Background/objectives: SETD2 is a dual-function methyltransferase important for methylation of histone H3 at lysine 36 and α-tubulin in spindle microtubules. Genetic inactivation of SETD2 during oncogenesis drives loss of H3K36me3, genomic instability, and cancer progression. This study asked if disruption of genomic stability was a canonical feature of SETD2 inactivation via different pathways.
Methods: We evaluated the impact of EPZ-719, a pharmacologic SETD2 inhibitor, and an H3.3K36M mutant histone ("oncohistone") that binds and sequesters SETD2, on methylation activity and genomic stability in human cell lines. Introduction: Sacubitril/valsartan (Sac/Val) represents a cornerstone of heart failure (HF) with reduced ejection fraction (HFrEF) management. This systematic review provides a comprehensive overview of real-world evidence (RWE) regarding the implementation, clinical effectiveness, and safety of Sac/Val in patients with HFrEF.
Methods: A systematic literature search of PubMed was conducted through March 2024 following PRISMA guidelines.
Results: The review included 45 manuscripts from 30 different studies, primarily from Europe (44%) and the US (30%). RWE confirmed that Sac/Val was associated with a lower risk of cardiovascular mortality (10%-16%), HF hospitalization (10%-38%), and all-cause mortality (10%-25%). Background: Ketone bodies (KB) are endogenous energy sources synthesized by the liver in response to metabolic stress. Their associations with atherosclerotic cardiovascular disease (ASCVD), heart failure (HF), and mortality and their potential beneficial or harmful effects have yet to be determined. This study aimed to examine the association between KB and incident cardiovascular outcomes and mortality in a large general population cohort free from ASCVD and HF at baseline.
Methods: This analysis included 90 987 participants (mean age 56.4 ± 8.1 years; 54.7% women) from the UK Biobank without prevalent ASCVD or HF. KB were measured by nuclear magnetic resonance …

Molecular profiling has identified 3 groups of meningiomas, with MenG C tumors exhibiting the vast majority of recurrences. Efforts to find effective treatments for recurrent meningiomas have remained elusive. Higher WHO-grade meningiomas have exhibited greater Programmed Death Ligand 1 (PD-L1) expression through various methods, but the prognostic value of PD-L1 expression has not been described in the context of molecular profiling. Additionally, trials investigating PD-1/PD-L1-targeted immunotherapies have produced disappointing results. Here, we find that PD-L1 positivity, while prevalent in MenG C tumors, does not predict recurrence in the benign MenG A and B tumors. PD-L1 positivity also occurs independently of …

Background/Objectives: Molecular subtyping of pediatric B-cell acute lymphoblastic leukemia (B-ALL) has improved patient outcomes through stratification and selection of targeted therapies. Despite extensive genomic and transcriptomic profiling of this cancer, few studies to date have characterized the proteomic landscape, although proteins are the direct targets of many therapeutic agents. Methods: In this study, we demonstrate the utility of multi-omic integration of global transcriptomic, proteomic, and phosphoproteomic profiles of samples from patients diagnosed with either of two B-ALL subtypes-Ph-like (BCR::ABL1-like) and ETV6::RUNX1. Objective.Accurate lung function assessment is essential for diagnosing and managing diseases like chronic obstructive pulmonary disorder, pulmonary emboli, and lung cancer. Single-photon emission computed tomography (SPECT) provides valuable 3D functional imaging of ventilation and perfusion, but is limited by low spatial resolution, availability, additional radiation, and cost. Alternative methods, including CT-based perfusion (CT-P) and deep learning models, require large datasets to validate results that are often scarce. Pulmonary function tests (PFTs) offer rapid and noninvasive global lung function measures and are clinically widely used. Povidone-iodine and citric acid are widely used antimicrobial agents. However, data on their comparative efficacy and the functional outcomes of their combination, particularly regarding the influence of common excipients like starch, is limited. Understanding these interactions is crucial for optimizing antiseptic formulations. This study aimed to evaluate and compare the antibacterial activity of povidone-iodine, citric acid (5% and 20%), and starch (5% and 20%), both individually and in combination, against clinical isolates of Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa. This paper introduces a quantum-hybrid-aware routing framework that incorporates the concepts of quantum computing with meta-cognitive learning to improve adaptability, reliability, and awareness in dynamic quantum networks. The proposed system accommodates a new IoT, edge computing and 6G system, where routing schemes should be flexible and robust and have cross-topology functionality. Conventional methods of quantum reinforcement learning can be characterized by a trade-off between exploration and decision stability. Maintaining the dependability and operational continuity of electric vehicle (EV) drivetrains has become crucial as the world's shift to electrified transportation picks up speed. This is a major obstacle for sustainable mobility. Using a high-capacity Artificial Neural Network (ANN), this study presents a data-driven multi-sensor framework for advanced fault diagnosis of induction motors in EV applications. Over several years, individual organizations in Jordan have been focused on increasing the participation of individuals with disabilities in the arts. Increasing inclusion can provide a powerful tool to create visibility, amplify marginalized voices and disrupt normative narratives. However, these isolated efforts had a limited impact until a cohesive effort borne by a burgeoning community of practice led to the first Amman Arts and Disability Summit. This study evaluates the biomechanical performance of a novel curved biodegradable ZK60 magnesium alloy locking plate for plate osteosynthesis in midshaft femoral fractures classified as AO/ASIF 32-A3 (Arbeitsgemeinschaft für Osteosynthesefragen/Association for the Study of Internal Fixation). Current metallic implants are associated with complications such as stress shielding, the need for secondary surgery, and limited support for biological healing. This study investigates the effect of Ag–PTFE nanocomposite coatings, prepared by radio frequency (RF) sputtering, on the surface and antimicrobial properties of orthodontic stainless steel archwires (SSW). The novelty lies in combining silver (Ag) and polytetrafluoroethylene (PTFE) nanoparticles into one coating, which has not been widely applied to orthodontics. The motivation is the need to reduce microbial adhesion and biofilm on archwires, which contribute to enamel demineralization, gingival disease, and infections. The construction industry’s reliance on ordinary Portland cement (OPC) contributes significantly to CO₂ emissions and resource depletion. Geopolymer concrete (GPC) offers a sustainable alternative, but natural aggregates (NAs) still harm the environment. This study investigates recycled coarse steel slag aggregate (RCSA), a metallurgical byproduct, as a partial replacement (10, 20, 30, and 40% by volume) for natural coarse aggregate (NCA) in metakaolin-based GPC. The geopolymer paste was formulated with optimal molar ratios of 3.65 for SiO₂/Al₂O₃, 2.9 for sodium hydroxide/ sodium silicate, and 0.62 for water/metakaolin. This study investigates the influence of small liquid metal gallium additions (1, 1.5, 2, and 2.5 wt%) on the antibacterial performance of Ti, Ti-15Mo, and Ti-45Zr alloys fabricated by the powder metallurgy method. The alloys were manufactured by sintering at 1250 °C to improve the antibacterial activity of Titanium and its alloys in biomaterial applications. Titanium and its alloys are used in biomedical applications due to their corrosion resistance and biocompatibility. This study examines how the amount of time spent sintering affects the density and corrosion resistance of Ti, Ti-6% Al, and Ti-6 % Al-2% HA (hydroxyapatite). Lightweight sandwich panels are mainly utilized in the aerospace and automobile industries, and are increasingly explored for sustainable construction. This study investigates the mechanical performance of sandwich panels composed of mortar and concrete facings with expanded polystyrene (EPS) and particle board (PB) cores. Sawdust-cement composites (SCC) are sustainable alternatives to conventional building materials, but they are underutilized by grassroots artisans due to the limited availability of reproducible methods in low-resource settings. This study develops a standardized, field-tested protocol for producing ceiling boards using mixed-species sawdust, cement, and mineral fillers with minimal technical requirements and without reliance on industrial infrastructure. The workflow from sourcing to manual hydraulic compaction was documented and validated under artisanal conditions to assess feasibility, consistency, and performance. This study evaluates the performance of henequen and sisal fibers in mortar to enhance their tensile properties and provide alternatives to synthetic fibers. Plastic pollution has become a global environmental crisis, with millions of tons of polyethylene terephthalate (PET) waste accumulating in landfills and ecosystems. One promising solution is the integration of waste PET into hot-mix asphalt (HMA), offering the dual benefits of sustainable waste management and enhanced pavement performance. This review critically examines the mechanical, environmental, and economic impacts of PET-modified asphalt. Studies indicate that PET improves rutting resistance, durability, and flexibility, with optimal performance observed at moderate PET dosages (4– 6%).
