Disc degeneration primarily contributes to chronic low back and neck pain. Consequently, there is an urgent need to understand the spectrum of disc degeneration phenotypes such as fibrosis, ectopic calcification, herniation, or mixed phenotypes. Amongst these phenotypes, disc calcification is the least studied. Ectopic calcification, by definition, is the pathological mineralization of soft tissues, widely studied in the context of conditions that afflict vasculature, skin, and cartilage. Clinically, disc calcification is associated with poor surgical outcomes and back pain refractory to conservative treatment. It is frequently seen as a consequence of disc aging and progressive degeneration but exhibits unique molecular …

Melanoma is the third most common type of skin cancer, characterized by its heterogeneity and propensity to metastasize to distant organs. Melanoma is a heterogeneous tumor, composed of genetically divergent subpopulations, including a small fraction of melanoma-initiating cancer stem-like cells (CSCs) and many non-cancer stem cells (non-CSCs). CSCs are characterized by their unique surface proteins associated with aberrant signaling pathways with a causal or consequential relationship with tumor progression, drug resistance, and recurrence. Melanomas also harbor significant alterations in functional genes (BRAF, CDKN2A, NRAS, TP53, and NF1). Of these, the most common are the BRAF and NRAS oncogenes, with 50% …

BACKGROUND: Research priorities are best defined through engagement with communities who will be impacted by the research and have lived experience of the topics to be studied. We aimed to establish a pediatric rare disease community stakeholder group and empower them in (1) eliciting perspectives from affected families in the wider region and (2) synthesizing collective ideas into a research agenda focused on shared ethical, legal, and social implications (ELSI) across rare disease.
METHODS: This two-year project utilized a community-centered approach to engage rare disease community members as equal partners in developing a research agenda for ELSI in rare disease. …

Rare DNA alterations that cause heritable diseases are only partially resolvable by clinical next-generation sequencing due to the difficulty of detecting structural variation (SV) in all genomic contexts. Long-read, high fidelity genome sequencing (HiFi-GS) detects SVs with increased sensitivity and enables assembling personal and graph genomes. We leverage standard reference genomes, public assemblies (n = 94) and a large collection of HiFi-GS data from a rare disease program (Genomic Answers for Kids, GA4K, n = 574 assemblies) to build a graph genome representing a unified SV callset in GA4K, identify common variation and prioritize SVs that are more likely to …

Existing descriptions of the treatment of avoidant/restrictive food intake disorder (ARFID) at higher levels of care (HLOC) for eating disorders are limited, despite HLOC settings frequently serving patients with ARFID. The purpose of this commentary is to expand on the preliminary literature that describes pediatric ARFID treatment at HLOC by describing two specific components of our approach to treating pediatric ARFID that may not yet have traction in the current literature. Specifically, we highlight the utility of (1) treatment accommodations that appropriately account for patients' neurodevelopmental needs (e.g., executive functioning, sensory processing) and (2) the adjunctive use of Dialectical Behavior …

p38 mitogen-activated protein kinases (MAPKs) participate in autophagic signaling; and previous reports suggest that pyridinyl imidazole p38 MAPK inhibitors, including SB203580 and SB202190, induce cell death in some cancer cell-types through unrestrained autophagy. Subsequent studies, however, have suggested that the associated cytoplasmic vacuolation resulted from off-target inhibition of an unidentified enzyme. Herein, we report that SB203580-induced vacuolation is rapid, reversible, and relies on the class III phosphatidylinositol 3-kinase (PIK3C3) complex and the production of phosphatidylinositol 3-phosphate [PI(3)P] but not on autophagy per se. Rather, vacuolation resulted from the accumulation of Rab7 on late endosome and lysosome (LEL) membranes, combined with …

Based on our previous success in using cyclobutanone derivatives as enzyme inhibitors, we have designed and prepared a 37-member library of α-aminocyclobutanone amides and sulfonamides, screened for inhibition of the bacterial enzyme diaminopimelate desuccinylase (DapE), which is a promising antibiotic target, and identified several inhibitors with micromolar inhibitory potency. Molecular docking suggests binding of the deprotonated hydrate of the strained cyclobutanone, and thermal shift analysis with the most potent inhibitor (3y, IC50 = 23.1 µM) enabled determination of a Ki value of 10.2 +/− 0.26 µM and observed two separate Tm values for H. influenzae DapE (HiDapE).

This multi-case study explores teachers’ experiences of the teacher evaluation process implemented in schools across the UAE. Data were collected using interviews and document analysis and covered the seven emirates using the same evaluation process; seventeen teachers—15 female and 2 male teachers––participated in online and face-to-face semi-structured interviews. Our objective was to examine teachers’ experiences in the yearly evaluation cycle enacted in public schools during the 2021–2022 academic year, from the formative evaluation process to the summative evaluation review. Objectives: Body size and shape are sexually dimorphic in humans, with men being characterized with larger upper bodies, while women typically having broader pelvises. Such sexually dimorphic traits, quantified as shoulder to hip ratio (SHR) in men and waist to hip ratio (WHR) in women, serve as cues of an individual’s genetic fitness, reproductive potential, health, and resource holding power, and, thereby, functioning as attractiveness cues to the opposite sex. Cryptococcus neoformans poses a threat to human health, but anticryptococcal therapy is hampered by the emergence of drug resistance, whose underlying mechanisms remain poorly understood. Herein, we discovered that Isw1, an imitation switch chromatin remodeling ATPase, functions as a master modulator of genes responsible for in vivo and in vitro multidrug resistance in C. neoformans. Cells with the disrupted ISW1 gene exhibited profound resistance to multiple antifungal drugs. Mass spectrometry analysis revealed that Isw1 is both acetylated and ubiquitinated, suggesting that an interplay between these two modification events exists to govern Isw1 function. The pursuit of an environmentally sustainable manufacturing process requires the substitution of less damaging and recyclable solutions for harmful reagents. This study aims to assess the effectiveness of using cellulose microfibrils synthesized via different hydrolysis reactions as reinforcing agents in polyvinyl alcohol (PVA) at varying concentrations. The investigation explores the morphology, thermal properties, and chemical behavior of the cellulose particles. The cellulose microfibrils (CMFs) produced using citric acid exhibited the highest yield and aspect ratio. Notably, particles from organic acids demonstrated greater thermal stability, with oxalic acid-derived particles displaying the maximum thermal degradation temperature. The oral health/systemic health connection has been an area of research interest that increased dramatically during the 1990s. Periodontal disease has been associated with a number of systemic conditions, including cardiovascular disease, diabetes, pre-term low-birth-weight infants, respiratory conditions, rheumatoid arthritis, cancer, and Alzheimer’s disease. Inflammation is the obvious link that connects periodontal disease with these conditions, but is this association casual or causal? We will address the biologic plausibility, evidence from human studies, evidence from animal studies, and therapeutic interventions as we review the current understanding of the link between periodontitis and Alzheimer’s disease.

This Article argues that a policy of containment directed at China could have disastrous consequences on the stability of the global system. The liberal international order, created to promote international coordination and structure global trade, comprises key institutions such as the United Nations, the World Trade Organization, the International Monetary Fund, and the World Bank. It is possible that much of the strength of these institutions stems from the fact that nations are "locked" into them simply because they are the only game in town. Put another way, the liberal order is, for lack of a better word, a "monopoly." …

Background: Habit formation can be a challenge for first-year students. Research has suggested that regardless of sleep knowledge, favorable sleep attitudes predict better sleep.
Aim: Our aim was to investigate whether sleep attitudes directly or indirectly predicted risk for metabolic syndrome via sleep.
Method: Students completed self-report and physiological measures. Participants wore wristwatches to collect sleep data. Path analyses investigated the direct or indirect effect of sleep attitude on risk for metabolic syndrome via subjective sleep (sleep quality, duration, risk for apnea) and objective sleep (sleep efficiency, duration, subjective risk for apnea).

The patient in this case study presented to a rural surgical clinic in Guatemala for surgery with characteristics of a probable difficult airway. In the United States, anesthesia providers will often use a video laryngoscope (VL) to manage an expected difficult airway requiring an endotracheal tube. However, many barriers exist towards this technology being adopted as common practice in low-middle income countries (LMICs) like Guatemala. These barriers include financial resources, education, and training. Kinase fusion genes are the most active fusion gene group in human cancer fusion genes. To help choose the clinically significant kinase so that the cancer patients that have fusion genes can be better diagnosed, we need a metric to infer the assessment of kinases in pan-cancer fusion genes rather than relying on the sample frequency expressed fusion genes. Most of all, multiple studies assessed human kinases as the drug targets using multiple types of genomic and clinical information, but none used the kinase fusion genes in their study. According to prior research, the average age at first marriage has steadily increased in the US due in large part to the significance that young adults place on marriage and their evolving conceptions of marital readiness. However, despite the nationwide legalization of same-sex marriage in 2015, there is a significant oversight of sexual minorities in this research. To address this, we draw upon qualitative data collected through an online survey to explore how our sample of unmarried sexual minority young adults (N=256) think about marital timing. Research has shown that during the COVID-19 pandemic, approximately 20% of children and adolescents in the United States experienced mental health issues that became a significant social concern. However, recent studies have demonstrated that the majority of adolescents maintain positive emotions despite the crisis. This cross-sectional and descriptive study delves into the emotional states of adolescents during the pandemic, considering the viewpoints of both adolescents and their parents, with a specific focus on Hispanic adolescents. Demyelination and failure of remyelination in the central nervous system (CNS) characterize a number of neurological disorders. Spontaneous remyelination in demyelinating diseases is limited, as oligodendrocyte precursor cells (OPCs), which are often present in demyelinated lesions in abundance, mostly fail to differentiate into oligodendrocytes, the myelinating cells in the CNS. In addition to OPCs, the lesions are assembled numbers of activated resident microglia/infiltrated macrophages; however, the mechanisms and potential role of interactions between the microglia/macrophages and OPCs are poorly understood.
