The dendritic cells of the CNS, the microglia (MG), are an initiation point of the immunological response within the post blood-brain barrier (BBB) compartment. Microglia morphology drastically changes in response to cell stress to a much different non-dendritic morphology. This investigation postulates that if the first MG responses to toxic injury are isolated and studied in greater morphological detail there is much to be learned about MG metamorphosis from a baseline (M2) to an activated (M1) state. Adeno-Associated Virus (AAV) is extensively utilized in various clinical trials and approved gene therapies. Despite its widespread use, the immune response to AAV vectors remains a significant limitation. Extensive research has been conducted to mitigate adaptive immune challenges, such as pre-existing antibodies and inhibiting the adaptive immune response. This study, however, centers on the innate immune response elicited by AAV, exploring the modulatory effects of insulin and DMA. Alternative splicing and adenosine-to-inosine (A-to-I) RNA editing are post-transcriptional mechanisms that promote proteomic diversity in the brain. The impact of RNA editing on brain physiology is underscored by its association with the etiology and pathogenesis of various neurological and neurodegenerative diseases. Alternative splicing of exon 43 within the CaV1.3 C-terminus produces long (CaV1.3L) and short (CaV1.343S) variants exhibiting distinct gating and pharmacological properties. While the physiological role of CaV1.3 A-to-I RNA editing in brain function is still unfolding, recent findings indicate that unedited CaV1.3 channels display larger calcium influx and enhanced neuronal excitability, suggesting that A-to-I RNA editing may act …

The resistance of cancer cells to multiple drugs, known as multidrug resistance (MDR), remains a significant obstacle that leads to the ineffectiveness of cancer chemotherapy. The overexpression of ATP-binding cassette (ABC) transporters, particularly ABCB1 and ABCG2, has been strongly correlated with MDR. There is an urgent need to explore and identify novel inhibitors targeting ABCB1 and ABCG2 to overcome the related MDR. Mobocertinib (TAK-788) is an orally administered, irreversible EGFR/HER2 inhibitor approved for the treatment of adult patients with locally advanced or metastatic non-small cell lung cancer (NSCLC) harboring epidermal growth factor receptor (EGFR) exon 20 insertion mutations. Stealing, Scavenging, Or Storing? Breeding Behaviors Of Unisexual Ambystoma Salamanders, Jane Venezia

The unisexual Ambystoma salamander complex is a lineage of all-female salamanders that use a unique form of reproduction: kleptogenesis. Kleptogenesis requires sperm from male salamanders of co-occurring sexual species, potentially putting unisexual and sexual females in breeding competition. We investigated if unisexual salamanders require courtship from sexual males or if they can use sperm left over from previous breeding events. We put unisexual salamanders into boxes containing spermatophores from a previous courtship and examined if they laid viable eggs. We also genotyped adults and embryos to assess parentage. Some of the unisexuals in the boxes produced viable eggs; however, the …

Chemotherapy poses a significant challenge for cancer patients due to drug-associated toxicity, which often results from their effects on both healthy (normal) and cancerous cells. While various options aim to reduce toxicity and optimize beneficial effects, a comprehensive solution remains elusive. Cyclotherapy is one such approach developed to protect normal cells from the toxic effects of chemotherapy drugs. The basic principle underlying cyclotherapy is p53- dependent cell cycle arrest of normal cells while killing cancer cells via a p53-independent mechanism using a second drug. Eastern diamondback rattlesnakes (Crotalus adamanteus; EDB) are an imperiled species native to the Southeastern United States longleaf pine (Pinus palustris) ecosystem. Several markrecapture methods have been used range-wide in hopes of increasing knowledge on their population numbers. I explored the efficacy of two mark-recapture methods (scale cauterization and dorsal head photography) using the Marine Corps Recruit Depot, Parris Island, SC EDB population in this multi-part study. Liaison librarians depend on communication and cooperation to be successful, so professional publications about liaison librarianship regularly include discussions of relationship building methods. As a department of experienced liaison librarians, we identified the relationship-building ideas from previous publications that were proactive in nature and had worked for us. Even though the advice has been helpful, we faced challenges or heard about challenges from other librarians when we discussed implementing the ideas. Ketamine has been shown to produce analgesia in various acute and chronic pain states; however, abuse liability concerns have limited its utility. The ketamine metabolite (2R,6R)-hydroxynorketamine (HNK) has been shown to produce antidepressant-like effects similar to ketamine without abuse liability concerns. (2R,6R)-HNK produces sustained analgesia in models of chronic pain, but has yet to be evaluated in models of acute pain. Habitat loss due to deforestation is a primary threat to global biodiversity. Clearing tropical rainforests for agriculture leads to forest fragmentation. Forest fragments contain fewer large trees and provide lower food availability for primates compared to continuous forests. Mantled howler monkeys inhabit the increasingly fragmented rainforests of Central and South America and may need to alter their activity and spatial cohesion to mitigate competition and preserve energy in fragments where there is lower quality food. Coastal ecosystems are degrading rapidly, and molecular analysis of environmental DNA (eDNA) has become an increasingly popular method to quantify changes in biodiversity, and as a powerful and affordable complement to monitor aquatic environments. Yet, side by side comparisons of biodiversity estimates from eDNA and other traditional methods are needed to understand the level of accuracy of the eDNA method. We used eDNA metabarcoding of the 12S mitochondrial gene to generate a multi-species fish biodiversity assessment of Narragansett Bay (NB). Current understandings of epigenetics offer some insights into the developmental origins of character virtues. Research on epigenetics has demonstrated that phenotypes depend on both genomic and non-genomic information for their development, and such is true for the development of character virtues as well. This conclusion offers significant support for the Developmental Systems perspective. Although researchers have not yet studied the role of molecular epigenetic factors in the development of character, the field of behavioral epigenetics has revealed how such factors contribute to psychological and other behavioral phenotypes in general. Purpose: To describe the development of a Compendium for estimating the energy costs of activities in adults ≥60 years (OA Compendium).

Methods: Physical activities (PAs) and their metabolic equivalent of task (MET) values were obtained from a systematic search of studies published in 4 sport and exercise databases (PubMed, Embase, SPORTDiscus (EBSCOhost), and Scopus) and a review of articles included in the 2011 Adult Compendium that measured PA in older adults. MET values were computed as the oxygen cost (VO2, mL/kg/min) during PA divided by 2.7 mL/kg/min (MET60+) to account for the lower resting metabolic rate …
