Variational Speculative Decoding: Rethinking Draft Training From Token Likelihood To Sequence Acceptance, Xiandong Zou, Jianshu Li, Jing Huang, Pan Zhou

Speculative decoding accelerates inference for (M)LLMs, yet a training-decoding discrepancy persists: while existing methods optimize single greedy trajectories, decoding involves verifying and ranking multiple sampled draft paths. We propose Variational Speculative Decoding (VSD), formulating draft training as variational inference over latent proposals (draft paths). VSD maximizes the marginal probability of target-model acceptance, yielding an ELBO that promotes high-quality latent proposals while minimizing divergence from the target distribution. To enhance quality and reduce variance, we incorporate a path-level utility and optimize via an Expectation-Maximization procedure. The E-step draws MCMC samples from an oracle-filtered posterior, while the M-step maximizes weighted likelihood using …

The widespread availability of large-scale code datasets has accelerated the development of code large language models (CodeLLMs), raising concerns about unauthorized dataset usage. Dataset poisoning offers a proactive defense by reducing the utility of such unauthorized training. However, existing poisoning methods often require full-dataset poisoning and introduce transformations that break code compilability. In this paper, we introduce FunPoison, a functionality-preserving poisoning approach that injects short, compilable weak-use fragments into executed code paths. Addressing itinerary modification is crucial for enhancing the travel experience as it is a frequent requirement during traveling. However, existing research mainly focuses on fixed itinerary planning, leaving modification underexplored due to the scarcity of shape need-to-modify itinerary data. To bridge this gap, we formally define the itinerary modification task and propose a general pipeline to construct the corresponding dataset, namely iTIMO. This pipeline frames the generation of shape need-to-modify itinerary data as an intent-driven perturbation task. Disentangled recommendation within the Variational Autoencoder (VAE) framework aims to capture multiple user interests. While effective, these VAEs are fundamentally constrained by their reliance on interaction data alone, lacking the rich external semantic knowledge needed to properly structure and separate latent interests. Meanwhile, Large Language Models (LLMs) excel at deriving profound user preference signals from textual data. Prevailing methods for integrating LLMs into recommendation, however, either focus on single-interest modeling or perform a shallow fusion by aligning LLM and VAE representation spaces. The Accidental Entrepreneur is the second narrative profile in the Reimagining Military Spouse Employment series, a companion to the initiative's data briefs. Through the story of a mid-career military spouse who builds a business after repeated career disruptions, it examines why many spouses turn to entrepreneurship and what stands in their way. The stages of mathematical modeling of complex technological processes using neural network technology were considered. English Abstract:
The article is devoted to the construction of a system of universal rules for the synthesis of modified verbs of the Russian language and ways of presenting them in a matrix table of forms. The aim of the study is to identify parameters of the sound process relevant for predicting the composition of modified sound verbs’ nests, and in developing a system of rules of word-formation synthesis. The research material consisted of 152 base verbs of sound in the Russian language and their 880 modified derivatives.

This paper introduces the concept of ‘seafood nationalism’ to describe how, since 2024–2025, the United States increasingly uses tariffs, trade remedies, food safety enforcement and origin-based rules to favour American seafood over foreign products. Federal actions include Executive Order 14276, Restoring American Seafood Competitiveness, which directs agencies to address unfair trade practices, unsafe imports and to develop a coordinated seafood trade strategy. Additional measures include a tariff package imposing a 10% duty on nearly all seafood imports and 30% on seafood from China, as well as new anti-dumping and countervailing duty orders on warm-water shrimp. The herpes simplex virus 1 (HSV-1) US3 is a multifunctional serine/threonine kinase that promotes HSV-1 replication and spread. But its role and the mechanisms by which US3 regulates actin cytoskeletal remodeling remain poorly defined. We combined flow cytometry, confocal microscopy, immunoprecipitation-mass spectrometry (IP-MS), protein complex mapping, and machine learning to characterize US3-mediated F-actin dynamics. Flow cytometry and confocal microscopy showed that wild-type HSV-1 induces significant F-actin remodeling, while the ΔUS3 mutant displays F-actin levels comparable to uninfected cells, identifying US3 as a key regulator. IP-MS identified 47 high-confidence US3 interactors enriched in conserved actin regulatory complexes, including Arp2/3 nucleation machinery, …

BACKGROUND: Hypertrophic cardiomyopathy (HCM) is a heterogeneous disease with diverse prognosis. The underlying mechanisms remain unknown, resulting in limited risk stratification and therapeutic strategies. This study aimed to elucidate molecular subtypes of HCM through integrated proteogenomic analysis and explore subtype-specific therapeutic strategies.
METHODS: We conducted an integrated proteogenomic analysis of 132 patients with HCM using myocardial samples, incorporating whole-exome sequencing, RNA sequencing, and proteomics. Unsupervised clustering was used to identify HCM subtypes, which were validated in heart tissues and human induced pluripotent stem cell-derived cardiomyocytes from 2 independent HCM subsets. Family offices manage substantial wealth and often undertake philanthropic initiatives as part of their broader objectives. These offices may assume a variety of legal forms, ranging from corporations, partnerships, trusts administered by private trust companies or a combination of these forms. This article is concerned with family offices which use trusts administered by private trust companies in offshore jurisdictions. BACKGROUND: Measurement of donor-derived cell-free DNA fraction (dd-cfDNA%) and donor quantity score (DQS) is a noninvasive biomarker of acute rejection in heart transplantation (HT). Its prognostic value beyond histopathology remains uncertain.
OBJECTIVES: This study sought to evaluate associations between donor-derived cell-free DNA (dd-cfDNA) and clinically meaningful outcomes using a composite endpoint of treated rejection, graft dysfunction, retransplantation, or death within 1 year post-HT.

BACKGROUND: Pearson syndrome (PS) is a rare multisystem mitochondrial disorder characterized by single large-scale mitochondrial DNA deletions (SLSMDs). It typically presents in infancy with refractory sideroblastic anemia, exocrine pancreatic insufficiency, and failure to thrive. Due to its heterogeneous manifestations and resemblance to other hematological conditions, early diagnosis remains a clinical challenge.

Asylum seekers are exposed to violence and traumatic events in their home countries at high rates, leading to mental health challenges such as posttraumatic stress disorder (PTSD). PTSD symptoms and comorbidities such as depression and alcohol misuse are associated with impacts to multiple domains of cognitive functioning among populations such as Veterans. However, the neurocognitive performance of Latinx asylum seekers who experience PTSD symptoms are under-documented in the current literature despite its meaningful impact on clinical care engagement and navigation of the legal process. They have been citizens of the United States since 1917, served in every major conflict since World War I, and have been influential members of American culture, politics, and society. Yet, Puerto Rico is not fully part of the United States. The island of 3.2 million people— more than the population of eighteen states—is a U.S. territory. Classic structuration theory explains how social actors use their personal and collective agency to contend with the structural forces they encounter in everyday life. The antonimous interface between structure and agency is analyzed from an evolutionary communication perspective as it applies to structuration theory generally and to media audiences specifically. The explanatory power of structuration theory is then reconsidered in light of new research on an eternally profound question: Is the power of human agency restricted, if not entirely eliminated, by a lack of free will that can be exercised in human endeavors? Background: Posttraumatic stress disorder (PTSD) is a mental disorder that may occur in the aftermath of severe psychological trauma. Epigenetic changes in the brain may play a critical role in understanding the neurobiology of PTSD by linking environmental traumatic stress exposure to lasting alterations in gene expression that shape neuronal function.
Methods: We examined 1,065,750 DNA methylation (DNAm) sites from 171 donors including neurotypical controls and PTSD and major depressive disorder (MDD) cases across 6 regions implicated in the fear circuitry of the brain. Purpose: This study aimed to apply cardiometabolic genetic testing in a community setting with a predominantly Hispanic population and assess feasibility and perspectives toward genetic testing.
Methods: A genome-sequencing-based genetic panel for cardiometabolic disorders (177 genes related to monogenic conditions, 2 LPA risk alleles, 2 pharmacogenomic loci, and ancestry-adjusted polygenic risk scores for type 2 diabetes and coronary artery disease) was deployed in community cardiology and endocrinology clinics in South Texas. A survey on perceptions toward genetic testing was administered after return of results.

Polygenic risk scores (PRS) estimate individuals' genetic risk for developing multifactorial conditions. Recent genome-wide association studies have enabled development of psychiatric PRS, which hold potential to streamline diagnosis and treatment of psychiatric conditions. As individuals with mental illness often identify concern about passing on psychiatric conditions as a key factor in family planning, some may also seek to use psychiatric PRS in reproductive decisions like childbearing or embryo selection for in vitro fertilization. Objectives: While maxillary sinus fungal balls (MSFB) can be associated with odontogenic conditions (MSFBO), MSFBO clinical and dental features have not been compared to odontogenic sinusitis (ODS). This multicenter study aimed to compare characteristics of MSFB, MSFBO, and ODS.
Methods: A multicenter international retrospective cohort study was conducted on adults with MSFBs and ODS who underwent sinus surgery. First in MSFBs, it was determined whether different dental conditions were more likely in FB versus non-FB sides. Second, clinical features and dental pathologies were compared between MSFBO and ODS. Purpose: Racial and ethnic differences in oral cavity cancer (OCC) stage at diagnosis are substantial, but the reasons remain unclear. We assessed insurance status and social neighborhood factors as potential pathways.
Methods: Using the Texas Cancer Registry, we identified adults diagnosed with OCC between 2010 and 2020. We examined the crude and multivariable associations of race and ethnicity, insurance status, and social neighborhood factors with stage at diagnosis. We investigated the mediating effects of insurance status and neighborhood-level social factors on racial and ethnic differences in stage at diagnosis using four-way decomposition with modified Poisson regression and robust standard errors. …
