Hydralazine (HYZ), a treatment for preeclampsia and hypertensive crisis, is listed by the World Health Organization as an essential medicine. Its mode of action has remained unknown through its seven decades of clinical use. Here, we identify 2-aminoethanethiol dioxygenase (ADO), a key mediator of targeted protein degradation, as a selective HYZ target. The drug chelates ADO's metallocofactor and can alkylate one of its ligands. The resultant inactivation stabilizes regulators of G protein signaling (RGS4 and RGS5) that ADO normally marks for proteolysis, explaining the drug's vasodilatory activity and comporting with observations of diminished RGS levels in both clinical preeclampsia and …

Skeletal muscle regeneration in chronic muscle diseases such as Duchenne Muscular Dystrophy (DMD) has remained clinically unsurmountable. Estrogen‐related receptor alpha (ERRα) plays a critical role in adult skeletal muscle metabolism and exercise fitness. Whether ERRα activation can drive muscle regeneration and mitigate dystrophy in DMD is not known. We have investigated ERRα signaling in pre‐clinical models of acute muscle injury and DMD. ERRα is induced in differentiating C2C12 myoblast and regenerating muscle. ERRα silencing suppressed proliferation and differentiation in C2C12 myoblasts. Background: The vast amount of natural language clinical notes about patients with cancer presents a challenge for efficient information extraction, standardization, and structuring. Traditional NLP methods require extensive annotation by domain experts for each type of named entity and necessitate model training, highlighting the need for an efficient and accurate extraction method.

Methods: This study introduces a tool based on the Large Language Model (LLM) for zero-shot information extraction from cancer-related clinical notes into structured data aligned with the minimal Common Oncology Data Elements (mCODE™) structure. International Journal of Aquatic Research and Education

Black youth in the United States disproportionately experience fatal drowning at rates up to five times higher than their white peers. Eliminating these inequities requires new approaches that address historical and structural barriers of learning to swim. The objective of this study was to evaluate the three-week long swimming component of a community-based discovery program to empower Black youth in Evanston, IL. Participants were Black youth ages 8-10 years enrolled in summer 2023 and 2024. Swim skills were assessed by instructors using the Pictorial Scale of Perceived Water Competence (PSPWC). The silent epidemic of childhood drowning, and the racial disparities associated with it require scalable and innovative approaches that both break cycles of exclusion and circumvent our societal tendency to overlook drowning as a matter in need of urgent resolution. No More Under, a Seattle-based water safety organization whose mission is to eradicate childhood drowning, has launched the Swim Safely Series. The series is a national program uniquely poised to overcome the various barriers associated with racial and socioeconomic drowning disparities by tapping into underutilized collegiate aquatic spaces and merging water safety with mentorship and social intervention.

Background: Visuospatial function is commonly impaired post-stroke and is associated with motor learning and recovery. A single, twenty-minute Corsi Block Tapping Task (CBTT) training session improved visuospatial function in young neurotypical adults; however, it is unclear whether this training would improve visuospatial function in adults with stroke.

Objective: To understand if a single, twenty-minute CBTT training session improved visuospatial function in adults with stroke compared to a no-training control group of adults with stroke.

Northern hardwood forests of the Lake Superior region face a series of novel disturbance pressures including canopy dieback. Previous studies have linked regional sugar-maple (Acer saccharum) canopy dieback to introduced earthworms, which may have coinciding impacts on the ground-layer plant community. Dieback–earthworm interactions may lead to important longer-term changes in forest structure and function, but these relationships but have not been characterized. We sampled ground-layer plant communities in five national forest units in Michigan, Wisconsin, and Minnesota in 2010, and again just over a decade later in 2021. Learning About Stability Of Risk Preferences, Levon Barseghyan, Francesca Molinari, Joshua C. Teitelbaum

Classical theories posit that risk preferences are stable across decision contexts. Two branches of empirical literature assess this claim. Structural approaches examine within-person consistency of model-based estimates of risk aversion, while “model-free” approaches examine within-person correlations of risky choices. We elucidate the latent structure underlying the model-free approach. Using this structure, we develop a new approach to assessing preference stability that has stronger testable implications, and we partially identify features of stability and heterogeneity of risk preferences. Our study illustrates a general principle: partial identification through minimal assumptions grounded in economic theory robustly bridges fully structural and model-free methods.

Multiplexed imaging allows multiple cell types to be simultaneously visualised in a single tissue sample, generating unprecedented amounts of spatially-resolved, biological data. In topological data analysis, persistent homology provides multiscale descriptors of “shape" suitable for the analysis of such spatial data. Here we propose a novel visualisation of persistent homology (PH) and fine-tune vectorisations thereof (exploring the effect of different weightings for persistence images, a prominent vectorisation of PH). These approaches offer new biological interpretations and promising avenues for improving the analysis of complex spatial biological data especially in multiple cell type data. In this study, a chitosan/copper (Ch/Cu²⁺) polymetallic complex was synthesized from chitosan obtained by the cryogenic method from Apis mellifera bees. The structural and physicochemical properties of the complex were investigated using FTIR, UV-Vis, XRD, TGA, and DLS analyses. FTIR spectra showed shifts in absorption bands at 1400-1339 cm⁻¹ and 1640-1550 cm⁻¹, confirming the coordination of Cu²⁺ ions with amino and hydroxyl groups. UV-Vis spectra revealed absorption peaks in the 410-540 nm range, while XRD analysis indicated new crystalline peaks at 2θ = 24°, 27°, and 37°. In the world, the air-dried food market is seeing rapid expansion, valued at USD 103.26 billion in 2021, out of which the Asia Pacific region consumed the maximum amount. The freeze-dried vegetable and fruit market is predicted to reach USD 132.7 billion by 2027. Processed vegetables are also exported to a large extent from India. Uzbekistan recorded excellent development in agriculture in 2024, with record harvests, better yields, and use of advanced machinery. China’S Turn To Domestic-Led Growth: Replacing Consumption Taxes With Land Taxes?, Yilin Hou, Michael Kumhof, Lei Shao

As part of its turn towards domestic-led growth China plans to stimulate consumption, including through a reduction in consumption taxes. We compare two model-based scenarios in which the revenue losses from lower consumption taxes are replaced by higher income taxes or higher land taxes. The model is carefully calibrated to recent Chinese data, including detailed fiscal data. We find that higher income taxes lead to significant losses in output and incomes, while higher land taxes lead to significant gains. These gains are increasing in the share of land in net wealth, a statistic for which better data would be invaluable.

BACKGROUND: The 2023-2024 influenza season included sustained elevated activity from December 2023 to February 2024 and continued activity through May 2024. Influenza A(H1N1), A(H3N2), and B viruses circulated during the season.

METHODS: During 1 September 2023 to 31 May 2024, a multistate sentinel surveillance network of 24 medical centers in 20 US states enrolled adults aged ≥18 years hospitalized with acute respiratory illness. Consistent with a test-negative design, cases tested positive for influenza viruses by molecular or antigen test, and controls tested negative for influenza viruses and severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). The Canadian government has a long history of regulation, exploitation, and violence against lesbian, gay, bisexual, trans, queer, and two-spirit (LGBTQ2S+) people. One of the most painful chapters in this history is the “LGBT Purge,” a term that refers to the expulsion of LGBTQ2S+ service members and employees from the Canadian Armed Forces, Royal Canadian Mounted Police, and Federal Public Service between 1955 and 1992. The LGBT Purge was the subject of a class action lawsuit filed in 2017 that resulted in a settlement agreement in 2018. Racial profiling is one of the most enduring problems in policing. Yet it remains largely under-theorized, which generates important theoretical and practical implications. Racial profiling tends to be construed as an arbitrary detention rather than a form of unconstitutional discrimination. For this reason, the section 15 Charter right to equality plays little to no role in most leading cases on racial profiling. The legal framework that governs racial profiling lacks clarity and can be applied inconsistently. And the remedial landscape associated with racial profiling claims has evolved minimally. The racial reckoning of 2020 and 2021 had profound effects on people. This study uses a retrospective design to examine differences in critical consciousness prior to 2020 and contemporarily among counseling students and professionals and whether critical consciousness predicts multicultural competence. Participants reported their current level of multicultural competence, and pre-2020 (retrospective) and current (contemporary) levels of critical reflection, critical motivation, critical action, and race-based critical consciousness. Participants reported higher levels on the contemporary critical consciousness measures than their retrospective assessments, suggesting that the high-impact events of 2020 raised their critical consciousness. In this workshop, we’ll talk about how mental health impacts our students’ abilities to engage in our classes and absorb class material/readings. Without requiring instructors to take on the emotional labor of managing students’ mental health, we’ll discuss things instructors can do to improve student learning by examining and reframing our pedagogical choices. OBJECTIVES: Antimicrobial resistance is a critical public health threat. Large language models (LLMs) show great capability for providing health information. This study evaluates the effectiveness of LLMs in providing information on antibiotic use and infection management.

METHODS: Using a mixed-method approach, responses to healthcare expert-designed scenarios from ChatGPT 3.5, ChatGPT 4.0, Claude 2.0 and Gemini 1.0, in both Italian and English, were analysed. Computational text analysis assessed readability, lexical diversity and sentiment, while content quality was assessed by three experts via DISCERN tool.
