Multivalent ligands hold promise for enhancing avidity and selectivity to simultaneously target multimeric proteins, as well as potentially modulating receptor signaling in pharmaceutical applications. Essential for these manipulations are nanosized scaffolds that precisely control ligand display patterns, which can be achieved by using polyproline oligo-helix macrocyclic nanoscaffolds via selective binding to protein oligomers and cell surface receptors. This work focuses on synthesis and structural characterization of different-sized polyproline tri-helix macrocyclic (PP3M) scaffolds. Through combined analysis of circular dichroism (CD), small- and wide-angle X-ray scattering (SWAXS), electron spin resonance (ESR) spectroscopy, and molecular modeling, a non-coplanar tri-helix loop structure with partially …

Vulvovaginal candidiasis (VVC), caused by is characterized by aberrant inflammation by polymorphonuclear neutrophils (PMNs) in the vaginal lumen. Data from the established murine model shows that despite potent antifungal properties, PMNs fail to clear due to local heparan sulfate that inhibits the interaction between PMNs and , resulting in chronic vaginal immunopathology. To understand the role of neutrophil extracellular traps (NETs) in defense against at the vaginal mucosa, we investigated the NET-forming capacity of PMNs in chronic VVC-susceptible (CVVC-S/C3H) and -resistant (CVVC-R/CD-1) mouse strains. Immunofluorescence revealed the formation of NETs (release of DNA with PMN-derived antimicrobial proteins) in PMN cocultures …

University Faculty Publications and Creative Works
We report an attractive update to the demonstration of acid−base buffer action that is appropriate for ease of use and getting attention in the classroom. The twist is based on new fluorophore information and the recent availability of UV flashlights serving as an ideal, portable excitation light source. The pH-dependent fluorescence comes from a choice among three natural sources, namely narra tree wood extract, kidneywood extract, and scopoletin, which is a purchasable coumarin, that all perform equally well. We provide practical details for performing the buffer demonstration at scales of 100 and 1000 mL and give background information for context

School of Nursing and Midwifery
Approaches to nurse education vary internationally. Our approach in the United Kingdom (UK), together with countries including Germany, Italy, and Republic of Ireland is unusual, where field-specific undergraduate nurse education programmes are standard. This approach allows nurses to register as children’s nurses without undertaking generalist training beforehand. Recently, arguments have been made for a move back towards generalist nurse education (Purssell and Sagoo, 2023). This has been met with passionate refutation from children’s (Fallon, 2023), learning disability (Cogher, 2023), and mental health (Warrender, 2022) nursing colleagues.Internationally, generalist nurse education is standard (van Kraaij et al., 2023). In most countries, nurses …

Mechanosensory hair cells of the mature mammalian organ of Corti do not regenerate; consequently, loss of hair cells leads to permanent hearing loss. Although nonmammalian vertebrates can regenerate hair cells from neighboring supporting cells, many humans with severe hearing loss lack both hair cells and supporting cells, with the organ of Corti being replaced by a flat epithelium of nonsensory cells. To determine whether the mature cochlea can produce hair cells in vivo, we reprogrammed nonsensory cells adjacent to the organ of Corti with three hair cell transcription factors: Gfi1, Atoh1, and Pou4f3. We generated numerous hair cell–like …
