A 72-Year-Old Man With Acute Lung Injury And Anti-Melanoma Differentiation-Associated Gene 5 Antibody: A Case Report, Irakli Lemonjava, Jose Manuel Martinez Manzano, Sahar Sultan, Rekha Bhat, Corrado Minimo, Zurab Azmaiparashvili, Sadia Benzaquen

Einstein Health Papers

Anti-melanoma differentiation-associated gene 5 antibody (anti-MDA-5 Ab) is associated with amyopathic dermatomyositis (DM). These patients are particularly at high-risk for developing acute and rapidly progressive interstitial lung disease (ILD). Given the lack of muscle-related symptoms, along with its sudden onset and rapid clinical progression, the diagnosis of anti-MDA-5 Ab + ILD represents a challenge for clinicians. Even after the diagnosis is established, prognosis remains dismal owing to a hyperinflammatory state, mimicking cytokine storm, commonly refractory to potent immunosuppressive therapy. Hence, we present an elderly African American man who developed acute and rapidly progressive ILD in the setting of positive anti-MDA5 …

Kindlin-2 is a cytoskeletal adapter protein that is present in many different cell types. By virtue of its interaction with multiple binding partners, Kindlin-2 intercalates into numerous signaling pathways and cytoskeletal nodes. A specific interaction of Kindlin-2 that is of paramount importance in many cellular responses is its direct binding to the cytoplasmic tails of integrins, an interaction that controls many of the adhesive, migratory and signaling responses mediated by members of the integrin family of cell-surface heterodimers. Kindlin-2 is highly expressed in many cancers and is particularly prominent in prostate cancer cells. CRISPR/cas9 was used as a primary approach …

We evaluate the impact of donor types on outcomes of hematopoietic cell transplantation (HCT) in myelofibrosis, using the Center for International Blood and Marrow Transplant Research registry data for HCTs done between 2013 and 2019. In all 1597 patients, the use of haploidentical donors increased from 3% in 2013 to 19% in 2019. In study-eligible 1032 patients who received peripheral blood grafts for chronic-phase myelofibrosis, 38% of recipients of haploidentical HCT were non-White/Caucasian. Matched sibling donor (MSD)-HCTs were associated with superior overall survival (OS) in the first 3 months (haploidentical hazard ratio [HR], 5.80 [95% confidence interval (CI), 2.52-13.35]; matched …

Osteosarcoma is the most common primary bone malignancy in children and young adults, and it has few treatment options. As a result, there has been little improvement in survival outcomes in the past few decades. The need for models to test novel therapies is especially great in this disease since it is both rare and does not respond to most therapies. To address this, an NCI-funded consortium has characterized and utilized a panel of patient-derived xenograft models of osteosarcoma for drug testing. The exomes, transcriptomes, and copy number landscapes of these models have been presented previously. This study now adds …

Underrepresentation of racial and ethnic subgroups in cancer clinical trials remains a persistent challenge. Restrictive clinical trial eligibility criteria have been shown to exacerbate this problem. We previously identified that up to 24% of patients treated with standard immunochemotherapy would have been excluded from recent first-line trials in diffuse large B-cell lymphoma (DLBCL) based on 5 laboratory-based criteria. These ineligible patients had worse clinical outcomes and increased deaths related to lymphoma progression, suggesting the potential exclusion of patients who could have benefited most from the novel therapies being evaluated. Using data from the prospectively enrolled Lymphoma Epidemiology Outcomes cohort study, …

We investigate JN.1-derived subvariants SLip, FLiRT, and KP.2 for neutralization by antibodies in vaccinated individuals, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2)-infected patients, or class III monoclonal antibody S309. Compared to JN.1, SLip, KP.2, and especially FLiRT exhibit increased resistance to bivalent-vaccinated and BA.2.86/JN.1-wave convalescent human sera. XBB.1.5 monovalent-vaccinated hamster sera robustly neutralize FLiRT and KP.2 but have reduced efficiency for SLip. All subvariants are resistant to S309 and show decreased infectivity, cell-cell fusion, and spike processing relative to JN.1. Modeling reveals that L455S and F456L in SLip reduce spike binding for ACE2, while R346T in FLiRT and KP.2 strengthens …

Selective Androgen Receptor Modulators Leading To Liver Injury: A Case Report, Michael R. Demangone, Karam R. Abi Karam, Joshua Li

Student Publications

Selective androgen receptor modulators (SARMs) have gained popularity for their alleged ability to selectively target androgen receptors, potentially offering muscle-building benefits with fewer side effects than traditional steroids. However, the safety profile of SARMs, including RAD-140, is not fully understood. This case report presents a 29-year-old male who developed liver injury after taking RAD-140. The patient experienced jaundice and elevated liver enzymes after three months of RAD-140 use. A liver ultrasound revealed hepatic steatosis and a hyperechoic lesion. Symptoms resolved after discontinuing RAD-140. Similar cases of liver injury associated with RAD-140 have been reported, highlighting the potential hepatotoxicity of this …

Assessing The Relationship Between Psychological Readiness And Physical Performance In Secondary School Physical Education, Nicholas Cullen-Carroll

Doctorate in Kinesiology

The purpose of this project was to analyze the relationship between a psychological preparedness survey and physical performance tests. Perceived preparedness is a valid measure for determining readiness to train, however, there is a lack of studies evaluating high school-aged students and the relationship these scales have with physical performance tests. 41 high school students were recruited and participated in a single session including a 6-point preparedness survey, an aerobic capacity test, and a power test. Demographic, perceptive, and performance data were recorded and analyzed. Pearson correlations were used to determine the relationship between the performance and preparedness variables. Significant …

Granulocyte colony-stimulating factor (G-CSF) is widely used to enhance myeloid recovery after chemotherapy and to mobilize hematopoietic stem cells (HSCs) for transplantation. Unfortunately, through the course of chemotherapy, cancer patients can acquire leukemogenic mutations that cause therapy-related myelodysplastic syndrome (MDS) or acute myeloid leukemia (AML). This raises the question of whether therapeutic G-CSF might potentiate therapy-related MDS/AML by disproportionately stimulating mutant HSCs and other myeloid progenitors. A common mutation in therapy-related MDS/AML involves chromosome 7 deletions that inactivate many tumor suppressor genes, including KMT2C. Here, we show that Kmt2c deletions hypersensitize murine HSCs and myeloid progenitors to G-CSF, as evidenced …

Baf155 Controls Hematopoietic Differentiation And Regeneration Through Chromatin Priming, Jun Wu, Changxu Fan, Ashraf Ul Kabir, Karen Krchma, Minseo Kim, Yoojung Kwon, Xiaoyun Xing, Ting Wang, Kyunghee Choi

2020-Current year OA Pubs

Chromatin priming promotes cell-type-specific gene expression, lineage differentiation, and development. The mechanism of chromatin priming has not been fully understood. Here, we report that mouse hematopoietic stem and progenitor cells (HSPCs) lacking the Baf155 subunit of the BAF (BRG1/BRM-associated factor) chromatin remodeling complex produce a significantly reduced number of mature blood cells, leading to a failure of hematopoietic regeneration upon transplantation and 5-fluorouracil (5-FU) injury. Baf155-deficient HSPCs generate particularly fewer neutrophils, B cells, and CD8

BRCA1/2 proteins function in genome stability by promoting repair of double-stranded DNA breaks through homologous recombination and by protecting stalled replication forks from nucleolytic degradation. In BRCA1/2-deficient cancer cells, extensively degraded replication forks can be rescued through distinct fork recovery mechanisms that also promote cell survival. Here, we identified a novel pathway mediated by the E3 ubiquitin ligase RAD18, the E2-conjugating enzyme UBC13, the recombination factor PALB2, the E3 ubiquitin ligase RNF168 and PCNA ubiquitination that promotes fork recovery in BRCA1- but not BRCA2-deficient cells. We show that this pathway does not promote fork recovery by preventing replication fork reversal …

Chikungunya virus (CHIKV) is a pathogenic arthritogenic alphavirus responsible for large-scale human epidemics for which a vaccine was recently approved for use. Mayaro virus (MAYV) is a related emerging alphavirus with epidemic potential with circulation overlap potential with CHIKV. We previously reported the ability of a non-replicating human adenovirus (AdV)-vectored vaccine expressing the MAYV structural polyprotein to protect against disease in mice following challenge with MAYV, CHIKV and UNAV. Herein, we evaluated mouse immunity and protective efficacy for an AdV-CHIKV full structural polyprotein vaccine in combination with heterologous AdV-MAYV prime/boost regimens versus vaccine coadministration. Heterologous prime/boost regimens skewed immunity toward …

2020-Current year OA Pubs

In order to facilitate cardiovascular research to develop non-invasive optical heart pacing methods, we have generated a double-transgenic Drosophila melanogaster (fruit fly) model suitable for optogenetic pacing. We created a fly stock with both excitatory H134R-ChR2 and inhibitory eNpHR2.0 opsin transgenes. Opsins were expressed in the fly heart using the Hand-GAL4 driver. Here we describe Hand > H134R-ChR2; eNpHR2.0 model characterization including bi-directional heart control (activation and inhibition) upon illumination of light with distinct wavelengths. Optical control and real-time visualization of the heart function were achieved non-invasively using an integrated light stimulation and optical coherence microscopy (OCM) system. OCM produced high-speed …

Experiences Of Queer Female Counselor Educators: An Intersectional Interpretive Phenomenological Analysis, Caryn Leonard-Wilde

Walden Dissertations and Doctoral Studies

The Council for Accreditation of Counselor Education and Related Programs and the American Counseling Association require counseling programs (CPs) to recruit, employ, and retain diverse faculty. Queer sexual orientation and gender identity is considered an aspect of diversity with which counselors must gain competency and against which counselors should not discriminate yet little is known about queer female counselor educators (QFCEs) with intersecting minority identities. The purpose of this interpretive phenomenological analysis (IPA) with both hermeneutic and intersectionality theory frameworks was to explore the experiences and meaning making of QFCEs and to describe the impact of these experiences on QFCE’s …

Aged-Out Foster Youths’ Exposure To Spiritual Foster Homes And Transition Into Adulthood, Dr. Valerie Elaine Sims Aug 2024

Aged-Out Foster Youths’ Exposure To Spiritual Foster Homes And Transition Into Adulthood, Dr. Valerie Elaine Sims

Walden Dissertations and Doctoral Studies

There are 438,000 children and youth in foster care in the United States; almost 46,000 are aged 16 or older, and over 20,000 “age out” of many child welfare services between ages 18 and 20 every year. This qualitative phenomenological study aimed to explore the lived experience of foster youth growing up in a spiritual foster home (incorporating religious belief, practice, higher power, Bible study, or youth groups) and aging out of the system through the lens of faith-based theory. This study had a sample size of eight participants, ages 18 to 25, who transitioned out of the foster care …

Constructive Engagement Between Ecological Thoughts Of Karl Marx And Lao Zi, Hongyin Zhou, Yuqi Chen

Comparative Philosophy

The global ecological crisis has posed unprecedented survival challenges to human being. Consequently, ecological issues have naturally become central concerns in philosophy. From the methodological perspective of constructive engagement in comparative philosophy, we have chosen to compare the ecological thoughts of two thinkers—Karl Marx and Lao Zi (老子)—who are separated by vast spans of time and cultural tradition. We argue that Karl Marx holds a mild anthropocentric ecological view, while Lao Zi holds a transcendental ecological view. In addressing ecological issues, Lao Zi’s transcendental methodological guiding principles can provide negative methodological constraints for Karl …

From Contemplation To Counseling: Exploring Philosophical Practice By Constructive-Engagement Strategy Of Comparative Philosophy, Xiaojun Ding, Chao Yang, Peter Harteloh, Feng Yu

Comparative Philosophy

This paper applies Bo Mou’s Constructive-Engagement Strategy of Comparative Philosophy (CESCP) to explore philosophical practice as a novel paradigm that applies philosophy to everyday life. The study advocates for a transformative methodology that seeks truth through critical engagement, joint contributions, and diverse methodological tools, advancing the discipline towards a comprehensive world philosophy. Philosophical practice, characterized by its engagement with human experience and existential inquiries, integrates Eastern and Western philosophies to guide individuals in the art of living. It offers a philosophical alternative to psychological counseling, focusing on reasoning to address life’s challenges and ethical dilemmas. The paper examines the integration …
