PURPOSE: To establish generalizable pointwise spatial relationship between structure and function through occlusion analysis of a deep-learning (DL) model for predicting the visual field (VF) sensitivities from 3-dimensional (3D) OCT scan.
DESIGN: Retrospective cross-sectional study.
PARTICIPANTS: A total of 2151 eyes from 1129 patients.
METHODS: A DL model was trained to predict 52 VF sensitivities of 24-2 standard automated perimetry from 3D spectral-domain OCT images of the optic nerve head (ONH) with 12 915 OCT-VF pairs. Appendiceal adenocarcinoma (AA) is unique from other gastrointestinal malignancies in that it almost exclusively metastasizes to the peritoneal cavity. However, few studies have investigated the molecular interaction of the peritoneal microenvironment and AA. Here, we use a multi-omics approach with orthotopic and flank-implanted patient-derived xenografts (PDX) to study the effect of the peritoneal microenvironment on AA. AA tumors implanted in the peritoneal microenvironment tended to grow faster and displayed greater nuclear expression of Ki-67 relative to the same tumors implanted in the flank. Metastatic disease remains the leading cause of death due to cancer, yet the mechanism(s) of metastasis and its timely detection remain to be elucidated. Neutrophil elastase (NE), a serine protease secreted by neutrophils, is a crucial mediator of chronic inflammation and tumor progression. In this study, we used the PyMT model (NE+/+ and NE-/-) of breast cancer to interrogate the tumor-intrinsic and -extrinsic mechanisms by which NE can promote metastasis. Our results showed that genetic ablation of NE significantly reduced lung metastasis and improved metastasis-free survival. Current cancer vaccines using T cell epitopes activate antitumor T cell immunity through dendritic cell/macrophage-mediated antigen presentation, but they lack the ability to promote B/CD4 T cell crosstalk, limiting their anticancer efficacy. We developed antigen-clustered nanovaccine (ACNVax) to achieve long-term tumor remission by promoting B/CD4 T cell crosstalk. The topographic features of ACNVax were achieved using an iron nanoparticle core attached with an optimal number of gold nanoparticles, where the clusters of HER2 B/CD4 T cell epitopes were conjugated on the gold surface with an optimal intercluster distance of 5-10 nm. Aberrant glycosylation is a crucial strategy employed by cancer cells to evade cellular immunity. However, it's unclear whether homologous recombination (HR) status-dependent glycosylation can be therapeutically explored. Here, we show that the inhibition of branched N-glycans sensitizes HR-proficient, but not HR-deficient, epithelial ovarian cancers (EOCs) to immune checkpoint blockade (ICB). In contrast to fucosylation whose inhibition sensitizes EOCs to anti-PD-L1 immunotherapy regardless of HR-status, we observe an enrichment of branched N-glycans on HR-proficient compared to HR-deficient EOCs. Mechanistically, BRCA1/2 transcriptionally promotes the expression of MGAT5, the enzyme responsible for catalyzing branched N-glycans. The branched N-glycans on HR-proficient tumors augment their …

Monitoring biomarkers related to inflammation and oxidative stress is critical in dogs because parvovirus causes both inflammatory and antioxidant alterations. The aim of this study was to investigate inflammatory and antioxidant changes caused by canine parvoviral enteritis to better understand the oxidative stress process related to this disease. Thus, the total thiol, native thiol, disulfide, and ischemia-modified albumin levels of Canine parvovirus infected symptomatic puppies and healthy puppies were examined. Nuclear factor κ-B (NFκB) is activated in iPSC-cardiac myocytes from patients with arrhythmogenic cardiomyopathy (ACM) under basal conditions, and inhibition of NFκB signaling prevents disease in Dsg2mut/mut mice, a robust mouse model of ACM. Here, we used genetic approaches and single-cell RNA-Seq to define the contributions of immune signaling in cardiac myocytes and macrophages in the natural progression of ACM using Dsg2mut/mut mice. We found that NFκB signaling in cardiac myocytes drives myocardial injury, contractile dysfunction, and arrhythmias in Dsg2mut/mut mice. NFκB signaling in cardiac myocytes mobilizes macrophages expressing C-C motif chemokine receptor-2 (CCR2+ cells) to affected areas within the …

Acronyms used and depended on by livestock producers are reviewed. Understanding the meaning of livestock acronyms is commonly overlooked. Fibromyalgia is a chronic, widespread pain disorder that is strongly represented across the affective and cognitive dimensions of pain, given that the underlying pathophysiology of the disorder is yet to be identified. These affective and cognitive deficits are crucial to understanding and treating the fibromyalgia pain experience as a whole but replicating this multidimensionality on a preclinical level is challenging. To understand the underlying mechanisms, animal models are used. Human infections caused by viral pathogens trigger a complex gamut of host responses that limit disease, resolve infection, generate immunity, and contribute to severe disease or death. Here, we present experimental methods and multi-omics data capture approaches representing the global host response to infection generated from 45 individual experiments involving human viruses from the Orthomyxoviridae, Filoviridae, Flaviviridae, and Coronaviridae families. Analogous experimental designs were implemented across human or mouse host model systems, longitudinal samples were collected over defined time courses, and global multi-omics data (transcriptomics, proteomics, metabolomics, and lipidomics) were acquired by microarray, RNA sequencing, or mass spectrometry analyses. Radically Feminist Or Monstrously Feminine?: Witches And Goddesses In Guadagnino's Suspiria (2018), Lindsay Macumber
Journal of Religion & Film
Guadagnino’s 2018 remake of Suspiria explicitly and implicitly incorporates two connected myths, witchcraft and goddess centered matriarchal prehistory. The fact that each of these myths have been claimed by feminists in myriad ways may explain Guadagnino’s claim that Suspiria is a great feminist film that escapes the male gaze. In this article, I argue that Guadagnino’s representation of these myths lays bare their misogynistic origins and perpetuates, rather than subverts, patriarchal power structures.

Spiritual Cinema: Agel, Merleau-Ponty And The Cinematic Real, Patrick O'Connor Dr

This article seeks to retrieve the work of Henri Agel, and his collaborator Amédée Ayfre, for our theoretical understanding of film-philosophy. I explore their distinctive contribution to thinking philosophically about film and assess the relative merits of their work for the phenomenology of film. While exceptionally valuable for religious and theological interpretations of film I proceed to argue that Agel and Ayfre’s work needs to be supplemented with the work of Maurice Merleau-Ponty’s film-phenomenology to adequately express the temporal and motional nature of film. Merleau-Ponty’s work I contend while exceptionally valuable is brief and underdeveloped, and therefore does not fully …

Effects of cadmium and obesity on accumulation of essential metals in mouse kidney were evaluated. Starting in utero and continuing until 24 weeks postweaning, male/female mice were exposed to cadmium (0, 0.5, or 5 ppm) and were fed on Normal Diet (ND)/High-Fat Diet (HFD) post weaning. ICP-MS analysis was performed to evaluate cadmium and essential metals absorbed in the kidneys of aforementioned mice. The pressing need for sustainable development solutions necessitates innovative data-driven tools. Machine learning (ML) offers significant potential, but faces challenges in centralized approaches, particularly concerning data privacy and resource constraints in geographically dispersed settings. Federated learning (FL) emerges as a transformative paradigm for sustainable development by decentralizing ML training to edge devices. However, communication bottlenecks hinder its scalability and sustainability. This paper introduces an innovative FL framework that enhances communication efficiency.
