A wide variety of CreERT2 driver lines are available for genetic manipulation of adult-born neurons in the mouse brain. These tools have been instrumental in studying fate potential, migration, circuit integration, and morphology of the stem cells supporting lifelong neurogenesis. Despite a wealth of tools, genetic manipulation of adult-born neurons for circuit and behavioral studies has been limited by poor specificity of many driver lines targeting early progenitor cells and by the inaccessibility of lines selective for later stages of neuronal maturation. We sought to address these limitations by creating a new CreERT2 driver line targeted to the endogenous mouse …

BACKGROUND: The risk of recurrence is overestimated by the Kaplan-Meier method when competing events, such as death without recurrence, are present. Such overestimation can be avoided by using the Aalen-Johansen method, which is a direct extension of Kaplan-Meier that accounts for competing events. Meningiomas commonly occur in older individuals and have slow-growing properties, thereby warranting competing risk analysis. The extent to which competing events are considered in meningioma literature is unknown, and the consequences of using incorrect methodologies in meningioma recurrence risk analysis have not been investigated.
METHODS: We surveyed articles indexed on PubMed since 2020 to assess the usage …

BACKGROUND: Respiratory viral infections are common in febrile infants ≤90 days. However, the detection of viruses other than enterovirus in the blood and cerebrospinal fluid (CSF) of young infants is not well defined. We sought to quantify the occurrence of respiratory viruses in the blood and CSF of febrile infants ≤90 days.
METHODS: We conducted a nested cohort study examining plasma and CSF samples from febrile infants 15-90 days via rtPCR. The samples were tested for respiratory viruses (respiratory syncytial virus, influenza, enterovirus, parechovirus, adenovirus, bocavirus). Clinical and laboratory data were also collected to determine the presence of serious bacterial …

In the past decade, Zika virus (ZIKV) emerged as a global public health concern. Although adult infections are typically mild, maternal infection can lead to adverse fetal outcomes. Understanding how ZIKV proteins disrupt development can provide insights into the molecular mechanisms of disease caused by this virus, which includes microcephaly. In this study, we generated a toolkit to ectopically express ZIKV proteins in vivo in Drosophila melanogaster in a tissue-specific manner using the GAL4/UAS system. We used this toolkit to identify phenotypes and potential host pathways targeted by the virus. Our work identified that expression of most ZIKV proteins caused …

Shotgun phosphoproteomics enables high-throughput analysis of phosphopeptides in biological samples. One of the primary challenges associated with this technology is the relatively low rate of phosphopeptide identification during data analysis. This limitation hampers the full realization of the potential offered by shotgun phosphoproteomics. Here we present DeepRescore2, a computational workflow that leverages deep learning-based retention time and fragment ion intensity predictions to improve phosphopeptide identification and phosphosite localization. Using a state-of-the-art computational workflow as a benchmark, DeepRescore2 increases the number of correctly identified peptide-spectrum matches by 17% in a synthetic dataset and identifies 19% to 46% more phosphopeptides in biological …

As global warming and urbanisation intensify unabated, a growing share of the human population is exposed to dangerous heat levels. Trees and forests can effectively mitigate such heat alongside numerous health co-benefits like improved mental wellbeing. Yet, which forest types are objectively and subjectively coolest to humans, and how thermal and mental wellbeing interact, remain understudied. We surveyed 223 participants in peri-urban forests with varying biodiversity levels in Austria, Belgium and Germany. Using microclimate sensors, questionnaires and saliva cortisol measures, we monitored intra-individual changes in thermal and mental states from non-forest baseline to forest conditions. Forests reduced daytime modified Physiologically …

Examining The Influence Of Adult Mentors On Fostering Prosocial Behaviors Among Teens, John H. Weaver Iii
Electronic Theses and Dissertations
This dissertation addressed a critical public health concern stemming from the prevalence of antisocial behavior among U.S. teenagers. The study specifically tackled the lack of information regarding the components of an effective risk-reduction mentoring program for teens. The primary aim was to explore how young adult alums of a central Florida mentoring program articulated the influence of mentoring relationships that have shaped their attitudes and perceptions concerning prosocial behavior during adolescence. As a bounded qualitative case study, this research employed semistructured interviews with 11 young adult mentees from a mentoring program. Additionally, a focus group of five adult mentors who …
Effectiveness Of Antibiotic Stewardship For Healthcare Providers At Urgent Care Clinics, Helen T. Adewole
Effectiveness Of Antibiotic Stewardship For Healthcare Providers At Urgent Care Clinics, Helen T. Adewole
Electronic Theses and Dissertations
Antibiotics have revolutionized modern medicine; however, the overuse in urgent care and primary care significantly contributes to the global burden of infections resistant to available antimicrobial medicines. Approximately 30% of the antimicrobials prescribed in acute care settings are unnecessary. Patient demand for antibiotics has seemingly skyrocketed following the recent COVID-19 pandemic. Practitioners must be knowledgeable about antibiotic stewardship initiatives, such as the wait-and-see approach, to reduce antibiotic overuse and improve the trajectory of antibiotic resistance and patient health outcomes. The gap in practice was the limited awareness of scientific evidence-based tools to partner with patients and improve antibiotic prescription patterns. …

Electronic Theses and Dissertations
This study examined why teachers write office discipline referrals on Black students, addressing a gap in the literature about the writing of office discipline referrals by teachers for disciplinary reasons. Few studies have office discipline referrals as a factor in the discipline gap between Black and White students. The researcher utilized a qualitative case study method to interview participants. Semi structured interviews were conducted and analyzed manually by the researcher. Fifteen classroom teachers participated: five from elementary, five from middle school, and five from high school. The study used the social-ecological theory framework to analyze the phenomenon of writing office …

Preventative measures taken by women of childbearing age before and during conception limit the risk of fetal defects or complications. One specific measure that is recommended by the Centers for Disease Control and Prevention is that all women of childbearing age take folic acid 400 mcg daily. Evidence-based research suggests that taking 400 mcg daily limits the risk of fetal neural tube defects if taken daily and started at least two months before conception. Fetal neural tube defects are classified as defects that damage the baby’s spine and brain. The researcher conducted an evidence-based practice project that sought to determine …

PURPOSE: The Ocular Hypertension Treatment Study (OHTS) identified risk factors for primary open-angle glaucoma (POAG) in patients with ocular hypertension, including pattern standard deviation (PSD). Archetypal analysis, an unsupervised machine learning method, may offer a more interpretable approach to risk stratification by identifying patterns in baseline visual fields (VFs).
METHODS: There were 3272 eyes available in the OHTS. Archetypal analysis was applied using 24-2 baseline VFs, and model selection was performed with cross-validation. Decomposition coefficients for archetypes (ATs) were calculated. A penalized Cox proportional hazards model was implemented to select discriminative ATs. The AT model was compared to the OHTS …

BACKGROUND: Treatments for multiple myeloma (MM) have evolved over time and improved MM survival. While racial differences in MM treatment and prognosis between non-Hispanic African American (NHAA) and non-Hispanic White (NHW) patients are well-established, it is unclear whether they have persisted after the introduction of novel agents.
METHODS: Using the Surveillance, Epidemiology, and End Results-Medicare linked database, our study investigated racial difference in the receipt of treatment within 1 year following diagnosis and assessed survival outcomes among Medicare beneficiaries (≥66 years) diagnosed with MM from 2007 to 2017. We applied multivariable Cox proportional hazards models to estimate the association between …
