Combinatorial Evaluation Of In Vivo Distribution Of Polyanhydride Particle-Based Platforms For Vaccine Delivery, Latrisha K. Petersen, Lucas M. Huntimer, Katharine Walz, Amanda E. Ramer-Tait, Michael Wannemueler, Balaji Narasimhan

Department of Food Science and Technology: Faculty Publications

Several challenges are associated with current vaccine strategies, including repeated immunizations, poor patient compliance, and limited approved routes for delivery, which may hinder induction of protective immunity. Thus, there is a need for new vaccine adjuvants capable of multi-route administration and prolonged antigen release at the site of administration by providing a depot within tissue. In this work, we designed a combinatorial platform to investigate the in vivo distribution, depot effect, and localized persistence of polyanhydride nanoparticles as a function of nanoparticle chemistry and administration route. Our observations indicated that the route of administration differentially affected tissue residence times. All …

Synthesis Of Adjacent-Bridged Benzo-Annelated Cyclam And Studies Of Mono- And Di-Benzo-Annelated Cyclam Derivatives, Amanuel B. Ghidey

Master's Theses and Capstones

A new synthetic methodology for the preparation of a novel adjacent-bridged benzoannelated cyclam is presented. This methodology utilizes reductive ring expansion followed by deallylation to give the desired adjacent-bridged benzo-annelated cyclam. This novel ligand is designed to complex metal cations with a trans coordination geometry. Derivatives of this ligand can be prepared by attaching pendant arms on the nitrogens which can aid as coordinating arms and linkers for potential use of this ligand as bifunctional chelator (BFC).

Coupling chemistry studies have been carried out in an attempt to increase the lipophilicity of a dibenzo-annelated tetracyclic bisaminal and to functionalize at …

The cleavage reactions of the dimers [(NHC)PdX_2]_2 with hydrophilic N-donors, L, afforded the mixed-ligand complexes of the type trans-[(NHC)LPdX_2] (X = Cl or Br; NHC = 1,3-dialkylbenzimidazol-2-ylidene (BIm) or bis(imino)acenaphthene-annulated bis(2,6-diisopropylphenyl)imidazol-2-ylidene (BIAN-IPr); L = diethanolamine (DEA), morpholine (MOR), and 3-pyridinecarboxylic acid (3-PCA)). The new complexes (1--3) were characterized by elemental analysis and spectroscopic methods and the molecular structure of 1a was determined by X-ray diffraction studies. These complexes were applied in the Suzuki--Miyaura cross-coupling reaction of phenylboronic acid with aryl halides in neat water. The activities of catalysts were monitored by gas chromatography--flame ionization detector and nuclear magnetic resonance. Whereas …

Effect Of Laser Ablation Depth In Otolith Life History Scans, Renée R. Hoover, Cynthia M. Jones Jan 2013

Effect Of Laser Ablation Depth In Otolith Life History Scans, Renée R. Hoover, Cynthia M. Jones

OES Faculty Publications

Life history scans of fish otoliths are bringing new insight into the structure, connectivity, and movement of fish populations. Data obtained from such scans, however, possess in-herent limitations that have not yet been fully addressed or understood. For example, several investigators have noted delays in otolith elemental uptake that do not appear to reflect habitat exposure. We hypothesized that the 3-dimensional structure of otoliths may produce sampling artifacts in the results obtained from laser ablation scans. To test this hypothesis, we sampled sagittal otoliths from juvenile Atlantic croaker Micropogonias undulatus with laser ablation inductively coupled plasma mass spectrometry to obtain …

Trace-Element And Physical Response To Melt Percolation In Summit (Greenland) Snow, Gifford J. Wong, Robert L. Hawley, Eric R. Lutz, Erich C. Osterberg Jan 2013

Trace-Element And Physical Response To Melt Percolation In Summit (Greenland) Snow, Gifford J. Wong, Robert L. Hawley, Eric R. Lutz, Erich C. Osterberg

Dartmouth Scholarship

Surface melt on a glacier can perturb the glaciochemical record beyond the natural variability. While the centre of the Greenland ice sheet is usually devoid of surface melt, many high- Arctic and alpine ice cores document frequent summertime melt events. Current hypotheses interpreting melt-affected ice-core chemistry rely on preferential elution of certain major ions. However, the precise nature of chemistry alteration is unknown because it is difficult to distinguish natural variability from melt effects in a perennially melt-affected site. We use eight trace-element snow chemistry records recovered from Summit, Greenland, to study spatial variability and melt effects on insoluble trace …

Robust Methods For Construction Of Global Potential Energy Surfaces, Phalgun Lolur Jan 2013

Robust Methods For Construction Of Global Potential Energy Surfaces, Phalgun Lolur

Masters Theses

"This thesis is about the development of robust methods for construction of global potential energy surfaces to study the spectroscopy and dynamics of molecular systems. A potential energy surface represents the electronic energy of a molecule as a function of its geometry. This is central to how chemists view molecular systems in terms of motion across a rich energy landscape where barriers separate wells corresponding to different stable structures. The range of molecular distortions defines the potential energy surface. Computing the potential energy surface of a molecule has become a fundamental operation in modern theoretical chemistry studies. The Born-Oppenheimer approximation …

Computational Chemistry Capacity Building In An Underprivileged Context: Challenges, Outcomes And Perspectives, L Mammino

Tanzania Journal of Science

Computational chemistry is a fast developing branch of modern chemistry, focusing on the study of molecules to enable better understanding of the properties of substances. Its applications comprise a variety of fields, from drug design to the design of compounds with desired properties (e.g., catalysts with specific actions) and to ample overlaps with nanotechnology. However, despite its relevance, the presence of computational chemistry research in Sub-Saharan Africa is still scarce, practically absent in many institutions. Bridging the gap with the other continents requires the identification of capacity building options that may enable a rapid growth. The recent experience at the …

Conservative And Accurate Data Remapping For Coupling Climate Models (Wrf And Cam), Ying Chen Dec 2012

Conservative And Accurate Data Remapping For Coupling Climate Models (Wrf And Cam), Ying Chen

Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)

The two-way coupling of WRF and CAM for regional climate modeling requires transferring data (such as wind velocities, temperature, moisture, etc.) between the grids of WRF and CAM. These grids are in general non-matching, and the data transfer should be both physical conservative and numerically accurate. Our method contains two parts: constructing the common refinement and conservative data transfer using the common refinement. The former computes the cell intersections and provides the necessary data structures for the latter. We project the grids of WRF and CAM onto a plain surface using their corresponding map projection methods (such as: Lambert conformal …

A Study Of Data Mining Trend Through The Optimized Bibliometric Methodology Based On Sci Database From 1993 To 2011, Lu Dai, Lixin Ding, Yunwen Lei, Yangge Tian

Applied Mathematics & Information Sciences

An optimized bibliometric method was applied in this work to evaluate global scientific production of data mining papers of the Science Citation Index (SCI). In compared with traditional bibliometric keyword analysis, a semantic words class was established by applying the text extraction mode to remove noise in the abstract and combining with the core relative phrases retrieved from keywords to get the sample for further experiment. The analysis shows a high correlation between title and keywords, and the title reports less information than keywords does. Also, keywords provide more positive guidance to know and be familiar with the status and …

Physical Environmental And Biological Correlates Of Otolith Chemistry Of Arctic Marine Fishes In The Chukchi Sea, Christine Marie Gleason Aug 2012

Physical Environmental And Biological Correlates Of Otolith Chemistry Of Arctic Marine Fishes In The Chukchi Sea, Christine Marie Gleason

Marine Biology

Life history movement patterns in marine fishes can be determined by otolith chemistry if environmental variables are reflected in the otoliths. Arctic cod (Boreogadus Saida), Arctic staghorn sculpin (Gymnocanthus tricuspis), and Bering flounder (Hippoglossoides robustus) are abundant Arctic fishes in the Chukchi Sea with overlapping distributions. Physical environmental data, demersal fishes, bottom seawater, and sediment interface seawater samples were collected from the Chukchi Sea Offshore Monitoring in Drilling Area (COMIDA) cruise on July 30, 2009 and the Russian American Long-term Census of the Arctic (RUSALCA) cruise from September 3 to 30, 2009 in the Chukchi Sea. Magnesium (Mg), strontium (Sr), …

In comparison with other primate species, humans have an extended juvenile period during which the brain is more plastic. In the current study we sought to examine gene expression in the cerebral cortex during development in the context of this adaptive plasticity. We introduce an approach designed to discriminate genes with variable as opposed to uniform patterns of gene expression and found that greater inter-individual variance is observed among children than among adults. For the 337 transcripts that show this pattern, we found a significant overrepresentation of genes annotated to the immune system process (pFDR>0). Moreover, genes known to …

Synthesis Of Novel Alkaline Earth Tosylate Complexes Through Donor Studies, Adam J. Brooks May 2012

Synthesis Of Novel Alkaline Earth Tosylate Complexes Through Donor Studies, Adam J. Brooks

Renée Crown University Honors Thesis Projects - All

Salt metathesis has become a critical reaction pathway in organometallic chemistry; however, for alkaline earth metals this process is hindered by the need of highly pure and expensive metal iodides as starting materials. This contribution highlights recent attempts to identify inexpensive reagents for salt metathesis. Alkaline earth-tosylates can be prepared cost effectively using p-toluenesulfonic acid in one-pot reactions in water. The coordinated water molecules can be removed by gentle heating under vacuum. The anhydrous tosylates, however, have major solubility limitations as the result of extensive aggregation networks. The goal of this study was to increase the solubility of the tosylates …

Nongenomic Mechanisms Of Pten Regulation, Jimmie E. Fata, Shawon Debnath, Edmund C. Jenkins, Marcia V. Fournier

A large amount of data supports the view that PTEN is a bona fide tumor suppressor gene. However, recent evidence suggests that derailment of cellular localization and expression levels of functional nonmutated PTEN is a determining force in inducing abnormal cellular and tissue outcomes. As the cellular mechanisms that regulate normal PTEN enzymatic activity resolve, it is evident that deregulation of these mechanisms can alter cellular processes and tissue architecture and ultimately lead to oncogenic transformation. Here we discuss PTEN ubiquitination, PTEN complex formation with components of the adherens junction, PTEN nuclear localization, and microRNA regulation of PTEN as essential …
