Background and Objective
Immune checkpoint inhibitors (ICIs) have been widely applied and studied in the treatment of gastrointestinal (GI) cancers, and have achieved good results. However, in clinical practice, it has been observed that only some patients respond well to ICIs, and some patients may experience various degrees of adverse reactions during the treatment. Timely evaluation of the potential therapeutic effects and adverse reactions of ICIs for patients has important clinical significance. This review aimed to summarize recent progress regarding efficacy-associated biomarkers for ICIs in GI cancer.

Oxytocin acts through the oxytocin receptor (OXTR) to modulate uterine contractility. We previously identified OXTR genetic variants and showed that, in HEK293T cells, two of the OXTR protein variants localized to the cell surface less than wild-type OXTR. Here, we sought to measure OXTR in the more native human myometrial smooth muscle cell (HMSMC) line on both the cell-surface and across the whole cell, and used CRISPR editing to add an HA tag to the endogenous OXTR gene for anti-HA measurement. CD20 is an established therapeutic target in B-cell malignancies. The CD20 × CD3 bispecific antibody mosunetuzumab has significant efficacy in B-cell non-Hodgkin lymphomas (NHLs). Because target antigen loss is a recognized mechanism of resistance, we evaluated CD20 expression relative to clinical response in patients with relapsed and/or refractory NHL in the phase 1/2 GO29781 trial investigating mosunetuzumab monotherapy. CD20 was studied using immunohistochemistry (IHC), RNA sequencing, and whole-exome sequencing performed centrally in biopsy specimens collected before treatment at predose, during treatment, or upon progression. Sushi, von Willebrand factor type A, EGF and pentraxin domain containing 1 (SVEP1) is an extracellular matrix protein that causally promotes cardiovascular disease in humans and mice. However, the receptor mediating the effect of SVEP1 on the development of disease remains unclear. We previously demonstrated that depleting either vascular smooth muscle cell (VSMC)- or myeloid cell-derived integrin α9β1, the first receptor that was identified to interact with SVEP1, did not phenocopy the disease-abrogating effect of depleting SVEP1. While CD4+ T-cell depletion is key to disease progression in people living with HIV and SIV-infected macaques, the mechanisms underlying this depletion remain incompletely understood, with most cell death involving uninfected cells. In contrast, SIV infection of “natural” hosts such as sooty mangabeys do not cause CD4+ depletion and AIDS despite high-level viremia. Here, we report that the CARD8 inflammasome is activated immediately after HIV entry by the viral protease encapsulated in incoming virions. Sensing of HIV protease activity by CARD8 leads to rapid pyroptosis of quiescent cells without productive infection, while T-cell activation abolishes CARD8 function and increases permissiveness …

Complete resistance to vitamin K antagonists is a rare but serious. It can complicate therapeutic management, especially when direct oral anticoagulants cannot be used. Some single mutations in the VKORC1 gene have been identified in patients partially or completely resistant to vitamin K antagonists. We report the cases of two women in their fifties who presented an unexplained peripheral venous thrombosis. The aetiological assessment did not show any abnormalities. Genetic testing showed that both patients had the VKORC1 5417GG genotype. The VKORC1 3673 genotype was GG in case 1 and GA in case 2. The two patients showed complete resistance …

An inherent genetic enzyme disorder in humans, known as glucose-6-phosphate dehydrogenase (G6PD) deficiency, arises due to specific mutations. While the prevailing approach for investigating G6PD variants involves biochemical analysis, the intricate structural details remain limited, impeding a comprehensive understanding of how different G6PD variants of varying classes impact their functionality. This study examined the dynamic properties of G6PD wild types and six G6PD variants from different classes using molecular dynamic simulation (MDS). Background: COVID-19 patients usually present multiple comorbidities and complications associated with severe forms of SARS-CoV-2 infection. This study aimed to assess the risk factors and prevalence of comorbidities and complications contributing to the severity of COVID-19.
Methods: This meta-analysis was performed according to PRISMA guidelines. We searched various databases, including PubMed, Google Scholar, and Scopus (between 2020 and 2023), for eligible studies for this meta-analysis.
Results: Thirty-three studies were eligible, including 85812 patients, of which 36% (30634/85812) had severe disease, whereas 64% (55178/85812) had non-severe disease. Severe cases were potentially correlated with the following factors: gender (male) (odd ratio (OR) …

Introduction: Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease with a steadily progressive course due to the death of central and peripheral motor neurons responsible for voluntary movements. Low-level laser therapy (LLLT) is a treatment method unique in its universality and efficacy, particularly for neurodegenerative diseases.
Methods: In this review, we discuss the effect and application of LLLT in the treatment of ALS. Induction is a critical phase for early career teachers (ECTs) because it is where they develop their sense of professional identity and often decide whether or not to stay in the profession. Substantial research has reported high rates of rural teacher retention during induction (e.g., Ingersoll & Strong, 2011), but few researchers have examined this pattern through a geographical lens. The purpose of this correlational study is to determine whether or not there is a relationship between the amount of time spent on the internet or social media sites and the frequency of pornography habits, and if gender has an effect on frequency of pornography habits. Participants were a convenience sample of 105 men and women between ages 18-24 among two universities in the Southeastern United States. Research has shown that graduate students experience a host of stressors as they navigate higher education. This study was a participant-generated visual method (PGVM) project with 14 doctoral students from one research university in the northeastern United States. The purpose of this study was to illuminate doctoral students’ experiences as the world was progressing toward a post-pandemic reality. Data sources included visual image solicitation, a focus group interview, and individual memoing over one semester. Several themes emerged, including stressors related to working while in graduate school, finances, and social challenges. This study offers insights to graduate programs seeking to reduce …

The impostor phenomenon (IP) includes five central factors: (a) a sense of fraudulence or phoniness; (b) a fear of failure and discovery; (c) compensatory perfectionism (i.e., procrastination and/or over-preparation); (d) interpersonal anxiety; and (e) externalized success and/or discounted positive feedback. After the final stage, the process starts over with reinforced vigor, creating a self-reinforcing cycle in which success is associated with psychological suffering. IP was initially used to describe the reports of high-achieving women, but recent studies have shown that IP is experienced across genders. Graduate students and universities continue to be challenged by the COVID-19 pandemic. This qualitative study explored how U.S. graduate students (n=19) experienced and navigated pandemic challenges to their education using by Pierre Bourdieu’s concepts of habitus, capital, and field. Respondents’ abilities to manage changes caused by the pandemic were largely dependent on the capital they or their informal networks had and the field where they positioned during pre-pandemic and pandemic. Institutions’ characteristics and supports greatly influenced students’ habitus and sense of belonging. The belief that more hours worked equate to higher levels of success has been institutionalized within sport. However, research has yet to interrogate this taken for granted notion. The current study examined the relationship between hours worked and athletic department success. Data were drawn from college coaches and athletic administrators in the United States and consisted of self-reported work hours, team postseason appearances, Directors’ Cup points, and Directors’ Cup rankings to determine if higher work hours were predictive of success. The purpose of this study was to increase the understanding of state laws governing collegiate student-athletes’ name, image, and likeness (NIL) rights and place the laws within the framework of the NCAA’s historic and current views on amateurism and commercialization. To accomplish such, a content analysis of the 30 existing NIL laws/executive orders was performed to identify trends in the statutes and establish what commonalities and differences exist across states. The analysis found five themes/sections common across state laws (i.e., definitions, basic NIL rules, limitations, disclosures and review processes, agent and representation rules, and required workshops). This focus group study explores the perceptions and experiences of college students working within an applied learning program during the COVID-19 pandemic. The program engages children from prekindergarten to sixth grade in local schools and early childhood education centers. The college students serve as teaching assistants in the SG Program hosted by an academic department in a northeastern university and lead philosophical inquiries as they read picture books. Informed by philosophical inquiry with children and the applied learning principles as the research framework, this focus group study invites five teaching assistants to three one-hour in-depth semi-structured interviews.
