Following metastatic spread, many hormone receptor positive (HR(+)) patients develop a more aggressive phenotype with an observed loss of the HRs estrogen receptor (ER) and progesterone receptor (PR). During metastasis, breast cancer cells are exposed to high magnitudes of fluid shear stress (FSS). Unfortunately, the role for FSS on the regulation of HR expression and function during metastasis is not fully understood. This study was designed to elucidate the impact of FSS on HR(+) breast cancer. Exploring the underwater world presents significant physical and financial barriers. Telepresence technologies offer a potential solution by a providing a more accessible version of an underwater experience. Our research involved a scoping review to consolidate previous findings on vision-based technologies that aim to recreate underwater experiences and their user evaluations. We searched 5 academic databases for papers describing or evaluating technologies providing visual underwater experiences without actual submersion. We systematically searched YouTube to include immersive experiences not documented in academic publications. BACKGROUND: Neutrophil to lymphocyte ratio (NLR) has been considered a prognostic biomarker of mortality and other major cardiac events. This study investigates NLR's efficacy in predicting in-hospital and long-term outcomes in patients with ST-segment elevated myocardial infarction (STEMI) undergoing percutaneous coronary intervention (PCI).
METHODS: Electronic databases (PUBMED, Cochrane CENTRAL, ERIC, Embase, Ovid, and Google Scholar) were searched till June 2022 to identify studies having STEMI patients who underwent PCI. Risk ratios and mean differences (MDs), along with their corresponding 95% confidence intervals (Cis) and standard deviations (SDs), were pooled using a random-effect model. Developing in situ spectroelectrochemistry methods, which can provide detailed information about species transformation during electrochemical reactions, is very important for studying electrode reaction mechanisms and improving battery performance. Rotator cuff repair surgeries fail frequently, with 20 to 94% of the 600,000 repairs performed annually in the United States resulting in retearing of the rotator cuff. The most common cause of failure is sutures tearing through tendons at grasping points. To address this issue, we drew inspiration from the specialized teeth of snakes of the Pythonoidea superfamily, which grasp soft tissues without tearing. To apply this nondamaging gripping approach to the surgical repair of tendon, we developed and optimized a python tooth-inspired device as an adjunct to current rotator cuff suture repair and found that it nearly doubled repair …

Extracorporeal membrane oxygenation support for respiratory failure in the intensive care unit continues to have an expanded role in select patients. While acute respiratory distress syndrome remains the most common indication, extracorporeal membrane oxygenation may be used in other causes of refractory hypoxemia and/or hypercapnia. The most common configuration is veno-venous extracorporeal membrane oxygenation; however, in specific cases of refractory hypoxemia or right ventricular failure, some patients may benefit from veno-pulmonary extracorporeal membrane oxygenation or veno-venoarterial extracorporeal membrane oxygenation. OBJECTIVE: The primary goal of this study is to evaluate the relationship between Body Mass Index (BMI) and muscle atrophy in individuals with rotator cuff tears.
METHODS: This study consists of patients with rotator cuff tears identified by MRI from two independent cohorts, the Rotator Cuff Outcomes Workgroup (ROW) and the Multicenter Orthopaedic Outcomes Network (MOON). Presence of atrophy (yes/no) and severity of atrophy (as an ordinal variable) were assessed on MRI by expert physicians. Wax deposition in pipelines and wells is one of the problems in the oil and gas industry. Wax deposition can lead to pipeline blockages, reduced flow capacity, and even production facility or pipeline closures. Various methods and techniques have been developed to mitigate wax deposition, including chemical, mechanical, and thermal methods. Inhibition of wax deposition using chemical methods involves using chemical additives to prevent or reduce wax buildup on pipelines or wellbores. Road traffic accidents are the primary source of injuries and fatalities worldwide, accounting for 20–50 million long term disabilities and 1.25 million deaths annually. Pavement conditions ranked as the second most common cause of traffic accidents. The combined effects of environmental factors and traffic loads lead to pavement discomfort. Ensuring proper pavement design and maintenance can assist in reducing accident rates and preventing long-term disability associated with drivers. This paper presents the dynamic modelling and tracking control of a holonomic 3-wheel omni-directional mobile robot. A proportional integral derivative (PID) controller is used, and its performance depends on the dynamic model’s accuracy. The controller design for the switched non-linear system is based on both a dynamic model and a kinematic design. A proposed dynamic model averages the contact radius of the 3-wheels. The resultant dynamic model, on average, is non-linear and smooth, serving as a potential solution for model-based control design. BACKGROUND: The lack of regular physical activity (PA) in individuals with spinal cord injury (SCI) in the United States is an ongoing health crisis. Regular PA and exercise-based interventions have been linked with improved outcomes and healthier lifestyles among those with SCI. Novel anticancer delivery vehicles are arising from engineered nanostructured materials. This work involved the synthesis, characterization, and release of novel hollow core polymeric nanoparticles (HCPNPs) for controlled drug delivery. Polyethyleneimine (PEI) was bound to AuNPs, forming polymer-shell nanoparticles with dissolution of the gold core via iodine forming HCPNPs with drug intercalated DNA bound to the surface. A high density (85 molecules/particle) of DNA intercalated with daunorubicin was conjugated. Particles were spherical with an average diameter of 105.7 ± 17.3 nm and zeta potential of 20.4 ± 3.54 mV. How can short-lived molecules selectively maintain the potentiation of activated synapses to sustain long-term memory? Here, we find kidney and brain expressed adaptor protein (KIBRA), a postsynaptic scaffolding protein genetically linked to human memory performance, complexes with protein kinase Mzeta (PKMζ), anchoring the kinase's potentiating action to maintain late-phase long-term potentiation (late-LTP) at activated synapses. Two structurally distinct antagonists of KIBRA-PKMζ dimerization disrupt established late-LTP and long-term spatial memory, yet neither measurably affects basal synaptic transmission. Neither antagonist affects PKMζ-independent LTP or memory that are maintained by compensating PKCs in ζ-knockout mice; thus, both agents require PKMζ for their effect. …

The biochemistry of human tissues can provide information for geoprofiling (geographic birthplace or migration patterns) of an unidentified decedent. Utilizing isotope analyses (both stable and radiogenic) from teeth, bone, nails, and/or hair reveals specific isotopic information that correlates to diverse geographic regions reflecting an individual’s birthplace and migration patterns. Isotope analysis of enamel and bone has been employed in bioarchaeology for decades to reconstruct the diet, health, and migration patterns of past populations. Isotope ratios measured from enamel can then be compared to isotope ratios measured from bone of the individual to track patterns of migration. The enamel from the …

This dissertation explores the application of location data in healthcare management, focusing on epidemic spread forecasts and fairness in healthcare access. It demonstrates the power of location data in understanding and forecasting epidemic spread with two examples, using mobility metrics derived from the location data. The second example also considers the effect of social vulnerability along with population mobility and extends a popular deep-learning algorithm to accept static anddynamic data. The dissertation also proposes health equity measures by evaluating the alignment between healthcare facility locations and people’s mobility patterns, considering demographic factors. Circadian rhythms are crucial in regulating various physiological processes, including mood regulation, and disruptions in these rhythms have been linked to several neuropsychiatric disorders, such as depression, bipolar disorder, and schizophrenia. FKBP51, a protein involved in stress responses and glucocorticoid receptor function has emerged as a potential regulator of the circadian clock. FKBP51’s interaction with glucocorticoid receptors impacts the transcriptional regulation of circadian-related genes. Efforts to develop targetable molecular bases for drug resistance for pancreatic ductal adenocarcinoma (PDAC) have been equivocally successful. Using RNA-seq and ingenuity pathway analysis we identified that the superpathway of cholesterol biosynthesis is upregulated in gemcitabine resistant (gemR) tumors using a unique PDAC PDX model with resistance to gemcitabine acquired in vivo. Analysis of additional in vitro and in vivo gemR PDAC models showed that HMG-CoA synthase 2 (HMGCS2), an enzyme involved in cholesterol biosynthesis and rate limiting in ketogenesis, is overexpressed in these models. Cutaneous wounds, both acute and chronic, begin with loss of the integrity, and thus barrier function, of the skin. Surgery and trauma produce acute wounds. There are 22 million surgical procedures per year in the United States alone, based on data from the American College of Surgeons, resulting in a prevalence of 6.67%. Acute traumatic wounds requiring repair total 8 million per year, 2.42% or 24.2 per 1000. The cost of wound care is increasing; it approached USD 100 billion for just Medicare in 2018. Little is known about risk factors for central nervous system (CNS) relapse in mature T-cell and natural killer cell neoplasms (MTNKNs). We aimed to describe the clinical epidemiology of CNS relapse in patients with MTNKN and developed the CNS relapse In T-cell lymphoma Index (CITI) to predict patients at the highest risk of CNS relapse. We reviewed data from 135 patients with MTNKN and CNS relapse from 19 North American institutions. After exclusion of leukemic and most cutaneous forms of MTNKNs, patients were pooled with non-CNS relapse control patients from a single institution to create a CNS relapse-enriched training set. …

This report mainly discusses the development of RET Proteolysis targeting chimera PROTAC molecules based on Selpercatinib (LOXO-292) and Sulfono-γ-AApeptide PROTAC. A PROTAC molecule is a bifunctional molecule composed of two active domains and a linker. It works by inducing selective protein intracellular degradation. This technology was first discussed by Kathleen Sakamoto, Ray Deshaies and Craig Crews in 2001. Using various E3 Ligase, such as CRBN, pVHL, beta-TrCP1, Mdm2, DCAF15, DCAF16, RNF114, and c-IAP1, the PROTAC technology has been applied to several novel drug development,. To understand the dynamics of cancer, mathematical oncologists have developed models of tumor growth and treatment response. Some models are mechanistic and approach tumor growth at the cell-scale, focusing on the evolution of cancerous cells within the ecology of normal tissue, and are often simulated with agent-based modeling. Other models are more clinically motivated and model tumor growth operating at the organ-scale, using patient data to predict treatment response, and are often simulated with partial differential equations. We developed the Hybrid Automata Library which includes both agent-based modeling and partial differential equations for modeling at either of these scales. We …
