The qualities of antibody (Ab) responses provided by B lymphocytes and their plasma cell (PC) descendants are crucial facets of responses to vaccines and microbes. Metabolic processes and products regulate aspects of B cell proliferation and differentiation into germinal center (GC) and PC states along with Ab diversification. However, there is little information about lymphoid-cell-intrinsic functions of enzymes that mediate ether lipid biosynthesis. Imaging mass spectrometry (IMS) results had indicated that concentrations of a number of these phospholipids were substantially enhanced in GC compared to the background average in spleens, but it was unclear if biosynthesis in B cells was …

Human babesiosis is an emerging tick-borne disease caused by Babesia parasites, most notably Babesia microti and Babesia duncani in North America and Babesia divergens in Europe. Infections can be severe or persistent or relapse despite treatment, and current therapeutic options remain limited, underscoring the urgent need for new and effective treatment options. Acridone derivatives originally developed as potent antimalarial agents against multiple life cycle stages of the malaria parasites were evaluated for their antibabesial activity using continuous in vitro culture systems of B. duncani and B. divergens. Purpose: The personal, physical, and financial toll of workplace injuries on workers is enormous, with many experiencing isolation and depressive symptoms.

Objective: This quality improvement project aimed to improve the Workers' Compensation (WC) program to enhance workforce retention, productivity, team member well-being and reduce liability.

Acropora cervicornis is a critically endangered Caribbean coral species facing a precipitous decline due to disease and thermal bleaching. To combat the loss of ecologically important reef-building corals, novel molecular approaches can be used to understand mechanisms of resilience and ultimately provide a means to modulate coral performance. In this study, we tested a recently developed method for gene knockdown in adult corals that utilizes short interfering RNAs (siRNAs) and the RNA interference pathway. Effective drought management depends not only on the accuracy of scientific data, but also on how that information is communicated and understood by diverse audiences. This dissertation examines drought communication through the lens of science communication, with a focus on improving readability, accessibility, understandability, and usability of drought information. Using a manuscript-style format, three studies investigate how drought information is written, delivered, and applied. Chapter 1 provides a brief overview and outline of this dissertation. As an academic coach in a large, Midwestern middle school, I noticed an influx of teachers with years of prior experience at the elementary or high school levels being hired to work with students in grades six through eight. Because they were already veteran teachers, they were not assigned mentorship, yet there were aspects of working in a middle-school setting that effectively “re-noviced” them (Hanuscin et al., 2020).

This dissertation studies how to build generalizable and reliable models for longitudinal experiential (LE) data. Such data arises in domains where human behavior, experience, and context evolve over time, including education, behavioral health, and related human-centered settings. These datasets are often heterogeneous, partially observed, temporally structured, and vulnerable to out-of-distribution (OOD) shift, making them difficult to model with conventional machine learning pipelines.

Rising costs and fluctuating availability of supplemental fats has led to increased interest in alternative fat sources or feed additives that might improve utilization of supplemental fat in feedlot diets. Two separate experiments were conducted to evaluate alternative strategies to overcome high fat prices. This dissertation explores the optimization of chicken feathers keratin hydrolysate (CFKH) production using alkaline hydrolysis (AH) and sub-critical water hydrolysis (SCWH), focusing on acquiring high keratin yield in the hydrolysate. Perinatal mental health conditions affect approximately one in five mothers and one in ten fathers in the United States, and caregivers of newborns admitted to the neonatal intensive care unit (NICU) are at an elevated risk for developing mental health conditions due to the significant stress associated with infant hospitalization. Depression and anxiety are leading causes of disability and are frequently underdiagnosed in outpatient psychiatric settings due to inconsistent screening and documentation practices. Evidence supports the use of standardized tools, such as the Patient Health Questionnaire-9 (PHQ-9) and Generalized Anxiety Disorder-7 (GAD-7), to improve early detection and clinical outcomes. This quality improvement (QI) project evaluated whether implementing standardized electronic health record (EHR) templates and provider education would improve documentation of PHQ-9 and GAD-7 screenings. Expectations And Realities In Primary Care Appointment Access In The United States: A National Study, Brigham Walker, Ethan Tsai, Alexandra Shimanovsky, Edel Doherty, Sarah E. Tinkler, Rajiv Sharma

Objective: While patients expect timely appointments, little is known about how accurately patient expectations reflect the realities of access to a primary care physician.

Methods: We deployed a national survey (N = 6119, conducted in 2021) to measure patient expectations and a separate national field experiment (N = 11,016, conducted between 2013 and 2016) where trained research assistants called physician offices seeking an appointment to measure physician availability.

Results: Using multivariate regression analyses, we find that patients expected to call 1.9 physicians to secure an appointment, consistent with an appointment offer rate of 49%. However, expectations of 5.4 days wait …

BACKGROUND: The use of glucagon-like peptide-1 receptor agonists (GLP-1 RAs) has remarkably increased in the past few years as they have demonstrated significant reduction in A1c and major cardiovascular outcomes and have led to weight loss with low risk of hypoglycemia. GLP-1 RA use has raised concerns about potential associations with increased gastric retention and adverse outcomes in upper endoscopy procedures.

Through an epistolary critical autoethnography and Latina and Chicana feminist framework, I explore the question of how international graduate students of color can overcome cultural, linguistic, ethnoracial, and social class isolation on predominantly white campuses and in college towns. Immigrant populations experience persistent challenges within the U.S. healthcare system, including marginalization, discrimination, and limited access to understandable health information, which contribute to health disparities. Factors such as low income, cultural differences, and intersecting identities related to gender and race further compound these challenges. This qualitative phenomenological study examined how these social determinants of health influenced the health literacy experiences of African immigrant mothers in Greensboro, North Carolina. Diagnostic service wait times affect veteran access to timely diagnoses and subsequent eligibility for disability benefits, vocational rehabilitation, and social services. The purpose of this integrative review was to identify evidence-based strategies to reduce diagnostic wait time within the VHA. This qualitative study explored how internalized RMA impacts survivors’ cognitive processing, coping strategies, and psychological agency, as observed by licensed psychotherapists who treat sexual assault survivors. Despite extensive literature on the societal impacts of RMA, less attention has been given to understanding how these cultural myths are internalized by survivors and manifest intrapsychically, influencing self-perception, self-blame, and identity reconstruction following trauma. Using a phenomenological approach, semi-structured interviews with therapists provided meta-level insights into how survivors’ narratives reflect internalized cultural stigma and misogynistic beliefs. Cell membranes display nanoscale heterogeneity in lipid composition and organization that regulates vital biological processes yet remain challenging to resolve with conventional imaging. We introduce spectral phasor single-molecule localization microscopy (SP-SMLM), a hyperspectral and super-resolution method that combines wavefront-like optical filtering with single-molecule imaging for simultaneous spatial and spectral analysis. A lab-built three-channel imager with sine/cosine filters encodes emission spectra of single molecules into the phasor space, enabling high-throughput, high-SNR mapping of membrane polarity at sub-50 nm spatial and 15 s temporal resolutions.
