The Von-Hippel-Lindau (VHL) gene, acting as a tumor suppressor, plays a crucial role in the tumorigenesis of clear cell renal cell carcinoma (ccRCC). Approximately 90% of individuals with advanced ccRCC exhibit somatic mutations in the VHL gene. Belzutifan, orally administered small-molecule inhibitor of hypoxia-induced factor-2α, has demonstrated promising efficacy in solid tumors associated with germline loss-of-function mutations in VHL, including ccRCC. However, its impact on cases with somatic or sporadic VHL mutations remains unclear. Here, we present 2 cases where belzutifan monotherapy was employed in patients with advanced ccRCC and somatic loss-of-function mutations in VHL. Both patients exhibited a swift …

The probiotic Limosilactobacillus reuteri DSM 17938 produces anti-inflammatory effects in scurfy (SF) mice, a model characterized by immune dysregulation, polyendocrinopathy, enteropathy, and X-linked inheritance (called IPEX syndrome in humans), caused by regulatory T cell (Treg) deficiency and is due to a Foxp3 gene mutation. Considering the pivotal role of lipids in autoimmune inflammatory processes, we investigated alterations in the relative abundance of lipid profiles in SF mice (± treatment with DSM 17938) compared to normal WT mice. We also examined the correlation between plasma lipids and gut microbiota and circulating inflammatory markers. We noted a significant upregulation of plasma lipids …

Background: The origin of variation is of central interest in evolutionary biology. Maternal mRNAs govern early embryogenesis in many animal species, and we investigated the possibility that heterogeneity in maternal mRNA provisioning of eggs can be modulated by environmental stimuli. Results: We employed two sibling species of the ascidian Ciona, called here types A and B, that are adapted to different temperature regimes and can be hybridized. Previous study showed that hybrids using type B eggs had higher susceptibility to thermal stress than hybrids using type A eggs. We conducted transcriptome analyses of multiple single eggs from crosses using eggs …

BACKGROUND: Children with relapsed central nervous system (CNS tumors), neuroblastoma, sarcomas, and other rare solid tumors face poor outcomes. This prospective clinical trial examined the feasibility of combining genomic and transcriptomic profiling of tumor samples with a molecular tumor board (MTB) approach to make real‑time treatment decisions for children with relapsed/refractory solid tumors.
METHODS: Subjects were divided into three strata: stratum 1-relapsed/refractory neuroblastoma; stratum 2-relapsed/refractory CNS tumors; and stratum 3-relapsed/refractory rare solid tumors. Tumor samples were sent for tumor/normal whole-exome (WES) and tumor whole-transcriptome (WTS) sequencing, and the genomic data were used in a multi-institutional MTB to make real‑time treatment …

Novice Kindergarten Through Second-Grade Teachers’ Perspectives Of Their Preparedness To Teach Students To Read, Shanell Lee-Angry
Walden Dissertations and Doctoral Studies
The majority of U.S fourth graders are not proficient in reading. With lawmakers in some U.S. states enacting “Read by Grade 3” laws, it is important for teachers to be well prepared to teach reading to their students. The purpose of this study was to explore novice kindergarten through second grade (K–2) teachers’ perspectives of their preparedness to teach students to read and their beliefs about what they needed to improve their strategies to teach reading. The conceptual framework was based on self-efficacy theory. The research questions addressed K–2 teachers’ perspectives on their preparedness to teach students to read and …

Making Customers More Loyal: The Role Of Value Cocreation In Branded Apps, Trang Phuc Tran, Adrienne Muldrow, Charlene Dadzie
AMTP Proceedings 2024
The paper develops a conceptual model that examines how value in use enhances value co-creation and then brand loyalty, and captures how value-in-use could change customer perception of branded app entertainment and perceived usefulness, leading to value co-creation. Data from 463 brand app users show that all hypothesized relationships are supported, except H5 (ENT -> COC). That means value co-creation is driven by value-in-use and perceived usefulness, but not by app entertainment. Moreover, value co-creation, in turn, has an impact on brand loyalty. Additionally, mediation effects exist in two paths: first, branded app entertainment and perceived usefulness mediate the effect …

Impact Of Ableist Microaggressions On Dis / Abled Youth: Perceptions Of School Climate And Academic Motivation, Nitza M. Torres Gonzalez
Theses and Dissertations
Ableist microaggressions are often subtle and covert negative attributions and behaviors towards people with dis/abilities that perpetuate stereotypes and inequities (Conover et al., 2021). Although limited research exists that has explored the impact of ableist microaggressions, some scholars have noted similar findings to studies to that have focused on racial microaggressions. Specifically, exposure to ableist microaggressions has been associated with negative mental health outcomes and increased negative affect, feelings of exclusion, and low academic performance and self-concept (Kattari, 2017; Kattari, 2020; Keller & Galgay, 2010; Lett et al., 2019; Nadal et al., 2014; Storey, 2007). Most of these findings have …

With technological advancements in diagnostic imaging, smart sensing, and wearables, a multitude of heterogeneous sources or modalities are available to proactively monitor the health of the elderly. Due to the increasing risks of falls among older adults, an early diagnosis tool is crucial to prevent future falls. However, during the early stage of diagnosis, there is often limited or no labeled data (expert-confirmed diagnostic information) available in the target domain (new cohort) to determine the proper treatment for older adults. Instead, there are multiple related but non-identical domain data with labels from the existing cohort or different institutions. Integrating different …

Background: There are currently no evidence-based guidelines addressing the optimal duration of follow-up after posterior spinal fusion (PSF) for adolescent idiopathic scoliosis (AIS). Despite the safety and efficacy of PSF for AIS, long-term complications exist, including infection, pseudoarthrosis, adjacent segment disease, deformity progression, persistent pain, and junctional deformities. In this study, we describe practice variation existing among surgeons regarding duration and intervals of patient follow-up after surgical treatment of AIS in addition to factors influencing provider recommendations for duration of radiographic and clinical follow-up. This investigation of practice variation is important, as opportunities exist for both limiting unnecessary radiation exposure, …

Small cell lung cancer (SCLC) is an aggressive malignancy composed of distinct transcriptional subtypes, but implementing subtyping in the clinic has remained challenging, particularly due to limited tissue availability. Given the known epigenetic regulation of critical SCLC transcriptional programs, we hypothesized that subtype-specific patterns of DNA methylation could be detected in tumor or blood from SCLC patients. Using genomic-wide reduced-representation bisulfite sequencing (RRBS) in two cohorts totaling 179 SCLC patients and using machine learning approaches, we report a highly accurate DNA methylation-based classifier (SCLC-DMC) that can distinguish SCLC subtypes. We further adjust the classifier for circulating-free DNA (cfDNA) to subtype …

Veggie Meat Alternatives: A Virtue-Ethical View, Carlo Alvaro
Publications and Research
This chapter discusses practical and ethical aspects concerning veggie meat alternatives. Upon consideration, it is argued that while the production, sale, and consumption of veggie meat alternatives seemingly contribute to a solution to environmental problems, we ought to consider veggie meat alternatives from a virtue-ethical perspective concerning human excellence and flourishing. Doing so reveals previously unexplored ethical reasons for not promoting the production, sale, and consumption of veggie meat alternatives. One reason in particular is that veggie meat alternatives stem from the lack of the virtue of temperance.

Examining The Feasibility And Acceptability Of An Online Mindfulness-Based Stress Reduction Program For Frontline Child Protection Social Workers In England, Oliver Beer, Lauren B. Mcinroy, Jennifer Hefner, Sandra Beer
School of Health Professions
This study explores the feasibility and acceptability of an online mindfulness-based intervention (O-MBI) for stress prevention and reduction among frontline child protection social workers (F-CPSWs) in England. In Phase I, a non-randomized, one-arm feasibility study (N=104) demonstrated post-O-MBI reductions in perceived stress and increased mindfulness. Phase II employed a phenomenological approach using semi-structured interviews (n=7). Participants suggested adjustments to content, delivery, and design, highlighting the need for customization. Despite promising results, the study underscores the importance of tailoring interventions to individual needs and circumstances. Future research should delve into O-MBI effectiveness with a more diverse F-CPSW sample.

ARID genes encode subunits of SWI/SNF chromatin remodeling complexes and are frequently mutated in human cancers. We investigated the correlation between ARID mutations, molecular features, and clinical outcomes in melanoma patients. Cutaneous melanoma samples (n = 1577) were analyzed by next-generation sequencing. Samples were stratified by pathogenic/likely pathogenic mutation in ARID genes (ARID1A/2/1B/5B). PD-L1 expression was assessed using IHC (SP142; positive (+): ≥ 1%). Tumor mutation burden (TMB)-high was defined as ≥ 10 mutations/Mb. Transcriptomic signatures predictive of response to immune checkpoint inhibitors-interferon gamma and T-cell inflamed score were calculated. Real-world overall survival (OS) information was obtained from insurance claims …
