Seniors at the University of Mississippi studying biomedical engineering complete a senior project that seeks to solve a problem in healthcare. Our team, composed of Sydney Rester, Shelby Berry, Andrew Ulmer, and Alex Bromley, sought to improve care associated with nasogastric feeding tubes. We identified two major problems: clogging and tube dislodgement. To address the problem of dislodgement, the team crafted a new feeding tube design with a balloon attachment. The attachment prevents a patient from pulling the feeding tube out and keeps the tube from dislodging due to normal patient movement. We further determined that our tube should be …

The overall goal of the annual Transdisciplinary Research in Energetics and Cancer (TREC) Training Workshop is to provide transdisciplinary training for scientists in energetics and cancer and clinical care. The 2022 Workshop included 27 early-to-mid career investigators (trainees) pursuing diverse TREC research areas in basic, clinical, and population sciences. The 2022 trainees participated in a gallery walk, an interactive qualitative program evaluation method, to summarize key takeaways related to program objectives. It is thought that African literature tends to be dominated by the masculine-oriented politics that also characterizes African public political life. In some cases, this is true, but there is a feminist movement in Africa, and many African women writers are using global feminist principles and global anti-colonial principles to write a different kind of literature. As a consequence, recent novels such as Yvonne Vera’s Nehanda (1993), set in Zimbabwe, and Petina Gappah’s Out of Darkness, Shining Light (2019), revise past, often male, African writers’ approaches to depicting the genders, even as they also criticize, implicitly or explicitly, still-widespread colonialist …

Alternative reproductive tactics (ARTs) refer to discrete variation in reproductive behavior between members of a sex and within a species. ARTs are present in Blue Ridge two-lined salamanders (Eurcyea cf. wilderae) in the form of male polymorphism. Two male morphs exist: “searching” males and “guarding” males. Each phenotype has a unique reproductive ecology and associated discrete morphological differences. Another trait that shows discrete variation in E. cf. wilderae is the larval period, which may be one or two years in length. Leucine-rich repeat-containing, G protein-coupled receptor 5 (LGR5) is highly expressed in colorectal cancer and cancer stem cells (CSCs) that play important roles in tumor initiation, progression, and metastasis. Loss of LGR5 has been shown to enhance therapy resistance. However, the molecular mechanisms that mediate this resistance remain elusive. In this study, we demonstrate conversion of LGR5+ colorectal cancer cells to an LGR5- state in response to chemotherapy, LGR5- targeted antibody-drug conjugates (ADCs), or LGR5 gene ablation led to activation of STAT3. Further investigation revealed increased STAT3 activation occurred as a result of increased mesenchymal epithelial transition (MET) factor receptor activity. …

BACKGROUND: Through the evolution of novel wing structures, bats (Order Chiroptera) became the only mammalian group to achieve powered flight. This achievement preceded the massive adaptive radiation of bats into diverse ecological niches. We investigate some of the developmental processes that underlie the origin and subsequent diversification of one of the novel membranes of the bat wing: the plagiopatagium, which connects the fore- and hind limb in all bat species.
RESULTS: Our results suggest that the plagiopatagium initially arises through novel outgrowths from the body flank that subsequently merge with the limbs to generate the wing airfoil. Our findings further …

Heparin-binding hemagglutinin (HBHA) and M. tuberculosis pili (MTP) are important antigens on the surface of Mycobacterium tuberculosis. To display these antigens effectively, the fusion protein HBHA-MTP with a molecular weight of 20 kD (L20) was inserted into the receptor-binding hemagglutinin (HA) fragment of influenza virus and was expressed along with matrix protein M1 in Sf9 insect cells to generate influenza virus-like particles (LV20 in short). The results showed that the insertion of L20 into the envelope of the influenza virus did not affect the self-assembly and morphology of LV20 VLPs. Change has become the urgency of our time, as the harrowing Anthropocene and ongoing socio-political crises are challenging the future of humanity and our planet. While systemic structural forces are often (and rightfully so) the target to blame, social activism is as much about our own path of inner change as it is about attempting to create a better world externally. State mandates imposed by governors during COVID-19 affected extracurricular activities for children and youth. To continue providing 4-H programming during quarantine, Clemson Extension 4-H Youth Development Agents created the SC 4-H@Home program. Daily activities were developed that could be completed at-home using common household items and were delivered via email to registered participants. A survey was conducted at the conclusion of the program that included open- and closed-ended questions to gauge the effectiveness of the program. Results indicate that, overall, the SC 4-H@Home program was beneficial to youth and contributed to their education during the COVID-19 crisis.

Alzheimer's disease (AD) is presumably caused by two neuropathological protein markers: amyloid beta (Aβ) and neuro fibrillary tangles. Alzheimer’s Disease Neuroimaging Initiative (ADNI) collects high quality patient data from standardized clinical trials using Magnetic Resonance Imaging (MRI), Positron Emission Tomography (PET), and Cerebral Spinal Fluid (CSF) biomarkers to diagnose and evaluate the effectiveness of treatment for AD. MRI and PET screening for Alzheimer’s Disease aim to find structural, functional, and neurochemical abnormalities causing or caused by AD brain diffusion tensor imaging (DTI) and Resting State functional MRI (RSfMRI) are emerging AD MRI tools. This thesis features a review of the body of literature surrounding teacher knowledge dyslexia and its interventions. Several studies show teachers (both preservice and inservice) do not have a strong understanding of neither the learning disability nor effective interventions for dyslexia. Students with dyslexia benefit from explicit and systematic teaching. The following lesson plans demonstrate how to lead both large- and small-groups using dyslexia-centered strategies as a general education teacher. The ten-day unit and accompanying intervention plans are designed to specifically accommodate the needs of students with dyslexia, though even students without dyslexia are known to benefit from systematic and …

Background: ALKBH5 is aberrantly activated and exerts critical roles in facilitating the development of glioblastoma. However, the underlying activation mechanism by which ALKBH5 protein is increased in glioblastoma is not completely understood. Our study aimed to elucidate the signaling pathways involved in mediating ALKBH5 protein stability.
Methods: The contribution of deubiquitinating enzymes (DUB) to the fluctuation of ALKBH5 protein expression was globally profiled with western blot analysis. Mass spectrometry and immunoprecipitation were performed to identify the USP36 and ALKBH5 interaction. Although midlobular hepatocytes in zone 2 are a recently identified cellular source for liver homeostasis and regeneration, these cells have not been exclusively fate mapped. We generated an Igfbp2-CreER knockin strain that specifically labels midlobular hepatocytes. During homeostasis over 1 year, zone 2 hepatocytes increased in abundance from occupying 21%-41% of the lobular area. After either pericentral injury with carbon tetrachloride or periportal injury with 3,5-diethoxycarbonyl-1,4-dihydrocollidine (DDC), IGFBP2+ cells replenished lost hepatocytes in zones 3 and 1, respectively. IGFBP2+ cells also preferentially contributed to regeneration after 70% partial hepatectomy, as well as liver growth during pregnancy. Because IGFBP2 labeling increased …

Noroviruses are the leading cause of severe childhood diarrhea and foodborne disease worldwide. While they are a major cause of disease in all age groups, infections in the very young can be quite severe, with annual estimates of 50,000-200,000 fatalities in children under 5 years old. In spite of the remarkable disease burden associated with norovirus infections, very little is known about the pathogenic mechanisms underlying norovirus diarrhea, principally because of the lack of tractable small animal models. Research has shown the metabolic role and therapeutic potential of the gut microbiome in modulating health and disease states. Microbial metabolites are known to be involved in various metabolic functions throughout the body. The purpose of this study was to determine if the microbial metabolite imidazole propionate, shown to be increased in obesity and type II diabetes, has an effect on key human T-cell functions. Two fasted blood samples were collected from 10 human subjects. PBMCs were isolated, and a subset were stimulated in the presence of ImP. The process of state formation in Egypt has been variously modeled. Lapidary tableaux from the eastern and western deserts of Egypt have been dated stylistically to the later period of state formation, the Late Predynastic. This thesis provides an overview of select theories on state formation, with particular focus on ancient Egypt. Artificial intelligence (AI) programs are beginning to generate art and design at levels once executed by trained graphic designers. Prior perceived threats have come from automation and the rise of computer-assisted graphic design, particularly Adobe Photoshop. Concerns associated with AI-based image generation programs and tools, such as DALL-E 2, Midjourney, and Stable Diffusion, present far more complex questions and rapidly changing conditions. By comparing three separate design activities, this thesis provides one perspective on the wider problems facing the graphic design field. As there is limited archeological evidence of Libyan society from Libya, assigning a social identity to an individual living in Egypt during the Third Intermediate Period (Dynasties 21-24, ca. 1076-723 BCE) is challenging. Comparison of textual and visual themes across a range of artifacts will allow further investigation into a connection between Libyan social identity and kinship. Case studies will cover genealogical texts, including texts on stelae and block statues, to provide insight into their function and significance to the Libyan people. Cancer-related alterations of the p53 tetramerization domain (TD) abrogate wild-type (WT) p53 function. They result in a protein that preferentially forms monomers or dimers, which are also normal p53 states under basal cellular conditions. However, their physiologic relevance is not well understood. We have established in vivo models for monomeric and dimeric p53, which model Li-Fraumeni syndrome patients with germline p53 TD alterations. p53 monomers are inactive forms of the protein. Unexpectedly, p53 dimers conferred some tumor suppression that is not mediated by canonical WT p53 activities. p53 dimers upregulate the PPAR pathway. The Integrator complex is a multi-subunit protein complex that regulates the processing of nascent RNAs transcribed by RNA polymerase II (RNAPII), including small nuclear RNAs, enhancer RNAs, telomeric RNAs, viral RNAs, and protein-coding mRNAs. Integrator subunit 11 (INTS11) is the catalytic subunit that cleaves nascent RNAs, but, to date, mutations in this subunit have not been linked to human disease. Here, we describe 15 individuals from 10 unrelated families with bi-allelic variants in INTS11 who present with global developmental and language delay, intellectual disability, impaired motor development, and brain atrophy. Exercise has the ability to rejuvenate stem cells and improve tissue regeneration in aging animals. However, the cellular and molecular changes elicited by exercise have not been systematically studied across a broad range of cell types in stem cell compartments. We subjected young and old mice to aerobic exercise and generated a single-cell transcriptomic atlas of muscle, neural, and hematopoietic stem cells with their niche cells and progeny, complemented by whole transcriptome analysis of single myofibers. BACKGROUND: Methylation of the p16 promoter resulting in epigenetic gene silencing-known as p16 epimutation-is frequently found in human colorectal cancer and is also common in normal-appearing colonic mucosa of aging individuals. Thus, to improve clinical care of colorectal cancer (CRC) patients, we explored the role of age-related p16 epimutation in intestinal tumorigenesis.
METHODS: We established a mouse model that replicates two common genetic and epigenetic events observed in human CRCs: Apc mutation and p16 epimutation. Remote tumors disrupt the bone marrow (BM) ecosystem (BME), eliciting overproduction of BM-derived immunosuppressive cells. However, the underlying mechanisms remain poorly understood. Herein, we characterized breast and lung cancer-induced BME shifts pre- and post-tumor removal. Remote tumors progressively lead to osteoprogenitors (OPs) expansion, hematopoietic stem cell dislocation and CD41− granulocyte-monocyte progenitor (GMP) aggregation. The tumor-entrained BME is characterized by co-localization between CD41− GMPs and OPs. OP ablation abolishes this effect and diminishes abnormal myeloid overproduction. Mechanistically, HTRA1 carried by tumor-derived small extracellular vesicles upregulates MMP-13 in OPs, which in turn induces the alterations in hematopoietic program. Gene-level associations obtained from mass-spectrometry-based cancer proteomics datasets represent a resource for identifying gene candidates for functional studies. When recently surveying proteomic correlates of tumor grade across multiple cancer types, we identified specific protein kinases having a functional impact on uterine endometrial cancer cells. This previously published study provides just one template for utilizing public molecular datasets to discover potential novel therapeutic targets and approaches for cancer patients. Proteomic profiling data combined with corresponding multi-omics data on human tumors and cell lines can be analyzed in various ways to prioritize genes of interest for interrogating biology.
