Life Without Oxygen: Cellular Mechanisms Enabling Genome Maintenance And Proliferation In Cells From An Anoxia-Tolerant Vertebrate, Riley Asa Roth-Carter

When exposed to the complete loss of oxygen (anoxia) most organisms undergo rapid cellular deterioration driven by a range of metabolic dysfunctions associated with the loss of the terminal electron acceptor for aerobic respiration. This loss of ability to create and maintain necessary levels of cellular ATP through oxidative phosphorylation contributes to loss of membrane potentials, increase in the accumulation of acidic by-products of anaerobic metabolism, and a decrease in the ability to support other molecular processes that rely on a steady supply of ATP. Background: Evaluating natural history and outcomes of rare inborn errors of immunity (IEIs) has been challenging due to the diverse disease manifestations and lack of standardized data.

Objective: To develop a set of standardized, quantitative, generalizable data collection modules to measure organ dysfunction in IEIs, modeled on the previously validated Common Terminology Criteria for Adverse Event system for grading treatment toxicity.

Methods: Disease conditions were organized by organ system. Objectives: To develop OMERACT core domain sets for chronic and recurrent calcium pyrophosphate deposition (CPPD) disease and acute calcium pyrophosphate (CPP) crystal arthritis.

Methods: Following OMERACT methodology, the CPPD Working Group (comprising of patient research partners, fellows, clinicians, and methodologists) defined the core domain sets. The development of the core sets occurred through a scoping review, qualitative study, Delphi surveys, and ranking exercises to identify the most relevant domains. A virtual e-module was developed to enable members of the OMERACT community to discuss and vote for endorsement of the OMERACT CPPD core domain sets.

Psychological stress can cause diminished immune response to infectious challenges. The extent this holds true during severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection and the modifying effect of vaccination status is untested. We explored these relationships in a non-random sample standardized to the US adult population. Across 2178 participants in the Aegis study (2021-2022), we identified 507 eligible laboratory-confirmed SARS-CoV-2 infections. We estimated the natural log of nucleocapsid-binding antibody absorbance (N-antibody) at study visits up to 225 days following a positive SARS-CoV-2 test. Accurate Adaptive Aperture (AA) leaf positioning is essential for some proton therapy systems to ensure precise dose delivery and patient safety. However, conventional quality assurance (QA) methods can be suboptimal or inefficient for routinely assessing the dynamic nature of AA in the Mevion S250i HYPERSCAN system. This study introduces a novel automated QA method to verify AA positions utilizing a scintillation detector. A specialized QA plan consisting of 7 rectangular fields was generated in the RayStation treatment planning system (TPS) and delivered using the Mevion S250i HYPERSCAN proton therapy system. Background: Preeclampsia is a multifactorial, pregnancy-related disorder characterized by new-onset hypertension and proteinuria, with distinct early- and late-onset forms. Although complement dysregulation has been implicated in the pathogenesis of preeclampsia, its causal role remains unclear. In mice, Crry (complement receptor 1-related protein Y) functions as a critical complement regulator at the fetal-maternal interface. Because complete Crry deficiency is embryonically lethal, direct in vivo investigation of complement activation in pregnancy has been challenging.

Lipoprotein(a) (Lp(a)) is a genetically determined causal risk factor for cardiovascular disease, with approximately 20% of the population exhibiting elevated levels. While there are promising drugs in development, there are currently no approved therapies specifically designed to lower Lp(a) levels. For high-risk individuals with extreme levels of Lp(a), liver-directed genome editing could be an effective one-time solution. Genome editing approaches such as CRISPR and TALENs can reduce Lp(a) in LPA-transgenic mouse models, but they frequently induce large and potentially harmful genomic deletions. Biologics, protein- and peptide-based drugs derived from living organisms or cell lines have emerged as effective therapies across a broad range of indications. However, parenteral administration and the need for frequent dosing increase costs and limit compliance, creating an urgent need for innovative platforms capable of continuously, safely, and efficiently delivering sustained biologics in situ within the host. Here, we describe a bioengineered hookworm platform to manufacture and deliver biologic therapeutics in vivo. As proof of concept, we engineer the Ancylostoma ceylanicum secretome by inserting a human single-chain variable fragment antibody (s16-HuScFv) into its genome. Drinking is intertwined with social processes, including social support and group belongingness. Although drinking can transiently enhance social functioning, escalating to heavy use can be detrimental to it. It is well-known that excessive drinking and social dysfunction exacerbate each other over time, and it follows that social support and belonging are key mechanisms of addiction recovery. Yet, less is known about how changes in belongingness throughout treatment dynamically affect drinking outcomes, and even less is known about how drinking outcomes and other social processes (e.g., therapeutic alliance, interpersonal drinking consequences) dynamically influence belongingness. Mining activities often cause mining-induced ground deformation, including subsidence and landslides, and related geo-environmental impacts, posing significant risks to infrastructure and safety. This study conducts a systematic assessment to identify, categorize, and evaluate AI-based methods (machine learning, deep learning, and hybrid models) for predicting and monitoring mining-induced ground deformation. The literature search was performed across major scientific databases, using predefined keywords and selection criteria, resulting in a final dataset of relevant peer-reviewed studies. The reviewed works were classified into three methodological groups: traditional machine learning, deep learning-based approaches, and hybrid methods. Congress has undertaken some of its broadest reforms of military courts-martial in decades. The creation of the Special Trial Counsels (STCs) intentionally removes nearly all courts-martial powers from military commanders for “covered offenses,” some of the most serious prosecutions. Many commentators have expressed concern that this change untethers the courts-martial system from its historical anchor of discipline within the military—an anchor that the Supreme Court has used to grant exceptional deference to Congress for its legislation pursuant to the Make Rules Clause. Journal of Law and Government

In his essay, Mark David Hall presents an overview of Christian scholarship on the question of whether the War for American Independence is biblical and just. He then considers Romans 13 and shows that Protestants, especially Calvinists, came to interpret these verses to permit active resistance to tyrannical rulers. Finally, he discusses the conflict in light of the just war tradition and contends that the patriots had excellent reasons to believe their active resistance was biblical and just.

Numerous indices measuring social capital and community challenges at the local level exist, but none attempt to offer a measure of community health as an independent variable in comparison to community challenges as a dependent variable. Therefore, we propose the Covenantal-Noncentralization Index (CNI) in an effort to provide a more nuanced understanding of social capital at the local level as well as to differentiate between an independent variable index consisting of family stability, religious adherence, non-profit activity, and charitable giving and dependent variables representing various community challenges: violent crime, childhood poverty, STD rate, high school diploma completion, GDP, and unemployment. …

Toward Liberation: Decolonizing Curriculum To Heal Double Consciousness In Students Of Color, Olivia Cimino

This paper explores how education can be reimagined as a form of liberation through the integration of interdisciplinary theories from political science, ethnic studies, and educational studies. Grounded in Antonio Gramsci’s theory of cultural hegemony and Louis Althusser’s concept of the Ideological State Apparatus, this paper introduces the concept of Reverse Hegemony as a framework to reclaim educational curriculum and mitigate systemic injustice. By applying this framework alongside liberatory pedagogies informed by Paulo Freire, Jarvis Givens, and Bryan Brayboy, the paper argues that education can heal the fractured sense of identity that W.E.B. This policy analysis examines the gentrification and displacement impacts resulting from the development of Seattle’s Link Light Rail in the Rainier Valley, one of the city’s most diverse and historically marginalized neighborhoods. The expansion of light rail aimed to improve transit access, reduce traffic congestion, and promote environmental sustainability. Black women in French Orientalist paintings have been portrayed as hypersexual, subservient, and used as props to highlight the presence of their white and Arab counterparts. These depictions of Black women represent early discriminatory ideas about Black Africans that emerged in the same era through racial science. Though partially deconstructed today, these early conceptions of Black women continue to be reinforced and impact modern perceptions of Black women in French society. Psychedelic-assisted therapy is a recently developed method for treating mental health disorders, including anxiety, depression, and post-traumatic stress disorder (PTSD). Research into the effectiveness of psychedelics in treating mental health has largely excluded Black Peoples, Indigenous Peoples, and People(s) of Color (BIPOC); this is problematic, as rates of these mental health disorders are proportionally higher in BIPOC individuals than in white individuals. Mental health issues experienced by BIPOC are, more often than not, rooted in experiences of racial discrimination (Schouler-Ocak et al., 2021). Starting with the infamous Ball of the 41, the perception of Mexican male queerness has been strongly affected by its visibility.
