BACKGROUND: Physical activity is associated with lower colorectal cancer (CRC) risk, but its association with molecular subtypes defined by genetic and epigenetic alterations of the disease is unclear. Such information may enhance the understanding of the mechanisms related to the benefits of physical activity.
METHODS: Pooled observational (cases: n = 5386; controls: n = 6798; studies n = 5) and genome-wide association data (cases: n = 8178; controls: n = 10 472; studies n = 5) were used. IMPORTANCE: The most common inherited blood disorder in the world, sickle cell disease (SCD) is a known global health concern. While survival of children into adulthood has improved significantly over the last few decades in high-resource countries, substantial gaps in care standards and limited practice harmonization persist.
OBJECTIVE: To provide up-to-date recommendations for preventive care and treatment of all people living with SCD and its related comorbidities.

Cantu syndrome (CS) is a rare disease caused by gain-of-function (GOF) mutations of Kir6.1 or SUR2 subunits of ATP-sensitive potassium (KATP) channels. CS patients with SUR2 and Kir6.1 variants display a similar constellation of symptoms, including muscle weakness and fatigue. The effects of CS mutations on skeletal muscle KATP channels, and any consequent direct effects on contractility, are currently unclear. Here, we used two knock-in mouse models of CS, respectively, carrying GOF mutations Kir6.1[V65M] or SUR2[A478V], to assess KATP channel properties and contractility in isolated fast-twitch extensor digitorum longus (EDL) and slow-twitch soleus (SOL) muscles. Inwardly rectifying potassium (Kir) channel activity is important in the control of membrane potentials in both excitable and non-excitable cells and is regulated through various ligands, including specific membrane lipids. Phosphatidyl-4,5-bisphosphate (PIP2) is required for activity of all Kir channels, binding to the cytoplasmic domain in a compact conformation tightly tethered to the transmembrane domain. Most Kir2 channel structures determined in complex with PIP2 molecules are nevertheless in a closed state, requiring additional conformational changes for channel opening. Mutations that initiate acute myeloid leukemia (AML) can cause clonal expansion without transformation (clonal hematopoiesis). Cooperating mutations, usually in signaling genes, are needed to cause overt disease, but these may require a specific fitness state to be tolerated. Here, we show that nearly all AMLs arising in a mouse model expressing 2 common AML-initiating mutations (Dnmt3aR878H and Npm1cA) acquired a single copy amplification of chromosome 7 (chr7), followed by activating mutations in signaling genes. We show that overexpression of a single gene on chr7 (Gab2, which coordinates signaling pathways) was tolerated in the presence of the Npm1cA mutation, could accelerate …

In the highly competitive Division I (DI) college sports system, coaches are expected to produce winning programs while developing their college athletes holistically. Recent changes in NCAA policies complicate the coach-athlete relationship, hindering coaches' ability to serve as effective holistic leaders. This study uses grounded theory to understand how DI coaches in high-profile sports perceive their roles as holistic leaders and manage external challenges to provide holistic college athlete experiences. Semi-structured interviews with 12 football and women’s basketball coaches revealed five main themes: 1) coaches are educators, 2) strategies for holistic coaching, 3) supporting Black college athletes, 4) college sports …

At the National Collegiate Athletic Association (NCAA) Division I (DI) level, senior athletic administrators (SAAs) play a critical role in hiring Mental Performance Consultants (MPCs). Researchers found that NCAA DI administrators generally believe MPCs are beneficial in helping student-athletes manage competition pressures and improve performance, the number of full-time MPCs in NCAA DI athletic departments is still limited. In this study, eleven NCAA DI SAAs with at least one full-time MPC in their athletic department that solely delivered mental performance services were interviewed about their perceptions of and experiences with MPCs and their services. In 2022-2023, across all Division I women's college sports teams, women only held 42% of all head coaching positions. Women's college basketball represents one of the outlier sports, as almost 70% of their head coaches for NCAA Division I women's basketball teams were women. While women head coaches are well represented in women's college basketball, Gerretsen et al. (2023) suggest inconsistencies exist in past experience, tenure length, and gender perceptions in women's college basketball hiring practices. Athletic administrators often assume a distinct stakeholder role that has the most immediate and frequent impact on college athletes’ well-being and retention. The implementation of name, image, and likeness (NIL) represents a historic transformation in college athletics. Utilizing a sport development framework, particularly the retention stage, to examine elite athlete development, how athletic administrators socially support college athletes in the new context of NIL and its impact on administrators’ work experiences were empirically assessed. International college athletes migrating to the United States for athletic competition encounter unique challenges such as language barriers, cultural adjustment, and familial detachment. These obstacles can hinder international college athletes’ transition compared to those of domestic athletes. While supportive programming and personnel have been illustrated to ease the transition and foster acculturation among international college athletes, the impact of the geographic location of an institution may impact the acculturation process for international college athletes. This paper examines the role that false memories play in eyewitness testimony. Though eyewitness testimonies are a direct source of evidence regarding a crime, they are often flawed due to the malleable and reconstructive nature of memory, leading to many wrongful convictions. This review synthesizes previous research on false memories and eyewitness testimony to analyze the factors that contribute to inaccuracies in eyewitness testimony. Background: Peer influence is central to adolescent smoking initiation, yet its impact varies depending on individual and contextual factors. Understanding which moderators (personality, contextual, cultural, and environmental traits) shape these processes can inform more effective prevention strategies. We investigated hypothesized moderators of peer influence for adolescent smoking/vaping norms and other smoking-related outcomes in high and low-middle-income countries (LMICs): Northern Ireland and Bogotá.

Userland memory forensics has become a critical component of smartphone investigations and incident response, enabling the recovery of volatile evidence such as deleted messages from end-to-end encrypted apps and cryptocurrency transactions. However, these forensics tools, particularly on Android, face significant challenges in adapting to different versions and maintaining reliability over time due to the constant evolution of low-level structures critical for evidence recovery and reconstruction. This Article explores the role of the Preamble in constitutional interpretation by looking at how it was perceived at the founding. It does this in three ways. First, it explores how preambles were used in eighteenth-century America. At the time, preambles were almost universally understood to be key to interpreting the laws and constitutions they preceded. This shared understanding forms the backdrop against which the Preamble was received by the founding generation. For decades, courts have grappled with the tension between compensating victims of mass harms and maintaining fairness to defendants when causation is difficult to prove. This Article argues that the Supreme Court’s due process jurisprudence provides a relevant framework for navigating this tension. We contend that the Court over the last three decades has established a consistent Fourteenth Amendment due process approach in punitive damages and personal jurisdiction cases, which is rooted in antecedents tracing to the nineteenth century and relies on a nexus of interests that balances individual rights, state interests, and federalism concerns. There is a large market for goods such as t-shirts, coffee mugs, posters, and baseball caps that are emblazoned with the names and symbols of culturally prominent institutions—universities, athletic teams, musical bands, automobile brands. We buy, wear, and display those goods to express our affinity with the referenced institution. The owners of the trade symbols that adorn these items, which this Article refers to as “affinity goods,” jealously guard what they claim is their sole right, under the trademark laws, to authorize the use of their symbols as decoration. Since 1887, Congress has created an impenetrable dike wall to stop appellate review of remand orders in 28 U.S.C. § 1447(d), which provides: “An order remanding a case to the State court from which it was removed is not reviewable on appeal or otherwise . . . .” Then, in 1976, the Supreme Court held “that § 1447(d) does not mean what it says” and, thereby, punctured not a tiny hole, but a big one in Congress’s dike wall that had been impenetrable for nearly 90 years. Background: Mothers experiencing homelessness often carry extensive trauma histories, yet little work examines how abuse and sexual violence shape day-to-day parenting inside communal shelter settings. This study explored how trauma histories intersect with shelter structures to influence caregiving and feelings surrounding parenting among homeless mothers.
Method: Using a qualitative design, four mothers (ages 30–51) residing in a large Midwestern homeless shelter completed approximately 30-minute, semi-structured interviews in a private room onsite. Data were audio-recorded, transcribed verbatim, and analyzed with inductive reflexive thematic analysis in Dedoose.

Autonomous robots are increasingly deployed on construction sites for tasks such as progress monitoring, inspection, and safety assessment. For these robots to operate effectively, they must perceive and interpret complex, dynamic environments populated by workers, machinery, and unstructured terrain. Achieving reliable perception depends on high performing semantic segmentation models trained on large volumes of annotated data—an expensive and logistically challenging requirement in construction due to privacy restrictions, variable site access, and slow digitalization. Paraprofessionals often assist teachers in providing instruction to students with autism spectrum disorder (ASD) and require training to help students achieve the goal of independence (Ledford et al., 2018). However, there is a lack of quality training among paraprofessionals when implementing evidence-based teaching practices (Jones & Zarcone, 2014). The problem explored is the lack of quality training paraprofessionals receive in the use of prompts when they implement transition plans for students with ASD (Brock, 2022).
