Still Searching For (Education For) Sustainable Development – Reflections On The Need, Challenges, And Chances Of Transforming Education In The 21st Century, Lars Keller
Jurnal Pendidikan Geografi: Kajian, Teori, dan Praktek dalam Bidang Pendidikan dan Ilmu Geografi
Scientific evidence has been put forward over and over again in the past decades that, during the Anthropocene, human impact on the various spheres of the Earth has grown to an extent that actually threatens the overall resilience of the Earth system and thus the basis for the existence of future societies. New concepts to tackle the problems ahead and to regain control about positive future development are urgently needed. Education – or rather the transformation of education – and the possible role of education as a key instrument to reach the Sustainable Development Goals could play a prominent role …
The Discursive Representations Of Africanness And Blackness In Chinese Cyberspace : A Critical Discourse-Historical Analysis, Jiapei Gu
Lingnan Theses (MPhil & PhD)
Recent years have seen deepened Sino-African ties. However, unlike friendly rhetoric at the state level, racial issues at the individual level are becoming increasingly salient. This study aims to enrich our current understanding of racial issues in China by examining the digital discursive representations of Africanness and Blackness produced by Chinese Weibo users. It investigates three African/Black people-related racial occurrences in China from 2020 to 2021. Those include the release of the draft of the Permanent Residence Law for Foreigners in 2020, the Okonkwonwoye case, where Okonkwonwoye physically assaulted a nurse during quarantine in 2020, and the Abdulmatten murder case …

Understanding and treating human diseases require valid animal models. Leveraging the genetic diversity in rhesus macaque populations across eight primate centers in the United States, we conduct targeted-sequencing on 1845 individuals for 374 genes linked to inherited human retinal and neurodevelopmental diseases. We identify over 47,000 single nucleotide variants, a substantial proportion of which are shared with human populations. By combining rhesus and human allele frequencies with established variant prediction methods, we develop a machine learning-based score that outperforms established methods in predicting missense variant pathogenicity. Remarkably, we find a marked number of loss-of-function variants and putative deleterious variants, which …
On The Intellectual Horizons Of Social Realism: A Response To Barton, Jim Hordern
On The Intellectual Horizons Of Social Realism: A Response To Barton, Jim Hordern
School of Law, Humanities and Social Sciences
This paper comments on the arguments made in ‘knowledge without disciplines: a critique of social realism’s disciplinary fixation’, authored by Keith Barton. It is suggested that Barton has not fully grasped how social realism views disciplines and curricula and that this reflects a wider tendency amongst commentators to overlook some of the thinking underpinning social realism. A different, more generous interpretation of social realism is outlined in respect of disciplinarity, the constitution of knowledge, the relationship between disciplines and subjects and the socio-epistemic formation that make knowledgeability possible. Social realism is seen as having already made a substantive impact, with …

Executive Summary Of The American Radium Society Appropriate Use Criteria For Brain Metastases In Epidermal Growth Factor Receptor Mutated-Mutated And Alk-Fusion Non-Small Cell Lung Cancer, Manmeet Ahluwalia
All Publications
The American Radium Society (ARS) Central Nervous System (CNS) committee reviewed literature on epidermal growth factor receptor mutated (EGFRm) and ALK-fusion (ALK+) tyrosine kinase inhibitors (TKIs) for the treatment of brain metastases (BrMs) from non-small cell lung cancers (NSCLC) to generate appropriate use guidelines addressing use of TKIs in conjunction with or in lieu of radiotherapy (RT). The panel developed three key questions to guide systematic review: can radiotherapy be deferred in patients receiving EGFR or ALK TKIs at (1) diagnosis or (2) recurrence? Should TKI be administered concurrently with RT (3)? Two literature searches were performed (May 2019 and …

Refugee women face a myriad of barriers to health after resettlement, including access to family planning services. They often have little knowledge of available family planning or contraception options, or where to obtain them after resettlement. As a result, refugee women in Tennessee suffer from high rates of unintended pregnancies, stemming from limited access to family planning services, and unmet family planning needs after resettlement. This dissertation combines the disciplines of history, public health, and public administration/policy to better understand why refugee women in Tennessee have had limited access to family planning services. To achieve this, it was necessary to …
Transcriptional Precision In Photoreceptor Development And Diseases - Lessons From The Retinal Transcription Factor Crx, Yiqiao Zheng
Transcriptional Precision In Photoreceptor Development And Diseases - Lessons From The Retinal Transcription Factor Crx, Yiqiao Zheng
Arts & Sciences Graduate Student Theses and Dissertations
Precise gene expression programs are foundational to organism development and homeostasis. Transcription factors play a central role in establishing and maintaining these programs. Large-scale transcriptomic studies have generated parts lists of transcription factors. However, these descriptive works rarely formulate a cohesive model of how these transcription factors work together to support a functional organism. Mutations that affect transcription factor functions can lead to a broad range of human diseases. Therefore, it is crucial to understand the principles that transcription factors achieve spatial, temporal, and quantitative precision in their regulated gene expression programs. I have investigated transcriptional regulation in rod and …

Opioid pain medications, such as morphine, remain the mainstay for treating severe and chronic pain. Prolonged morphine use, however, triggers analgesic tolerance and hyperalgesia (OIH), which can last for a long period after morphine withdrawal. How morphine induces these detrimental side effects remains unclear. Here, we show that morphine tolerance and OIH are mediated by Tiam1-coordinated synaptic structural and functional plasticity in the spinal nociceptive network. Tiam1 is a Rac1 GTPase guanine nucleotide exchange factor that promotes excitatory synaptogenesis by modulating actin cytoskeletal dynamics. We found that prolonged morphine treatment activated Tiam1 in the spinal dorsal horn and Tiam1 ablation …

CRISPR/Cas9 gene editing represents an exciting avenue to study genes of unknown function and can be combined with genetically encoded tools such as fluorescent proteins, channelrhodopsins, DREADDs, and various biosensors to more deeply probe the function of these genes in different cell types. However, current strategies to also manipulate or visualize edited cells are challenging due to the large size of Cas9 proteins and the limited packaging capacity of adeno-associated viruses (AAVs). To overcome these constraints, we developed an alternative gene editing strategy using a single AAV vector and mouse lines that express Cre-dependent Cas9 to achieve efficient cell-type specific …

This mixed methods paper reports findings from three studies examining the overlap between autism and hobbyist board gaming. The first was a quantitative survey of over 1600 board gamers, showing that autistic individuals are overrepresented in this hobby compared to the general population and that autistic traits measured by the AQ are significantly elevated amongst board gamers. Study 1 also assessed gamers’ motivations and preferences and reported key differences as well as similarities between autistic and non-autistic gamers. The second was a qualitative study that reported the results of 13 interviews with autistic individuals who are hobbyist board gamers. Using …
Regional And Ethnic Inequality In Burkina Faso, Michael J. Kevane
Regional And Ethnic Inequality In Burkina Faso, Michael J. Kevane
Economics
This chapter summarizes data on regional and ethnic inequality in Burkina Faso. The Sahel and Est regions appear to be the worst-off, according to a variety of indicators. There is less data and research on ethnic inequality, but it is likely that the Peul and Gourma ethnic groups (residing predominantly in the Sahel and Est regions) are also worse off. There has been little systematic analysis of how government investment in public goods have affected regional and ethnic inequality. Some of the important drivers of inequality in Burkina Faso are likely similar to those identified for other African countries. These …

Human leukocyte antigens (HLAs) are polymorphic glycoproteins expressed on the cell surface of nucleated cells and consist of two classes, HLA class I and HLA class II. In contrast, in mice, these molecules, known as H-2, are expressed on both nucleated cells and erythrocytes. HLA-I molecules (Face-1) are heterodimers consisting of a polypeptide heavy chain (HC) and a light chain, B2-microglobulin (B2m). The heterodimers bind to antigenic peptides and present them to the T-cell receptors of CD8+ cytotoxic T lymphocytes. The HCs can also independently emerge on the cell surface as B2m-free HC monomers without peptides (Face-2). Early investigators suggested …

BACKGROUND: OX40 has been widely studied as a target for immunotherapy with agonist antibodies taken forward into clinical trials for cancer where they are yet to show substantial efficacy. Here, we investigated potential mechanisms of action of anti-mouse (m) OX40 and anti-human (h) OX40 antibodies, including a clinically relevant monoclonal antibody (mAb) (GSK3174998) and evaluated how isotype can alter those mechanisms with the aim to develop improved antibodies for use in rational combination treatments for cancer.
METHODS: Anti-mOX40 and anti-hOX40 mAbs were evaluated in a number of in vivo models, including an OT-I adoptive transfer immunization model in hOX40 knock-in …

T-BOX factors belong to an evolutionarily conserved family of transcription factors. T-BOX factors not only play key roles in growth and development but are also involved in immunity, cancer initiation, and progression. Moreover, the same T-BOX molecule exhibits different or even opposite effects in various developmental processes and tumor microenvironments. Understanding the multiple roles of context-dependent T-BOX factors in malignancies is vital for uncovering the potential of T-BOX-targeted cancer therapy. We summarize the physiological roles of T-BOX factors in different developmental processes and their pathological roles observed when their expression is dysregulated. We also discuss their regulatory roles in tumor …

Gastric cancer is an aggressive and multifactorial disease. Helicobacter pylori (H. pylori) is identified as a significant etiological factor in gastric cancer. Although only a fraction of patients infected with H. pylori progresses to gastric cancer, bacterial infection is critical in the pathology and development of this malignancy. The pathogenic mechanisms of this bacterium involve the disruption of the gastric epithelial barrier and the induction of chronic inflammation, oxidative stress, angiogenesis and metastasis. Adherence molecules, virulence (CagA and VacA) and colonization (urease) factors are important in its pathogenicity. On the other hand, resveratrol is a natural polyphenol with …

Protein SUMOylation is a prevalent stress-response posttranslational modification crucial for maintaining cellular homeostasis. Herein, we report that protein SUMOylation modulates cellular signaling mediated by cAMP, an ancient and universal stress-response second messenger. We identify K561 as a primary SUMOylation site in exchange protein directly activated by cAMP (EPAC1) via site-specific mapping of SUMOylation using mass spectrometry. Sequence and site-directed mutagenesis analyses reveal that a functional SUMO-interacting motif in EPAC1 is required for the binding of SUMO-conjugating enzyme UBC9, formation of EPAC1 nuclear condensate, and EPAC1 cellular SUMOylation. Heat shock-induced SUMO modification of EPAC1 promotes Rap1/2 activation in a cAMP-independent manner. …

Optimizing Associative Memory In Autistic And Non-Autistic Adults: Retrieval Practice, Post-Retrieval Feedback, And Gesture, Brianna E. Cairney
LSU Doctoral Dissertations
Associative memory—or the ability to form a memory representation linking two previously unrelated concepts—is a critical aspect of learning. This work provides insight into strategies that can be used to support associative memory, their underlying mechanisms, and their effectiveness in autistic and non-autistic learners. Chapter 1 demonstrates that iconic gestures can be used as a tool to enhance associative memory by increasing word concreteness and imageability. This effect was eliminated when the iconic gestures were interspersed with ambiguous nonsense gestures, suggesting a boundary on these benefits that depends on listener expectations regarding gesture clarity. However, it remains unknown whether gestures …

BACKGROUND: T cells play a central role in the antitumor response. However, they often face numerous hurdles in the tumor microenvironment, including the scarcity of available essential metabolites such as glucose and amino acids. Moreover, cancer cells can monopolize these resources to thrive and proliferate by upregulating metabolite transporters and maintaining a high metabolic rate, thereby outcompeting T cells.
METHODS: Herein, we sought to improve T-cell antitumor function in the tumor vicinity by enhancing their glycolytic capacity to better compete with tumor cells. To achieve this, we engineered human T cells to express a key glycolysis enzyme, phosphofructokinase, in conjunction …

Some herbal extracts contain relatively high amounts of lipopolysaccharide (LPS). Because orally administered LPS activates innate immunity without inducing inflammation, it plays a role as an active ingredient in herbal extracts. However, the LPS content in herbal extracts remains extensively unevaluated. This study aimed to create a database of LPS content in herbal extracts; therefore, the LPS content of 414 herbal extracts was measured and the macrophage activation potential was evaluated. The LPS content of these hot water extracts was determined using the kinetic-turbidimetric method. The LPS concentration ranged from a few ng/g to hundreds of μg/g (Standard Escherichia coli …

Ghost fishing is a global issue that can be addressed using fishing gear materials that do not persist in the marine environment. However, for these alternatives to be widely adopted, they must meet the same mechanical specifications as current commercial materials while degrading without any negative impact. The objective of this study was to compare a conventional gillnet made of polyamide 6 (PA6) with an alternative made of poly(butylene succinate-co-adipate-co-terephthalate) (PBSAT) at three different scales: monofilament, knot, and net. While the PBSAT monofilament's strength was half that of the conventional PA6 net, knot and net losses were even more significant. …

Mitogen-activated protein kinase kinase kinase 4 (MAP3K4) promotes fetal and placental growth by regulating growth-promoting pathways. Mice lacking MAP3K4 kinase activity exhibit placental insufficiency and fetal growth restriction. Upstream pathways activating MAP3K4 may represent key mechanisms to induce growth. We have previously shown that the MAP3K4 binding partner tumor necrosis factor receptor-associated factor 4 (TRAF4) promotes MAP3K4 activity in the embryo. However, the role of TRAF4 and its association with MAP3K4 in the placenta is unknown. Our analysis of single-cell RNA-Seq data showed that Traf4 is one of the most abundant Trafs in the mouse placenta and is coexpressed with …

Mutations that decrease or increase the activity of the tyrosine phosphatase, SHP2 (encoded by PTPN11), promotes developmental disorders and several malignancies by varying phosphatase activity. We uncovered that SHP2 is a distinct class of an epigenetic enzyme; upon phosphorylation by the kinase ACK1/TNK2, pSHP2 was escorted by androgen receptor (AR) to chromatin, erasing hitherto unidentified pY54-H3 (phosphorylation of histones H3 at Tyr54) epigenetic marks to trigger a transcriptional program of AR. Noonan Syndrome with Multiple Lentigines (NSML) patients, SHP2 knock-in mice, and ACK1 knockout mice presented dramatic increase in pY54-H3, leading to loss of AR transcriptome. In contrast, prostate tumors …

Automated image acquisition can significantly improve the throughput of serial section scanning electron microscopy (ssSEM). However, image quality can vary from image to image depending on autofocusing and beam stigmation. Automatically evaluating the quality of images is, therefore, important for efficiently generating high-quality serial section scanning electron microscopy (ssSEM) datasets. We tested several convolutional neural networks for their ability to reproduce user-generated evaluations of ssSEM image quality. We found that a modification of ResNet-50 that we term quality evaluation Network (QEN) reliably predicts user-generated quality scores. Running QEN in parallel to ssSEM image acquisition therefore allows users to quickly identify …

Over one third of sharks, rays and chimaeras (chondrichthyans) are threatened with extinction globally. In the Mediterranean Sea, more than half of chondrichthyan species face this risk, although a variety of international, regional, and national rules and regulations apply directly and indirectly targeting management and conservation for these species. In this work, we provide an overview of relevant legislation and policies in the region, which regulate, inter alia, commercial fisheries, while highlighting through cases studies how implementation in practice at national level looks like. Horizontal gaps and inconsistencies that hinder chondrichthyan management in the region are also illustrated. Furthermore, we …

Essays in Education
This article begins by examining the adverse effects of ostracization on students' mental health and academic performance. It identifies deficit thinking within educational institutions as contributing to the social exclusion experienced by students, leading to their psychological need for belonging going unmet. Deficit thinking, fuelled by racist and classist ideologies, theorizes that the blame for a student’s failure in school results from their inherent deficiencies. Deficit thinking oppresses and marginalizes students by justifying harmful practices such as school segregation and scientific racism. Schools serving communities of color are disproportionately impacted, compromising students’ educational achievement and prospects. To disrupt deficit thinking, …
The Vanishing Sound Grammar: Agamben's Critique Of Sound Politics, Jian Guo
The Vanishing Sound Grammar: Agamben's Critique Of Sound Politics, Jian Guo
Theoretical Studies in Literature and Art
The construction of sound theory is still in its initial stage, requiring exploration of existing intellectual resources. Agamben's insights into sound not only inform sound studies theoretically but also offer a new direction in connecting his early linguistic philosophy with later political philosophy. Agamben's awareness of the problematics lies in the fact that, despite Derrida's deconstruction of the present illusion of sound, a more covert transcendental illusion, the Voice, persists. The Voice, as sound and metaphysical grammar, resolves philosophical binary opposition through transformation. Nevertheless, achieving such unification requires that the Voice, endowed with an embedded function of “dividing and connecting,” …
