The global climate crisis represents the most urgent problem facing the planet, impacting social, cultural, political, economic, and environmental dimensions of life. The agr quorum-sensing system links Staphylococcus aureus metabolism to virulence, in part by increasing bacterial survival during exposure to lethal concentrations of H2O2, a crucial host defense against S. aureus. We now report that protection by agr surprisingly extends beyond post-exponential growth to the exit from stationary phase when the agr system is no longer turned on. Thus, agr can be considered a constitutive protective factor. Deletion of agr resulted in decreased ATP levels and growth, despite increased rates of respiration or fermentation at appropriate oxygen tensions, suggesting that Δagr cells undergo a shift towards a hyperactive metabolic …

Computational models and simulation are important tools to investigate the mechanisms underlying traumatic brain injury (TBI); their accuracy depends strongly on accurate model parameters, including the mechanical properties of the brain and its interactions with the skull. However, most existing TBI modeling studies are performed using material properties derived from animal tissue obtained after death. Magnetic resonance elastography (MRE) provides a non-invasive method for estimating the mechanical properties of soft tissue. The prevalence of wasting under five in Indonesia in 2022 is 7.7%; according to WHO, this wasting problem is a serious public health problem. Wasting is a nutritional problem in toddlers that has an impact on morbidity and mortality. The research objective was to determine the factors associated with wasting in toddlers aged 6–59 months in Cimpaeun Village in 2023. Objective: To achieve rapid, quantitative and high-throughput detection of aminoglycoside antibiotic residues in pork. Methods: Square-array aligned SERS substrates based on Au nanoflowers (AuNFs) were prepared using PP synthetic paper as a substrate. Pork samples were pre-treated by QuEChERs method and subjected to SRES. Results: Using 4-mercaptobenzoic acid as the SERS probe molecule, the substrate exhibited good homogeneity, SERS enhancement effect, reproducibility and stability. the SERS signal intensities at the characteristic peaks at 475, 619 cm-1 showed good linear relationships with the logarithms of the concentrations of gentamicin sulphate and neomycin sulphate, respectively (R2 of 0.991 6 …

Fatty acid synthesis has been extensively investigated as a therapeutic target in cancers, including colorectal cancer (CRC). Fatty acid synthase (FASN), a key enzyme of de novo lipid synthesis, is significantly upregulated in CRC, and therapeutic approaches of targeting this enzyme are currently being tested in multiple clinical trials. However, the mechanisms behind the pro- oncogenic action of FASN are still not completely understood. Here, for the first time, we show that overexpression of FASN increases the expression of glutamine–fructose-6-phosphate transaminase 1 (GFPT1) and O-linked N-acetylglucosamine transferase (OGT), enzymes involved in hexosamine metabolism, and the level of O-GlcNAcylation in vitro …

This research explores the evolving perceptions of English teachers regarding the integration of Technological Pedagogical Content Knowledge (TPACK) and its application in English Language Teaching (ELT) in the post-Covid-19 educational landscape. Using a mixed methods approach, this research draws from educators' experiences in innovatively incorporating TPACK to increase student engagement, facilitate personalized learning experiences, and improve communication. A total of seventeen English teachers in different schools in North Sumatra, Medan. Findings highlight the influence of TPACK on pedagogical strategies in response to the dynamic post-pandemic educational landscape.

Jurnal Pendidikan dan Pembelajaran

This study analyzes the impact of exciting and interactive learning message design on the learning motivation of equality package B students at the Pontianak SKB SPNF. Using a qualitative approach to case studies, data is collected through interviews, observations, and document analysis. The results showed that conventional learning methods were less effective, while students wanted more dynamic learning. Observations identify the limitations of learning media use. Document analysis highlights the need for a learning design focusing more on student characteristics. Implications and recommendations are discussed to improve student learning quality and motivation.

Background: The prediabetic stage exhibits common symptoms; hence, the risk of developing diabetes can be reduced by identifying these symptoms in the most affected population. This study aimed to examine the prevalence of the main symptoms of insulin resistance in healthy adults aged 18 years and above.

Methods: A cross-sectional study was conducted on 1,514 Saudi adults using an online questionnaire collected from 13th November 2020 to 23rd March 2021. Following parturition, the uterus must undergo significant changes in size and structure through a process called postpartum remodeling. This process is known to have both pro- and anti- inflammatory components. However, how various immune cells contribute to remodeling and the pathways that promote inflammation are not well understood. We hypothesize that immune cells that infiltrate the uterus postpartum modulate inflammation to promote postpartum remodeling. Our preliminary data suggests that inflammatory cell death may contribute to the inflammation present in the early postpartum period, and thus further studies will analyze caspase-1, gasdermin D, and IL-1β and IL-18. Background: The cavernous nerve (CN) is frequently damaged in prostatectomy and diabetic patients with erectile dysfunction (ED), initiating changes in penile morphology including an acute and intense phase of apoptosis in penile smooth muscle and increased collagen, which alter penile architecture and make corpora cavernosa smooth muscle less able to relax in response to neurotransmitters, resulting in ED.

Enteropathogenic E. coli (EPEC) are enteric pathogens that are non-invasive, lack Shiga toxin, and use a type 3 secretion system encoded on the locus of enterocyte effacement (LEE) pathogenicity island to translocate bacterial effector proteins into host intestinal epithelial cells leading to diarrhea. Atypical EPEC (aEPEC), in contrast to typical EPEC (tEPEC), lack bundle-forming pili, likely altering adherence and pathogenicity. Detection of aEPEC with co-infecting pathogens and in some asymptomatic individuals leads to questions regarding aEPEC virulence, especially in adults. Polyamines are a class of metabolites consisting of small carbon chains that contain amine groups and are positively charged at cellular pH. They play many important roles within the cell and are necessary for cell growth, membrane function, transcription, and translation. They are also required by a myriad of viruses for a productive infection. This includes the non-polio enterovirus Coxsackievirus B3 (CVB3). CVB3 is a cardiotropic virus that can readily infect the heart and cause irreversible damage which can eventually lead to dilated cardiomyopathy and the need for a heart transplant. The project focused on the production of bio-fertilizer from fruit and garden waste, with a specific emphasis on utilizing pineapple tops in the composting process. The objective was to produce and assess the properties of the bio fertilizer obtained from this process and evaluate its potential application in agriculture. In this study, the composting process was done in a 0.7 m long, 0.85 wide and 1.1 m high vessel, and 0.238 m3 pineapple top wastes, resulting in 0.35 m3 of biofertilizer. This paper describes the encounters that Italian travellers, explorers, and traders had with the peoples of the Malay world at the turn of the century. In particular, it focuses on the linguistic descriptions and observations made by Italian explorers of the languages spoken in the places they visited and included in their travel writings. This article explores cross-cultural encounters and identities discourses in selected Polish and Russian travelogues about the Netherlands East Indies. Poles and Russians could travel to the Netherlands East Indies thanks to advantages afforded Europeans by the colonial system. Their occupations (for example, a privileged tourist, colonial scientist, diplomat) often made them suitable imperial agents. They defined themselves as Europeans but, as Eastern Europeans, they occupied an ambiguous position: Russians came from a land-based, economically backward “empire of the periphery“ (Boris Kagarlitsky 2008); Poles came from a semi-peripheral European nation subjected to foreign rule and, from their common experience of subjugation, …

The goal of this study is to analyse the travelogues of Protestant evangelists in Wonogiri, a topic so far barely touched upon. Despite being a predominantly Muslim community, Wonogiri was one of the areas in the principalities of Surakarta targeted by the zending. Therefore, this study explores various aspects, including the purpose behind the visits of Protestant missionaries, the discourses presented in the travelogues, and the perspectives of these evangelists on the belief system prevalent in the community in the early twentieth century. In the first year after World War II, there was a power vacuum in Indonesia. The Indonesians had declared their independence. The Allies had assigned the administration of the former Dutch colony to the British. The Dutch thought they could continue their colonial ambitions. It was a year of utter chaos and extreme violence. While most Dutch journalists remained in Jakarta, three went to the war zones: two of them as reporters and the other as a soldier. The quality of water used for the irrigation of leafy vegetables in urban settings is in doubt due to contamination associated with poor hygiene and water pollution. This study examined the microbiological quality of irrigation water and leafy vegetables at Chang’ombe in Dar es Salaam, Tanzania as well as the perceptions and level of awareness of nearby consumers. Water and leafy vegetable samples were analysed for faecal coliform (FC) using standard techniques. Perception and awareness of consumers were collected using a questionnaire survey. The results showed an average FC count of 406.1 ± 16.40 MPN/100 ml for the irrigation water. …

Yam (Dioscorea spp.) is a tuber crop cultivated in over 30% of all regions in Tanzania, contributing to the carbohydrate needs of many households. The morphological and genetic diversity of water yam (D. alata L.) local cultivars collected from Pwani region and Unguja Island were evaluated using 26 agro-morphological characters and 10 Short Sequence Repeat (SSR) markers. Cluster analysis using morphological characters classified nine water yam local cultivars into two main clusters with three subclusters. Malaria is a significant public health concern, especially in tropical regions. It remains endemic in many areas where malaria mosquitoes are prevalent. This study investigates the viability of insecticide zooprophylaxis (IZ) in conjunction with Long Lasting Insecticidal Nets (LLINs) as a strategic approach to combat malaria transmission, particularly in regions dominated by zoophagic vectors. Through the presentation and analysis of a mathematical model that integrates IZ and LLINs for the control of Anopheles arabiensis (An. arabiensis), the research underscores IZ's role in reducing zoophagic vector populations and enhancing malaria control efforts. In recent years, the term “trauma” has become increasingly controversial. Many of these controversies revolve around the definition of trauma, and the way that this definition varies across contexts. What kinds of experience can plausibly be considered “traumatic”? In some contexts, the standards are incredibly permissive, while others are much more restrictive. These different ways of understanding “trauma” prompts an important question: Who has the authority to define trauma? In other words, who decides which conception of trauma is the most “legitimate” – who decides when an experience is or is not traumatic? My project attempts to answer these questions …
