Diverse factors contribute to the limited clinical response to radiotherapy (RT) and immunotherapy in metastatic non-small cell lung cancer (NSCLC), among which is the ability of these tumors to recruit a retinue of suppressive immune cells-such as M2 tumor-associated macrophages (TAM)-thereby establishing an immunosuppressive tumor microenvironment that contributes to tumor progression and radio resistance. M2 TAMs are activated by the STAT6 signaling pathway. PURPOSE: Cisplatin (CDDP)-based chemotherapy is a first-line treatment for patients with advanced head and neck squamous cell carcinomas (HNSCC), despite a high rate of treatment failures, acquired resistance, and subsequent aggressive behavior. The purpose of this study was to study the mechanism of CDDP resistance and metastasis in HNSCC. We investigated the role of NRF2 pathway activation as a driven event for tumor progression and metastasis of HNSCC.

Hypersecretory malignant cells underlie therapeutic resistance, metastasis, and poor clinical outcomes. However, the molecular basis for malignant hypersecretion remains obscure. Here, we showed that epithelial-mesenchymal transition (EMT) initiates exocytic and endocytic vesicular trafficking programs in lung cancer. The EMT-activating transcription factor zinc finger E-box-binding homeobox 1 (ZEB1) executed a PI4KIIIβ-to-PI4KIIα (PI4K2A) dependency switch that drove PI4P synthesis in the Golgi and endosomes. EMT enhanced the vulnerability of lung cancer cells to PI4K2A small-molecule antagonists. PI4K2A formed a MYOIIA-containing protein complex that facilitated secretory vesicle biogenesis in the Golgi, thereby establishing a hypersecretory state involving osteopontin (SPP1) and other prometastatic ligands. …

Myeloid cells comprise the majority of immune cells in tumors, contributing to tumor growth and therapeutic resistance. Incomplete understanding of myeloid cells response to tumor driver mutation and therapeutic intervention impedes effective therapeutic design. Here, by leveraging CRISPR/Cas9-based genome editing, we generate a mouse model that is deficient of all monocyte chemoattractant proteins. Using this strain, we effectively abolish monocyte infiltration in genetically engineered murine models of de novo glioblastoma (GBM) and hepatocellular carcinoma (HCC), which show differential enrichment patterns for monocytes and neutrophils. We examined the association between caffeine and coffee intake and the community composition and structure of colonic microbiota. A total of 34 polyp-free adults donated 97 colonic biopsies. Microbial DNA was sequenced for the 16S rRNA gene V4 region. The amplicon sequence variant was assigned using DADA2 and SILVA. Food consumption was ascertained using a food frequency questionnaire. We compared the relative abundance of taxonomies by low (<82.9 mg) vs. high (≥82.9 mg) caffeine intake and by never or <2 cups vs. 2 cups vs. ≥3 cups coffee intake. False discovery rate-adjusted p values (q values) <0.05 indicated statistical significance. Multivariable negative binomial regression models were used to estimate the incidence rate ratio and its 95% confidence interval of having a non-zero count of certain bacteria by intake level. Higher caffeine and coffee intake was related to higher alpha diversity (Shannon index p < 0.001), higher relative abundance of Faecalibacterium and Alistipes, and lower relative abundance of Erysipelatoclostridium (q values < 0.05). After adjustment of vitamin B2 in multivariate analysis, the significant inverse association between Erysipelatoclostridium count and caffeine intake remained statistically significant. Glioblastoma multiforme (GBM) is a common and fatal brain tumour in the central nervous system with a poor survival rate and a median survival time of 15 months only. The transcription factor Prdm16 functions as a potent suppressor of transforming growth factor-beta (TGF-β) signaling, whose inactivation is deemed essential to the progression of pancreatic ductal adenocarcinoma (PDAC). Using the KrasG12D-based mouse model of human PDAC, we surprisingly found that ablating Prdm16 did not block but instead accelerated PDAC formation and progression, suggesting that Prdm16 might function as a tumor suppressor in this malignancy. Subsequent genetic experiments showed that ablating Prdm16 along with Smad4 resulted in a shift from a well-differentiated and confined neoplasm to a highly aggressive and metastatic disease, which was associated with a striking deviation in the …

Background: Decoding is a foundational skill for reading, contributing to both reading fluency and comprehension (Lyon et al., 2003). Visual enhancements of alphabetic letters such as shaping letters to resemble words beginning with that sound (e.g., “f” drawn as a flower) (Handler & Fierson, 2011) and associating photographs of lips producing the sounds (Lindamood & Lindamood, 1998) have been shown to improve decoding skills. FLASH radiation therapy (RT) is a promising new paradigm in radiation oncology. However, a major question that remains is the robustness and reproducibility of the FLASH effect when different irradiators are used on animals or patients with different genetic backgrounds, diets, and microbiomes, all of which can influence the effects of radiation on normal tissues. Purpose: Current study proposed to estimate the anticarcinogenic influence of Selenium Nanoparticles (SeNps) on induced squamous cell carcinoma utilizing caspase3 as an apoptotic marker. Purpose: This study was carried out to evaluate the effect of two different regenerative materials on periodontal Ligament Model Defect through histological examination of rabbit’s mandible. Using Diamond's Five point framework for how societies have collapsed in the past allow us in the present to recognize these signs in modern societies. We have the gift of hindsight in which we now know that factors such as climate change, impact on the environment, decline on friendly contact, increase in hostile contact, and a conservative outlook have all contributed to past societies' collapse. We now recognize these issues in modern societies such as China, Australia, Rwanda, and Haiti. This allows these societies to reevaluate their current environmental standards on whether they are compatible with their survival or collapse. …

Since the spring of 2022, the MSU sheep flock has been an Innovation Flock for the Sheep GEMS project, managed by the University of Nebraska-Lincoln. The GEMS project is a multi-breed, national project evaluating longevity of ewes within flocks. Our part as a participant in the project involves collecting and sending raw data which is compiled into 1 very large data set. The research discussed here uses our preliminary data from the 2022 and 2023 lambing seasons collected from our Katahdin ewes (n =38; 1-4.5 years of age). This thesis is an exploration of Quantum Computing applied to Pell’s equation in an attempt to find solutions to the Billiard Ball Problem. Pell’s equation is a Diophantine equation in the form of x2 − ny2 = 1, where n is a given positive nonsquare integer, and integer solutions are sought for x and y. We will be applying Hallgren’s algorithm for finding irrational periods in functions, in the context of billiard balls and their movement on a friction-less unit square billiard table. In places globally and locally, people will likely find some sense of division, and/or violence in the world they live. There might be conflict in their homes, workplaces, schools, and places of worship but not limited to those areas. In the 1960’s, psychologist Marshall Rosenberg first developed Nonviolent Communication (NVC) amid the United States of America, starting the work with youth and then spreading to include school integration. NVC has found success in places with histories of violence – among families, prisons, warring tribes, and ethnic/racial groups. Early Christian thinkers developed the widespread linguistic cosmology of the Roman Mediterranean in a novel way in order to advance a specific bibliographic project: aligning the emergent fourfold Gospel with the structure of the physical cosmos. Employing interlocking concepts from the disciplines of meteorology, geography, music, mathematics, and astronomy, a number of figures—including Irenaeus, Origen, Ephrem, Eusebius, Fortunatianus, Augustine, and Maximus—imagined a Gospel corpus consisting of precisely four texts.
