Pexidartinib (PEX, TURALIO), a selective and potent inhibitor of the macrophage colony-stimulating factor-1 receptor, has been approved for the treatment of tenosynovial giant cell tumor. However, frequent and severe adverse effects have been reported in the clinic, resulting in a boxed warning on PEX for its risk of liver injury. The mechanisms underlying PEX-related hepatotoxicity, particularly metabolism-related toxicity, remain unknown. In the current study, the metabolic activation of PEX was investigated in human/mouse liver microsomes (HLM/MLM) and primary human hepatocytes (PHH) using glutathione (GSH) and methoxyamine (NH2OMe) as trapping reagents. A total of 11 PEX-GSH and 7 PEX-NH2OMe adducts were …

School of Biological and Marine Sciences
Three Odontaspis ferox (confirmed by mtDNA barcoding) were found in the English Channel and Celtic Sea in 2023 at Lepe, UK (50.7846, −1.3508), Kilmore Quay, Ireland (52.1714, −6.5937), and Lyme Bay, UK (50.6448, −2.9302). These are the first records of O. ferox in either country, and extend the species' range by over three degrees of latitude, to >52° N. They were ~275 (female), 433 (female), and 293 cm (male) total length, respectively. These continue a series of new records, possibly indicative of a climate change−induced shift in the species' range.

The distribution of avian haemosporidians of the genus Leucocytozoon in the Neotropics remains poorly understood. Recent studies confirmed their presence in the region using molecular techniques alone, but evidence for gametocytes and data on putative competent hosts for Leucocytozoon are still lacking outside highland areas. We combined morphological and molecular data to characterize a new Leucocytozoon species infecting a non-migratory red-legged seriema (Cariama cristata), the first report of a competent host for Leucocytozoon in Brazil. Leucocytozoon cariamae n. sp. is distinguished from the Leucocytozoon fringillinarum group by its microgametocytes that are not strongly appressed to the host cell …

In desert environments, unique communities depend on groundwater from springs, including a diverse radiation of small (<5 >mm) snails found in the desert across the southwestern United States. Nearly all springsnail species are considered critically imperiled with their existence depending on maintenance of spring-flows in regions of declining water availability. Extant, endemic springsnails in the Trans-Pecos region of Texas include one species of Pseudotryonia Hershler, 2001, five nominal Tryonia W. Stimpson, 1865 (Cochliopidae) and seven Pyrgulopsis Call & Pilsbry, 1886 (Hydrobiidae). Four of these are classified as endangered under the US Endangered Species Act. Surveys for springsnails were conducted at …5>

Organization of extracellular matrix (ECM) components, including collagens, proteoglycans, and elastin, is essential for maintaining the structure and function of heart valves throughout life. Mutations in ECM genes cause connective tissue disorders, including Osteogenesis Imperfecta (OI), and progressive debilitating heart valve dysfunction is common in these patients. Despite this, effective treatment options are limited to end-stage interventions. Mice with a homozygous frameshift mutation in col1a2 serve as a murine model of OI (oim/oim), and therefore, they were used in this study to examine the pathobiology of aortic valve (AoV) disease in this patient population at structural, functional, and molecular levels. …

In Silico Prediction Of Acute Chemical Toxicity Of Biocides In Marine Crustaceans Using Machine Learning, Rama Krishnan, Ian S. Howard, Sean Comber, Awadhesh N. Jha
School of Engineering, Computing and Mathematics
Biocides are a heterogeneous group of chemical substances intended to control the growth or kill undesired organisms. Due to their extensive use, they enter marine ecosystems via non-point sources and may pose a threat to ecologically important non-target organisms. Consequently, industries and regulatory agencies have recognized the ecotoxicological hazard potential of biocides. However, the prediction of biocide chemical toxicity on marine crustaceans has not been previously evaluated. This study aims to provide in silico models capable of classifying structurally diverse biocidal chemicals into different toxicity categories and predict acute chemical toxicity (LC50) in marine crustaceans using a set of calculated …

Investigating The Salinity Tolerance Of The Swordtail (Xiphophorus Hellerii), Alexandra Megan Krak
College of Science and Health Theses and Dissertations
The swordtail, Xiphophorus hellerii, is a freshwater fish species native to Mexico and Central America and is commonly used in the tropical aquarium trade. Swordtails have been shown to have limited survival in 6 ‰ salinity (Nanda et al., 2016), suggesting they may have a greater salinity tolerance than previously expected. Using a gradual acclimation method, we examined the salinity tolerance and swim performance of this species. Freshwater-reared female swordtails were housed in 29-gallon aquaria. A control group was held in freshwater throughout the experiment. For the experimental group, the salinity was increased by 2 ‰ every week until 30 …

Impact Of Retinoic Acid Receptor Alpha Conditional Knockout On Ovarian Follicle Development In The Mouse, Zlata Bogin
College of Science and Health Theses and Dissertations
When there is abnormal endocrine signaling in the follicles, infertility, and ovarian diseases such as polycystic ovarian syndrome (PCOS), premature ovarian failure (POF), and different types of ovarian cancers may occur. Retinoic acid (RA), a biologically active derivative of vitamin A, is a critical signaling molecule that plays a vital role in various physiological processes, including embryonic development, tissue differentiation, and reproductive function. RA has been observed to influence granulosa cell proliferation through a cell signaling cascade involving Retinoic Acid Receptors (RARs), which have three isoforms alpha, beta, and gamma. We characterized a new retinoic acid receptor alpha (RARA) conditional …

Characterization Of The Immune Deficiency Pathway During Female Meiosis In Drosophila Melanogaster, Sarah Mashburn
College of Science and Health Theses and Dissertations
Organisms can defend against pathogens by significantly increasing the diversity of their progeny, so that some progeny are more likely to survive infection. This led to the prediction that infection should cause an increase in recombination rates which was seen in a previous study. However, the mechanism that regulates recombination rates in response to infection is not understood and in that study they did not provide a mechanism for how that can occur. In an RNAi screen, our lab found the gene mustard (mtd) is required for normal recombination rates. This gene was previously shown to be part of the …
Targeting S. Maltophilia Biofilms With Phage Bfi2, Lydia Walther
Targeting S. Maltophilia Biofilms With Phage Bfi2, Lydia Walther
College of Science and Health Theses and Dissertations
Stenotrophomonas maltophilia is a multidrug resistant bacterium which can cause severe infections of the respiratory tract, soft tissue, and bloodstream, and its ability to form biofilms on medical implants and devices contributes to its role as a hospital-acquired pathogen (Brooke, 2021; Flores-Treviño et al., 2019). The rise of antibiotic resistance has prompted the search for new treatments against bacteria. Bacteriophages, or phages, are a promising alternative to antibiotics because they infect and kill bacteria (Ling et al., 2022). Phage therapy has gained recent attention as a life-saving treatment for antibiotic resistant infections (Racenis et al., 2023; Schooley et al., 2017). …

Verifying Empirical Predictive Modeling Of Societal Vulnerability To Hazardous Events: A Monte Carlo Experimental Approach, Yi Victor Wang, Seung Hee Kim, Menas C. Kafatos
Institute for ECHO Articles and Research
With the emergence of large amounts of historical records on adverse impacts of hazardous events, empirical predictive modeling has been revived as a foundational paradigm for quantifying disaster vulnerability of societal systems. This paradigm models societal vulnerability to hazardous events as a vulnerability curve indicating an expected loss rate of a societal system with respect to a possible spectrum of intensity measure (IM) of an event. Although the empirical predictive models (EPMs) of societal vulnerability are calibrated on historical data, they should not be experimentally tested with data derived from field experiments on any societal system. Alternatively, in this paper, …

Artificial intelligence and machine learning (ML) have exploded in use, accessibility, and awareness in the past few years, particularly with the release of ChatGPT in late 2022. Advances in end-user ML tools are accelerating the development of ML applications, lowering the technical barrier of entry for users outside of the computer science (CS) community. Access to ML education within STEM is mostly limited to upper-level computer science courses that have deep pre-requisite requirements or to introductory workshops that yield limited ML skills. Despite the critical need for ML education, there is a lack of guidance in instructional design for applied …

Transcranial Focused Ultrasound-Induced Blood‒Brain Barrier Opening In Mice Without Shaving Hairs, Lu Xu, Yan Gong, Chih-Yen Chien, Eric Leuthardt, Hong Chen
2020-Current year OA Pubs
Acoustic coupling through hairs remains a challenge to performing transcranial-focused ultrasound procedures. Here, we demonstrated that this challenge could be addressed by using oil as the coupling medium, leveraging oil's high affinity to hairs due to their inherent hydrophobicity. We compared focused ultrasound-induced blood-brain barrier opening (FUS-BBBO) outcomes in mice under three coupling conditions: oil with hairs ("oil + hairs"), ultrasound gel with hair shaving ("ultrasound gel + no hair"), and ultrasound gel with hairs ("ultrasound gel + hairs"). The quality of the coupling was evaluated by [Formula: see text]-weighted magnetic resonance imaging (MRI) and passive cavitation detection (PCD). The …

Mycobacterium avium (M. avium), a type of nontuberculous mycobacteria (NTM), poses a risk for pulmonary infections and disseminated infections in immunocompromised individuals. Conventional treatment consists of a 12-month regimen of the first-line antibiotics rifampicin and azithromycin. However, the treatment duration and low antibiotic tolerability present challenges in the treatment of M. avium infection. Furthermore, the emergence of multidrug-resistant mycobacterium strains prompts a need for novel treatments against M. avium infection. This study aims to test the efficacy of a novel antimicrobial peptide, cyclic [R4W4], alongside the first-line antibiotics azithromycin and rifampicin in reducing M. avium survival. Colony-forming unit (CFU) …
