An Advanced Way To Ensure Data Quality Of Retrieval Augmented Generation (Rag) Pipelines, Kautuk Pandey

Defensive Publications Series

The present disclosure provides a method of an automated way to calculate and evaluate quality factor of metadata provided to Retrieval Augmented Generation (RAG) pipelines for ensuring data quality of RAG pipelines used in LLM apps for Generative Artificial Intelligence (AI) application. The method includes capturing metadata of vectors/ embeddings while receiving vectors/ embeddings, applying Anomaly Detection models to generate threshold for expected metadata, applying quality score based filtering based on metadata on the Approximate Nearest Neighbour (ANN) search while generating output response in response to user query, providing output response based on the filtered vectors

Efficacy And Mechanism Of Action Of Cd40 Ligand And Interleukin-12 Plasmid Electroporation Intratumoral Immunotherapy, Gregory W. Ho Sep 2024

Efficacy And Mechanism Of Action Of Cd40 Ligand And Interleukin-12 Plasmid Electroporation Intratumoral Immunotherapy, Gregory W. Ho

Dartmouth College Ph.D Dissertations

Cancer is a disease in which normal cells have become altered such that they divide uncontrollably, ultimately leading to tissue destruction. While efficacious cancer therapies have been developed in recent decades, many of them come with drawbacks such as cost and systemic toxicity. Additionally, solid tumors remain difficult to treat due to the immunosuppressive tumor microenvironment. Intratumoral immunotherapy (ITIT) strives to generate effective antitumor immunity by directly administering immunostimulatory adjuvants into tumors to reverse local tumor-mediated immune suppression and generate systemic immune responses against metastases. Intratumoral immunotherapy utilizing in vivo expression of interleukin-12 (IL-12) via plasmid electroporation has been investigated …

Life In The Margins: The Effect Of Immersion/Emersion And Tidal Cycle On The North Atlantic Limpet Patella Vulgata Protein Synthesis Rates, Ignacio Alvarez-Cienfuegos, Benjamin Ciotti, Richard Billington, Paul Sutton, Keiron Fraser Sep 2024

Life In The Margins: The Effect Of Immersion/Emersion And Tidal Cycle On The North Atlantic Limpet Patella Vulgata Protein Synthesis Rates, Ignacio Alvarez-Cienfuegos, Benjamin Ciotti, Richard Billington, Paul Sutton, Keiron Fraser

School of Biological and Marine Sciences

Biological processes in intertidal species follow tidal rhythms that enhance survival and fitness. Whereas tidal effects on behaviour and metabolic rates have been widely studied, impacts on other key process such as protein synthesis are still poorly understood. To date, no studies have examined the effect of immersion/emersion and tidal cycles on protein synthesis rates (k s). Patella vulgata is an intertidal limpet present in North-Eastern Atlantic rocky shores from high to low shore. Previously reported P. vulgata respiration and heart rate measurements suggest aerobic metabolism is maintained during emersion and growth rates increase from high to low shore, …

Myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML) belong to a continuous disease spectrum of myeloid malignancies with poor prognosis in the relapsed/refractory setting necessitating novel therapies. Natural killer (NK) cells from patients with myeloid malignancies display global dysfunction with impaired killing capacity, altered metabolism and exhausted phenotype at the single cell transcriptomic and proteomic levels. In this study we identified that this dysfunction was mediated through a crosstalk between NK cells and myeloid blasts necessitating cell-cell contact. NK cell dysfunction could be prevented by targeting the αvβ integrin/TGF-β/SMAD pathway but, once established, was persistent due to profound epigenetic reprogramming. …

Acute lower gastrointestinal GVHD (aLGI-GVHD) is a serious complication of allogeneic hematopoietic stem cell transplantation. Although the intestinal microbiota is associated with the incidence of aLGI-GVHD, how the intestinal microbiota impacts treatment responses in aLGI-GVHD has not been thoroughly studied. In a cohort of patients with aLGI-GVHD (n = 37), we found that non-response to standard therapy with corticosteroids was associated with prior treatment with carbapenem antibiotics and a disrupted fecal microbiome characterized by reduced abundances of Bacteroides ovatus. In a murine GVHD model aggravated by carbapenem antibiotics, introducing B. ovatus reduced GVHD severity and improved survival. These beneficial effects …
