The scope of RNA-based therapeutics continues to expand as more siRNA, mRNA, and CRISPR-based treatments are approved for various medical applications. Purification of synthetic RNA drugs is inherently challenging and requires considerable time and resources. New cost-effective and efficient purification strategies will expedite drug development process and will lower the drug costs. In this thesis I describe two generations of photolabile linkers for the non-chromatographic purification of therapeutic RNAs. In addition, the photolabile linker was utilized for regulation CRISPR-Cas9 gene editing. As climate-related disasters are becoming more frequent and severe, local governments face growing pressure to manage disaster-related fiscal shocks. Although disaster mitigation and preparedness are widely recognized as cost-effective strategies for reducing future losses, local governments often lack the financial, political, and technical capacity to invest in them. Despite the importance of pre-disaster investment, mitigation and preparedness efforts continue to lag at the local level, and concerns remain that federal mitigation funding is not distributed equitably across communities. The first chapter decomposes aggregate productivity growth into a pure-technology contribution and an allocative-efficiency contribution for Japan and the United States, applying a single estimation methodology symmetrically to comparable firm-level panels. Production functions are estimated by the Gandhi, Navarro, and Rivers (2020) gross-output procedure under monopolistic competition (GNR-MC), which recovers firm-level price markups and wage markdowns, and these enter the Baqaee–Farhi (2020) decomposition through the closed form of Aoki et al. (2024). Both panels are drawn from Compustat with identical pre-processing and estimation, and we close the markup-level identification gap that GNR leave open with a normalization anchored on the …

Graduate education is often organized around norms of uninterrupted productivity, flexibility, and professional availability, making graduate study especially challenging for students who are simultaneously raising children. Although prior research has documented the structural barriers faced by graduate student parents (GSPs), less attention has been paid to how resilience is communicatively enacted within the intersection of academic work and family life. This dissertation examines how GSPs communicatively constitute resilience while navigating work-life disruptions. Clinical trials suggest potential therapeutic utility of psilocybin-assisted psychotherapy (PAP) for nicotine cessation. In response to promising outcomes, research into PAP for nicotine dependence is growing. However, attitudes towards PAP among the general public, especially daily nicotine users, remains understudied. To address this knowledge gap, we recruited daily nicotine users (N=534) to complete an anonymous online survey with questions on demographics, nicotine use, prior history with psychedelics, and attitudes towards PAP and standard interventions for nicotine cessation. Generative AI (GenAI) and Large Language Models (LLMs) have made large strides in coding task capabilities, with many software developers integrating agentic engineering into their workflow. While GenAI has largely benefited professional software engineers who can automate their work, it has also created room for those with little coding expertise to also create fully fledged programs and applications. It is commonly noted that GenAI is trained with two major goals in mind: to be as helpful as possible, and be as harmless as possible. Per- and polyfluoroalkyl substance (PFAS) contamination in Hoosick Falls, New York, resulted from historical industrial activities, with groundwater transport identified as the primary pathway impacting the municipal wellfield within the Village of Hoosick Falls, whereas atmospheric deposition remains a potential source of private well contamination. This study employed two complementary methodologies to evaluate historical municipal and private well contamination: (i) groundwater flow and transport modeling of PFOA, PFOS, PFHxS, and PFNA within the village, and (ii) Geographic Information Systems (GIS)–based inverse distance weighting (IDW) interpolation of PFOA concentrations in private wells across the broader Town of Hoosick.

Tropical cyclone (TC) intensity change in moderate vertical wind shear (VWS) is difficult to predict. Specifically, ventilation, the intrusion of dry and/or cool (i.e., low equivalent potential temperature (theta-e) air inside the TC, can have differing effects on TCs. Downdraft ventilation is the downward transport of below-mean theta-e air by downdrafts, and radial ventilation is the inward transport of below-mean theta-e air by storm-relative radial winds. If this low theta-e air is able to get ingested by updrafts, it can result in a reduction of convection in the TC, thereby reducing intensity. Tropical cyclones (TCs) are challenging to forecast when under vertical wind shear (VWS). VWS tilts a TC’s vortex, creating asymmetries in its structure that impedes intensification. Although work has been done to conceptualize and understand how TCs evolve under shear and how they axisymmetrize and intensify, vortex alignment remains a complex and unresolved topic. Extreme precipitation events in the South American Altiplano present significant hazards to communities in the Andean region, yet their future characteristics under anthropogenic climate change remain uncertain because of complex terrain and limitations of course-resolution climate models. This study evaluates the ability of South America Affinity Group Weather Research and Forecasting (SAAG-WRF) model with 4-km grid-spacing to reproduce 2000-2015 temperature, precipitation, and extreme precipitation events over the Altiplano. Examination of projected future changes is completed using the SAAG-WRF pseudo global warming (WRF PGW) simulation representing an approximately 3°C warmer global climate. The alarming rise of carbapenem-resistant bacteria poses an urgent global health threat, driven in large part by the horizontal spread of carbapenemase-encoding plasmids (Ma et al., 2023). These plasmids are often highly mobile and transmissible, enabling rapid dissemination of resistance genes across diverse bacterial populations. Their mobility is largely mediated by conjugation, a process dependent on the formation of pili, which serve as physical bridges for plasmid transfer between cells (Murray et al. 2022; San Millan and MacLean 2017; Virolle et al. 2020). This study examined how K–12 public school teachers in Florida and New York perceived the adequacy of curriculum standards and instructional materials for teaching American slavery, how they responded when those materials were perceived as limited, and how the standards and materials aligned with Shuster’s (2018) Ten Key Concepts for Teaching American slavery. Using a qualitative multi-method design, the study combined qualitative survey research with document analysis. Teachers’ open-ended survey responses were analyzed through a Modified Grounded Theory approach, while curriculum standards and instructional materials were analyzed in relation to Shuster’s framework. This dissertation examines the consequences of nursing home closures for access to care, local economic conditions, and resident health in the United States. As the long-term care system shifts toward home- and community-based services and the nursing home sector contracts, facility closures have become an increasingly important policy concern. Although closures may reflect necessary market restructuring in some settings, they may also reduce access, weaken local economies, and place pressure on surviving facilities. Within the study of cross-linguistic influence, research on cognate facilitation has consistently shown that proficient bilinguals process cognates (e.g., English “sock” and German “Socke”) more efficiently than non-cognates. However, less is known about how novice adult learners may benefit from these similarities during the earliest stages of second language acquisition or how instructional interventions might facilitate this process. This dissertation addressed this gap through experiments with English-speaking adults learning German. Despite the growing understanding of therapist factors in client outcomes, there remains limited understanding of the skills that distinguish effective family therapists. The aim of the present study was to assess the interrater reliabilities and factorial structure of a Spanish version of the Facilitative Systemic Intervention Skills (FSIS; Jones et al., 2023a), an observational rating system for evaluating the skillfulness of family therapists in delivering systemic interventions. This doctoral dissertation on the foundations of quantum theory tells the story of a conceptual protagonist I have called “Epistemic Constraint.” Here, epistemic constraints are the definite, intersubjectively agreeable, ordinary-language conditions under which experiments are described.
The usual formulation of the quantum measurement problem, which I call the Schrodingerian measurement problem, has the structure of an anomaly: if we take quantum theory at face value, we expect no definite values, and yet we see definite values in experiments. This dissertation asks how multicultural adolescents in South Korea develop their attitudes toward crossing group boundaries in social and intimate life—whom they would accept as a close friend, a romantic partner, and a spouse—and what factors shape these attitudes. Korea’s immigrant population has grown more than seventyfold since 1990, largely through marriage migration, and the children of those unions are now reaching adulthood. Research on these adolescents has focused on how well they adjust, leaving open how they themselves come to view relationships across group lines.

This study uses atmospheric reanalysis, precipitation and snow water equivalent (SWE) analyses, mesonet observations, and streamflow records to examine the impact of an atmospheric river storm that produced heavy rainfall and snowpack ablation across the Catskill Mountains of New York on 24–25 December 2020. Earlier in December, an antecedent storm produced SWE values in the Catskills above the 90th percentile for that point in the season. The atmospheric river event followed with strong southerly warm-air advection ahead of an anomalously deep trough, temperatures and dewpoints that exceeded 10°C, and heavy orographically enhanced rainfall and rapid snowmelt. Public education in the United States is socially and politically constructed, and ongoing reforms increasingly require collaborative engagement among diverse stakeholders amid heightened divisiveness. This mixed methods comparative case study examined how educational stakeholders make sense of one another’s perspectives during district-level collaborative work and the conditions that may foster conscientization, the critical reflection on one’s positionality. How do pathways between childhood victimization, drug use, and offending behavior vary for recordholders of different races and genders? Prior literature and theory indicates that there are distinct processes that can link childhood victimization to drug use and other offending behavior. These processes are rooted at least partially in experiences of racism/discrimination, placement relative to sociocultural power structures, and norms established over one’s childhood through peer and familial influences. Extreme rainfall is a persistent hazard in the Hawaiian Islands, where steep terrain and short catchments convert heavy rain into damaging floods with little warning. The trade wind inversion, a subsidence-driven stable layer that caps the marine boundary layer over the subtropical North Pacific, limits the depth of convection and is therefore expected to govern whether available moisture can be converted into extreme rainfall. Precipitation from landfalling tropical cyclones (TCs) poses a significant risk to life and property in both coastal and inland communities. Early warning systems can help mitigate the impacts of TC-induced flooding; however, this requires accurate quantitative precipitation forecasts (QPFs). Forecasting precipitation from landfalling TCs is a challenge, as the intensity, duration, and location of precipitation can be dependent on numerous storm-related and environmental factors. This dissertation examines the persistence and spatial concentration of gender-based educational inequality in Afghanistan's public school system from 2002 to 2021–– a period spanning post-2001 reconstruction, the expansion of the Republic era, and the reversal under the second Taliban regime. Despite substantial international investment and two decades of repeated commitments to gender parity, access to and progression through public schooling remained profoundly uneven across the country’s thirty-four provinces. The Gender Parity Index values differ sharply between provinces, and those with the lowest overall enrollment tend to exhibit the widest gender gaps. This thesis details the development of multivalence-nanostructured vanadium oxide (VOₓ) thin films for uncooled microbolometer applications, with a systematic optimization of magnetron-sputtering parameters. The temperature coefficient of resistance (TCR), resistivity, and optical response of VOₓ thin-film sensing layers are controlled by valence composition, grain growth, and surface morphology. The primary goal was to achieve a high TCR with low resistivity to improve thermal detector performance.

Transportation agencies make critical operational decisions during hazardous weather events, including assessment of road conditions and resource allocation. In this study, machine learning models are developed to provide additional support for the New York State Department of Transportation (NYSDOT) by automatically classifying road conditions across the state. Convolutional neural networks and random forests are trained on NYSDOT roadside camera images and weather data to predict road surface conditions. Opportunity youth—young people aged 16–24 who are disconnected from both school and employment—experience significant barriers to successful transitions into adulthood. Existing research has largely focused on deficits, risk factors, and outcomes associated with disconnection, often overlooking the strengths, resilience, and aspirations of these young people. This dissertation studies the eighteenth-century English novel and expands the understanding of realism by rereading realist novels through Gothic representations of the masses. The conventional understanding of realism is shaped by Ian Watt’s canonical concept of “formal realism” in The Rise of the Novel and Jürgen Habermas’s conception of the bourgeois public sphere. It identifies realism with a vision of civility grounded in the modern individual and bourgeois social reality. High-temperature water flowing through the coolant circuits in pressurized water reactors (PWRs) creates a harsh environment, deteriorating these materials. This corrosion results in the formation of oxide layer(s) primarily composed of nickel ferrites, iron oxides, nickel oxides, and other nickel-iron-chrome spinel oxides. Being exposed to fluid with high flow rates and pressure, these oxide layers can be eroded, resulting in particulate fouling due to the release of corrosion products (Chalk River Unidentified Deposits, CRUD) into the coolant circuit.
