Vcp Suppresses Proteopathic Seeding In Neurons, Jiang Zhu, Sara Pittman, Dhruva Dhavale, Jessica N Patterson, Mohamed Salman Kaleelurrrahuman, William J Buscher, Paul Kotzbauer, Albert A Davis, Conrad Weihl, Et Al. Apr 2022

Vcp Suppresses Proteopathic Seeding In Neurons, Jiang Zhu, Sara Pittman, Dhruva Dhavale, Jessica N Patterson, Mohamed Salman Kaleelurrrahuman, William J Buscher, Paul Kotzbauer, Albert A Davis, Conrad Weihl, Et Al.

2020-Current year OA Pubs

BACKGROUND: Neuronal uptake and subsequent spread of proteopathic seeds, such as αS (alpha-synuclein), Tau, and TDP-43, contribute to neurodegeneration. The cellular machinery participating in this process is poorly understood. One proteinopathy called multisystem proteinopathy (MSP) is associated with dominant mutations in Valosin Containing Protein (VCP). MSP patients have muscle and neuronal degeneration characterized by aggregate pathology that can include αS, Tau and TDP-43.

METHODS: We performed a fluorescent cell sorting based genome-wide CRISPR-Cas9 screen in αS biosensors. αS and TDP-43 seeding activity under varied conditions was assessed using FRET/Flow biosensor cells or immunofluorescence for phosphorylated αS or TDP-43 in primary …

Revisiting Concepts Of Evidence In Implementation Science, Ross C Brownson, Rachel C Shelton, Elvin H Geng, Russell E Glasgow

2020-Current year OA Pubs

BACKGROUND: Evidence, in multiple forms, is a foundation of implementation science. For public health and clinical practice, evidence includes the following: type 1 evidence on etiology and burden; type 2 evidence on effectiveness of interventions; and type 3: evidence on dissemination and implementation (D&I) within context. To support a vision for development and use of evidence in D&I science that is more comprehensive and equitable (particularly for type 3 evidence), this article aims to clarify concepts of evidence, summarize ongoing debates about evidence, and provide a set of recommendations and tools/resources for addressing the "how-to" in filling evidence gaps most …

Secondary central nervous system large B-cell lymphoma (SCNSL) is rare, with a generally poor prognosis. There is limited data about the role of autologous stem cell transplantation (ASCT) in these high-risk patients. We explored in this study treatment outcomes and prognostic factors for patients with SCNSL who underwent ASCT. We included all consecutive patients who underwent ASCT at our institution. Primary endpoints were progression-free survival (PFS) and overall survival (OS). One-hundred two patients were identified. Median age at transplant was 56 (range, 21-71) years. With a median follow-up of 56 (range, 1-256) months, the median PFS and OS were 40 …

Identification Of Small Regions Of Overlap From Copy Number Variable Regions In Patients With Hypospadias, Carter H Scott, Ina E Amarillo

Open Access Publications

Hypospadias is a common form of congenital atypical sex development that is often associated with other congenital comorbidities. Many genes have been associated with the condition, most commonly single sequence variations. Further investigations of recurrent and overlapping copy number variations (CNVs) have resulted in the identification of genes and chromosome regions associated with various conditions, including differences of sex development (DSD). In this retrospective study, we investigated the DECIPHER database, as well as an internal institutional database, to identify small recurrent CNVs among individuals with isolated and syndromic hypospadias. We further investigated these overlapping recurrent CNVs to identify 75 smallest …

From Self-Efficacy To Imposter Syndrome: The Intrapersonal Traits Of Surgeons, Alexandra Medline, Helyn Grissom, Ndéye F Guissé, Victoria Kravets, Sandra Hobson, Julie Balch Samora, Mara Schenker

Open Access Publications

BACKGROUND: Demographic differences among surgical trainees regarding intrapersonal traits, such as imposter syndrome and assertiveness, have become widely acknowledged. However, many of these characteristics have not been examined in tandem, nor among both trainees and surgeons in practice. This study aimed to address these knowledge gaps.

METHODS: This was an anonymous, voluntary survey study comprised of validated measures of (1) self-efficacy, (2) imposter syndrome, (3) assertiveness, (4) perfectionism, and (5) self-rated likeability. A multimodal recruitment strategy was used and surgeons across all subspecialties were eligible for inclusion.

RESULTS: A total of 296 participants were included, with 54% identifying as female …

Assessing Thematic Correlations Between Extremist Political Organizations Of The Past And Present, Andrew C. Eunice Apr 2022

Assessing Thematic Correlations Between Extremist Political Organizations Of The Past And Present, Andrew C. Eunice

Honors College Theses

The Unite the Right rally in Charlottesville, Virginia, saw more than five hundred protestors teamed with dozens of militia groups, all representing anti-government and white nationalist groups. Also present were over one thousand counter protestors, many representing far-left and antifascist groups, leading to a violent confrontation between the two sides. As we have developed our core themes of what defines the United States of America, we have also fostered a strong division along the way, enough to the point where some individuals feel the necessity to act with violence to “save our nation” so to speak. This conviction exists within …

Faculty, Staff and Student Publications

Immune checkpoint inhibitors are associated with immune-related adverse events (irAEs), including arthritis (arthritis-irAE). Management of arthritis-irAE is challenging because immunomodulatory therapy for arthritis should not impede antitumor immunity. Understanding of the mechanisms of arthritis-irAE is critical to overcome this challenge, but the pathophysiology remains unknown. Here, we comprehensively analyze peripheral blood and/or synovial fluid samples from 20 patients with arthritis-irAE, and unmask a prominent Th1-CD8

Cngpld: Case-Control Copy-Number Analysis Using Gaussian Process Latent Difference, David J H Shih, Ruoxing Li, Peter Müller, W Jim Zheng, Kim-Anh Do, Shiaw-Yih Lin, Scott L Carter

Faculty, Staff and Student Publications

MOTIVATION: Cross-sectional analyses of primary cancer genomes have identified regions of recurrent somatic copy-number alteration, many of which result from positive selection during cancer formation and contain driver genes. However, no effective approach exists for identifying genomic loci under significantly different degrees of selection in cancers of different subtypes, anatomic sites or disease stages.

RESULTS: CNGPLD is a new tool for performing case-control somatic copy-number analysis that facilitates the discovery of differentially amplified or deleted copy-number aberrations in a case group of cancer compared with a control group of cancer. This tool uses a Gaussian process statistical framework in order …

Surface Urban Heat Island In South Korea’S New Towns With Different Urban Planning, Kyungil Lee, Yoonji Kim, Hyun Chan Sung, Seung Hee Kim, Seong Woo Jeon

Institute for ECHO Articles and Research

A new town is strategically built within a short period compared to naturally developed cities. It is considered an appropriate study area for analyzing the urban climate problems such as surface urban heat islands (SUHIs) that is differently generated according to urban planning and development. In this study, we suggest comprehensive method for determining and comparing changes in surface UHI distribution during 1989–2048 in two new towns with different urban planning. First, a substantial increase in built-up areas was observed from 1989 (< 5%) to 2018 (> 40%) in both new towns. However, SUHI phenomenon-increasing patterns were different of about 12.25% depending on urban …
