Objectives: Oral cavity head and neck squamous cell carcinoma (HNSCC) represents a major global health burden, with over 40,000 new cases annually in the United States and >500,000 worldwide. Despite advances in surgery, radiation therapy, chemotherapy, and immunotherapy, overall survival rates have remained largely stagnant over the past three decades, and over half of patients ultimately die from the disease. Although immune checkpoint inhibitors (ICIs) have improved outcomes for a subset of patients with recurrent or metastatic HNSCC, the majority gain limited benefit. Isolation and characterization of rare cell types in non-model organisms present a challenge. Here, we provide a protocol to purify specific cells from planaria for downstream RNA sequencing. We employ a one-step dissociation and fixation method that avoids RNA-degrading formaldehyde. We label cells using RNA fluorescence in situ hybridization (RNA FISH) and flow sort to enrich sequencing reads from cells of interest for deeper biological insight.

Age-related decline in oocyte quality increases the risk of infertility, miscarriage, and birth defects. Mitochondrial dysfunction is a key contributor to this decline. Here, we report that oocyte-specific deletion of Uba3, which encodes the catalytic subunit of the E1 NEDDylation-activating complex, causes sterility in mice. Fully grown, germinal vesicle–stage Uba3 conditional knockout oocytes exhibit mitochondrial dysfunction, including elevated reactive oxygen species, impaired oxidative phosphorylation, and depletion of mitochondrially encoded RNA transcripts. Proteomic analysis identified alterations in mitochondrial-associated proteins, including enrichment of mitochondrial matrix and respiratory chain components and reduced abundance of electron transport chain complexes. Wnt proteins are lipid-modified morphogens fundamental in development and disease. During Wnt biogenesis, the G-protein-coupled receptor (GPCR)-like transporter Wntless (WLS) escorts lipidated Wnts from the endoplasmic reticulum to the plasma membrane, then transfers them to extracellular carriers, forming active and soluble morphogen-carrier complexes. To dissect the mechanisms involved, we solve cryo-EM structures of Wnt-bound WLS and unliganded WLS, and perform structure-guided functional experiments. Wnts engage WLS via three conserved hairpins, which are all required for Wnt trafficking to the cell surface and carrier-mediated secretion. Mitochondria are hubs of metabolism and signaling. We previously demonstrated the importance of mitochondrial structure and function in chemotherapy-refractory triple-negative breast cancer (TNBC). Herein, we present the first 3D analysis of mitochondrial networks in human tumor tissues. Using serial block face scanning electron microscopy, we reconstructed 3,750 mitochondria and 800 lipid droplets (LDs) in naive and residual tumors persisting after conventional chemotherapies in two orthotopic patient-derived xenografts (PDX). Chemotherapies administered as monotherapy or in combination produced residual tumors that harbored mitochondria with significantly increased areas, volumes, and perimeters. Aggressive-variant prostate cancers (AVPCs) respond poorly to anti-androgen therapy but show sensitivity to taxane-platinum chemotherapy, though outcomes remain poor. We conducted a phase 2 trial testing induction cabazitaxel plus carboplatin (CabCarb) followed by olaparib maintenance versus observation in men with AVPC. The primary endpoint of improved progression-free survival (PFS) was not met, likely due to the study being underpowered after 38.5% of patients experienced early progression (ChemoPD) prior to randomization. No genomic alterations predicted ChemoPD; however, transcriptomic analysis revealed the enrichment of metabolic pathways, including arginine metabolism, in ChemoPD tumors. These findings were supported by metabolomics data from preclinical models. …

Social anxiety disorder (SAD) is thought to persist through self-reinforcing cycles linking threat perception, fear of evaluation, avoidance, and social behavior. This dissertation examined whether the nature of these daily processes generalize across individuals, cluster into subgroups, or vary by social anxiety severity. Two ecological momentary assessment (EMA) studies were conducted: a community sample with and without SAD (Study 1: N = 96; five prompts per day across two 14-day bursts) and an undergraduate sample (Study 2: N = 129; six prompts per day across 14 days). Background: Drug importation plays a critical role in Thailand’s healthcare system; however, barriers affecting its regulatory control and their public health implications remain inadequately examined. This study compared international drug importation procedures and evaluated Thailand’s control system with strategies for improvement.
Methods: Conducted in Bangkok between 2020 and 2023, this two-phase study employed a qualitative design. Phase 1 involved content analysis comparing drug importation procedures across Thailand, Singapore, Australia and the United States to identify best practices and potential solutions. Adjunct clinical instructors are essential in helping nursing students connect classroom learning to real-world patient care. A needs assessment within an associate degree nursing (ADN) program identified that adjunct clinical instructors received only a brief overview of policies, contributing to role uncertainty and inconsistent preparation for clinical teaching responsibilities. Current literature supports the use of structured orientation and mentorship to better prepare nurses as they transition into clinical educator roles. In this study, it was aimed to predict Türkiye’s furniture exports for the period from January 2010 to May 2025 using machine learning methods based on macroeconomic indicators. The dataset consisted of 10 economic variables, including the furniture industry production index, capacity utilization rate, import volume, total exports, real effective exchange rate, producer price index, money supply, and oil prices, with 185 observations per variable and a total of 2035 data points. The aim of the present research was to evaluate, for the first time, the effect of ozonated water treatments on the postharvest quality of capia pepper (Capsicum annuum L. ‘Kaptan’). The peppers were harvested at the optimum time and transported to the laboratory immediately. Uniform and quality fruit was selected and precooled. After precooling, the fruit was divided into four groups. The first three groups were immersed in ozonated water at three different concentrations (1.0, 2.0 and 3.0 ppm) for 15 min. An Untapped Resource: Exploring The Role Of Spirituality In The Psychological Healing Of African Asylum-Seekers From A Counselor’S Perspective, Brittany A. Duncan

This qualitative study explored how counselors perceive and integrate spirituality into the psychological healing of African asylum-seekers, a population facing compounded cultural, legal, and existential challenges. Using interpretative phenomenological analysis, the study examined the lived experiences of eight counselors working with this population. Findings revealed that counselors’ personal sense of religion influenced their comfort and approach in broaching spiritual topics, with both positive (e.g., spirituality as a source of support and community) and negative (e.g., religion as a source of suffering) associations shaping clinical engagement. Dr. John Hibbing was awarded the ORCA in 1993. John was born in 1953 in Paullina, Iowa. He earned a bachelor's in secondary education from Dana College in Blair, Nebraska, but then realized that teaching high school wasn't for him. Instead, he attended the University of Iowa, where he earned a master's and doctorate in political science. In 1981, he joined the University of Nebraska-Lincoln as an assistant professor of political science and rose quickly through the ranks. Pediatric acute lymphoblastic leukemia (ALL) survivors face treatment-related risk for late-emerging cognitive difficulties. Chemotherapy can disrupt white-matter structure leading survivors to experience subtle but persistent challenges with processing speed, working memory, and executive functioning. However, cognitive outcomes vary widely even among survivors with similar treatment exposures. To better understand this variability, this study examines how the brain adapts after treatment and how that adaptation is shaped by the environment. Violations of consent and sexual assault occurred frequently within the genre of romance popular across medieval England and France during the late Middle Ages. While many of the characters who experienced sexual assault in these pieces of literature were women, “Subjection to Chivalry” investigates two masculine characters whose consent is violated within their respective romances. Writing center literature often focuses on the documents used to justify a center’s role in the larger institution while largely ignoring internal documents that aid in training. By also answering Babcock & Thonus (2012) and Driscoll & Perdue (2014)’s call for more replicable, aggregable, and data-based (RAD) research in writing centers, this study addresses this literature gap by conducting a qualitative study about a newly-implemented handbook at a public R1 university. As populations age and chronic illness increasingly characterizes the final stages of life in the United States, end-of-life doulas (EOLDs) have emerged as non-medical practitioners who support dying people and their families through education, companionship, advocacy, and ritualized care. Drawing on a thematic analysis of 18 semi-structured interviews with practicing EOLDs across the United States, this thesis examines how EOLDs conceptualize a “good death,” navigate emotionally intensive work, and sustain themselves in an unregulated and often marginal professional landscape. Cerebral Palsy (CP) and Spina Bifida (SB) are among the most prevalent pediatric neuromotor impairments, posing significant challenges to independent mobility and dynamic balance. Although arising from different neurological origins, CP from non-progressive brain disruptions during development and SB from lower motor neuron lesions in the neural tube, both the conditions often lead to similar functional limitations, including muscle weakness, impaired gait and limited mobility. Affected children typically exhibit compromised postural control and an increased dependence on assistive devices. High precision lightning localization is essential for atmospheric research and public safety, yet the high cost of traditional VLF/LF systems restricts high resolution detection primarily to well funded agencies. Furthermore, conventional low frequency receivers often overlook microwave frequency emissions associated with lightning leader development. This thesis proposes a compact, low cost microwave frequency localization system operating in the 1.42 GHz band to provide an accessible alternative for high resolution mapping. Between 2021 and 2025, PEN America documented over 22,000 book bans across the United States (US). Yet, with only 451 districts banning out of over 13,000, 97% of US school districts are not engaging in banning. The 451 banning districts represent only 3% of US school districts. While most research examines individual stakeholders, such as librarians and teachers, this study focused on district and community data which do not change as fast as microsystem variables. IMPORTANCE: Thyroid radiofrequency ablation (RFA) is a minimally invasive alternative to surgery for thyroid nodules. Despite strong international evidence, thyroid RFA was only approved by Health Canada in April 2023. Understanding the experiences of early adopters can inform the broader adoption of RFA in Canada and other healthcare systems.
OBJECTIVE: To describe the implementation experiences of early adopters of thyroid RFA in Canada and identify barriers and facilitators to adoption.
DESIGN: Multiple methods.
SETTING: All 8 listed RFA facilities in Canada.
PARTICIPANTS: Physicians performing thyroid RFA (n = 9).

BACKGROUND: Tobacco dependence is well established to impair bone healing and increase postoperative complications. However, the independent effects of nicotine, separate from combustion-related factors, remain unclear. This study aimed to evaluate the impact of non-tobacco nicotine dependence (NTND) following open reduction and internal fixation (ORIF) of clavicle fractures, including midshaft and lateral fracture subtypes.
METHODS: A retrospective cohort study was performed using a large national database. Patients undergoing ORIF of clavicle fractures were identified using procedural and diagnostic codes. OBJECTIVE: To evaluate which of the different sonographic definitions of isolated polyhydramnios (elevated amniotic fluid index [AFI] alone, elevated maximum vertical pocket [MVP] alone, or both elevated AFI and MVP) is best associated with adverse perinatal outcomes.
STUDY DESIGN: This retrospective case-control study included singleton pregnancies undergoing sonographic surveillance at a tertiary referral center between February 2021 and January 2026. Cases of isolated polyhydramnios identified during this time-period were compared with pregnancies with normal amniotic fluid volume from 2024.
