Reactive Amine Functionalized Microelectrode Arrays Provide Short-Term Benefit But Long-Term Detriment To In Vivo Recording Performance, Lindsey N. Druschel, Pierce E. Boucher, George F. Hoeferlin, Dhariyat M. Menendez, Jonathan L. Duncan, Jeffrey R. Capadona

Faculty Scholarship

Intracortical microelectrode arrays (MEAs) are used for recording neural signals. However, indwelling devices result in chronic neuroinflammation, which leads to decreased recording performance through degradation of the device and surrounding tissue. Coating the MEAs with bioactive molecules is being explored to mitigate neuroinflammation. Such approaches often require an intermediate functionalization step such as (3-aminopropyl)triethoxysilane (APTES), which serves as a linker. However, the standalone effect of this intermediate step has not been previously characterized. Here, we investigated the effect of coating MEAs with APTES by comparing APTES-coated to uncoated controls in vivo and ex vivo. First, we measured water contact angles …

Myd88 In Myofibroblasts Enhances Nonalcoholic Fatty Liver Disease-Related Hepatocarcinogenesis Via Promoting Macrophage M2 Polarization, Yu Liu, Haiqiang Chen, Xuanxuan Yan, Jie Zhang, Zhenzhong Deng, Maosheng Huang, Jianchun Gu, Jinhua Zhang

Faculty, Staff and Student Publications

Background: Nonalcoholic fatty liver disease (NAFLD) is a major cause of chronic liver diseases and has emerged as the leading factor in the pathogenesis of hepatocellular carcinoma (HCC). MyD88 contributes to the development of HCC. However, the underlying mechanism by which MyD88 in myofibroblasts regulates NAFLD-associated liver cancer development remains unknown.

Results: Myofibroblast MyD88-deficient (SMAMyD88-/-) mice were protected from diet-induced obesity and developed fewer and smaller liver tumors. MyD88 deficiency in myofibroblasts attenuated macrophage M2 polarization and fat accumulation in HCC tissues. Mechanistically, MyD88 signaling in myofibroblasts enhanced CCL9 secretion, thereby promoting macrophage M2 polarization. This process may depend on …

Hydrophobic Surface Patch Disruption To Produce Water-Soluble G-Protein Coupled Receptors, Christy Lynn Dyer

Electronic Theses and Dissertations Archive

G-Protein-Coupled Receptors (GPCR) are membrane protein receptors involved in most eukaryotic signaling processes. GPCR are the largest pharmaceutically relevant membrane protein group, serving as targets for approximately one-third of all drugs currently approved by the FDA and many drug candidates presently undergoing clinical trials. Still, only about one-fourth of all known GPCR are current pharmaceutical targets, leaving vast untapped potential. Chapter 1 describes the conserved structure of GPCR, and briefly introduces their diverse ligands and functions. The pharmaceutical relevance of GPCR is highlighted, along with some of the challenges associated with their structure determination that have hindered our understanding of …

Nest/Ing: An Emergent (Un)Methodology For Becoming Otherwise, Jayne Osgood, Claire Walsh

School of Law, Humanities and Social Sciences

A dense, pungent, brown-green, intricately woven, itchy-silky, moss-strewn, twig- ridden ball brought us together.Since then, nest(ing)[i] has become a shared methodology - nest/ing has offered a praxis of getting to know each other, getting to know nests (better) and it has become a capacious writing methodology. Taking nest(ing) seriously has drawn into sharp focus the perils of human exceptionalism. Nesting has gifted opportunities to wallow in porous boundaries and to luxuriate in modes of liminal reading/writing/experimenting, informed by a feminist politics to imagine the world differently (Despret, 2016). It is through lively storytelling, involving passing patterns back and forth that …

Enhancing Antibody Responses By Multivalent Antigen Display On Thymus-Independent Dna Origami Scaffolds, Eike-Christian Wamhoff, James Brett Case, Shilpa Sanapala, Michael S Diamond, Et Al.

2020-Current year OA Pubs

Protein-based virus-like particles (P-VLPs) are commonly used to spatially organize antigens and enhance humoral immunity through multivalent antigen display. However, P-VLPs are thymus-dependent antigens that are themselves immunogenic and can induce B cell responses that may neutralize the platform. Here, we investigate thymus-independent DNA origami as an alternative material for multivalent antigen display using the receptor binding domain (RBD) of the SARS-CoV-2 spike protein, the primary target of neutralizing antibody responses. Sequential immunization of mice with DNA-based VLPs (DNA-VLPs) elicits protective neutralizing antibodies to SARS-CoV-2 in a manner that depends on the valency of the antigen displayed and on T …

Expression Of Atoh1, Gfi1, And Pou4f3 In The Mature Cochlea Reprograms Nonsensory Cells Into Hair Cells, Melissa M Mcgovern, Ishwar V Hosamani, Yichi Niu, Ken Y Nguyen, Chenghang Zong, Andrew K Groves

Faculty, Staff and Students Publications

Mechanosensory hair cells of the mature mammalian organ of Corti do not regenerate; consequently, loss of hair cells leads to permanent hearing loss. Although nonmammalian vertebrates can regenerate hair cells from neighboring supporting cells, many humans with severe hearing loss lack both hair cells and supporting cells, with the organ of Corti being replaced by a flat epithelium of nonsensory cells. To determine whether the mature cochlea can produce hair cells in vivo, we reprogrammed nonsensory cells adjacent to the organ of Corti with three hair cell transcription factors: Gfi1, Atoh1, and Pou4f3. We generated numerous hair cell–like …

Hala Aswang!: A Zine On The Injustices During The Duterte Administration Foretold Through The Aswang Folklore, Eritch Angela Torrato

DLSU Senior High School Research Congress

The Duterte administration, which lasted from 2016 to 2022, incited a multitude of human rights violations against the marginalized groups of Philippine society, namely women, and the poor. It was discovered and established through literature that these injustices can primarily be attributed to the former president’s hypermasculinity, a manifestation of hegemonic masculinity in the contemporary era. Given this, this research then seeked to investigate the aforementioned phenomenon and communicate this using art, more specifically through the Aswang folklore and visual communication, to make these information accessible, especially to the masses; to foster social consciousness among its audience as a means …

California’s Proposition 12, which outlaws the use of cages in the egg production process, went into effect on January 1, 2022. The main focus of this study is to follow the trend of weekly Californian and national egg price changes before and after the implementation of Proposition 12. Using the price series reported by the United States Department of Agriculture (USDA), structural break tests and Ordinary Least Squares (OLS) regressions are carried out to directly measure the economic impacts of this new regulation on egg prices. With different models used, this study shows that California consumers now pay an additional …

Machine Learning Of Big Data: A Gaussian Regression Model To Predict The Spatiotemporal Distribution Of Ground Ozone, Jerry Gu Jan 2024

Machine Learning Of Big Data: A Gaussian Regression Model To Predict The Spatiotemporal Distribution Of Ground Ozone, Jerry Gu

The Journal of Purdue Undergraduate Research

Tracking pollution levels on the ground is important to the environment and public health. One of the pollutants of concern is ozone, which, at high concentrations, can cause respiratory and cardiovascular problems. The National Center for Atmospheric Research (NCAR) has published valuable ozone data obtained from ground-based sensors installed at selected locations. Because it is unfeasible to measure the exact ozone levels everywhere at any time, it would be valuable to predict the temporal-spatial distributions of ozone concentration based on existing data. This would help us better understand the patterns and trends in the data and make better decisions to …
