Given the central role of the androgen receptor (AR) in prostate cancer cell biology, AR-targeted therapies have been the backbone of prostate cancer treatment for over 50 years. New data indicate that AR is expressed in additional cell types within the tumor microenvironment. Moreover, targeting AR for the treatment of prostate cancer has established side effects such as bone complications and an increased risk of developing cardiometabolic disease, indicating broader roles for AR. With the advent of novel technologies, such as single-cell approaches and advances in preclinical modeling, AR has been identified to have clinically significant functions in other cell …

The androgen receptor (AR) is one of the oldest therapeutic targets in oncology and continues to dominate the treatment landscape for advanced prostate cancer, where nearly all treatment regimens include some form of AR modulation. In this regard, AR remains the central driver of prostate cancer cell biology. Emerging preclinical and clinical data implicate key roles for AR in additional cancer types, thereby expanding the importance of this drug target beyond prostate cancer. In this mini-review, new roles for AR in other cancer types are discussed as well as their potential for treatment with AR-targeted agents. Our understanding of these …

Mitochondria are hubs where bioenergetics, redox homeostasis, and anabolic metabolism pathways integrate through a tightly coordinated flux of metabolites. The contributions of mitochondrial metabolism to tumor growth and therapy resistance are evident, but drugs targeting mitochondrial metabolism have repeatedly failed in the clinic. Our study in pancreatic ductal adenocarcinoma (PDAC) finds that cellular and mitochondrial lipid composition influence cancer cell sensitivity to pharmacological inhibition of electron transport chain complex I. Profiling of patient-derived PDAC models revealed that monounsaturated fatty acids (MUFAs) and MUFA-linked ether phospholipids play a critical role in maintaining ROS homeostasis. We show that ether phospholipids support mitochondrial …

Despite extensive research on astrocytic Ca2+ in synaptic transmission, its contribution to the modulation of sensory transmission during different brain states remains largely unknown. Here, by using two-photon microscopy and whole-cell recordings, we show two distinct astrocytic Ca2+ signals in the murine barrel cortex: a small, long-lasting Ca2+ increase during sleep and a large, widespread but short-lasting Ca2+ spike when aroused. The large Ca2+ wave in aroused mice was inositol trisphosphate (IP3)-dependent, evoked by the locus coeruleus-norepinephrine system, and enhanced sensory input, contributing to reliable sensory transmission. However, the small Ca2+ transient was IP3-independent and contributed to decreased extracellular K+, …

No Refuge From The Fields Vol. 2: Revitalization Of Farmworker Housing Research, Manda Laporte
Capstone Projects – Politics and Government
Farmworkers in the U.S. are often subjected to substandard living situations. However, since they are primarily seasonal, migrant, or temporary work visa holders, limited attention is focused on their quality of life. Although farmworkers are some of the hardest working people, they are typically exposed to extreme weather and working conditions, have the highest percentage of people among the working poor, face restrictive access to education and health care, etc. While service providers, such as housing developers and healthcare specialists, are concerned about these individuals’ well-being, there is still minimal research available about current farmworker trends. This report seeks to …

Embodied cognition is a paradigm in cognitive science that emphasizes the fundamental role of bodies, environmental interactions, situational contexts, and sensory and motor systems in cognitive processing. Much of the research in embodied cognition has focused on online cognitive activities (such as reactive, overt behaviors) that are directly coupled to environmental stimuli. Such research fails to explain offline cognitive activities (such as planning, deliberation, and mental imagery) that are spatially or temporally decoupled from one’s immediate sensory experiences. Embodied, simulation-based theories of cognition attempt to address this shortcoming by proposing a mechanism—modal simulations—by which sensory and motor systems can directly …

Whole -genome sequencing projects of millions of subjects contain enormous genotypes, entailing a huge memory burden and time for computation. Here, we present GBC, a toolkit for rapidly compressing large-scale genotypes into highly addressable byte-encoding blocks under an optimized parallel framework. We demonstrate that GBC is up to 1000 times faster than state-of-the-art methods to access and manage compressed large-scale genotypes while maintaining a competitive compression ratio. We also showed that conventional analysis would be substantially sped up if built on GBC to access genotypes of a large population. GBC's data structure and algorithms are valuable for accelerating large-scale genomic …

With an ever-increasing human population, food security remains a central issue for the coming years. The magnitude of the environmental impacts of food production has motivated the assessment of the environmental and health benefits of shifting diets, from meat to fish and seafood. One of the main concerns for the sustainable development of aquaculture is the emergence and spread of infectious animal diseases in a warming climate. We conducted a meta-analysis to investigate the influence of global warming on mortality due to viral infections in farmed aquatic animals. We found a positive trend between increasing temperature and increasing viral virulence, …

A primary goal of the Affordable Care Act (ACA) was to increase health insurance coverage, reduce health care costs, and improve quality of care. Insurance coverage expanded, however, the effect of the ACA on barriers to accessing care and quality of care is less apparent. Outcome variables related to barriers (having a usual source of care, delaying care due to cost, forgoing care due to cost) and quality (whether healthcare providers always: showed respect, spent enough time listened, explained; and an overall rating of providers) from the Medical Expenditure Panel Survey (MEPS) were examined to evaluate whether barriers to healthcare …

The rapid development of electronic material and sensing technology has enabled research to be conducted on liquid metal-based soft sensors. The application of soft sensors is widespread and has many applications in soft robotics, smart prosthetics, and human-machine interfaces, where these sensors can be integrated for precise and sensitive monitoring. Soft sensors can be easily integrated for soft robotic applications, where traditional sensors are incompatible with robotic applications as these types of sensors show large deformation and very flexible. These liquid-metal-based sensors have been widely used for biomedical, agricultural and underwater applications. In this research, we have designed and fabricated …

Although traditional ryegrass pastures are still commonly used, they require intense management and input and do not perform well during the summer months. Multispecies systems have been recognised as more sustainable, needing less fertiliser and being tolerant to drought and heat. While data on monoculture system fat composition exist, information for multispecies is scarce. The present study compared the fatty acid (FA), carotenoid and fat-soluble composition of a multispecies system (consisting of perennial ryegrass, timothy, white clover, red clover, chicory and plantain) (MULTI) with two other conventional grazing systems (monoculture perennial ryegrass (PRG) and a binary mixture of perennial ryegrass …

Lysosomal inhibition elicited by palmitoyl-protein thioesterase 1 (PPT1) inhibitors such as DC661 can produce cell death, but the mechanism for this is not completely understood. Programmed cell death pathways (autophagy, apoptosis, necroptosis, ferroptosis, and pyroptosis) were not required to achieve the cytotoxic effect of DC661. Inhibition of cathepsins, or iron or calcium chelation, did not rescue DC661-induced cytotoxicity. PPT1 inhibition induced lysosomal lipid peroxidation (LLP), which led to lysosomal membrane permeabilization and cell death that could be reversed by the antioxidant N-acetylcysteine (NAC) but not by other lipid peroxidation antioxidants. The lysosomal cysteine transporter MFSD12 was required for intralysosomal transport …

Theses and Dissertations
Scholars have associated the portrayal of idealized images of the body on social media with the negative body image of users, which manifests as several mental health problems such as eating disorders, body dissatisfaction, body dysmorphic disorder, depression, and anxiety in users. To tackle this issue, body-positive (BoPo) influencers on Instagram, having achieved a state of positive body image, post verbal and visual messages to advocate against normative ideologies that foster the negative body image of social media users. These messages have some implications for health communication and body image research. This study analyzes body-positive influencers’ self-assertion and self-expression in …
