Steroid receptor coactivator 3 (SRC-3) is most strongly expressed in regulatory T cells (Tregs) and B cells, suggesting that it plays an important role in the regulation of Treg function. Using an aggressive E0771 mouse breast cell line syngeneic immune-intact murine model, we observed that breast tumors were "permanently eradicated" in a genetically engineered tamoxifen-inducible Treg-cell-specific SRC-3 knockout (KO) female mouse that does not possess a systemic autoimmune pathological phenotype. A similar eradication of tumor was noted in a syngeneic model of prostate cancer. A subsequent injection of additional E0771 cancer cells into these mice showed continued resistance to tumor …

Obstructive Sleep Apnea-Induced Hypertension Is Associated With Increased Gut And Neuroinflammation, Sriram Ayyaswamy, Huanan Shi, Bojun Zhang, Robert M Bryan, David J Durgan

Faculty, Staff and Students Publications

Background

Obstructive sleep apnea (OSA) is an independent risk factor for the development of hypertension. We have demonstrated that OSA induces gut dysbiosis, and this dysbiotic microbiota contributes to hypertension. However, the mechanisms linking gut dysbiosis to blood pressure regulation remain unclear. Recent studies demonstrate that gut dysbiosis can induce a proinflammatory response of the host resulting in peripheral and neuroinflammation, key factors in the development of hypertension. We hypothesized that OSA induces inflammation in the gut that contributes to neuroinflammation and hypertension.

Methods and Results

OSA was induced in 8‐week‐old male rats. After 2 weeks of apneas, lymphocytes were …

Mapping Human Norovirus Antigens During Infection Reveals The Breadth Of The Humoral Immune Response, Lynn Su, Wanzhi Huang, Frederick H Neill, Mary K Estes, Robert L Atmar, Timothy Palzkill

Faculty, Staff and Students Publications

Human noroviruses (HuNoV) are the leading cause of acute gastroenteritis worldwide. The humoral immune response plays an important role in clearing HuNoV infections and elucidating the antigenic landscape of HuNoV during an infection can shed light on antibody targets to inform vaccine design. Here, we utilized Jun-Fos-assisted phage display of a HuNoV genogroup GI.1 genomic library and deep sequencing to simultaneously map the epitopes of serum antibodies of six individuals infected with GI.1 HuNoV. We found both unique and common epitopes that were widely distributed among both nonstructural proteins and the major capsid protein. Recurring epitope profiles suggest immunodominant antibody …

Sustained Alternate-Day Fasting Potentiates Doxorubicin Cardiotoxicity, Mualla Ozcan, Zhen Guo, Carla Valenzuela Ripoll, Ahmed Diab, Antonino Picataggi, David Rawnsley, Aynaz Lotfinaghsh, Carmen Bergom, Jeff Szymanski, Daniel Hwang, Jie Zheng, Robert J Hayashi, Pamela K Woodard, Attila Kovacs, Joel Schilling, Babak Razani, Abhinav Diwan, Ali Javaheri, Et Al.

2020-Current year OA Pubs

Fasting strategies are under active clinical investigation in patients receiving chemotherapy. Prior murine studies suggest that alternate-day fasting may attenuate doxorubicin cardiotoxicity and stimulate nuclear translocation of transcription factor EB (TFEB), a master regulator of autophagy and lysosomal biogenesis. In this study, human heart tissue from patients with doxorubicin-induced heart failure demonstrated increased nuclear TFEB protein. In mice treated with doxorubicin, alternate-day fasting or viral TFEB transduction increased mortality and impaired cardiac function. Mice randomized to alternate-day fasting plus doxorubicin exhibited increased TFEB nuclear translocation in the myocardium. When combined with doxorubicin, cardiomyocyte-specific TFEB overexpression provoked cardiac remodeling, while systemic …

What Could A Trans Book History Look Like? Toward Trans Codicology, J D. Sargan Jun 2023

What Could A Trans Book History Look Like? Toward Trans Codicology, J D. Sargan

Criticism

This article draws on critical trans studies and queer archival practice to propose a book historical mode that extends what we know about the premodern trans experience beyond the recovery of individual biographies. Instead of turning to textual sources for the identification of transness, the author looks to Susan Stryker’s call for the “recuperat[ion of] embodied knowing as a formally legitimated basis of knowledge production.” Bibliography, he suggests, makes claims of objectivity that engender a particular reluctance to respond to such calls. But the lived reality of archival research is one of affective embodiment. Affect theory is an area that, …

Making Mary Ann Waters Is A Free Black Woman: Critical Fabrication As Bibliographic Method, Kadin Henningsen Jun 2023

Making Mary Ann Waters Is A Free Black Woman: Critical Fabrication As Bibliographic Method, Kadin Henningsen

Criticism

This article discusses the process of creating an artist’s book on Mary Ann Waters, a Black trans woman and sex worker in Antebellum Baltimore. In addition, informed by the author’s experience of making the artist’s book, he proposes a process of critical fabrication—which brings together Saidiya Hartman’s method of “critical fabulation” with Natalie Loveless’s articulation of “research-creation”—as a method for critical bibliography.

Listening To Bamewawagezhikaquay’S Teachers: Jane Johnston Schoolcraft’S Citational Cosmopolitics, Shelby Johnson Jun 2023

Listening To Bamewawagezhikaquay’S Teachers: Jane Johnston Schoolcraft’S Citational Cosmopolitics, Shelby Johnson

Criticism

This article argues that Bamewawagezhikaquay, or Jane Johnston Schoolcraft, offers vital citations of Anishinaabe cosmologies, including lineages of human and nonhuman teachers and perspectives on animate archives that should inflect new approaches to textual studies. Bamewawagezhikaquay’s writings express a citational cosmopolitics, a practice where Bamewawagezhikaquay invokes, and occasionally translates into English, human and more-than-human agents in the cocreation of Anishinaabe knowledge. In her descriptions of Anishinaabe plants and geographies, she models a citational praxis that intersects with resurgent frameworks on orienting to Anishinaabe writings, including birchbark maps and cliff paintings, not as inert objects but as dynamic nodal points in …

Barbara Grier’S Enumerative Bibliographies: Iterating Communal Lesbian Identities, Julie R. Enszer Jun 2023

Barbara Grier’S Enumerative Bibliographies: Iterating Communal Lesbian Identities, Julie R. Enszer

Criticism

Barbara Grier, best known for her publishing work with the Naiad Press, started her literary life in the pages of The Ladder, the magazine of the Daughters of Bilitis. Working initially under the tutelage of Jeanette Howard Foster, Grier cataloged and categorized work by and about lesbians during the repressive decades of the 1950s and 1960s. By tracing Grier’s work in three major bibliographic projects, the Lesbiana column in The Ladder, the Lesbian in Literature (published in three separate editions), and Lesbiana (a book Grier published from her columns), Grier’s bibliographic practices, enumerative and annotative, emerge as tools …

Surface Reading Paper As Feminist Bibliography, Georgina Wilson Jun 2023

Surface Reading Paper As Feminist Bibliography, Georgina Wilson

Criticism

This article models a mode of feminist bibliography by “surface reading” paper. Taking Ben Jonson’s Sejanus His Fall (1605) as a case study, this article reads watermarks as reminders of paper’s three-dimensional materiality, whose surfaces and depths model the more and less legible forms of labor which contribute to paper’s making. Watermarks here become a creative and critical prompt to recover the interventions of John Spilman (the papermaker whose output was used for Sejanus), Spilman’s workers, and especially his female ragpickers. This article fuses close reading of literary texts and archival sources with bibliography and theory to demonstrate fresh …

Beast And Man In India: Undoing John Lockwood Kipling’S Imperial Citation, Oishani Sengupta Jun 2023

Beast And Man In India: Undoing John Lockwood Kipling’S Imperial Citation, Oishani Sengupta

Criticism

This article posits that John Lockwood Kipling’s Beast and Man in India (1891), the illustrated compendium on animals that mixes discussions of colonial cross-species entanglements with personal reflections on transforming local arts and crafts in India in the service of imperial power, is a multiauthored book. Centering the presence of Indian illustrators as central to Beast and Man’s texture, this essay uses the term “imperial citation” to highlight the range of strategies Kipling uses to overtly and covertly appropriate the labor of Indigenous creators within the fabric of this volume. By placing the material text within the context of colonial …

Critique! Critique! Critique! Black Labor In The Early American Book Trade, John J. Garcia Jun 2023

Critique! Critique! Critique! Black Labor In The Early American Book Trade, John J. Garcia

Criticism

This article pursues two lines of inquiry: first, recovering the presence of Black labor in the history of the book in colonial North America, the British Caribbean, and the early United States, with a second and complementary discussion of why critique must be foregrounded in the field formation of critical bibliography. Free and enslaved Black men and women helped make early American books possible. Their presences are to be found at the edges and vicinities of print cultural production, in roles such as papermaking, wagon driving, and forms of domestic labor that extended to the libraries and reading practices of …

Trees And Texts: Indigenous History, Material Media, And The Logan Elm, Mark Alan Mattes Jun 2023

Trees And Texts: Indigenous History, Material Media, And The Logan Elm, Mark Alan Mattes

Criticism

Settler accounts of the Cayuga Native American Soyeghtowa (Logan), such as Thomas Jefferson’s Notes on the State of Virginia, interpret his famous mourning speech, “Logan’s Lament,” as the words of a melancholic, noble savage and vanishing Indian. This essay decolonizes settler accounts of Logan’s words and deeds such as Jefferson’s book by considering Indigenous relationships to a once-living memorial on Shawnee land in central Ohio, the Logan Elm, which nineteenth-century settlers apocryphally identified as the site of Logan’s speech. Drawing on scholarly work on Indigenous writing and historical media by Native American and settler intellectuals, as well as local …

Warp/Weft/Word: Inscriptive Materiality, Epistemological Violence, And The Inka Khipu, Travis Sharp Jun 2023

Warp/Weft/Word: Inscriptive Materiality, Epistemological Violence, And The Inka Khipu, Travis Sharp

Criticism

Many competing theories of the Indigenous inscription practice known as the khipu have been offered, from L. Leland Locke’s long-standing postulation that khipus are accounting devices, to Walter Ong’s description of them as aide-mémoire, to Gary Urton’s more experimental theory that they constituted an early form of binary composition. Just as fraught is the history of the khipu, which were utilized by the Inka, intermediated by Spanish and Catholic authorities in their legal and religious systems, and, finally, banned and burned as seditious and sacrilegious. Contemporary khipus are primarily limited to those used by herders, but Chilean American poet-artist Cecilia …

Pipestone Books: Indigenous Materialisms And Bibliographical Methods, Daniel Radus Jun 2023

Pipestone Books: Indigenous Materialisms And Bibliographical Methods, Daniel Radus

Criticism

This article argues that the methods of bibliographical scholarship would be enriched with further attention to Indigenous ontological traditions. It does so via an analysis of a small tablet of pipestone that in 1957 was carved into the shape of an open book by Ephraim Taylor, a Dakota artist. The article first establishes that, for the Dakotas, pipestone is a vibrant and animate material, a sentient trace of ancestral kin. The article then aligns the pipestone sculpture with an archive of books whose material traits have been altered by Indigenous readers, arguing ultimately that a sophisticated account of these books—and …

Hyperthermic Extracorporeal Applied Tumor Therapy (Heatt®) In Advanced Unresponsive Cancer, Joseph B. Zwischenberger, Peyton Skaggs, Katherine Breetz, Jan Winetz, Roger Vertrees

Journal of Shock and Hemodynamics

Hyperthermia has been utilized for cancer therapy, including metastatic cancer, for decades with isolated success. Previous research has indicated that the elevated temperature of 42°C induces cell death, apoptosis, or senescence of responsive cancers, providing a mechanism for tumor destruction and management. Veno-venous perfusion-induced systemic hyperthermia (V-V PISH) may be the key to improving advanced tumor responsiveness to previously failed chemotherapy and/or radiation as combination therapy. The most recent iteration of V-V PISH, Hyperthermic Extracorporeal Applied Tumor Therapy (HEATT®), provides homogeneous heating of all tissues with electrolyte and pH control and continues to prove safe and effective. The utilization of …

Batrachochytrium salamandrivorans (Bsal) is a fungal pathogen of amphibians that is emerging in Europe and could be introduced to North America through international trade or other pathways. To evaluate the risk of Bsal invasion to amphibian biodiversity, we performed dose-response experiments on 35 North American species from 10 families, including larvae from five species. We discovered that Bsal caused infection in 74% and mortality in 35% of species tested. Both salamanders and frogs became infected and developed Bsal chytridiomycosis. Based on our host susceptibility results, environmental suitability conditions for Bsal, and geographic ranges of salamanders in the …

Mitochondria play an important role in both normal heart function and disease etiology. We report analysis of common genetic variations contributing to mitochondrial and heart functions using an integrative proteomics approach in a panel of inbred mouse strains called the Hybrid Mouse Diversity Panel (HMDP). We performed a whole heart proteome study in the HMDP (72 strains, n=2-3 mice) and retrieved 848 mitochondrial proteins (quantified in ≥50 strains). High- resolution association mapping on their relative abundance levels revealed three trans-acting genetic loci on chromosomes (chr) 7, 13 and 17 that regulate distinct classes of mitochondrial proteins as well as cardiac …

Cyclic regression of the ovarian corpus luteum, the endocrine gland responsible for progesterone production, involves rapid matrix remodeling. Despite fibroblasts in other systems being known for producing and maintaining extracellular matrix, little is known about fibroblasts in the functional or regressing corpus luteum. Vast transcriptomic changes occur in the regressing corpus luteum, among which are reduced levels of vascular endothelial growth factor A (VEGFA) and increased expression of fibroblast growth factor 2 (FGF2) after 4 and 12 h of induced regression, when progesterone is declining and the microvasculature is destabilizing. We hypothesized that FGF2 activates luteal …

Landscape Interventions For Pedestrian Safety Around Urban Parks: Learning From City Of Dallas, Texas, Ann Mariya Thuruthy

Landscape Architecture Masters & Design Theses - Archive

Urban parks and their immediate surroundings provide access to nature in urbanized areas. However, they can also be crime hotspots if not properly kept or monitored or provide a safe environment for positive activities (Jones & Robinson, 2008). The local community respects a safe and secure neighborhood urban park as proximity to green spaces (McCormack et al., 2010), which enhances living quality (Iqbal & Ceccato, 2015). Nevertheless, a neighborhood urban park and its context have a negative impact on the quality of life if they cater to criminal activity. Crime rates have increased in urban public parks and buffer zones …
