Acetaminophen (APAP) is a leading cause of acute liver failure. The effect of APAP metabolite's effects in the periphery are well characterized; however, associated consequences in the brain remain poorly understood. Animal studies on this subject are few and reveal that frequent APAP intake can trigger cerebral abnormalities that vary depending on the subject's age. Alarmingly, experimental efforts have yet to examine associated consequences in elderly hosts, who correspond to the highest risk of medication overload, impaired drug clearance, and cognitive deficits. Brace root architecture is a critical determinant of maize's stalk anchorage and nutrition uptake, influencing root lodging resistance, stress tolerance, and plant growth. To identify the key microRNAs (miRNAs) in control of maize brace root growth, we performed small RNA sequencing using brace root samples at emergence and growth stages. We focused on the genetic modulation of brace root development in maize through manipulation of miR390 and its downstream regulated (). Reconceiving Virtue: A Mengzian Adaptation Of Eudaimonic Virtue Ethics In Response To Contemporary Criticisms, Gina Elizabeth Lebkuecher

The primary question my dissertation aims to answer is: how might eudaimonic virtue ethics be reimagined to respond to contemporary criticisms from disability scholars, feminists, and empirical psychology? To answer this, I introduce the Eudaimonic View of Virtue, or EV, and propose a Mengzian adaptation of the EV (EV-M) in response to these criticisms. The EV captures the four core claims to which eudaimonic virtue ethical theories are committed: (i) virtues, in the sense of excellent character traits or dispositions, are the foundation of ethics or ethical action (i.e., aretaicism); (ii) virtue is at least partially constitutive of human well-being/flourishing …

Despite sharing largely similar genomes, females and males of the same species exhibit morphological, physiological, and behavioral differences. Sex-role reversal stems from a unique case of sexual selection in which females compete for mates and males conduct the majority of parental duties. We examined the extent to which aggression and parenting may be mediated by different neurogenomic mechanisms by comparing females versus males in the northern jacana (Jacana spinosa), a sex-role reversed shorebird. Glioblastoma, the most common primary brain tumor, has a 6.8% survival rate 5 years post diagnosis. Our team developed an oncolytic adenovirus with an OX-40L expression cassette named Delta-24-RGDOX. While studies have revealed the interaction between the gut microbiota and immunotherapy agents, there are no studies linking the gut microbiota with viroimmunotherapy efficacy. We hypothesize that gut bacterial signatures will be associated with oncolytic viral therapy efficacy. An array of sauropod taxa coexisted in Late Jurassic North America. How so many giant genera coexisted without competing for food is puzzling considering their potential metabolic requirements. One aspect of this question is neck posture, which has been much contested, both in terms of the intrinsic curvature of the neck and the resultant elevation of the head. Psoriasis is an immune-mediated skin disease associated with neurogenic inflammation, but the underlying molecular mechanism remains unclear. We demonstrate here that acid-sensing ion channel 3 (ASIC3) exacerbates psoriatic inflammation through a sensory neurogenic pathway. Global or nociceptor-specific Asic3 knockout (KO) in female mice alleviates imiquimod-induced psoriatic acanthosis and type 17 inflammation to the same extent as nociceptor ablation. However, ASIC3 is dispensable for IL-23-induced psoriatic inflammation that bypasses the need for nociceptors. Mechanistically, ASIC3 activation induces the activity-dependent release of calcitonin gene-related peptide (CGRP) from sensory neurons to promote neurogenic inflammation. In the decimated and silent post-apocalyptic setting of Cormac McCarthy's The Road, the main characters spend a significant amount of time listening to silence, listening for signs of danger, but also listening for the voice of God. Many have discussed the hopeless nihilism juxtaposed with the heavy use of Christian iconography in the novel classifying it within one of these dichotomies or somewhere on the spectrum in between. Cytokine-Supplemented Maturation Medium Enhances Cytoplasmic And Nuclear Maturation In Bovine Oocytes, Renata Blocher, Ying Liu, Tayler Patrick, Irina A. Polejaeva

Bovine in vitro oocyte maturation (IVM) is an easy way to obtain oocytes for subsequent assisted reproductive techniques but is inefficient compared to in vivo maturation. Supplementation of three cytokines, fibroblast growth factor 2 (FGF2), leukemia inhibitory factor (LIF), and insulin-like growth factor 1 (IGF1), or FLI, has increased oocyte maturation and embryo development in multiple species, but studies have not explored the oocyte differences caused by FLI IVM supplementation. This study aimed to assess important nuclear and cytoplasmic maturation events in high-quality oocytes. FLI-supplemented oocytes had a decreased GV (3.0% vs. 13.7%, p < 0.01) and increased telophase I incidence (34.6% vs. 17.6%, p < 0.05) after IVM, increased normal meiotic spindles (68.8% vs. 50.0%, p < 0.001), and an increased nuclear maturation rate (75.1% vs. 66.8%, p < 0.001). Moreover, in metaphase II oocytes, the percentage of FLI-treated oocytes with a diffuse mitochondrial distribution was higher (87.7% vs. 77.5%, p < 0.05) and with a cortical mitochondrial distribution was lower (11.6% vs. 17.4%, p < 0.05). Additionally, FLI-supplemented oocytes had more pattern I cortical granules (21.3% vs. 14.4%, p < 0.05). These data suggest that FLI supplementation in bovine in vitro maturation medium coordinates nuclear and cytoplasmic maturation to produce higher-quality oocytes.

Individuals with uncontrolled diabetes are highly susceptible to tuberculosis (TB) caused by Mycobacterium tuberculosis (M. tb) infection. Novel treatments for TB are needed to address the increased antibiotic resistance and hepatoxicity. Previous studies showed that the administration of liposomal glutathione (L-GSH) can mitigate oxidative stress, bolster a granulomatous response, and diminish the M. tb burden in the lungs of M. tb-infected mice. Nonetheless, the impact of combining L-GSH with conventional TB treatment (RIF) on the cytokine levels and granuloma formation in the livers of diabetic mice remains unexplored. Navigating Undergraduate Medical Education: The Impact Of Enhanced Mentorship Pairing At A New Medical School, Shawn Izadi

Introduction

Transitioning into medical school is challenging, particularly in the first year, with a notable support gap. This study aimed to evaluate a mentorship program at a new medical school.

Methods

Initiated in 2017 at the University of Texas Rio Grande Valley School of Medicine, the mentorship program had two iterations: initial random pairings and subsequent formative pairings based on matching criteria. A mixed-methods approach assessed its effectiveness in supporting first-year students.

Results

Of 109 first-year students, 76% completed a 6-month survey. Both classes primarily had male mentees with varied interests in primary or specialty care. No significant demographic differences …

This study examined the invertebrate fauna from three deep wells located at the Uvalde National Fish Hatchery in south-central Texas. These wells provide a water source from geological strata of the southern boundary of the Edwards Aquifer ranging in age from Lower Cretaceous to Recent. Forty-seven collections, sampled on a continuous basis from November 2020 to January 2022, produced a total of 738 specimens comprising 15 different species. These collections reveal a diverse crustacean assemblage along with dominant numbers of the stygobiotic snail Balconorbis uvaldensis. With increased fire activity, the presence of fire refugia is critical to support forest resiliency and mitigate large-scale vegetation conversion, however, the persistence of fire refugia within burn perimeters over time is uncertain. This study examines the dynamic nature of fire refugia by mapping the decline in forest cover over time, identifying spatiotemporal patch- and landscape-scale patterns and the biophysical drivers of delayed mortality following the 2020 Labor Day Fires occurring in Oregon’s western Cascades. Chimeric antigen receptor (CAR)-redirected immune cells hold significant therapeutic potential for oncology, autoimmune diseases, transplant medicine, and infections. All approved CAR-T therapies rely on personalized manufacturing using undirected viral gene transfer, which results in nonphysiological regulation of CAR-signaling and limits their accessibility due to logistical challenges, high costs and biosafety requirements. Random gene transfer modalities pose a risk of malignant transformation by insertional mutagenesis. Here, we propose a novel approach utilizing CRISPR-Cas gene editing to redirect T cells and natural killer (NK) cells with CARs. Monitoring trends in wildlife communities is integral to making informed land management decisions and applying conservation strategies. Birds inhabit most niches in every environment and because of this they are widely accepted as an indicator species for environmental health. Traditionally, point counts are the common method to survey bird populations, however, passive acoustic monitoring approaches using autonomous recording units have been shown to be cost-effective alternatives to point count surveys. Using 1.5 mm inner diameter (i.d.) columns to bridge the gap between routinely used 2.1 mm i.d. columns and capillary bore columns allows for a sequential but significant increase in performance without the need for specialized equipment associated with very low dispersion liquid chromatography (LC) systems. These 1.5 mm i.d. columns balance an increase in sensitivity compared to 2.1 mm i.d. columns (theoretically doubling the time-domain peak area in mass sensitive detectors for the same mass load), while mitigating the efficiency losses due to extra-column dispersion effects that are commonly observed with 1.0 mm i.d. columns. Preciado into conversations with contemporary philosophies of religion and interrogates a hegemonic Christian theological political matrix. The mitigation of deleterious arcing and sparking onboard spacecraft depends upon a thorough understanding of the electrical properties of the materials involved. This includes not only understanding steady-state parameters such as dark conductivity, but responses to the space environment as well. For example, radiation-induced conductivity drastically changes the conductivity of a material due to the presence of radiation. This technology uses artificial intelligence to create digital replicas with hyperreal accuracy.
