later life, using data from two nationally representative surveys in China. The study finds that childhood hunger exposure significantly increases the risks of depression and anxiety in later life. These effects are most pronounced among individuals exposed after age 5, suggesting that autobiographical memory formation plays an important role beyond socioeconomic and health pathways. Fetal congenital anomalies and genetic disorders complicate 3%-5% of pregnancies and can have a significant impact on pregnancy outcomes. Precise and individualized prenatal diagnosis is crucial for effective counseling and management. Cerebral hemodynamic imaging is increasingly used as a non-invasive technique to study brain function and dysfunction in both research and clinical settings. Recent advances in imaging technologies have enabled high-speed, high-resolution, whole-brain measurements of blood flow and oxygenation. Improving the interpretation of these cerebral hemodynamic signals through a mechanistic understanding of their cellular drivers would enable more accurate characterization of brain function and pathology. The number of reliable and non-invasive techniques to characterize ocular blood flow is limited. Here, we present a protocol for identifying a soft tissue window in the orbital aperture using pulsed-Doppler ultrasonography to offer real-time, non-invasive blood flow velocity monitoring in the mouse ophthalmic artery. We describe steps for preparing a mouse, positioning the ultrasound probe, and acquiring and recording signals. We then detail procedures for analyzing results using a workstation equipped with Doppler flow velocity software. Age-related hearing loss (ARHL) is a major public health concern, driven by the interplay of multiple factors. Here, we reveal spatiotemporal metabolic shifts in murine inner ears mirroring erythrocytes and plasma. Isotope-labeled glucose tracing demonstrates a metabolic rerouting favoring glycolysis over the pentose phosphate pathway, alongside downregulation of the tricarboxylic acid cycle, indicating impaired energy production and redox homeostasis. Accumulation of medium- and long-chain acylcarnitines further exacerbates lipotoxicity. Notably, age-dependent depletion of arginine, lysine, proline, and glycine disrupts the arginine-polyamine-urea cycle. Translationally, UK Biobank plasma metabolomics links omega-6 fatty acids, linoleic acid, glycine, and albumin to ARHL resilience, while branched-chain …

Acute coronary syndrome (ACS) is a major cause of morbidity and mortality in the United States (US). However, the trends and disparities in ACS mortality are not well understood. This study aimed to analyze the ACS mortality in the US from 1999 to 2020 using the Centers for Disease Control and Prevention Wide-Ranging Online Data for Epidemiologic Research database. The age-adjusted mortality rates (AAMR) due to ACS were calculated for each year from 1999 to 2020, stratified by sex, race, urbanization, and state.
