Turkish Journal of Zoology

Anurans are the most suitable vertebrate group to see the development and also evolution in the skull, compared to other terrestrial vertebrates as they show highly derived morphology. For this reason, the cranium of anurans has been widely used for analyzing e.g., development and integration, evolutionary history, phylogenetic relationships, sexual dimorphism; and for taxon determination. We applied geometric morphometric techniques and then multivariate statistical analyses to see the skull size and shape variations in closely related Bufo species (B. bufo and B. verrucosissimus) inhabiting Turkey. Females have larger skull than males for both species. Ventral skull size of males differed …

Biomedical and Pharmaceutical Sciences Faculty Publications

Oleanolic acid (OA) is a natural cosmeceutical compound with various skin beneficial activities including inhibitory effect on hyaluronidase but the anti-hyaluronidase activity and mechanisms of action of its synthetic analogues remain unclear. Herein, a series of OA derivatives were synthesised and evaluated for their inhibitory effects on hyaluronidase. Compared to OA, an induction of fluorinated (6c) and chlorinated (6g) indole moieties led to enhanced anti-hyaluronidase activity (IC50 = 80.3 vs. 9.97 and 9.57 µg/mL, respectively). Furthermore, spectroscopic and computational studies revealed that 6c and 6g can bind to hyaluronidase protein and alter its secondary structure leading …

Photoassisted Synthesis Of Complex Polyheterocycles Via Rational Design Of Tailored Photoprecursors, Tina Alanna Holt Jan 2021

Photoassisted Synthesis Of Complex Polyheterocycles Via Rational Design Of Tailored Photoprecursors, Tina Alanna Holt

Electronic Theses and Dissertations

Natural products remain the most common source for drug leads, although diversity-oriented synthesis of unnatural complex molecules is gaining momentum. Our lab has been developing experimentally simple and straightforward approaches to complex molecular architectures via photochemical reactions involving a minimal number of experimental steps. In order to achieve this, photoprecursors were assembled via high yielding reactions. Next, we deployed photoinduced cycloadditions to create complex 3-dimentional structures. Lastly, with this obtained primary photoproduct, post-photochemical transformations were used to further grow its scaffold complexity.

One method to increase scaffold complexity is the [4+2] intramolecular cycloadditions of azaxylylenes, which are developed through standard …

A Biophysical Investigation Of Hiv-1 Envelope Cytoplasmic-Tail Mediated Retention In The Gag Lattice, Nicholas Scott Groves Jan 2021

A Biophysical Investigation Of Hiv-1 Envelope Cytoplasmic-Tail Mediated Retention In The Gag Lattice, Nicholas Scott Groves

Electronic Theses and Dissertations

The infectivity of Human Immunodeficiency Virus is critically dependent on successful incorporation of the trimeric viral envelope glycoprotein, Envelope (Env), into Gag lattice assembly sites on the host-cell plasma membrane during virus biogenesis. The mechanistic nature of interaction between Gag, the structural protein, and Env, the viral spike, are dependent on the Gag matrix (MA) and Env cytoplasmic tail (CT) domains. Env laterally diffuses on the host cell plasma membrane until encountering a budding Gag lattice, where it is retained until particle scission and release. The mechanism of Env entry and retention in the lattice remains elusive and enigmatic. Here …

Faculty Research 2021

Multiple mutations have been described in the human GBA1 gene, which encodes the lysosomal enzyme beta-glucocerebrosidase (GCase) that degrades glucosylceramide and is pivotal in glycosphingolipid substrate metabolism. Depletion of GCase, typically by homozygous mutations in GBA1, is linked to the lysosomal storage disorder Gaucher's disease (GD) and distinct or heterozygous mutations in GBA1 are associated with increased Parkinson's disease (PD) risk. While numerous genes have been linked to heritable PD, GBA1 mutations in aggregate are the single greatest risk factor for development of idiopathic PD. The importance of GCase in PD necessitates preclinical models in which to study GCase-related mechanisms …

Identification Of Loci Influencing Teosinte Crossing Barrier 1 (Tcb1) Efficacy In Maize By Quantitative Trait Loci (Qtl) Mapping And Genome-Wide Association Study (Gwas), Namrata Maharjan

Electronic Theses and Dissertations

Pollen cross-contamination has been a major problem for maize breeders. Various mechanical methods applied to avoid the contaminations are ineffective. The genetic factors related to maize fertilization can be used to develop an effective method to prevent pollen contamination. Pollen rejection ability controlled by Teosinte crossing barrier 1 (Tcb1) is such a genetic system. Silks possessing dominant Tcb1-s reject pollen possessing the recessive allele (tcb1). Successful fertilization occurs when Tcb1-s pollen falls upon tcb1 silks. The efficacy of dominant Tcb1-s was, however, reduced when repeatedly backcross with maize inbred lines, which suggests that there are modifiers to Tcb1-s. To find …

Multi-Functional Fluorescence Microscopy Via Psf Engineering For High-Throughput Super-Resolution Imaging, Jinhan Ren

Electronic Theses and Dissertations, 2020-2023

Image-based single cell analysis is essential to study gene expression levels and subcellular functions with preserving the native spatial locations of biomolecules. However, its low throughput has prevented its wide use to fundamental biology and biomedical applications which require large cellular populations in a rapid and efficient fashion. Here, we report a 2.5D microcopy (2.5DM) that significantly improves the image acquisition rate while maintaining high-resolution and single molecule sensitivity. Unlike serial z-scanning in conventional approaches, volumetric information is simultaneously projected onto a 2D image plane in a single shot by engineering the fluorescence light using a novel phase pattern. The …

Effect Of 5-O-(N-Boc-L-Alanine)-Renieramycin T In The Suppression Of Lung Cancer Stem Cells, Darinthip Suksamai

Chulalongkorn University Theses and Dissertations (Chula ETD)

Cancer stem cells (CSCs) drive aggressiveness and metastasis by utilizing stem cell-related signals. In this study, 5-O-(N-Boc-L-alanine)-renieramycin T (OBA-RT) was demonstrated to suppress CSC signals and induce apoptosis. OBA-RT exerted cytotoxic effects with a half-maximal inhibitory concentration of approximately 7 µM and mediated apoptosis as detected by annexin V/PI and nuclear staining assays. Mechanistically, OBA-RT exerted dual roles, activating p53-dependent apoptosis and concomitantly suppressing CSC signals. A p53-dependent pathway was indicated by the induction of p53 and the depletion of anti-apoptotic Mcl-1 and Bcl-2 proteins. Cleaved PARP was detected in OBA-RT-treated cells. Interestingly, OBA-RT exerted strong CSC-suppressing activity, reducing the …

Isolation Of Marker Compounds From Copaiba Essential Oil And Mimosa Pigra For Quality Evaluation Of Dietary Supplements, Mohammed Hawwal

Electronic Theses and Dissertations

Natural products have been used as a primary means of treating illness since ancient times, and they remain an important pool of U.S. FDA approved drugs. Approximately 33% of all current pharmaceutical medications discovered between 1981 and 2014 are either natural products or derived from natural products (Newman and Cragg 2016). The process of isolating specialized metabolites from medicinal plants has led to the development of particularly important medicines worldwide (Rakotomalala, Agard et al. 2013) such as Taxol® from Taxus brevifolia Nutt. (Taxaceae) and salicylic acid from the bark of Salix spp. This research is composed of five chapters; the …

Material Engagement Theory And Extensive Enactivism Within The 4e Cognitive Debate: A Phenomenological Approach To Material Agency And Application To Current Technology, Israel Quirit

Theses

4E Cognition is a fairly new field of study within cognitive science, cognitive psychology, and cognitive philosophy. The various approaches to cognition namely embodied, embedded, extended and enacted cognition; provide multi-faceted approaches to cognition. A common claim of these approaches is that cognition may have various contributing factors such as the role of the brain, body and its environments. Thus challenges the traditional idea that cognition exists purely mentally within the confines of the brain and skull. However, even within the 4E Cognition family, some nuanced arguments are hotly debated, particularly among the proponents of Embedded Theories (EmT) and Extended …

Probing Interactions Of Single-Walled Carbon Nanotubes At The Nano-Bio Interface, Mitchell Gravely Jan 2021

Probing Interactions Of Single-Walled Carbon Nanotubes At The Nano-Bio Interface, Mitchell Gravely

Open Access Dissertations

Non-covalent hybrids of single-stranded DNA and single-walled carbon nanotubes (DNA-SWCNTs) have demonstrated applications in biomedical imaging and sensing due to their enhanced biocompatibility and photostable, environmentally-responsive near-infrared (NIR) fluorescence. Significant progress has been made in developing robust biological probes based on the sensitive optical properties of SWCNTs, however biological environments introduce complex and dynamic conditions that interact with, and ultimately modulate, their intrinsic properties. These fundamental interactions within biological settings can determine whether a nanomaterial is biocompatible and robust, or cytotoxic and disruptive to biological processes. Thus, a mechanistic understanding of such interactions and their effect on cellular function is …

Photoreceptor Phosphodiesterase (Pde6): Activation And Inactivation Mechanisms During Visual Transduction In Rods And Cones, Rick H. Cote Jan 2021

Photoreceptor Phosphodiesterase (Pde6): Activation And Inactivation Mechanisms During Visual Transduction In Rods And Cones, Rick H. Cote

Faculty Publications

Rod and cone photoreceptors of the vertebrate retina utilize cGMP as the primary

intracellular messenger for the visual signaling pathway that converts a light stimulus into an electrical response. cGMP metabolism in the signal-transducing photoreceptor outer segment reflects the balance of cGMP synthesis (catalyzed by guanylyl cyclase) and degradation (catalyzed by the photoreceptor phosphodiesterase, PDE6). Upon light stimulation, rapid activation of PDE6 by the heterotrimeric G-protein (transducin) triggers a dramatic drop in cGMP levels that lead to cell hyperpolarization. Following cessation of the light stimulus, the lifetime of activated PDE6 is also precisely regulated by additional processes. This review summarizes …

Interdisciplinary Stem Undergraduate Programs And The Effectiveness Of Computing Competencies Within The Curriculum, Katherine G. Herbert, Thomas J. Marlowe, Kees Leune, Robert M. Siegfried, Jeanette Wilmanski

Department of Computer Science Faculty Scholarship and Creative Works

Undergraduate interdisciplinary, multidisciplinary and transdisciplinary computing-related STEM programs are proliferating extensively. Each of these programs requires a robust computing component to be integrated into the curriculum. However, including an introductory, programmingoriented sequence designed for CS majors is not always the best fit for these multifaceted programs. In this work in progress paper, we set out to investigate possibilities and issues for the computing component, focusing on three fields: bioinformatics, data science, and cybersecurity. We explore commonalities and differences, and discuss initial plans to test our hypotheses. In doing so, we consider data organization, data acquisition, and a preliminary survey design.
