Clonal hematopoiesis (CH) is detectable in upwards of 20% of patients with solid tumors and is associated with worsened prognosis; however, its role in tumor immunology and immune checkpoint therapy (ICT) is unknown. Using a bone marrow chimera model of Tet2+/mut CH in mice with solid tumors, we found the Tet2-mutant myeloid cells are abundant in the tumor microenvironment and contributed to an improved response to ICT. Mechanistically, Tet2+/mut macrophages inside the tumor act as immunogenic antigen-presenting cells that more effectively cross-prime naive CD8+ T cells in response to IFNγ. Background/objectives: Progression of pancreatic ductal adenocarcinoma (PDAC) and other carcinomas relies on cancer-associated fibroblasts (CAFs). A subset of CAFs is derived from adipose stromal cells (ASCs) recruited by tumors and the ASC-CAF conversion has been associated with invasiveness and poor prognosis.

Methods: To explore the underlying molecular mechanisms, we used a model based on primary ASCs derived from human visceral adipose tissue co-cultured with human PDAC cell line Capan-1. To investigate cancer progression in vivo, we also used mice orthotopically grafted with mouse KPC cells.

Objectives: This study aimed to determine the incidence, outcomes and risk factors of arrhythmia in children undergoing congenital heart disease (CHD) surgery in Oman. Methods: This prospective cohort study was conducted between 2023 and 2024 at the National Heart Centre, Muscat, Oman. Paediatric patients who underwent CHD surgery were included; those with preoperative arrhythmia or permanent pacemakers were excluded. Demographic, intraoperative and postoperative variables were collected. Univariate and multivariate logistic regression analyses were performed to identify independent predictors of postoperative arrhythmia. Introduction: Hepatocellular carcinoma (HCC) can develop in individuals with metabolic dysfunction-associated steatotic liver disease (MASLD) without cirrhosis, especially in those with elevated liver enzymes. However, there is currently no low-cost, scalable screening tool for this population. To address this need, we investigated the association between serial changes in FIB-4 (fibrosis-4) scores over a 3-year period and the risk of developing HCC in a large, diverse cohort of patients with MASLD without cirrhosis.

Aging is associated with changes in sensorimotor control that contribute to functional decline, mobility limitations, and increased fall risk. Traditional motor assessments often rely on subjective measures, highlighting the need for objective, quantitative tools. We developed three robot-based tasks using the vBOT planar manipulandum to evaluate sensorimotor performance in healthy young (<35 years) and older (>60 years) adults. These tasks uniquely combined bimanual control and altered dynamic conditions to assess age-related differences. The first task required bimanual coordination to control a virtual 2D arm over 400 center-out and return trials, targeting de novo motor learning. Individuals with history of chemo- or radiotherapy frequently exhibit somatic mosaicism in the blood, often involving mutations in genes responsible for DNA damage responses (DDR), such as CHEK2. However, the mechanisms by which CHEK2 mutations promote the expansion of mutant cells following chemo- or radiotherapy remain poorly understood. Here, we demonstrate that loss of CHEK2 confers resistance to chemotherapy in hematopoietic stem and progenitor cells (HSPCs). Through a CRISPR-based screen, we identified CHEK2 as a gene whose loss enhances resistance to cytotoxic chemotherapies. In this fact sheet, we examine the drought management options available to cattle operations on the Navajo Nation, the largest Native American reservation in the U.S. Specifically, we discuss the results of a study evaluating the economic outcomes of two common drought response strategies: herd reduction and supplemental hay purchases. We also provide recommendations for ranchers on optimal strategies, i.e., those that maximize profits, for two rancher types, differentiated by their willingness to accept (risk-neutral) or avoid risk (risk-averse).

Heterozygosity at human leukocyte antigen (HLA) loci may improve lung cancer immunosurveillance by increasing recognition of the tumor by the immune system. Previous studies utilizing data from population-level biobanks, such as the United Kingdom Biobank and FinnGen, have identified an association between germline HLA class-II heterozygosity and reduced lung cancer risk in smokers. In the present study, we evaluate the association between HLA heterozygosity and lung cancer in a large case-control study (15,302 cases; 14,580 controls) with imputed HLA allele-type information, comparing differences in HLA heterozygosity between smokers and non-smokers, amongst lung cancer subtypes, and at 2- and 4-digit HLA …

The state and federally endangered light-footed Ridgway’s rail (LFRR; Rallus obsoletus levipes) is central to regional conservation, yet the drivers of its population fluctuations remain poorly understood. The fragmented nature of coastal wetlands in Southern California is threatening the existence of endemic species like the LFRR. Tidal inundation, severe storms, and overpredation threaten the survival of LFRR. This study addresses population fluctuations by analyzing the spatial and temporal drivers of LFRR populations to inform effective, site-specific management.

BACKGROUND: Parkinson's disease (PD) is the second most common neurodegenerative disease, characterized by both motor and non-motor symptoms. Falls and fractures are common in advanced PD but the risk of fractures in newly diagnosed people with PD and the impact of treatment with levodopa is understudied.

METHODS: We investigated the risk of fractures among Medicare beneficiaries with incident PD age ≥ 67 compared to controls. Among PD cases only with continuous Part D medication coverage, we examined the effect of levodopa treatment on fracture risk. Long-distance travel is often celebrated as an opportunity for growth, but its physiological and cognitive consequences are rarely addressed in academic contexts. PURPOSE: This study investigated the physiological and cognitive effects of long-distance travel on university students journeying from the Southeastern U.S. to Hawaii. METHODS: Using a repeated measures design, participants were assessed at baseline (pre-travel) and across the first four days on island (post-travel). RESULTS: Results from a repeated measures ANOVA revealed significant disruptions in cardiovascular and stress markers. While diversity, equity, and inclusivity (DEI) principles support ethically responsible, cost-effective, evidence-supported health promotion and performance interventions, they are rarely addressed within published biomedical/behavioral science research, including in kinesiology. PURPOSE: Because journal submission guidelines could increase integration of DEI principles’ application, we sought to develop a valid and reliable procedure to quantify DEI statements within kinesiology-related journal submission guidelines. METHODS: The pilot study was conducted between 2023 and 2025. Sodium bicarbonate (SB) is a widely used supplement that can enhance repeat sprint performance; however, a common side effect is gastrointestinal (GI) distress. Exercise in the heat can cause intestinal injury and endotoxemia, but no study has examined whether SB heightens this injury. PURPOSE: To determine whether SB ingestion exacerbates intestinal discomfort, injury, and endotoxemia during repeated sprints in the heat. The efficacy of dietary nitrate supplementation for enhancing resistance exercise remains unclear, and nitrate dosing guidelines are relatively unexplored. PURPOSE: To determine whether acute and chronic dietary nitrate supplementation, provided in the form of nitrate-rich beetroot juice (BR), can improve resistance exercise performance in resistance-trained males. METHODS: Thirty-six resistance-trained males and females will be recruited. Participants will complete a standardized one-repetition maximum (1RM) test in barbell back squat and barbell bench press and a familiarization prior to experimental visits. Removing the element of anticipation has previously shown to lead to worse performance on the Landing Error Scoring System (LESS) field test. It is unknown if sex or athletic status contributes to this phenomenon. Visceral adipose tissue (VAT) is strongly associated with metabolic disease risk; however, commonly used anthropometric measures, such as body mass index (BMI), waist circumference (WC), and percentage of body fat (%BF), may not accurately estimate VAT. Chronic exposure to tobacco cigarette smoke (CS) leads to pulmonary damage, inflammation, exercise intolerance, and skeletal muscle contractile and repair dysfunction, as our group has shown that CS exposure also delays muscle repair after eccentric contractions. This study investigated whether chronic CS exposure increases the susceptibility of locomotor muscles to injury from a session of eccentric contractions, as previous studies have shown that smokers have a greater risk of muscle injuries.
