DNA polymerase θ (Pol θ)-mediated end-joining (TMEJ), one of several pathways for repairing DNA double-strand breaks, is traditionally thought to initiate via anchoring at short, consecutive, and perfectly matched microhomologies (MHs). Emerging evidence indicates that Pol θ can utilize MHs containing mismatches both in vitro and in vivo. This revised definition of MH provides a mechanistic explanation for a broader spectrum of Pol θ-dependent repair outcomes. Here, we summarize recent findings on the revised definition of MHs utilized by Pol θ, assess the applicability of this concept across species, and compare TMEJ with other (micro)hom(e)ology-mediated repair pathways. RATIONALE & OBJECTIVE: Idiopathic nephrotic syndrome (INS) is viewed as a podocyte-specific disease. Recent reports indicate endothelial involvement, but its significance is unclear. Here, we investigated the relationship between the glomerular expression of selected genes relevant to endothelial health and clinical markers of disease severity.

STUDY DESIGN: A cross-sectional study.

SETTING & PARTICIPANTS: Patients with INS (n = 70 minimal change disease and n = 83 focal segmental glomerulosclerosis) from the Nephrotic Syndrome Study Network cohort study and 53 control participants. Validation studies, including animal and cell culture experiments, were performed.

Background: The global obesity crisis and the limited success of current treatments underscore the need to identify novel regulatory pathways. While central administration of α-Klotho exerts anti-obesity effects in rodents through AgRP neurons, the intracellular signaling mechanisms that mediate this process remain undefined.

Methods: To define the role of FGFR1 within the α-Klotho signaling pathway in AgRP neurons, we performed a targeted deletion of the receptor in adult mice using an AAV-mediated CRISPR/Cas9 system alongside transgenic models.

Antibody-drug conjugates (ADCs) have revolutionized breast cancer therapy. HER2 is the only validated biomarker guiding ADC therapy in breast cancer. However, their clinical benefit remains confined to HER2- and TROP2-targeted therapies. Tumor heterogeneity and acquired resistance often limit durable responses, underscoring the need for novel, tumor-specific ADC targets. CD44 variant isoform 9 (CD44v9), a splice variant of the CD44 family, is largely absent in normal tissues but enriched in aggressive breast cancers, where it contributes to stemness, redox regulation, and therapy resistance. Peripheral Arterial Occlusive Disease (PAOD) affects millions of people in the United States, creating pain, discomfort, and limited mobility due to restricted blood flow from obstructed arteries. While current treatments include lifestyle changes and revascularization procedures, these options are often insufficient or inaccessible because of procedural pain and socioeconomic barriers. In some cases, this can lead to critical limb ischemia and amputation as a last resort. Low-coverage whole genome sequencing (lcWGS) combined with genotype imputation provides a cost-efficient alternative to high-coverage sequencing for large-scale genotyping. Although widely implemented in human and livestock genomics, this strategy has not yet been systematically optimized for insects of industrial importance. The black soldier fly (BSF, Hermetia illucens ) is increasingly used in global waste bioconversion and sustainable protein production, but genomic resources remain limited. The failure to prevent brain tumors, including both recurrent primary and metastatic brain tumors, is the primary cause of patients' mortality. We developed a novel whole tumor-cell vaccine to rapidly induce long-duration brain-resident memory T (TRM) cells that prevent brain tumor progression. Ten Fgl2-KO primary and metastatic tumor cell lines, generated via CRISPR/Cas9, were used to vaccinate mice and for intracranial challenges with the WT tumor cells. Not only did vaccinated mice reject these tumor cell challenges, but also more than half of these mice became long-duration survivors. Background: Understanding the median nerve's position relative to surrounding anatomy is essential; however, there are many variations among individuals. This study assesses differences in median nerve position with or without palmaris longus (PL). We hypothesize that PL presence alters median nerve position, resulting in a greater distance to the skin volar surface, a decreased distance to the radius volar surface, and an increased distance to the flexor carpi radialis (FCR).

Community philanthropy is increasingly promoted as an alternative to institutional, top-down philanthropy, yet the term remains contested. In the Global North, it is typically associated with community foundations, while in the Global South the emphasis is on community itself.

This article offers a definition of community philanthropy as a practice rooted in context and power rather than a single institutional form. It identifies five transformative characteristics that distinguish community philanthropy from conventional giving and translates these principles into seven dimensions that can be used to identify and assess community philanthropy in practice.

Despite rising interest in supporting local causes, many donors continue to give to large, nationally recognized nonprofits. This reflects ongoing structural barriers such as limited nonprofit visibility, decision fatigue, and search environments that prioritize brand familiarity.

At the same time, small-dollar giving has declined for three consecutive years, highlighting the need for more transparent, accessible, and trustworthy tools to support donor decision-making (Childress, 2024; Lilly Family School of Philanthropy, 2023).

The study offered a 5-week summer immersion program to address the shortage of students from underrepresented populations enrolling in degree programs and seeking careers in science, technology, engineering, and mathematics (STEM). The study also addressed the importance of offering summer immersion programs to close the achievement gap. A quantitative descriptive design was used to gather data from participants related to STEM lessons during the program. Gender, grade level, and race served as the demographic variables. Black feminist theory has identified sexuality as a site of oppression for Black women, however, there has been limited attention to Black women’s pleasure. We suggest that WAP by Cardi B and Meg the Stallion provides a conceptual lens through which Black women's sexuality can be more humanely envisioned. WAP, in the tradition established by Black women in the blues, represents Black feminist theorizing about sexuality. Madwoman Makings: The Modern Girl As An Object Of Fear And Fascination In Unno Jūza’S "Mrs. Hirumi's Refrigeration Bag", Eliza Joy Mortimer

This thesis argues that Unno Jūza's 1937 short story "Mrs. Hirumi's Refrigeration Bag" uses the sensational tropes of deviant detective fiction to both stage and interrogate interwar Japanese anxieties around gender, science, and modernity. Focusing on the figure of Mrs. Hirumi—a scientifically empowered and sexually assertive female plastic surgeon who dismembers and cryogenically preserves her husband—the author reads her as a radical iteration of the Japanese Modern Girl character. By drawing on scholarship on deviant detective fiction, imperial modernity, and Kazue Harada's concept of (re)productivity, this thesis demonstrates how the short story both reproduces misogynistic criminological discourse and unsettles heteronormative …

This capstone review examines the development of AI Fishbowl, a public-facing, interactive artificial intelligence system, as a case study in how Agile methods evolve from a project management tool into a design philosophy under real-world constraints. Although the project adopted an Agile workflow early on through a Kanban-style task management approach, the initial system design and architecture were still shaped by a largely plan-first mindset. This created a mismatch between flexible process and rigid design assumptions, which became increasingly apparent as the team moved from high-level architecture into implementation.

OBJECTIVE: Periodontal disease is associated with increased risk of preterm birth. Effective management during pregnancy may reduce preterm birth, though clinical trial evidence has been inconsistent. This study aimed to assess whether treating dental disease during pregnancy reduces preterm birth.

DATA SOURCES: A systematic search was performed of EMBASE, MEDLINE, PubMed, Cochrane Library, and trial registries up to December 2023 and re-run in January 2025.

STUDY ELIGIBILITY CRITERIA: Randomized controlled trials (RCTs) enrolling pregnant persons with any dental disease randomized to receive dental treatment vs no treatment were included. No language restriction was applied.

Youth urban agriculture programs connect youth with nature and support positive youth development. In this qualitative study, we use photovoice to amplify youth voices and explore 16 high school youths’ perceptions of how participation in a youth gardening program supported their confidence, connection, and sense of belonging. Using qualitative thematic analysis of photos, posters, and interviews, we identify several program processes that supported confidence and belonging, including meaningful relationships with peers and adults, connections with nature, and relationships with more-than-human beings. The neuronal cell adhesion molecule NrCAM is widely expressed in the nervous system across the lifespan and has important physiological functions in the development of neuronal circuits through axonal growth and guidance and formation and maintenance of synapses in the cortex. NrCAM gene polymorphisms are associated with vulnerability to neuropsychiatric disorders such as schizophrenia, as well as vulnerability to substance use disorders. Societal Impact Statement: Plants provide oxygen, food, shelter, medicines and environmental services, without which human society could not exist. Tackling pressing and global challenges requires well-trained plant scientists and plant-aware individuals. This manifesto provides a practical evidence-based vision to strengthen plant science education, focused on five strategic priorities. It is relevant to all stakeholders within plant science and beyond: from frontline educators to institutional leaders; from commercial or charitable professionals to entrepreneurs and donors; from individual community members to their legislative representatives. Jumonji domain-containing 1A (JMJD1A, also known as KDM3A) is a histone demethylase that specifically demethylates H3K9me1/2 to enhance gene expression. The roles of JMJD1A in many physiological and pathological processes have been revealed. However, it is unclear whether JMJD1A is involved in host defense against enteric pathogen infection. In this study, we found that enteric infection with C. rodentium induced JMJD1A expression in colonic epithelial cells at the transcriptional level partly mediated by IRF1. After C. rodentium infection, JMJD1A-/- mice exhibited increased mortality, colonic injury, and C. rodentium load and systemic spread, suggesting that JMJD1A protects host against C. rodentium …

Nitric oxide (NO) is a critical signalling molecule in cardiovascular, metabolic, and muscular function. Endogenous NO production occurs via two primary metabolic pathways: 1) the classical nitric oxide synthases (NOS) pathway, and 2) the alternative (nitrate-nitrite-NO) pathway, in which inorganic nitrate (NO 3 -) is sequentially reduced to nitrite (NO 2 -) and other NO intermediates (e.g., S-nitrosothiol). The latter pathway relies heavily on the oral microbiota, which catalyze the two-electron partial reduction of NO 3 - to NO 2 -, which is influenced by oral physiology, microbial composition and salivary flow. Aging research relies on varied statistical methods, and applying these methods appropriately is important for scientific rigor. However, proper use of these statistical techniques is a challenge. We discuss two categories of statistical methods in aging research: (a) emerging methods requiring further validation, including techniques to examine compression of morbidity, maximum lifespan, immortal time bias, molecular aging clocks, and treatment response heterogeneity, and (b) classic and existing methods needing reconsideration and improvement, such as stepwise regression, generalized linear models, methods for accounting for clustering and nesting effects, methods for testing for group differences, methods for mediation …

Tumor evolution involves genetic, transcriptional, and phenotypic alterations that shape cancer cell behavior and interactions with the microenvironment. While single‐cell technologies have advanced our understanding of this process, spatial dynamics remain incompletely characterized. Here, whole‐exome sequencing (WES), imaging mass cytometry (IMC), and spatial transcriptomics (ST) were integrated to study molecular evolution and immune responses in two lung adenocarcinoma (LUAD) mouse models: a genetically engineered model (129S4/Sv‐KrasLSL‐G12D, termed 129S4 K) and a carcinogen‐induced precancer model (129S4 U). Stepwise regression remains widely used for model selection, yet the field lacks a comprehensive, well-documented tool that supports diverse model families and selection strategies, implements multiple information criteria, and addresses overfitting and post-selection inference. We present StepReg, an R package that unifies stepwise selection across linear, generalized linear (e.g., logistic, Poisson, Gamma, negative binomial), and Cox models, supporting forward, backward, bidirectional, and best-subset search under various information criteria. StepReg also supports multivariate multiple linear stepwise regression, enabling simultaneous modeling of multiple dependent variables. Appetite self-regulation (ASR) among children is thought to have a fundamental role in shaping the development of healthy eating behaviors, dietary intake, and growth during childhood. Parallel to developmental frameworks for understanding "general" self-regulation among children, ASR has been described as involving children's use of "top-down" cognitive processes to moderate "bottom-up" biological drives around food approach and avoidance in the interest of achieving desired eating behaviors or outcomes. Cardiovascular disease remains the leading cause of death worldwide with low density lipoprotein being a major, yet modifiable, risk factor. Proprotein convertase subtilisin/kexin type 9 (PCSK9) plays a central role in regulating low density lipoprotein (LDL) receptor expression. Naturally occurring loss-of-function variants in the PCSK9 gene result in lifelong lower LDL cholesterol (LDL-C) levels and significantly lower risk of atherosclerotic cardiovascular disease (ASCVD), providing strong genetic validation of PCSK9 as a therapeutic target. Background: To investigate the proteomic signatures of heart failure (HF) in diabetes. The underlying mechanisms of the elevated risk of HF in diabetes are unknown.

Methods: In 10 189 ARIC study (Atherosclerosis Risk in Communities) participants free of HF (mean age 57±7 years, 56% women, 22% Black adults, 14% with diabetes), we conducted discovery and internal validation for the associations of 4955 plasma proteins with HF by diabetes status. We performed (1) Cox regression to identify proteins associated with HF by diabetes status, (2) external validation in the MESA study (Multi-Ethnic Study of Atherosclerosis, n=5233, 633 with diabetes), and (3) …
