The central nervous system plays a vital role in the homeostatic control of body weight and metabolism. Among brain nuclei, the ventromedial hypothalamus (VMH) is an important hypothalamic nucleus known to regulate body weight, energy balance, and counterregulatory response. However, the pathways for VMH neurons to regulate lipid metabolism and the mechanisms by which VMH neurons sense nutrients and hormones remain obscure. O-linked β-D-N-acetylglucosamine modification (O-GlcNAcylation), catalyzed by O-GlcNAc transferase (OGT), is considered a cellular sensor of stress, nutrients, and hormones, regulating a variety of cellular activities. Nitrogen heterocycles are present in almost two-thirds of U.S. FDA approved pharmaceuticals, as well as biologically active natural products. The first two chapters discuss the application of C-H activation methodology to explore novel, nitrogen heterocycle containing molecules as potent and selective ligands towards different protein targets. The third chapter depicts the design of new N-heterocyclic covalent molecular probes that uniquely labels a serine hydrolase protein in a chemoproteomics screen. Chapter 1 details an investigation using structure-based docking with a virtual library of tetrahydropyridines (THPs) to identify unique agonists for the serotonin 5-HT2A receptor (5-HT2AR). The burden of pain is significant and current pain treatments are often ineffective. Opiate use has led to an epidemic, requiring alternative pain medications that are more effective and non-addictive. Voltage-gated sodium channel NaV1.7 is preferentially expressed in pain-sensing neurons. Mutations in NaV1.7 can cause disorders ranging from intense pain to complete painlessness in humans, suggesting that its inhibition could provide analgesia without CNS side-effects or addictive potential. Indeed, existing non-selective NaV channels blockers prevent pain, but their utility is limited due to serious side-effects caused by inhibition of NaV isoforms expressed in the heart and brain. Social evaluation refers to the process of using observed social behavior to determine who will be a suitable social partner. While this phenomenon is well-documented in this human species, less is known regarding how we acquired the cognitive abilities needed to make this assessment of with whom we should affiliate. To better understand the evolutionary origins of social evaluation abilities in the human species, researchers have explored nonhuman animals’ capacities for social evaluation. Researchers are beginning to explore the impact of the COVID-19 pandemic on island populations, but little is known about the experiences of the residents of Nantucket and Martha’s Vineyard in the Northeast region of the United States. In this study, islanders (N=41) provide insight into how spending time in nature influenced their well-being, especially their mental well-being, during this unprecedented time. For the majority of participants, the influence of nature on their well-being increased during the pandemic. Additionally, participants shared benefits of island living such as isolation from the mainland, as well as challenges such as food and supply shortages. …

Elizabeth Acevedo, an author and poet, uses her beautiful piece of slam poetry to explore how language can empower marginalized voices, speaking on behalf of those who are usually silenced. The Poet X is a verse novel that narrates the powerful story of a young Afro-Latina girl, Xiomara Batista, who navigates the intersecting pressures of school bullies, family issues, religious conservatism, patriarchal rules, and racialized body politics in her Harlem community. A common reprise among labor activists and scholars has been that for the fortunes of labor to change, the law must change. Prompted perhaps by a seeming surge in labor movement activity over the past few years, including headline-grabbing strikes and recent union victories at several U.S. Starbucks locations, various labor law activists and scholars have called to seize the moment and proposed the enactment of comprehensive labor law reform. On the Great Plains, the Middle Holocene (8200–5000 yr BP) was a warm period where foraging peoples expanded into new food resources and developed new food processing technologies. It is unclear precisely what drove these cultural changes, and environmental explanations tend to assume that climatically mediated causes occurred along latitudinal gradients, with the southern Plains experiencing severe water shortages and declining abundances of important resources relative to the northern Plains. A Randomized Waitlist-Controlled Pilot Trial Of Online-Delivered Comprehensive Behavioral Intervention For Tics-Enhanced, Jordan Stiede

Although studies demonstrate that comprehensive behavioral intervention for tics (CBIT) is more effective than nonspecific supportive therapy in reducing tics and related impairment, there is room for improvement, as up to 47% of treatment seeking youth do not show adequate response to treatment. Accordingly, the current randomized, waitlist-controlled pilot trial examined the preliminary efficacy, acceptability, and durability of online-delivered CBIT-Enhanced (CBIT-E), which included the standard CBIT protocol, plus specific and targeted instrumental conditioning procedures focused on directly reinforcing the use of inhibitory competing responses. Feasibility Of Sewer Overflow Treatment Using Chemical Oxidation After Rapid Solids Removal, Paige Peters

Sewer overflows discharge 850 billion gallons of untreated sewage into U.S. lakes and rivers every year during high-intensity precipitation events. Common approaches include storage tanks or rapid solids removal followed by chlorine disinfection. These approaches can be capital intensive and only achieve partial treatment. A decentralized, end-of-pipe sewer overflow treatment system would eliminate detrimental overflow effects while handling highly variable flow and pollutant loading. Specifically, the rapid reaction rate of chemical oxidation can achieve soluble organics removal and disinfection of chlorine-resistant pathogens in a small footprint. Nonprofit scholars and practitioners alike adhere to a long-held assumption that nonprofit work is, and will remain, inherently meaningful work. The long-term marketization of the nonprofit sector coupled with the influence of the COVID-19 pandemic has undercut this narrative. Our research on meaningful nonprofit work indicates that while many nonprofit workers do find their work meaningful, pay, flexibility, and work/life balance are increasingly important to them. This commentary suggests that nonprofit leaders can no longer presume that workers motivated by prosocial values will seek out and stay with nonprofit work, satisfied with the “psychic income” that comes from doing good …

Diverse mouse strains have different health and life spans, mimicking the diversity among humans. To capture conserved aging signatures, we studied long-lived C57BL/6J and short-lived NZO/HILtJ mouse strains by profiling transcriptomes and epigenomes of immune cells from peripheral blood and the spleen from young and old mice. Transcriptional activation of the AP-1 transcription factor complex, particularly Fos, Junb, and Jun genes, was the most significant and conserved aging signature across tissues and strains. ATAC-seq data analyses showed that the chromatin around these genes was more accessible with age and there were significantly more binding sites for these TFs with age …

Ultimately, this Article has three goals. The first is to offer an analysis of users’ rights under copyright law from four commonly used theoretical perspectives. These are labor, personality, economic and utilitarian theories. In doing so, this Article will demonstrate that the philosophies that underpin modern copyright law support a broad and liberal set of rights for derivative creativity. It will argue that current treatment of derivative works is unnecessarily conservative from a theoretical perspective. Non-fungible tokens (NFTs) are unique tokens stored on a digital ledger – the blockchain. They are meant to represent unique, non-interchangeable digital assets, as there is only one token with that exact data. Moreover, the information attached to the token cannot be altered as on a regular database. While copies of these digital items are available to all, NFTs are tracked on blockchains to provide the owner with proof of ownership. This possibility of buying and owning digital assets can be attractive to many individuals.
