Soil Carbon And Nitrogen After Eight Years Of Rotational Grazing In The Nebraska Sandhills Meadows, Gandura O. Abagandura, Martha Mamo, W. H. Schacht, Aaron Shropshire, Jerry Volesky
Department of Agronomy and Horticulture: Faculty Publications
Grassland provides many ecosystem services; therefore, sustainable management practices of grassland are crucial for maintaining and enhancing its ecosystem health and resilience. Rotational grazing at a high stocking density (a.k.a., ultrahigh stocking density) is purported to sequester greater amounts of carbon (C) in grassland soils than rotational grazing at low stocking densities. This study was conducted in the Nebraska Sandhills meadows for eight years to evaluate how rotational grazing with different stocking densities can affect soil C and total nitrogen (TN) in bulk soils, soil organic matter fractions, and sequestration rate. The grazing management included a high stocking density with …

Background: Increasing evidence implicates microbiome involvement in the development and progression of pancreatic ductal adenocarcinoma (PDAC). Studies suggest that reflux of gut or oral microbiota can lead to colonization in the pancreas, resulting in dysbiosis that culminates in release of microbial toxins and metabolites that potentiate an inflammatory response and increase susceptibility to PDAC. Moreover, microbe-derived metabolites can exert direct effector functions on precursors and cancer cells, as well as other cell types, to either promote or attenuate tumor development and modulate treatment response.
Content: The occurrence of microbial metabolites in biofluids thereby enables risk assessment and prognostication of PDAC, …

We explored the association between macronutrient intake and postprandial glucose variability in a large sample of youth living with T1D and consuming free-living meals. In the Type 1 Diabetes Exercise Initiative Pediatric (T1DEXIP) Study, youth took photographs before and after their meals on 3 days during a 10 day observation period. We used the remote food photograph method to obtain the macronutrient content of youth's meals. We also collected physical activity, continuous glucose monitoring, and insulin use data. We measured glycemic variability using standard deviation (SD) and coefficient of variation (CV) of glucose for up to 3 h after meals. …

Knowing what happens over time to the lifeways of people in contemporary small-scale non-industrial societies of the rural Global South matters because it helps assess changes in the quality of life of underrepresented groups. It has been hard to answer the question because longitudinal information is rarely collected in such settings. A longitudinal dataset of nine years (2002–2010) from a horticultural-foraging society of Indigenous People in the Bolivian Amazon (Tsimane’) is used for an exploratory analysis of micro-level trends in indicators of well-being and economic inequalities. We selected 13 Tsimane’ villages (from ∼ 100) that varied in proximity to town …

PPFIA3 encodes the protein-tyrosine phosphatase, receptor-type, F-polypeptide-interacting-protein-alpha-3 (PPFIA3), which is a member of the LAR-protein-tyrosine phosphatase-interacting-protein (liprin) family involved in synapse formation and function, synaptic vesicle transport, and presynaptic active zone assembly. The protein structure and function are evolutionarily well conserved, but human diseases related to PPFIA3 dysfunction are not yet reported in OMIM. Here, we report 20 individuals with rare PPFIA3 variants (19 heterozygous and 1 compound heterozygous) presenting with developmental delay, intellectual disability, hypotonia, dysmorphisms, microcephaly or macrocephaly, autistic features, and epilepsy with reduced penetrance. Seventeen unique PPFIA3 variants were detected in 18 families. To determine the pathogenicity …

Cytokines employ downstream Janus kinases (JAKs) to promote chronic inflammatory diseases. JAK1-dependent type 2 cytokines drive allergic inflammation, and patients with JAK1 gain-of-function (GoF) variants develop atopic dermatitis (AD) and asthma. To explore tissue-specific functions, we inserted a human JAK1 GoF variant (JAK1

Members of the low-density lipoprotein receptor (LDLR) family, including LDLRAD3, VLDLR, and ApoER2, were recently described as entry factors for different alphaviruses. However, based on studies with gene edited cells and knockout mice, blockade or abrogation of these receptors does not fully inhibit alphavirus infection, indicating the existence of additional uncharacterized entry factors. Here, we perform a CRISPR-Cas9 genome-wide loss-of-function screen in mouse neuronal cells with a chimeric alphavirus expressing the Eastern equine encephalitis virus (EEEV) structural proteins and identify LDLR as a candidate receptor. Expression of LDLR on the surface of neuronal or non-neuronal cells facilitates binding and infection …

Stroke results in local neural disconnection and brain-wide neuronal network dysfunction leading to neurological deficits. Beyond the hyper-acute phase of ischaemic stroke, there is no clinically-approved pharmacological treatment that alleviates sensorimotor impairments. Functional recovery after stroke involves the formation of new or alternative neuronal circuits including existing neural connections. The type-5 metabotropic glutamate receptor (mGluR5) has been shown to modulate brain plasticity and function and is a therapeutic target in neurological diseases outside of stroke. We investigated whether mGluR5 influences functional recovery and network reorganization rodent models of focal ischaemia. Using multiple behavioural tests, we observed that treatment with negative …

Patients with diabetes have a high risk of developing skeletal diseases accompanied by diabetic peripheral neuropathy (DPN). In this study, we isolated the role of DPN in skeletal disease with global and conditional knockout models of sterile-α and TIR-motif-containing protein-1 (Sarm1). SARM1, an NADase highly expressed in the nervous system, regulates axon degeneration upon a range of insults, including DPN. Global knockout of Sarm1 prevented DPN, but not skeletal disease, in male mice with type 1 diabetes (T1D). Female wild-type mice also developed diabetic bone disease but without DPN. Unexpectedly, global Sarm1 knockout completely protected female mice from T1D-associated bone …

Document Productivity Cycle (Study Case Of Samudera Raksa Ship Museum), Ciwuk Musiana Yudhawasthi, Lydia Christiani
Proceedings from the Document Academy
The study aims to discuss document productivity in the case of the Samudera Raksa Ship Museum. To answer this, the researchers made a productivity document study based on (1) Blasius Sudarsono's axiom, which states that "In the beginning, it was the human will to express what he thought and/or felt;" (2) Sudarsono's thoughts regarding documents as processes and products; (3) Lund’s concept of document creation; (4) Sabine Roux's thoughts on the rhizome concept in the document productivity process; and (5) the concept of museum communication by Yudhawasthi. Based on these theoretical frameworks, an analysis of the document productivity in the …

Document Theory And Document Design, Arthur Perret
Proceedings from the Document Academy
This paper reports on an experiment which connects document design with document theory. Document design is about what documents should do. We use the example of document graphs and of Cosma, a visualization program created during the HyperOtlet research programme, to show how theory can inform design decisions and help conceptualize results.

Making-To-Be: Documents, Facta, And Material-Discursive Agency, Elliott Hauser
Proceedings from the Document Academy
This paper presents the performative analysis of agency within and surrounding documents as a path towards uniting the otherwise incompatible insights of both meaningcentric and materialcentric approaches. I contrast the terms agentical, providing agency, and agentic, possessing agency, to help clarify the apparent incompatibilities of prior approaches. I argue that a relational conception of agency, wherein the agentical/agentic distinction is blurred, preserves important virtues of both meaning and materialcentric approaches to documents. This paves the way for a unified materialdiscursive account of documents and a cure for document studies’ inherited duality malady. Extending prior work on capta (Drucker, 2011) and …
Powerful Particulars As “Autodocuments” In Documentality, Ronald E. Day
Powerful Particulars As “Autodocuments” In Documentality, Ronald E. Day
Proceedings from the Document Academy
The purpose of this short paper is to sketch the problem of whether documentality, in the sense of the appearance of evidence, must always take the form of a type-token relationship. In contrast to a type-token epistemology common in the Library and Information Science tradition, the paper argues that there is precedence for a theory of documentality that views evidentiality as a product of the powers of particulars to make themselves present. To make this argument, it appeals to Robert Pagès theory of documents and, over a half century later, Bernd Frohmann’s proposal for a philosophy of information, “Documentality.” Such …

Using Poetics and Technology to Inform Pre-Service Educators’ Transformative Pedagogy via Innovative Sociocultural Interactivities
During the HICE workshop my colleague and I will demonstrate and engage HICE participants through learner centered practices we developed and studied during the Covid pandemic. We embedded oral and written poetic expression with a variety of technological practices to offer pre-service educators creative opportunities to learn and participate in the course through unique expressive movements, music, and media during on-line instruction. These approaches will show how to create a culture of care and Social Justice in a synchronous online classroom.

Therapeutic resistance remains a major obstacle to successful clinical management of diffuse intrinsic pontine glioma (DIPG), a high-grade pediatric tumor of the brain stem. In nearly all patients, available therapies fail to prevent progression. Innovative combinatorial therapies that penetrate the blood-brain barrier and lead to long-term control of tumor growth are desperately needed. We identified mechanisms of resistance to radiotherapy, the standard of care for DIPG. On the basis of these findings, we rationally designed a brain-penetrant small molecule, MTX-241F, that is a highly selective inhibitor of EGFR and PI3 kinase family members, including the DNA repair protein DNA-PK. Preliminary …

Here, we report a previously unrecognized syndromic neurodevelopmental disorder associated with biallelic loss-of-function variants in the RBM42 gene. The patient is a 2-year-old female with severe central nervous system (CNS) abnormalities, hypotonia, hearing loss, congenital heart defects, and dysmorphic facial features. Familial whole-exome sequencing (WES) reveals that the patient has two compound heterozygous variants, c.304C>T (p.R102*) and c.1312G>A (p.A438T), in the RBM42 gene which encodes an integral component of splicing complex in the RNA-binding motif protein family. The p.A438T variant is in the RRM domain which impairs RBM42 protein stability in vivo. Additionally, p.A438T disrupts the interaction of …

Dry bean is a nutrient-dense food targeted in biofortification programs to increase seed iron and zinc levels. The underlying assumption of breeding for higher mineral content is that enhanced iron and zinc levels will deliver health benefits to the consumers of these biofortified foods. This study characterized a diversity panel of 275 genotypes comprising the Yellow Bean Collection (YBC) for seed Fe and Zn concentration, Fe bioavailability (FeBio), and seed yield across 2 years in two field locations. The genetic architecture of each trait was elucidated via genome-wide association studies (GWAS) and the efficacy of genomic prediction (GP) was assessed. …

There are limited therapeutic options for patients with advanced prostate cancer (PCa). We previously found that heat shock factor 1 (HSF1) expression is increased in PCa and is an actionable target. In this manuscript, we identify that HSF1 regulates the conversion of homocysteine to cystathionine in the transsulfuration pathway by altering levels of cystathionine-β-synthase (CBS). We find that HSF1 directly binds the CBS gene and upregulates CBS mRNA levels. Targeting CBS decreases PCa growth and induces tumor cell death while benign prostate cells are largely unaffected. Combined inhibition of HSF1 and CBS results in more pronounced inhibition of PCa cell …
