Reassessing Transketolase Assays: Methodological Considerations For Detecting Functional Thiamine Deficiency, Katie A. Edwards
Pharmacy Faculty Scholarship
Thiamine (vitamin B1) deficiency due to degradation, inadequate intake, impaired absorption, or increased metabolic demand remains prevalent in both human and animal populations. In its diphosphate form (thiamine diphosphate, TDP), thiamine serves as an essential cofactor for metabolic enzymes, including transketolase (TKT), a rate-limiting enzyme of the pentose-phosphate pathway. Measurement of TKT activity with and without exogenous TDP provides a functional assessment of thiamine utilization that serves as a surrogate for or complements direct quantification of thiamine forms. Beyond its historical role as a deficiency-screening tool, the TKT assay offers opportunities to interrogate enzyme function and cofactor dependence. Strategic variation …

A New Tool For Handling Multiracial And Multi-Identity Data In Health Research, Gabriel J. Merrin
Population Health Research Brief Series
When surveys ask about race or ethnicity, a growing number of Americans select more than one category. The multiracial population now represents over 10% of the U.S. population and is the fastest growing racial group in the country. Yet researchers routinely collapse these individuals into an “other race” category for statistical analysis, rendering specific subgroups invisible. This brief introduces CATAcode, a free software tool that helps researchers systematically explore, document, and prepare check-all-that-apply demographic data for statistical modeling. In a demonstration with over 8,000 high school students, CATAcode revealed 85 distinct racial identity combinations from just eight response options. The …

What Goals? Which Point? Whose Purpose? A Critical Engagement With Sport Internalism, Francisco Javier Lopez Frias

In this article, I critique the dominance of internalism in sport philosophy and outline an alternative theoretical framework that integrates internal and external elements while striving to transcend the dichotomous language used to conceptualize sport. The analysis begins by claiming that internalism conflates three fundamental teleological aspects of sport: goals, point, and purposes. I argue that this conflation limits internalism’s ability to explain the complexity of sporting practices. By carefully distinguishing these elements, I illustrate their distinct roles in shaping sport and explore how they interact. This report presents pre- and post-training survey results for Phase 1 of the Nebraska Department of Transportation (NDOT) grant. Law enforcement officers regularly interact with individuals who have experienced trauma. These encounters can be complex, unpredictable, and high stress. Trauma-informed policing training is intended to give officers practical tools to recognize trauma responses, communicate more effectively, and reduce unnecessary escalation during these interactions.

This mixed-methods action research study examined how implementing the CKLA, Core Knowledge Language Arts, curriculum, founded in the science of reading, enhanced the reading achievement among first-grade African American students. The purpose of the study was to explore how phonics instruction, specifically phonemic awareness, and multicultural literature would improve literacy. The study was guided by Ivankova’s six-phased mixed methods model integrating quantitative and qualitative data. The quantitative data collected included the DIBELS, Dynamic Indicators of Basic Early Literacy Skills, benchmark and progress monitoring assessments. While the field of brain aging research continues to grow at a rapid pace, there is still much to learn about how certain factors influence brain aging as well as when and how these factors can be intervened upon. Cerebrovascular and neurodegenerative diseases continue to pose a major obstacle to healthy brain aging and the better we understand the risk factors of these diseases, the greater chance we have at mitigation strategies and prevention. Healthcare systems today face the dual challenge of escalating costs and shrinking budgets, forcing administrators to scrutinize every investment in technology and medication. Even as innovations continue to advance patient safety, financial sustainability demands that hospitals allocate resources strategically to achieve the greatest impact. In the case examined, a 67-year-old man with extensive comorbidities including paraplegia, coronary artery disease, and obstructive sleep apnea, underwent sacral flap reconstruction requiring deep neuromuscular blockade. Quantitative electromyography monitoring (qNMM) with a TetraGraph device was applied concurrently with a traditional qualitative peripheral nerve stimulator (PNS). BACKGROUND: Reproductive factors are key breast cancer risk factors, yet contemporary generational patterns remain unclear. We aimed to evaluate trends in reproductive factors among US women born between 1910 and 2000.
METHODS: We conducted a serial, cross-sectional analysis of reproductive factors using data from the US National Health and Nutrition Examination Survey (NHANES) collected between 1999 and 2020. Female participants were grouped by birth cohorts (1910-1930, then by 10-year to 2000). Self-reported data were used to derive age at menarche, prevalence of age at menarche < 12 years, age at natural menopausal, prevalence of first live birth after age 30, and lifetime number of live births.

UNLABELLED: Developing rapid, non-sputum-based tests is a global priority to improve the diagnosis and timely treatment of tuberculosis (TB). In this study, we evaluated the detection limit, precision, stability, and lot-to-lot variation of a next-generation rapid diagnostic test (RDT), the plasmonic fluor (PF)-enhanced urinary lipoarabinomannan lateral flow assay (PF-LAM). We also assessed the diagnostic performance of PF-LAM, Alere Determine TB LAM Ag (AlereLAM), and electrochemiluminescence LAM assay (EclLAM), a highly sensitive laboratory-based test by comparing with microbiology reference standard (MRS) that includes molecular testing (Xpert MTB/RIF) and culture. BACKGROUND: Mitral annular calcification (MAC) is common and associated with increased cardiovascular risk and, when severe, mitral stenosis (MS). MAC-related MS differs anatomically and hemodynamically from rheumatic MS (RMS), challenging standard diagnostic methods. This study compares structural and flow characteristics, including kinetic energy losses, across MAC-related MS, RMS, and normal mitral valves, and evaluates the applicability of conventional diagnostic metrics in MAC.
METHODS: Three-dimensional transesophageal echocardiographic data sets from 70 patients (22 normal mitral valves, 26 RMS valves, 22 MAC valves) were used to obtain linear, area, and volumetric measurements for valve comparison. Representative valves from each group were converted …

Grass-legume cover crop mixtures are promoted to balance ecosystem services, but predicting their decomposition and nutrient release remains a challenge. A two-year litterbag study investigated the decomposition of cereal rye, crimson clover, their field mixture, and a gradient of biomass proportions in a no-till corn system. Monocultures exhibited a strong functional dichotomy where rye decomposed slowly with a large recalcitrant fraction and clover decomposed rapidly. The field mixture yielded high biomass similar to rye and exhibited intermediate decomposition kinetics. Crucially, analysis of non-additive effects revealed a fundamental decoupling as the residue mass loss showed context-dependent synergistic acceleration but nitrogen release …

Importance
Fibromyalgia (FM) is a chronic pain condition associated with substantial quality-of-life impairments and economic burden. Although multiple pharmacologic options are recommended in clinical guidelines, the relative cost-effectiveness of off-label amitriptyline compared with FDA-approved treatments remains poorly defined due to a scarcity of direct economic comparisons and the use of heterogenous outcome measures in the extant literature.
Objective
To evaluate the cost-effectiveness of pregabalin, duloxetine, and milnacipran compared with amitriptyline in adults with moderate to severe FM.
