BACKGROUND: NODAL signaling plays a critical role in embryonic patterning and heart development in vertebrates. Genetic variants resulting in perturbations of the TGF-β/NODAL signaling pathway have reproducibly been shown to cause laterality defects in humans. To further explore this association and improve genetic diagnosis, the study aims to identify and characterize a broader range of NODAL variants in a large number of individuals with laterality defects.

METHODS: We re-analyzed a cohort of 321 proband-only exomes of individuals with clinically diagnosed laterality congenital heart disease (CHD) using family-based, rare variant genomic analyses. To this cohort we added 12 affected subjects with …

Improving Implicit Communication In Remote Collaboration Through Augmented Reality And Digital Twins, Nicholas Levergne Apr 2024

Improving Implicit Communication In Remote Collaboration Through Augmented Reality And Digital Twins, Nicholas Levergne

LSU Master's Theses

Large scale digital twinning projects are beginning to emerge across the tech industry. Within these projects is a desire to integrate augmented reality capabilities into industrial workflows. However, research on augmented reality technology for remote collaboration lacks ecologically valid studies of real world scenarios. Additionally, prior remote collaboration literature is focused on white-collar applications instead of blue-collar field work. Prior AR collaboration software is similarly limited, with most software allowing mixed camera views and annotation that requires participants to be stationary. This thesis introduces SpectAR, an augmented reality and desktop remote collaboration software suite developed in Unreal Engine 5.1.1. SpectAR …

Speech And Language Errors In An 8-Year-Old Spanish-English Bilingual Speaker: A Case Study, Alaniss Heredia Apr 2024

Speech And Language Errors In An 8-Year-Old Spanish-English Bilingual Speaker: A Case Study, Alaniss Heredia

LSU Master's Theses

Despite the growing population of Spanish-English bilingual speakers with developmental language disorders and speech sound disorders (SSD) in the United States, there is limited research on assessment for this population. Research suggests that the linguistic environment in which the Spanish-English bilingual speakers are brought up should be considered when assessing and treating this population since the Spanish-English bilingual population is diverse. One result of such environments is differences in degrees of language exposure. For example, a child can be exposed to two languages but have more exposure in one than the other. This might lead to better proficiency in one …

Chagas disease (CD) (American trypanosomiasis caused by Trypanosoma cruzi) is a parasitic disease endemic in 21 countries in South America, with increasing global spread. When administered late in the infection, the current antiparasitic drugs do not prevent the onset of cardiac illness leading to chronic Chagasic cardiomyopathy. Therefore, new therapeutic vaccines or immunotherapies are under development using multiple platforms. In this study, we assessed the feasibility of developing an mRNA-based therapeutic CD vaccine targeting two known T. cruzi vaccine antigens (Tc24—a flagellar antigen and ASP-2—an amastigote antigen). We present the mRNA engineering steps, preparation, and stability of the lipid …

An increasing number of pathogenic variants in presynaptic proteins involved in the synaptic vesicle cycle are being discovered in neurodevelopmental disorders. The clinical features of these synaptic vesicle cycle disorders are diverse, but the most prevalent phenotypes include intellectual disability, epilepsy, movement disorders, cerebral visual impairment, and psychiatric symptoms ( Verhage and Sørensen, 2020; Bonnycastle et al., 2021; John et al., 2021; Melland et al., 2021). Among this growing list of synaptic vesicle cycle disorders, the most frequent is STXBP1 encephalopathy caused by de novo heterozygous pathogenic variants in syntaxin-binding protein 1 (STXBP1, also known as …

Neuromodulation Of Mouse Auditory Cortex Using Focused Ultrasound, Jacob Hehir Apr 2024

Neuromodulation Of Mouse Auditory Cortex Using Focused Ultrasound, Jacob Hehir

NYMC Student Theses and Dissertations

Neurological diseases impose a large health and financial burden for nations worldwide. Current treatments for many conditions are limited to pharmacological or invasive surgical approaches. Neuromodulation is an alternative therapy wherein electrical currents, magnetic fields, or electromagnetic waves are used to noninvasively modify brain function. Focused ultrasound (FUS) has been shown to alter neural activity, theoretically with much higher spatial resolution than other neuromodulation techniques. However, these predictions assume that neuromodulatory effects are constrained to the acoustic focus. Given that the architecture of neural circuitry in the cerebral cortex supports delicately balanced lateral processing on spatial scales that may exceed …

Regulation Of Fatty Acid Delivery To Metastases By Tumor Endothelium, Deanna N Edwards, Shan Wang, Wenqiang Song, Laura C Kim, Verra M Ngwa, Yoonha Hwang, Kevin C Ess, Mark R Boothby, Jin Chen

Faculty, Staff and Students Publications

Tumor metastasis, the main cause of death in cancer patients, requires outgrowth of tumor cells after their dissemination and residence in microscopic niches. Nutrient sufficiency is a determinant of such outgrowth1. Fatty acids (FA) can be metabolized by cancer cells for their energetic and anabolic needs but impair the cytotoxicity of T cells in the tumor microenvironment (TME)2,3, thereby supporting metastatic progression. However, despite the important role of FA in metastatic outgrowth, the regulation of intratumoral FA is poorly understood. In this report, we show that tumor endothelium actively promotes tumor growth and restricts …

Can A Classical Christian Education Help Break Generational Poverty In Urban Students?, Elizabeth P. Porter Apr 2024

Can A Classical Christian Education Help Break Generational Poverty In Urban Students?, Elizabeth P. Porter

Master of Arts in Classical Studies

Although there has been a surge in the number of classical Christian schools in the last decade, there remains a dearth of classical Christian schools serving urban students. Unlike their suburban peers who tend to be more affluent, urban students often suffer physically, academically, and socioemotionally from the ills of poverty. The current study explores whether a classical Christian education (CCE) can help break generational poverty in urban students. Data from three CCE schools will be analyzed to assess quantitative and qualitative markers of success. The former include high school graduation rates, chronic absenteeism rates, and standardized test scores. The …

Maldi-Msi Identification Of Tissue-Level N-Glycomic And Proteomic Molecular Biomarkers Of Aggressive Prostate Cancer, Jordan Paige Hartig Apr 2024

Maldi-Msi Identification Of Tissue-Level N-Glycomic And Proteomic Molecular Biomarkers Of Aggressive Prostate Cancer, Jordan Paige Hartig

MUSC Theses and Dissertations

Prostate Cancer (PCa) poses a significant clinical challenge, characterized by debates on screening efficacy and continuous issues with distinguishing aggressive from non-aggressive tumors. Despite treatment advancements, poor patient outcome at late stages and clinical overtreatment of indolent disease emphasizes the urgent need for markers of PCa aggression. Aberrant glycosylation and extracellular matrix (ECM) remodeling are two complex molecular processes necessary for PCa progression, highlighting a novel area for biomarker discovery. Leveraging formalin-fixed paraffin-embedded tissues, tissue microarrays (TMAs), and matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI), distinct N-glycan and ECM profiles are identified. Elevated bisecting, multi-antennary, and fucosylated N-glycans indicate early …

Impact Of Fads Genotype On Polyunsaturated Fatty Acid Content In Human Milk Extracellular Vesicles: A Genetic Association Study, John J. Miklavcic, Natalie Paterson, Jennifer Hahn-Holbrook, Laura M. Glynn

Food Science Faculty Articles and Research

Background

Extracellular vesicles in human milk are critical in supporting newborn growth and development. Bioavailability of dietary extracellular vesicles may depend on the composition of membrane lipids. Single-nucleotide polymorphisms (SNPs) in the fatty acid desaturase gene cluster impact the content of long-chain polyunsaturated fatty acids in human milk phospholipids. This study investigated the relation between variation in FADS1 and FADS2 with the content of polyunsaturated fatty acids in extracellular vesicles from human milk.

Methods

Milk was obtained from a cohort of mothers (N = 70) at 2–4 weeks of lactation. SNPs in the FADS gene locus were determined using …

We have observed that as the theoretical landscape in behavioral science continues to proliferate and diversify, it is not uncommon that distance between subspecialties emerge, artificially accentuating differences between camps. This may occur at the risk of losing sight of the common ground, and worsening communication between subspecialties. In this paper, we propose conceptual “crosswalking” as a means to preserve interdisciplinary communication within the behavioral sciences. This is illustrated using Bronfenbrenner's Person-Process-Context-Time (PPCT) Model, mapped onto therapeutic modalities with a focus on ecological systems. Recommendations for future crosswalking are made.

Secrets, Soviets, And Sverdlovsk: Critiques Of The Biological Weapons Convention And Biosecurity In The 1970s And 1980s, Morgan Kelley

Departmental Honors & Graduate Capstone Projects

The Biological Weapons Convention, initially ratified in 1975, banned the production and stockpiling of biological weapons; however, it has faced considerable modern criticism for being unenforceable and not strong enough to ensure states' compliance. These modern critiques are based on the knowledge that the Soviet Union was in violation of the Convention, which was not confirmed until 1989. By analyzing the reactions to the Biological Weapons Convention by scholars and scientists, American intelligence officials, and American news media, it becomes clear that concerns about the Convention did exist prior to 1989, even when for many it was not certain that …

Faculty, Staff and Student Publications

Appendiceal adenocarcinoma (AA) is unique from other gastrointestinal malignancies in that it almost exclusively metastasizes to the peritoneal cavity. However, few studies have investigated the molecular interaction of the peritoneal microenvironment and AA. Here, we use a multi-omics approach with orthotopic and flank-implanted patient-derived xenografts (PDX) to study the effect of the peritoneal microenvironment on AA. AA tumors implanted in the peritoneal microenvironment tended to grow faster and displayed greater nuclear expression of Ki-67 relative to the same tumors implanted in the flank. Comparing the tumor-specific transcriptome (excluding stromal transcription), the peritoneal microenvironment relatively upregulated genes related to proliferation, including …

Metastatic disease remains the leading cause of death due to cancer, yet the mechanism(s) of metastasis and its timely detection remain to be elucidated. Neutrophil elastase (NE), a serine protease secreted by neutrophils, is a crucial mediator of chronic inflammation and tumor progression. In this study, we used the PyMT model (NE+/+ and NE-/-) of breast cancer to interrogate the tumor-intrinsic and -extrinsic mechanisms by which NE can promote metastasis. Our results showed that genetic ablation of NE significantly reduced lung metastasis and improved metastasis-free survival. RNA-sequencing analysis of primary tumors indicated differential regulation of tumor-intrinsic actin cytoskeleton signaling pathways …

Current cancer vaccines using T cell epitopes activate antitumor T cell immunity through dendritic cell/macrophage-mediated antigen presentation, but they lack the ability to promote B/CD4 T cell crosstalk, limiting their anticancer efficacy. We developed antigen-clustered nanovaccine (ACNVax) to achieve long-term tumor remission by promoting B/CD4 T cell crosstalk. The topographic features of ACNVax were achieved using an iron nanoparticle core attached with an optimal number of gold nanoparticles, where the clusters of HER2 B/CD4 T cell epitopes were conjugated on the gold surface with an optimal intercluster distance of 5-10 nm. ACNVax effectively trafficked to lymph nodes and cross-linked with …
