Clear cell renal cell carcinoma (ccRCC) is the most prevalent kidney neoplasm; bone metastasis (BM) develops in 35% to 40% of metastatic patients and results in substantial morbidity and mortality, as well as medical costs. A key feature of ccRCC is the loss of function of the von Hippel-Lindau protein, which enhances angiogenesis via vascular endothelial growth factor release. Consequently, antiangiogenic tyrosine kinase inhibitors (TKI) emerged as a treatment for ccRCC. However, limited data about their efficacy in BM is available, and no systematic comparisons have been performed. We developed mouse models of bone and lung ccRCC tumors and compared …
A Novel Deep Learning Model For Tool Wear Estimation Of Cutting Tools, Ahmed Darwish
A Novel Deep Learning Model For Tool Wear Estimation Of Cutting Tools, Ahmed Darwish
Sustainable Machine Intelligence Journal
In cutting tools, tool wear may result in low-quality surfaces and precision in dimensional measurement. Tool wear may additionally result in fractures and harm to the workpieces over operations. It's critical to track tool wear in life in order to avoid these situations. This research proposes a data-driven deep learning strategy to improve the accuracy of cutting tool wear estimates. This technique combines a Gated Recurrent Unit (GRU) with a Convolutional Neural Network (CNN). In order to circumvent the problems associated with single-domain features that lack appropriate tool status data, a multi-domain combination technique was used to develop candidate features. …

Sustainable Machine Intelligence Journal
Handwritten Arabic character recognition remains a challenging task in pattern recognition due to the inherent complexities of the cursive script and visual similarities between characters. While deep learning techniques have demonstrated promising results in this domain, further enhancements to the model architecture can drive even greater performance improvements. This study introduces a hybrid deep learning approach that combines Convolutional Neural Networks (CNNs) with Bidirectional Recurrent Neural Networks, specifically Bidirectional Long Short-Term Memory (Bi-LSTM) and Bidirectional Gated Recurrent Units (Bi-GRU). By leveraging the strengths of both convolutional and recurrent neural network components, the proposed models are able to effectively capture spatial …

The incidence of metabolic dysfunction-associated steatohepatitis (MASH) is on the rise, and with limited pharmacological therapy available, identification of new metabolic targets is urgently needed. Oxalate is a terminal metabolite produced from glyoxylate by hepatic lactate dehydrogenase (LDHA). The liver-specific alanine-glyoxylate aminotransferase (AGXT) detoxifies glyoxylate, preventing oxalate accumulation. Here we show that AGXT is suppressed and LDHA is activated in livers from patients and mice with MASH, leading to oxalate overproduction. In turn, oxalate promotes steatosis in hepatocytes by inhibiting peroxisome proliferator-activated receptor-α (PPARα) transcription and fatty acid β-oxidation and induces monocyte chemotaxis via C-C motif chemokine ligand 2. In …

T cell is an indispensable component of the immune system and its multifaceted functions are shaped by the distinct T cell types and their various states. Although multiple computational models exist for predicting the abundance of diverse T cell types, tools for assessing their states to characterize their degree of resting, activation, and suppression are lacking. To address this gap, a robust and nuanced scoring tool called T cell state identifier (TCellSI) leveraging Mann-Whitney

Temporal ordering of cellular events offers fundamental insights into biological phenomena. Although this is traditionally achieved through continuous direct observations1,2, an alternative solution leverages irreversible genetic changes, such as naturally occurring mutations, to create indelible marks that enables retrospective temporal ordering3–5. Using a multipurpose, single-cell CRISPR platform, we developed a molecular clock approach to record the timing of cellular events and clonality in vivo, with incorporation of cell state and lineage information. Using this approach, we uncovered precise timing of tissue-specific cell expansion during mouse embryonic development, unconventional developmental relationships between cell …

Invasive Forest Pest Education Through Identification, Reporting, And Children’S Introduction Brochures, Lauren E. Keefe
Department of Entomology: Distance Master of Science Projects
Forest pest insects, whether native or non-native, can be extremely detrimental to the health of a landscape, and can contribute to massive ecological, economic, and cultural losses. Unless directly exposed to the damage these pests cause, many people do not seek to become informed on the insects. A series of brochures was thus created as a collaboration between the online UNL M.S. in Entomology requirements and a need for updated Nebraska Forest Service Forest Health brochures. These brochures focus on invasive forest insect identification and reporting, as well as introducing the concepts of the invasives to children.

The Cancer Genome Atlas (TCGA) and the Cancer Cell Line Encyclopedia (CCLE) are foundational resources in cancer research, providing extensive molecular and phenotypic data. However, large-scale proteomic data across various cancer types for these cohorts remain limited. Here, we expand upon our previous work to generate high-quality protein expression data for approximately 8,000 TCGA patient samples and around 900 CCLE cell line samples, covering 447 clinically relevant proteins, using reverse-phase protein arrays. These protein expression profiles offer profound insights into intertumor heterogeneity and cancer dependency and serve as sensitive functional readouts for somatic alterations. We develop a systematic protein-centered strategy …

Multiple Sclerosis (MS) is a chronic autoimmune demyelinating disease of the central nervous system (CNS) characterized by inflammation, demyelination, and axonal damage. Early recognition and treatment are important for preventing or minimizing the long-term effects of the disease. Current gold standard modalities of diagnosis (e.g., CSF and MRI) are invasive and expensive in nature, warranting alternative methods of detection and screening. Oculomics, the interdisciplinary combination of ophthalmology, genetics, and bioinformatics to study the molecular basis of eye diseases, has seen rapid development through various technologies that detect structural, functional, and visual changes in the eye. Ophthalmic biomarkers (e.g., tear composition, …

The rapid rise of generative AI represents a palpable paradigm shift: models like GPT-3 (and its subsequent iterations), DALL-E, and AlphaCode are becoming foundation models for many other AI-based applications. Foundation models may be used for many different tasks with minimal modification and are positioned to replace task-specific models of AI. This holds tremendous promise in society such as advancing scientific research, rethinking education and training, and creating new types of art and expression. At the same time, however, generative AI has its dark side such as accuracy risk, the threat of large-scale unemployment, potential security vulnerabilities, and AI misbehaviour …

The Origins Of The Major Questions Doctrine, Rachel Rothschild
The Origins Of The Major Questions Doctrine, Rachel Rothschild
Articles
In a series of recent cases, the Supreme Court has invoked the newly named “major questions doctrine” to strike down agency regulations that protect public health and the environment. Several Justices have argued that while the name “major questions” may be new, these decisions are simply the latest iteration in a longstanding effort of the courts to curtail the explosive growth of the administrative state since 1970. The first paradigmatic example of this line of cases is the 1980 “Benzene” case, in which the Supreme Court set aside the Occupational Safety and Health Administration (OSHA)’s new workplace standards for the …

Information Exchange Through Social Networks Links To Adoption Of Child Feeding Practices In Nepal, Mohammad Masudur Rahman
Theses and Dissertations
While there have been notable decreases in child undernutrition (i.e., stunting) over the past few decades, it remains a major concern in Nepal, with 25% of children under the age of five experiencing chronic undernourishment. There is considerable scope for improving nutrition-related behaviors to address this ongoing issue. In Nepal, knowledge regarding appropriate child feeding practices is often inadequate, and thus additional sources of accurate information are required. Information through multiple sources (i.e., interpersonal communication, mass media, and mobile SMS) provides opportunity to reach households who inadequately practice childcare. Information improves knowledge and information exchange (i.e., receiving or sharing) usually …

Sickle cell disease (SCD) causes inflammation that contributes to poor clinical outcomes, including cerebrovascular disease (CVD). CVD can take several forms in SCD, including neurocognitive deficits without visible cerebral infarction. The role of chronic inflammation in cognitive development in SCD has received little attention to-date. We aimed to study novel markers of systemic inflammation: neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte ratio (PLR), and the systemic inflammation index (SII) to describe developmental trends and relationships to cognitive outcomes in SCD at early elementary school-age. Sixty-one children with SCD and no history of stroke were identified who completed an early elementary school age cognitive …
Vascular Calcification In A Mouse Model Of Chronic Kidney Disease And In Human Peripheral Arterial Disease, Breanna Pederson
Vascular Calcification In A Mouse Model Of Chronic Kidney Disease And In Human Peripheral Arterial Disease, Breanna Pederson
Theses and Dissertations
Vascular calcification is a process in which calcium and phosphate crystallize to form hydroxyapatite in the extracellular matrix of blood vessels. There are two forms of arterial calcification: intimal and medial, which occur via different etiologies. Intimal calcification is associated with atherosclerosis and occurs in the layer of endothelial cells adjacent to the artery lumen. Medial arterial calcification (MAC) is independent of atherosclerosis and occurs in the middle, muscular layer of the artery. MAC is strongly associated with aging, diabetes mellitus, and chronic kidney disease (CKD). MAC is frequently in patients with peripheral arterial disease (PAD). Most studies to date …

Impact Of A Mathematics Review On Students' Chemistry Self-Efficacy And Problem-Solving Skills In An Undergraduate Chemistry Course, Antoinette Marie Gayle
Theses and Dissertations
Mathematics is an integral part of chemistry courses, and the application of arithmetic and algebraic knowledge is important for solving calculation-based problems in undergraduate general chemistry. Students begin undergraduate chemistry with varying levels of mathematics skills and experience which may impact their ability to solve problems and understand the content in chemistry courses. Utilizing a convergent mixed methodology, this action research study evaluated the impact of an in-class mathematics review intervention on students’ chemistry self-efficacy and problem-solving abilities in general chemistry. Mathematics review sessions were conducted in undergraduate general chemistry classes with 40 first-year students at a small university college …
Teacher Perception Of International Baccalaureate Middle Years Programme Implementation, Lorene Michelle Lloyd-Peay
Teacher Perception Of International Baccalaureate Middle Years Programme Implementation, Lorene Michelle Lloyd-Peay
Theses and Dissertations
The rural, Title I, underperforming, priority schools in the study encounter several challenges when implementing the International Baccalaureate Middle Years Program (IBMYP) framework. Successful implementation is contingent on how the teachers perceive these challenges. This research aims to understand how teachers' perspective on the IBMYP affects programme implementation. Understanding the experiences and perspectives of teachers increased my and the school district’s ability to support the effective implementation of the IBMYP framework. The research questions focused on gathering data to determine teacher perspectives about the IBMYP, how those perspectives affect programme implementation, and the factors that appear to enhance or impede …
