Dnp Final Report: An Evidence-Based Initiative To Reduce Risk Of Ventilator-Associated Events, Bailey Dee Burge
Dnp Final Report: An Evidence-Based Initiative To Reduce Risk Of Ventilator-Associated Events, Bailey Dee Burge
DNP Final Reports
Ventilator-associated events have been considered an outcome metric for quality patient care. Ventilator-associated events are associated with unnecessary medical expenses. These events are multifactorial, yet bedside nurses can help decrease this nosocomial infection through proper oral care. A PICOT question was constructed to search the evidence systematically. The systematic literature search evidence was appraised to provide recommendations for the bundle of care process regarding ventilator-associated event prevention, specifically oral care. The body of evidence was comprised of five articles reporting statically significant outcomes. The evidence-based oral care protocol was implemented for a quarter of a year in patients with mechanical …
Forest Understory Plant Community Response To Prescribed Fire And Mechanical Thinning Treatments, Noelle V. Zenger
Forest Understory Plant Community Response To Prescribed Fire And Mechanical Thinning Treatments, Noelle V. Zenger
Theses and Dissertations
Much of the biodiversity in a forest comes from the understory vegetation. However, centuries of fire suppression in the western United States have made human-implemented disturbance necessary for reinstating natural disturbance cycles. However, the mechanical thinning and prescribed burning treatments used by land managers may differentially affect the response of the understory community. This study aimed to compare how prescribed fire and mechanical thinning treatments, with the size and severity of those treatments, affect understory plant communities in aspen-conifer forests over time. We sampled the understory community in burned, mechanically thinned, and untreated forest stands one, three, or five years …

With an ever-increasing human population, food security remains a central issue for the coming years. The magnitude of the environmental impacts of food production has motivated the assessment of the environmental and health benefits of shifting diets, from meat to fish and seafood. One of the main concerns for the sustainable development of aquaculture is the emergence and spread of infectious animal diseases in a warming climate. We conducted a meta-analysis to investigate the influence of global warming on mortality due to viral infections in farmed aquatic animals. We found a positive trend between increasing temperature and increasing viral virulence, …

Treatment failure in joint infections is associated with fibrinous, antibiotic-resistant, floating and tissue-associated Staphylococcus aureus aggregates formed in synovial fluid (SynF). We explore whether antibiotic activity could be increased against Staphylococcus aureus aggregates using ultrasound-triggered microbubble destruction (UTMD), in vitro and in a porcine model of septic arthritis. In vitro, when bacterially laden SynF is diluted, akin to the dilution achieved clinically with lavage and local injection of antibiotics, amikacin and ultrasound application result in increased bacterial metabolism, aggregate permeabilization, and a 4-5 log decrease in colony forming units, independent of microbubble destruction. Without SynF dilution, amikacin + UTMD does …

Cancer-Specific Perturbations To Arginine Metabolism Blunt Replication And Performance Of Oncolytic Myxoma Virus, Parker Dryja
Cancer-Specific Perturbations To Arginine Metabolism Blunt Replication And Performance Of Oncolytic Myxoma Virus, Parker Dryja
MUSC Theses and Dissertations
Oncolytic virotherapy (OV) is a class of immunotherapy for treatment of malignancy. Using viruses that exhibit natural coincidental tropisms for cancer, or others that have been engineered to the same effect, intentional infection of lesions leads to two therapeutically beneficial effects: (1) direct destruction of the infected tumor through virally-mediated cell lysis, and (2) recruitment of an otherwise blunted or absent anti-cancer immune response to affect both local and disseminated disease. A surfeit of cancer-specific changes are accumulated during progression from first genetic insult to clinical detection, presenting a dramatically altered underlying biology of cell and tissue. The viruses employed …

Haptic technology uses forces, vibrations, and movements to simulate a sense of touch. In the context of spacesuits, proposals to use haptic systems are scant despite evidence of their efficacy in other domains. Existing review studies have sought to summarize existing haptic system applications. Despite their contributions to the body of knowledge, existing studies have not assessed the applicability of existing haptic systems in spacesuit design to meet contemporary challenges. This study asks, “What can we learn from existing haptic technologies to create spacesuits?”. As such, we examine academic and commercial haptic systems to address this issue and draw insights …

Biparental care involves both parents contributing to offspring, though each parent does not necessarily contribute equally or in the same manner. While offspring benefit from parental care, sexual conflict arises within parental pairs because each sex would benefit from a partner’s greater contribution to the young. Nevertheless, each parent may be better suited or have preferences for the type and intensity of care they provide. Here, we examined parental pairs of Texas Cichlids in a naturally occurring population in southwest Texas as they defended their free-swimming fry against potential conspecific and heterospecific predators. Sex differences were apparent, with females generally …

Floral Visitors Of A Colorado Endemic Chasmophyte, Telesonix Jamesii (Saxifragaceae), Andrew G. Gaier, Erin Manzitto-Tripp, Julian Resasco
Western North American Naturalist
Telesonix jamesii, a rare and imperiled species of perennial saxifrage, is restricted to rocky habitats at high elevations across 21 isolated, known populations in the southern Rocky Mountains of Colorado and New Mexico. Despite its imperiled conservation status, very little is known about the natural history of T. jamesii. We studied pollination of this species during the summers of 2019–2021 at multiple locations on Pikes Peak, Colorado. We conducted a total of 899 min of pollinator surveys, identifying all floral visitors during this time period. We then examined floral visitors for the presence of T. jamesii pollen to …

Advancing Wildfire Preparedness And Planning In Anchorage: Wildfire Exposure And Egress Study, Jennifer Schmidt, John See
ISER Reports
Advancing Wildfire Preparedness and Planning takes an in-depth look at the dynamic factors that are impacting wildfire occurrence for the most populated geographic area in the 49th State of Alaska, the Municipality of Anchorage (MOA). The length and severity of recent fire seasons pose a threat to those who have chosen a niche in the Wildland Urban Interface (WUI) area to call “home.” This report is a “call to action” in many ways, delving into some of the swings in weather patterns caused by climatic change. These subtle changes are at the root of the evolving wildfire danger and its …

Permian Trilobites And The Applicability Of The “Living Fossil” Concept To Extinct Clades, Melanie J. Hopkins, Peter Wagner, Katherine J. Jordan
Department of Earth and Atmospheric Sciences: Faculty Publications
Some taxa occupy our imaginations as “living fossils” because they were known from the fossil record before being discovered alive today. Other taxa are considered “living fossils” because modern relatives bear a strong morphological resemblance to fossil relatives, or because they occupy a contracted geographic range or have less diversity now than in the past, or because they represent phylogenetic diversity that requires conservation. A characterizing feature of living fossils–and thus an implicit assumption of all criteria–is that the “living fossil” of interest is extant. However, the general research questions that “living fossils” inspire–Why do rates of evolution vary across …

Mitochondria are hubs where bioenergetics, redox homeostasis, and anabolic metabolism pathways integrate through a tightly coordinated flux of metabolites. The contributions of mitochondrial metabolism to tumor growth and therapy resistance are evident, but drugs targeting mitochondrial metabolism have repeatedly failed in the clinic. Our study in pancreatic ductal adenocarcinoma (PDAC) finds that cellular and mitochondrial lipid composition influence cancer cell sensitivity to pharmacological inhibition of electron transport chain complex I. Profiling of patient-derived PDAC models revealed that monounsaturated fatty acids (MUFAs) and MUFA-linked ether phospholipids play a critical role in maintaining ROS homeostasis. We show that ether phospholipids support mitochondrial …

Given the central role of the androgen receptor (AR) in prostate cancer cell biology, AR-targeted therapies have been the backbone of prostate cancer treatment for over 50 years. New data indicate that AR is expressed in additional cell types within the tumor microenvironment. Moreover, targeting AR for the treatment of prostate cancer has established side effects such as bone complications and an increased risk of developing cardiometabolic disease, indicating broader roles for AR. With the advent of novel technologies, such as single-cell approaches and advances in preclinical modeling, AR has been identified to have clinically significant functions in other cell …

Despite extensive research on astrocytic Ca2+ in synaptic transmission, its contribution to the modulation of sensory transmission during different brain states remains largely unknown. Here, by using two-photon microscopy and whole-cell recordings, we show two distinct astrocytic Ca2+ signals in the murine barrel cortex: a small, long-lasting Ca2+ increase during sleep and a large, widespread but short-lasting Ca2+ spike when aroused. The large Ca2+ wave in aroused mice was inositol trisphosphate (IP3)-dependent, evoked by the locus coeruleus-norepinephrine system, and enhanced sensory input, contributing to reliable sensory transmission. However, the small Ca2+ transient was IP3-independent and contributed to decreased extracellular K+, …

As computer science (CS) education becomes more prevalent in K-12 instruction, it is critical for educators, researchers, and curriculum developers to identify culturally responsive and pedagogically inclusive approaches that can increase participation, access, and feelings of belonging for students from historically marginalized communities. In response, we developed an equity-centered curricular framework and illustrative crosswalk that synchronizes three distinct pedagogical approaches: culturally responsive pedagogy (CRP), Universal Design for Learning (UDL), and project-based learning (PBL). We describe the framework’s theoretical underpinnings and explain how this framework informed the development of an integrated elementary science+CS curricular unit and provide examples of its implementation. …

The melanocortin action is well perceived for its ability to regulate body weight bidirectionally with its gain of function reducing body weight and loss of function promoting obesity. However, this notion cannot explain the difficulty in identifying effective therapeutics toward treating general obesity via activation of the melanocortin action. Here, we provide evidence that altered melanocortin action is only able to cause one-directional obesity development. We demonstrate that chronic inhibition of arcuate neurons expressing proopiomelanocortin (POMC) or paraventricular hypothalamic neurons expressing melanocortin receptor 4 (MC4R) causes massive obesity. However, chronic activation of these neuronal populations failed to reduce body weight. …

2020-Current year OA Pubs
Inactivation of tetracycline antibiotics by tetracycline destructases (TDases) remains a clinical and agricultural threat. TDases can be classified as type 1 Tet(X)-like TDases and type 2 soil-derived TDases. Type 1 TDases are widely identified in clinical pathogens. A combination therapy of tetracycline and a TDase inhibitor is much needed to rescue the clinical efficacy of tetracyclines. Anhydrotetracycline is a pan-TDase inhibitor that inhibits both type 1 and type 2 TDases. Here, we present structural, biochemical, and phenotypic evidence that anhydrotetracycline binds in a substrate-like orientation and competitively inhibits the type 1 TDase Tet(X6) to rescue tetracycline antibiotic activity as a …

Introduction: Bipolar disorder (BD) is a chronic mental illness characterized by recurrent episodes of mania and depression and associated with social and cognitive disturbances. Environmental factors, such as maternal smoking and childhood trauma, are believed to modulate risk genotypes and contribute to the pathogenesis of BD, suggesting a key role in epigenetic regulation during neurodevelopment. 5-hydroxymethylcytosine (5hmC) is an epigenetic variant of particular interest, as it is highly expressed in the brain and is implicated in neurodevelopment, and psychiatric and neurological disorders.
Methods: Induced pluripotent stem cells (iPSCs) were generated from the white blood cells of two adolescent patients with …

Rhythmic intraorgan communication coordinates environmental signals and the cell-intrinsic clock to maintain organ homeostasis. Hepatocyte-specific KO of core components of the molecular clock Rev-erbα and -β (Reverb-hDKO) alters cholesterol and lipid metabolism in hepatocytes as well as rhythmic gene expression in nonparenchymal cells (NPCs) of the liver. Here, we report that in fatty liver caused by diet-induced obesity (DIO), hepatocyte SREBP cleavage-activating protein (SCAP) was required for Reverb-hDKO-induced diurnal rhythmic remodeling and epigenomic reprogramming in liver macrophages (LMs). Integrative analyses of isolated hepatocytes and LMs revealed that SCAP-dependent lipidomic changes in REV-ERB-depleted hepatocytes led to the enhancement of LM metabolic …
