The public has expressed growing concern for the well-being of fishes, including popular pet species such as the Siamese fighting fish (Betta splendens). In captivity, male Bettas behave aggressively, often causing injuries and death if housed together. As a result, they are typically isolated in small fishbowls, which has been widely criticised as cruel. To investigate the impact of keeping Bettas in these conditions, we recorded the behaviour of individual males in containers of different sizes that were either bare or enriched with gravel, large rocks, and live plants. BACKGROUND: Cardiac metabolic dysfunction is a hallmark of heart failure (HF). Estrogen-related receptors ERRα and ERRγ are essential regulators of cardiac metabolism. Therefore, activation of ERR could be a potential therapeutic intervention for HF. However, in vivo studies demonstrating the potential usefulness of ERR agonist for HF treatment are lacking, because compounds with pharmacokinetics appropriate for in vivo use have not been available.

Protein kinases are frequently dysregulated and/or mutated in cancer and represent essential targets for therapy. Accurate quantification is essential. For breast cancer treatment, the identification and quantification of the protein kinase ERBB2 is critical for therapeutic decisions. While immunohistochemistry (IHC) is the current clinical diagnostic approach, it is only semiquantitative. Mass spectrometry-based proteomics offers quantitative assays that, unlike IHC, can be used to accurately evaluate hundreds of kinases simultaneously. The enrichment of less abundant kinase targets for quantification, along with depletion of interfering proteins, improves sensitivity and thus promotes more effective downstream analyses. AMP-activated protein kinase (AMPK) plays a crucial role in maintaining ATP homeostasis in photoreceptor neurons. AMPK is a heterotrimeric protein consisting of α, β, and γ subunits. The independent functions of the 2 isoforms of the catalytic α subunit, PRKAA1 and PRKAA2, are uncharacterized in specialized neurons, such as photoreceptors. Here, we demonstrate in mice that rod photoreceptors lacking PRKAA2, but not PRKAA1, showed altered levels of cGMP, GTP, and ATP, suggesting isoform-specific regulation of photoreceptor metabolism. Furthermore, PRKAA2-deficient mice displayed visual functional deficits on electroretinography and photoreceptor outer segment structural abnormalities on transmission electron microscopy consistent with neuronal dysfunction, …

Abdominal aortic aneurysms (AAAs) are prevalent with aging, and AAA rupture is associated with increased mortality. There is currently no effective medical therapy to prevent AAA rupture. The monocyte chemoattractant protein (MCP-1)/C-C chemokine receptor type 2 (CCR2) axis critically regulates AAA inflammation, matrix-metalloproteinase (MMP) production, and extracellular matrix (ECM) stability. We therefore hypothesized that a diet intervention that can modulate CCR2 axis may therapeutically impact AAA risk of rupture. Since ketone bodies (KBs) can trigger repair mechanisms in response to inflammation, we evaluated whether systemic ketosis in vivo could reduce CCR2 and AAA progression. Age-related macular degeneration (AMD) is a leading cause of blindness featuring pathogenic neovascularization of the choroidal vasculature (CNV). Although systemic immunity plays a role in AMD, the ocular signals that recruit and activate immune cells remain poorly defined. Using single-cell RNA sequencing, we prospectively profile peripheral blood mononuclear cells from 65 individuals including AMD and controls, which we integrate with existing choroid data. We generate a network of choroid-peripheral immune interactions dysregulated in AMD, including known AMD-relevant gene vascular endothelial growth factor (VEGF) receptor 2. Additionally, we find CYR61 is upregulated in choroidal veins and may signal to circulating monocytes. …

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease characterized by motor neuron loss. Importantly, non-neuronal cell types such as astrocytes also play significant roles in disease pathogenesis. However, mechanisms of astrocyte contribution to ALS remain incompletely understood. Astrocyte involvement suggests that transcellular signaling may play a role in disease. We examined contribution of transmembrane signaling molecule ephrinB2 to ALS pathogenesis, in particular its role in driving motor neuron damage by spinal cord astrocytes. In symptomatic SOD1G93A mice (a well-established ALS model), ephrinB2 expression was dramatically increased in ventral horn astrocytes. We examine social-environmental justice in forest governance by asking who is problematized as drivers of deforestation and forest degradation. We adapt Bacchi’s “What is the problem represented to be” approach to the community forest (CAF) model in Burkina Faso and the Payment for Forest Environmental Services (PFES) in Vietnam and examine the implementation of these policies in specific sites through disaggregated focus group discussions (men, women, youth, ethnic minorities). A Pilot Model Of Community-Based Forest Management In Xuan Nha Nature Reserve, Son La Province, Vietnam, Thuy Thi Phan, Duong Huu Nong

This study evaluated the implementation of a pilot model for promoting community-based conservation through a contracting program in Xuan Nha Nature Reserve, located in Son La province, northern Vietnam, from 2014 to 2022. To assess the effectiveness of the program, in-depth interviews were conducted with 26 officials and 100 individuals residing in three villages. The findings revealed that the program successfully facilitated community-based conservation by involving local communities in participatory land use planning and forest protection at the village level. The Tau Taa Vana people live in the Bulang Highlands, Tojo Una-Una, in Central Sulawesi Province. The region's development has shaped the marginalization of forest-dwelling and forest-adjacent communities. From the 1980s to the 1990s, illegal logging networks served as the power holders, backed by Indonesia’s authoritarian regime of that time. Illegal logging destroyed a large part of the Tau Taa Vana's sacred forest (pengale kapali). As part of the massive logging agenda, the government launched many legal programs that further isolated the Tau Taa Vana people from their land. Mapping the demand for ecosystem services (ES) is a profoundly debated research topic that needs to be investigated further to overcome spatial discrepancies between supply and demand in the literature. This study proposes a holistic approach to valuing ES, which is demanded by local people living adjacent to Sabah’s protected area in the southwestern part of Sabah, Malaysia. This study analyses dimensions of participatory forest landscape governance of the Essential Ecosystem Area (EEA) of Taman Kili-Kili, Indonesia. The voices of marginalized communities, and especially women, are rarely incorporated into forest landscape governance and conservation policies. The recently established Indonesian EEA policy mandates a participatory approach, with explicit requirements to involve marginalized groups and gendered perspectives. However, on a practical level, policy formulation and application unfold in very different ways. Char is any accretion in a river course that extends or establishes new land. A well-planned integrated land-use system combining woody perennials (agroforestry) can ensure sustainable, environmentally friendly climate resilience land-use systems and livelihood options on charland. To date, no systemic investigation of charland land agroforestry has been undertaken. A comprehensive study of the potential of agroforestry systems in charland areas was conducted using qualitative and quantitative methodologies. Two methods were employed: structured questionnaire interviews and botanical surveys. In this study we evaluated feed and temporal effects on prokaryotic (16S) and eukaryotic (18S) microbiome dynamics in an outdoor mixotrophic biofloc system used to grow hybrid tilapia (Oreochromis aureus x O. niloticus) to market size. The dicalcium phosphate -free practical feed treated with 6-phytase (3000 FTU/kg; P-Free+ Phytase) and positive (Control) and negative (P-Free) control practical feeds used were formulated to contain 27.7% digestible protein and 4% lipid. BACKGROUND: QiDiTangShen granules (QDTS), a traditional Chinese medicine (TCM) compound prescription, have remarkable efficacy in diabetic nephropathy (DN) patients, and their pharmacological mechanism needs further exploration.

METHODS: According to the active ingredients and targets of the QDTS in the TCMSP database, the network pharmacology of QDTS was investigated. The potential active ingredients were chosen based on the oral bioavailability and the drug similarity index. At the same time, targets for DN-related disease were obtained from GeneCards, OMIM, PharmGKB, TTD, and DrugBank. Placental development involves coordinated expansion and differentiation of trophoblast cell lineages possessing specialized functions. Among the differentiated trophoblast cell lineages are invasive trophoblast cells, which exit the placenta and invade the uterus, where they restructure the uterine parenchyma and facilitate remodeling of uterine spiral arteries. The rat exhibits deep intrauterine trophoblast cell invasion, a feature shared with human placentation, and is also amenable to gene manipulation using genome-editing techniques. This project investigated small-scale habitat use patterns of harbor seals (Phoca vitulina). The goals were to provide an updated overview of the harbor seal population in Elkhorn Slough, California, USA, identify haul-out areas that are seasonally relevant for pupping and molting, and analyze environmental factors that may influence the harbor seals’ abundance and distribution in the slough. Seal counts were conducted by Elkhorn Slough volunteers throughout the entire slough June 2020-May 2023. The COVID-19 pandemic was associated with an increased global use of traditional medicines, including Ayurvedic herbal preparations. Due to their growing demand, their processed nature, and the complexity of the global supply chain, there is an increased risk of adulteration in these products.

Hematopoietic stem cells (HSCs) are capable of regenerating the blood system, but the instructive cues that direct HSCs to regenerate particular lineages lost to the injury remain elusive. Here, we show that iron is increasingly taken up by HSCs during anemia and induces erythroid gene expression and regeneration in a Tet2-dependent manner. Lineage tracing of HSCs reveals that HSCs respond to hemolytic anemia by increasing erythroid output. The number of HSCs in the spleen, but not bone marrow, increases upon anemia and these HSCs exhibit enhanced proliferation, erythroid differentiation, iron uptake, and TET2 protein expression. Heart failure (HF) causes substantial morbidity and mortality but its pathobiology is incompletely understood. The proteome is a promising intermediate phenotype for discovery of novel mechanisms. We measured 4877 plasma proteins in 13,900 HF-free individuals across three analysis sets with diverse age, geography, and HF ascertainment to identify circulating proteins and protein networks associated with HF development. Parallel analyses in Atherosclerosis Risk in Communities study participants in mid-life and late-life and in Trøndelag Health Study participants identified 37 proteins consistently associated with incident HF independent of traditional risk factors. Alternative feeds are critical for the sustainable expansion of the marine finfish aquaculture industry. The industry uses wild-caught forage fish as a primary ingredient in farmed fish feeds. Alternative ingredients are needed to safeguard fisheries' sustainability and future aquaculture development. BACKGROUND: Neural tube defects (NTDs) are caused by genetic and environmental factors. ARMC5 is part of a novel ubiquitin ligase specific for POLR2A, the largest subunit of RNA polymerase II (Pol II).

RESULTS: We find that ARMC5 knockout mice have increased incidence of NTDs, such as spina bifida and exencephaly. Surprisingly, the absence of ARMC5 causes the accumulation of not only POLR2A but also most of the other 11 Pol II subunits, indicating that the degradation of the whole Pol II complex is compromised. This research explores the role of native plants in enhancing the distinctiveness of cities in Saudi Arabia, focusing on a specific case study of Al-Samra Park. The study aims to investigate the prevalence and effects of non-native and native plants in the park and their contribution to the uniqueness of the landscape. A questionnaire was designed and implemented to gather data, and the park's plant composition was examined.
