Stratified medicine holds great promise to tailor treatment to the needs of individual patients. While genetics holds great potential to aid patient stratification, it remains a major challenge to operationalize complex genetic risk factor profiles to deconstruct clinical heterogeneity. Contemporary approaches to this problem rely on polygenic risk scores (PRS), which provide only limited clinical utility and lack a clear biological foundation. To overcome these limitations, we develop the CASTom-iGEx approach to stratify individuals based on the aggregated impact of their genetic risk factor profiles on tissue specific gene expression levels. Purpose: Volumetric-modulated arc therapy (VMAT) is a widely accepted treatment method for head and neck (HN) and cervical cancers; however, creating contours and plan optimization for VMAT plans is a time-consuming process. Our group has created an automated treatment planning tool, the Radiation Planning Assistant (RPA), that uses deep learning models to generate organs at risk (OARs), planning structures and automates plan optimization. This study quantitatively evaluates the quality of contours generated by the RPA tool.

Background and purpose: In many clinics, positron-emission tomography is unavailable and clinician time extremely limited. Here we describe a deep-learning model for autocontouring gross disease for patients undergoing palliative radiotherapy for primary lung lesions and/or hilar/mediastinal nodal disease, based only on computed tomography (CT) images.

Materials and methods: An autocontouring model (nnU-Net) was trained to contour gross disease in 379 cases (352 training, 27 test); 11 further test cases from an external centre were also included. Anchor-point-based post-processing was applied to remove extraneous autocontoured regions. Theta-mediated end joining (TMEJ) is critical for survival of cancer cells when other DNA double-stranded break repair pathways are impaired. Human DNA polymerase theta (Pol θ) can extend ssDNA oligonucleotides, but little is known about preferred substrates and mechanism. We show that Pol θ can extend both ssDNA and RNA substrates by unimolecular stem-loop synthesis initiated by only two 3' terminal base pairs. Given sufficient time, Pol θ uses alternative pairing configurations that greatly expand the repertoire of sequence outcomes. Further primer-template adjustments yield low-fidelity outcomes when the nucleotide pool is imbalanced. Unimolecular stem-loop synthesis competes with bimolecular end joining, …

Mitosis is a fundamental process that ensures the equal distribution of replicated chromosomes, which carry an organism's genetic material, into two daughter cells. The fidelity of mitosis is paramount, as errors in chromosome segregation result in aberrant chromosome numbers, including gains or losses, a state termed aneuploidy. Aneuploidy impacts a broad range of human conditions, including being one of the major hurdles in early human development where it is the primary cause of miscarriages and birth defects. The mechanisms that govern cell size have long been topics of study in the field of cell biology. In eukaryotic cells this size control is tied to checkpoints, a set threshold of minimum necessary growth linked to cyclin dependent kinase activity regulation. In the fission yeast Schizosaccharomyces pombe, the Cdk1 regulatory network is conserved, and G2/M represents the major size checkpoint. Prior to mitosis, Cdk1 is inhibited by phosphorylation applied by Wee1 during G2 phase. Once S. pombe cells have satisfied the size checkpoint, Cdk1 is activated through dephosphorylation by Cdc25. Resilience: Understanding The Role Of Coping In A Racialized Society Among Black Women, Danielle Pendarvis

The present study explored the intricate relationships between race-based stress, coping strategies (Emotion-focused, Problem-focused, Avoidant coping), the Black Superwoman Schema, resilience, and health outcomes among Black women. Employing a mixed-methods design, an online survey (N = 97) and in-depth semi-structured interviews (N = 10) were conducted, with five transcripts selected for detailed thematic analysis. Quantitative results revealed significant relationships between race-based stress, coping strategies, and adherence to the Black Superwoman Schema. Specifically, race-based stress predicted lower levels of general health, while adherence to the Black Superwoman Schema was associated with diverse coping strategies and higher levels of race-based resilience. Diet quality during early childhood is a strong predictor of mental and physical outcomes, as well as future diet quality. Although many factors determine diet quality during childhood, food insecurity is a significant predictor of lower diet quality. On the other hand, participation in nutrition assistant programs, such as the Special Supplemental Nutrition Program for Women, Infants, and Children (WIC) is a significant predictor of higher diet quality. New York courts are showing impatience with local governments that withhold or stretch out approval of projects without valid environmental grounds. In seven cases last year under the State Environmental Quality Review Act (SEQRA),courts overturned municipal actions that rejected projects or delayed approvals. In all, the courts decided 46 cases under SEQRA in 2023. Where the government agency had decided that an environmental impact statement (EIS) was not needed, that choice was upheld in 23 cases and overturned in eight. Where an EIS had been prepared, the EIS was upheld in 11 and found inadequate in only one. The remaining …

Dynamics of species persistence in habitat fragments is often understood in the framework of island biogeography theory (IBT), with habitat fragments being analogous to oceanic islands. Pseudomonas aeruginosa are prevalent opportunistic bacterial pathogens that cause nosocomial infections in at-risk individuals. The rise of multidrug-resistant P. aeruginosa poses an immense threat to hospitals. Additionally, P. aeruginosa are common food spoilage organisms that contribute to food waste. Alternative effective treatment methods are needed to improve public health and reduce food waste. An estimated 85-99% of alarms in the intensive care unit (ICU) do not require nursing intervention. This overabundance of non-actionable alarms can contribute to sensory overload and is a significant factor in alarm-related safety events. The aim of this project was to reduce the prevalence of non-actionable alarms through the implementation of an evidence-based alarm management bundle. The CEASE Bundle, a comprehensive five-step protocol encompassing Communication, Electrodes, Appropriateness, Setup, and Education, was implemented in an 11-bed surgical intensive care unit. In recent years, the relationship between registered voters of minority groups and the government of the states they reside in has become increasingly complex due to controversial legislation proposed and often passed in these states. Specifically, some states have passed laws that make it more difficult for Hispanic/Latinx residents to live and work. In particular, Florida has enacted legislation that makes it more difficult for members of the minority groups to vote, drive, and gain access to healthcare. The purpose of this study is to explore the experiences and perceptions of teachers who use arts-integrated instruction and their directors of K-2 students before, during, and after the unique period of online instruction during the COVID-19 pandemic, which began in March of 2020. Arts integration is a method of teaching in which students are taught comprehension of subject areas (i.e. – math, science, reading, social studies) through art forms. These art forms include music, theatre, dance, art, and writing. I collected he qualitative data for this research through virtual interviews with five female participants. Improvement Of Triboelectric Nanogenerator By As-Prepared Lithium Niobate For Energy Harvesting And Sensing Applications, Jahid Inam Chowdhury

One readily available green energy source that is used to address the small-scale energy requirement is mechanical energy. To harvest mechanical energy, supply energy for upcoming electronic devices, and enable flexible health monitoring, triboelectric nanogenerators composed of perovskite and lightweight polymers have garnered significant interest. A material named lithium niobate (LiNbO3) was produced in two different phases and used to make a flexible triboelectric nanogenerator (TENG). Furthermore, lightweight polydimethylsiloxane (PDMS), Polyvinyl Alcohol (PVA), and LiNbO3 nanoparticles were used to produce a small triboelectric nanogenerator that successfully converts mechanical energy into electricity.
