Molecular Determinant Of Low-Voltage Dependence Of Human Nav1.7 Inactivation Revealed By Efficacy-Based Nav1.7 Selective Inhibitor, Fang Zhao, Chuchu Xi, Jie Li, Kerui Ren, Qinglian Tang, Huaduan Liang, Shilong Yang, Michael X Zhu, Zhengyu Cao

Nav1.7 is a voltage-gated sodium channel (VGSC) subtype predominantly expressed in sensory neurons, amplifying threshold currents. Here, we identify that Uvarigranol D (UGD) suppresses human (h) Nav1.7 with a much greater maximal inhibition than other VGSC subtypes, despite having similar apparent affinities. We demonstrate that Thr1398 determines the greater inhibitory efficacy of UGD, the leftward shift in voltage-dependence and faster inactivation kinetics of hNav1.7. UGD binds to the inactivated state, with Gln360, Ile394, Lys1395, Phe1737, and Tyr1744 being critically involved. Channel Gating In Bacteriorhodopsin-Like Channelrhodopsins With Contrasting K+ And Na+ Selectivity, Oleg A Sineshchekov, Elena G Govorunova, Hai Li, Yumei Wang, John L Spudich

Two closely homologous channelrhodopsins from Hyphochytrium catenoides, known as HcKCR1 (kalium channelrhodopsin 1) and HcCCR (cation channelrhodopsin), exhibit >100 times different K+/Na+ relative permeabilities and are emerging optogenetic tools for controlling neurons and cardiomyocytes. Key residue motifs and trimeric organization relate them structurally to bacteriorhodopsin-like cation channelrhodopsins (BCCRs). Here, we demonstrate that they utilize a specific gating mechanism previously suggested for the earlier-discovered BCCR from cryptophytes. Using a comparative analysis of transient absorption changes and photocurrents under single-turnover conditions, we identified an early, far-UV-absorbing photocycle intermediate that precedes channel opening in wild-type HcKCR1 and HcCCR. Avian influenza virus (AIV) remains a major challenge for global poultry health, food production, and agricultural economies. Both low-pathogenic (LPAIV) and highly pathogenic (HPAIV) strains continue to circulate and evolve, creating an ongoing need for vaccines that can be updated quickly and provide broad protection. Although DNA vaccines offer a safe and flexible approach to rapidly incorporate new antigens, their use in poultry has been limited by poor delivery efficiency and the vulnerability of naked DNA to degradation. Objectives: This study aimed to determine associations between hypothyroidism during pregnancy and selected maternal and neonatal outcomes. Methods: A retrospective, cohort study was conducted between January 2018 and December 2020 at the two main tertiary hospitals in Muscat, Oman. The study included 408 Omani pregnant women aged 18–45 years. Participants were divided into two groups: those diagnosed with overt or subclinical hypothyroidism (n = 201; exposed group) and those with normal thyroid function (n = 207; unexposed group), matched for relevant variables. This study investigates how Egypt's Takaful and Karama Program (TKP) can enhance long-term poverty reduction and program sustainability through the inclusion of economicempowerment interventions. Based on the frameworks of Social Risk Management 2.0 and Transformative Social Protection, the study places Egypt's experience in the context of international discussions about conditional cash transfers, multidimensional poverty, and graduation models. The study uses a qualitative methodology, conducting semi-structured interviews with academics, civil society experts, and policymakers. The interviews' findings are supported by content analysis of policy documents and impact evaluations on TKP and Forsa. According to the research analysis, cash transfers help alleviate …

Artificial intelligence (AI) is increasingly used for documentation purposes in clinical practice, yet guidance for resident use is limited. Given the substantial documentation burden on medical trainees, AI-powered scribing tools may offer benefits, but their integration into residency training raises educational, supervisory, and patient safety considerations. This study aimed to assess the availability of resident-specific guidance on AI scribe use from major medical and specialty organizations and to summarize current evidence on AI scribes in residency. Sleep disturbances are prominent across neurodevelopmental disorders (NDDs) and may reflect specific abnormalities in brain development and function. Overnight polysomnography (PSG) allows for detailed investigation of sleep architecture, offering a unique window into neurocircuit function. Analysis of existing pediatric PSGs from clinical studies could enhance the availability of sleep studies in pediatric patients with NDDs towards a better understanding of mechanisms underlying abnormal development in NDDs. Here, we introduce and characterize a retrospective collection of 1527 clinical pediatric overnight PSGs across five different sites. The 3D images derived from non-contrast Computed Tomography (NCCT) scans provide a glimpse into the intra- and extracranial structures, based on the ability of electromagnetic X-rays to penetrate through various tissue densities. The gray matter, for example, possesses a higher density than the white matter, absorbs more X-rays, allows less penetration (attenuation), and thus has larger Hounsfield units (HU), the mathematical value of attenuation coefficient. The CT scanner’s wide availability and ease of use allow for quick assessment of acute neurosurgical abnormalities, such as hemorrhages, mass effect, hydrocephalus, and traumatic injuries. This study investigated the phytochemical constituents and biological activities of Horsfieldia macrothyrsa leaves to identify their bioactive compounds. Methanol extracts were fractionated using n-hexane and ethyl acetate, followed by silica gel column chromatography with a stepwise polarity gradient. From 100 g of powdered leaves, three major compounds were successfully isolated from the ethyl acetate fraction: 1-(2,4,6-trihydroxyphenyl)dodecan-1-one (1), sesamin (2), and β-sitosterol (3). Their chemical structures were confirmed using UV, IR, LC–MS/MS, and NMR spectroscopy.
