OBJECTIVES: Heart failure (HF) is a global health concern with rising incidence and poor prognosis. While the essential role of nutritional and dietary strategies in HF patients is acknowledged in the existing scientific guidelines and clinical practice, there are no comprehensive nutritional recommendations for optimal dietary management of HF.
METHODS: In this review, we discuss results from recent studies on the obesity paradox and the effects of calorie restriction and weight loss, intermittent fasting, the Western diet, the Mediterranean diet, the ketogenic diet, and the DASH diet on HF progression.

Fairy Tales In Service Of Political Projects: “Hansel And Gretel” In Changing Political Regimes, Polona Tratnik
Marvels & Tales
The production of fairy tales depends on political interests. The tales’ regimes of production and anticipated reception condition their inscribed ideological inclinations and erasures. I examine how Kinder- und Hausmärchen by the brothers Grimm contributed to nation-building and, focusing on “Hansel and Gretel,” how the changing regimes of production required omissions in the translations of the Grimms’ tales into Slovenian. I argue that the sui generis appropriation of the tale titled “Janko and Metka” by the writer France Bevk helped build a socialist nation. Finally, the article comments on some issues in capitalist regimes of fairy-tale production.

Ganoderma lucidum is a mushroom forming fungus that has been used in traditional Chinese medicine for over 2,000 years. G. lucidum, also known as lingzhi or reishi, is consumed in several Asian cultures to promote health and longevity. Bioactive molecules with anticancer properties have been isolated and characterized primarily from the species Ganoderma lucidum, and the cytotoxic potential of other species in the genus Ganoderma remains relatively unclear. Bioactive molecules isolated from mushroom fruiting bodies are demonstrated to have cytotoxicity against different cancer cell lines. Endometriosis negatively impacts the health-related quality of life of 190 million women worldwide. Novel advances in nonhormonal treatments for this debilitating condition are desperately needed. Macrophages play a vital role in the pathophysi- ology of endometriosis and represent a promising therapeutic target. In the current study, we revealed the full transcriptomic complexity of endometriosis-associated macrophage subpopulations using single-cell analyses in a preclinical mouse model of experimental endometriosis. We have identified two key lesion-resident populations that resemble i) tumor-associated macrophages (characterized by expression of Folr2, Mrc1, Gas6, and Ccl8+) that promoted expression of Col1a1 and Tgfb1 in human endometrial stromal cells …

As we face multiple environmental crises (e.g. climate change, nutrient pollution, freshwater scarcity), there is often a disconnect between an entity’s activities and the pollution resulting from that activity. Footprint tools help address this disconnect. They are effective in educating people, institutions and communities on how their resource use results in environmental pollution and what we can do to moderate that pollution. These tools connect an entity’s activities with the associated pollution. Footprint tools are also constantly evolving. As we better understand how to best estimate emissions—especially from sources further removed like those from the supply chain—the guidance and methods …

Pancreatic cancer is associated with an oncogenic KRAS mutation in approximately 90% of cases. However, a non-negligible proportion of pancreatic cancer cases harbor wild-type KRAS (KRAS-WT). Metabolic dysfunction-associated steatotic liver disease (MASLD) prevalence is increasing annually and affects over a third of US adults. MASLD can progress to metabolic dysfunction-associated steatohepatitis (MASH), characterized by severe hepatocyte injury, inflammation, and eventual advanced fibrosis or cirrhosis. MASH is predicted to become the primary cause of liver transplant by 2030. Although the etiology of MASLD/MASH is incompletely understood, dysregulated fatty acid oxidation is implicated in disease pathogenesis. Mexican Americans are disproportionally affected by metabolic dysfunction-associated steatotic liver disease (MASLD), which often co-occurs with diabetes. Despite extensive evidence on the causative role of the gut microbiome in MASLD, studies determining the involvement of the gut phageome are scarce. In this cross-sectional study, we characterized the gut phageome in Mexican Americans of South Texas by stool shotgun metagenomic sequencing of 340 subjects, concurrently screened for liver steatosis by transient elastography. Inter-individual variations in the phageome were associated with gender, country of birth, diabetes, and liver steatosis. Early changes in IVD degeneration include reduced cellularity and altered phenotype in nucleus pulposus (NP) cells, leading to diminished capacity for extracellular matrix (ECM) and glycosaminoglycan production. Cell supplementation, particularly with bone marrow-derived human mesenchymal stem cells (MSCs), has been explored to promote ECM synthesis and repair. MSCs offer potential for either direct cell replacement or by secreting factors to support disc repair by restoring biosynthetic activity of remaining endogenous NP cells and ameliorating inflammation. Increasing anthropogenic stressors will likely result in many coral reefs undergoing compositional and functional benthic changes, including towards non-hard coral dominated communities. The decline of reef building species and the increase of alternative (i.e., non reef-building) organisms profoundly affect the functioning of coral reefs and the services provided, but this remains poorly understood, especially for non-algal alternative communities. Skull morphogenesis is a complex, dynamic process involving two different germ layers and progressing to the coordinated, directional growth of individual bones. The mechanisms underlying directional growth toward the apex are not completely understood. Here, a microfluidic chip-based approach is utilized to test whether calvarial osteoblast progenitors undergo haptotaxis on a gradient of Fibronectin1 (FN1) via lamellipodia. Mimicking the embryonic cranial mesenchyme's FN1 pattern, FN1 gradients is established in the chip using computer modeling and fluorescent labeling. Primary mouse calvarial osteoblast progenitors are plated in the chip along an array of segmented gradients of adsorbed FN1. The study performs single-cell …

Tumor-associated neutrophils (TANs) have been shown to promote immunosuppression and tumor progression, and a high TAN frequency predicts poor prognosis in triple-negative breast cancer (TNBC). Dysregulation of CREB-binding protein (CBP)/P300 function has been observed with multiple cancer types. The bromodomain (BRD) of CBP/P300 has been shown to regulate its activity. In this study, we found that IACS-70654, a selective CBP/P300 BRD inhibitor, reduced TANs and inhibited the growth of neutrophil-enriched TNBC models. In the bone marrow, CBP/P300 BRD inhibition reduced the tumor-driven abnormal differentiation and proliferation of neutrophil progenitors. Autophagy is an essential self-degradative and recycling mechanism that maintains cellular homeostasis. Estrogen receptor-related orphan receptors (ERRs) are fundamental in regulating cardiac metabolism and function. Previously, we showed that ERR agonists improve cardiac function in models of heart failure and induce autophagy. Central noradrenergic (NA) neurons are key constituents of the respiratory homeostatic network. NA dysfunction is implicated in several developmental respiratory disorders including Congenital Central Hyperventilation Syndrome (CCHS), Sudden Infant Death Syndrome (SIDS), and Rett Syndrome. The current unchallenged paradigm in the field, supported by multiple studies, is that glutamate co-transmission in subsets of central NA neurons plays a role in breathing control. If true, NA-glutamate co-transmission may also be mechanistically important in respiratory disorders. However, the requirement of NA-derived glutamate in breathing has not been directly tested and the extent of glutamate co-transmission in the central NA system remains uncharacterized. …

Fibrosis represents the uncontrolled replacement of parenchymal tissue with extracellular matrix (ECM) produced by myofibroblasts. While genetic fate-tracing and single-cell RNA-Seq technologies have helped elucidate fibroblast heterogeneity and ontogeny beyond fibroblast to myofibroblast differentiation, newly identified fibroblast populations remain ill defined, with respect to both the molecular cues driving their differentiation and their subsequent role in fibrosis. Using an unbiased approach, we identified the metalloprotease ADAMTS12 as a fibroblast-specific gene that is strongly upregulated during active fibrogenesis in humans and mice. Functional in vivo KO studies in mice confirmed that Adamts12 was critical during fibrogenesis in both heart and kidney. …
