Breast cancer is the most common cancer type and the major cause of death in females worldwide. For effective breast cancer treatment, systems with promising delivery of anticancer agents is an important strategy. Peptide 18 (P18), a tumor homing peptide, shows high binding affinity towards breast cancer cells. Nanoliposomes are known to have enhanced accumulation ability in tumors with longer systemic circulation. Poly (2-ethyl-2-oxazoline) (PEtOx) polymers conjugated with DOPE were used to prepare PEtOx-DOPE nanoliposomes. BikDD is a mutant form of Bik gene, a member of the BH3-only proapoptotic genes, mimics the constitutively phosphorylated form of the gene. To the …

The definitive demonstration of protein localization on primary cilia has been a challenge for cilia biologists. Primary cilia are solitary thread-like projections that have a specialized protein composition, but as the ciliary structure overlays the cell membrane and other cell parts, the identity of ciliary proteins are difficult to ascertain by conventional imaging approaches like immunofluorescence microscopy. Surface scanning electron microscopy combined with immunolabeling (immuno-SEM) bypasses some of these indeterminacies by unambiguously showing protein expression in the context of the three-dimensional ultrastructure of the cilium. Here, we apply immuno-SEM to specifically identify proteins on the primary cilia of mouse and …

Dysfunctional adipose tissue is believed to promote the development of hepatic steatosis and systemic insulin resistance, but many of the mechanisms involved are still unclear. Lipin 1 catalyzes the conversion of phosphatidic acid to diacylglycerol, the penultimate step of triglyceride synthesis, which is essential for lipid storage. Herein we found that adipose tissue LPIN1 expression is decreased in people with obesity compared with lean subjects, and low LPIN1 expression correlated with multi-tissue insulin resistance and increased rates of hepatic de novo lipogenesis. Comprehensive metabolic and multiomic phenotyping demonstrated that adipocyte-specific Lpin1-/- mice had a metabolically unhealthy phenotype, including liver and …

The disposable soma theory (DST) posits that organisms age and die because of a direct trade-off in resource allocation between reproduction and somatic maintenance. DST predicts that investments in reproduction accentuate somatic damage which increase senescence and shortens lifespan. Here, we directly tested DST predictions in breeding and nonbreeding female C57BL/6J mice. We measured reproductive outputs, body composition, daily energy expenditure, and oxidative stress at peak lactation and over lifetime. We found that reproduction had an immediate and negative effect on survival due to problems encountered during parturition for some females. However, there was no statistically significant residual effect on …

BACKGROUND: The discovery of viruses in small mammalian populations, particularly rodents, has expanded the family Paramyxoviridae. The overlap in habitats between rodents and humans increases the risk of zoonotic events, underscoring the importance of active surveillance. Rodent species, such as Apodemus agrarius, are natural hosts for Paramyxoviridae in the Republic of Korea (ROK). However, it is unknown whether Paramyxoviridae is present in Micromys minutus, another common rodent.
METHOD: Here, we screened M. minutus collected from the Gangwon Province in the ROK for paramyxoviruses using nested polymerase chain reaction and confirm positive samples by next-generation metagenomic sequencing. Complete paramyxovirus genomes were …

Assessing The Climate Readiness Of Physician Education Leaders In Graduate Medical Education, Deborah Simpson, Anne Getzin, Aaron A. Levy, Victoria Gillet
Journal of Patient-Centered Research and Reviews
Climate change affects patient health through an array of exposures, including increasing heatwaves, extreme weather events, poor air quality, and expanding vector-borne illnesses. Physicians are at the forefront of addressing the health consequences of these exposures with patients, and environmental sustainability has become a priority for health care organizations. Accordingly, climate change and health is becoming a critical area for graduate medical education (GME). As GME leaders design and drive education in residency and fellowship programs, understanding those leaders’ baseline knowledge on this topic and its alignment with their organizations’ priorities is an essential step in the development of climate …

Biallelic loss of cyclin-dependent kinase 12 (CDK12) defines a metastatic castration-resistant prostate cancer (mCRPC) subtype. It remains unclear, however, whether CDK12 loss drives prostate cancer (PCa) development or uncovers pharmacologic vulnerabilities. Here, we show Cdk12 ablation in murine prostate epithelium is sufficient to induce preneoplastic lesions with lymphocytic infiltration. In allograft-based CRISPR screening, Cdk12 loss associates positively with Trp53 inactivation but negatively with Pten inactivation. Moreover, concurrent Cdk12/Trp53 ablation promotes proliferation of prostate-derived organoids, while Cdk12 knockout in Pten-null mice abrogates prostate tumor growth. In syngeneic systems, Cdk12/Trp53-null allografts exhibit luminal morphology and immune checkpoint blockade sensitivity. Mechanistically, Cdk12 inactivation …

Purpose: Large cell neuroendocrine carcinoma (LCNEC) is a high-grade neuroendocrine malignancy that, like small cell lung cancer (SCLC), is associated with the absence of druggable oncogenic drivers and dismal prognosis. In contrast to SCLC, however, there is little evidence to guide optimal treatment strategies, which are often adapted from SCLC and non-small cell lung cancer approaches.
Experimental design: To better define the biology of LCNEC, we analyzed cell line and patient genomic data and performed IHC and single-cell RNA sequencing of core needle biopsies from patients with LCNEC and preclinical models.
Results: In this study, we demonstrate that the presence …

ATP-grasp superfamily enzymes contain a hand-like ATP-binding fold and catalyze a variety of reactions using a similar catalytic mechanism. More than 30 protein families are categorized in this superfamily, and they are involved in a plethora of cellular processes and human diseases. Here, we identify C12orf29 (RLIG1) as an atypical ATP-grasp enzyme that ligates RNA. Human RLIG1 and its homologs autoadenylate on an active site Lys residue as part of a reaction intermediate that specifically ligates RNA halves containing a 5’-phosphate and a 3’-hydroxyl. RLIG1 binds tRNA in cells and can ligate tRNA within the anticodon loop in vitro. Transcriptomic …

The Catholic Imaginations Of J.R.R. Tolkien And Oscar Wilde, Frazier A. Johnson
Mythlore: A Journal of J.R.R. Tolkien, C.S. Lewis, Charles Williams, and Mythopoeic Literature
This paper uses terminology from The Catholic Imagination by Andrew Greeley to explore the influence of Catholicism on the writings of Oscar Wilde and J.R.R. Tolkien. Greeley asserts that Catholics see metaphors for the transcendent in matter, value community within a well-ordered hierarchy, and sanctify suffering—qualities that abound in the writings of Wilde and Tolkien. I begin with short biographical sketches of both authors, paying special attention to Wilde’s and Tolkien’s varied involvement with orthodox Catholicism. I then move into a discussion about both authors’ aesthetic philosophies and the similar value they see in art. Next, I detail the authors’ …

At first sight, That Hideous Strength (1945) by C.S. Lewis and the MaddAddam trilogy (2003, 2009, 2013) by Margaret Atwood seem to have little in common. Yet, despite differences of time and place, and approach of the authors, there are startling and generative similarities between the two communities they depict, the Company at St. Anne’s and the God’s Gardeners. In considering the groups' opposition to reductionist technocracy and a shared set of principles, Atwood's MaddAddam trilogy seems to be carrying on the heart of Lewis' dystopic That Hideous Strength as effective ecological and ethical speculative fiction.

Applying C.S. Lewis' metaphor of authors being cathedral-builders to understand intertextual writing (combining influences with innovation) provides a fascinating lens to understand T.H. White's Arthurian work. Exploring how White's The Once and Future King influenced J.K. Rowling's Harry Potter series (particularly her characters Vernon and Dudley Dursley) provides a particularly interesting example of how fantasy authors use intertextuality to create original works.

Factors Affecting Plant Sulphur Requirements In New Zealand Pastoral And Arable Systems, M L. Nguyen, K M. Goh
Factors Affecting Plant Sulphur Requirements In New Zealand Pastoral And Arable Systems, M L. Nguyen, K M. Goh
IGC Proceedings (1977-2023)
This paper reviews major factors affecting sulphur (S) cycling in New Zealand pastoral and arable· systems and identifies areas of research required to improve the efficient use of S fertilisers and soil S reserves. In the grazed pastoral system, S removal in animal products constitutes a small fraction of total S output while S leaching losses and excreta! S transfer losses to stock camps are substantial. Although grazing animals may cause a considerable S loss from the system, they enhance the oxidation of elemental S. Accumulated soil organic S due to the return of animal excreta and long-term annual applications …
An Overview Of The New Zealand Experience In Establishing, Maintaining, And Utilizing White Clover Based Pastures, M B. O'Connor, A M. Bryant, R A. Prestidge
An Overview Of The New Zealand Experience In Establishing, Maintaining, And Utilizing White Clover Based Pastures, M B. O'Connor, A M. Bryant, R A. Prestidge
IGC Proceedings (1977-2023)
Establishment of white clover (Trifolium repens L.) involves the provision of all major and minor elements necessary for clover growth and N fixation (particularly P, S, K, Mo). Nitrogen fertiliser is not necessary. Inoculation of seed with rhizobium is required where white clover has not been grown previously and in difficult environments. Pest control where necessary is enhanced by insecticidal and nematicidal seed coatings on the white clover seed, Barty management is critical and involves regulnr close grazing during the first 6 months. Maintenance of white clover pastures involves annual inputs of fertiliser (P, S, K) with lime at …
Recent Considerations Of Grassland "Soil Quality" In Temperate Regions, S P. Mcgrath, S C. Jarvis
Recent Considerations Of Grassland "Soil Quality" In Temperate Regions, S P. Mcgrath, S C. Jarvis
IGC Proceedings (1977-2023)
The term "soil quality" is increasingly being used in Europe, albeit in an ill-defined way. Perceived quality depends critically on the use being made of the soil and the tolerance of other parts of the environment to substances moving from soils. This paper focuses on issues relevant to the maintenance of quality in temperate grassland soils. Soils are under threat from heavy metals, organic contaminants and radionuclides because they are sinks for many of these substances, thus lending to accumulation. The impact of these contaminants is assessed, along with the management options and relevant legislation to minimise their effects. Most …

Carcinogenicity Of Bracken Fern (Pteridium Spp.) In Relationship To Grazing Ruminants, B L. Smith, D R. Lauren, P P. Embling, M Agnew
IGC Proceedings (1977-2023)
Bracken fern (Pteridium spp.) is the only higher plant which has been shown to cause cancer naturally in animals. Bracken contains ptaquiloside (Pta) a known clastogen and mutagen which is believed to be the major carcinogen involved in the urinary bladder neoplasia. In New Zealand, bracken fern from an area where bovine enzootic haematuria occurs has been shown to have much higher concentrations of Pia than an area where the disease has not been identified. The difference was shown to be related to the bracken rather than the environment and was widespread for the plants in the two areas …

A Slow Release Intra-Ruminal Device For Protection Against Facial Eczema In Sheep, R Munday, E A. Fowke, A M. Thompson, C Wesselink, N R. Towers
IGC Proceedings (1977-2023)
Facial eczema (FE) is a hepatogenous photosensitization disease of ruminants which causes severe losses in agricultural revenue in New Zealand. It is caused by sprophytic, a myotoxin produced by the saprophytic pastoral fungus Pilllomyces chartarum. Zinc (Zn) salts, given to animals before exposure to the toxin, protect against the harmful effects of sporidesmin. Large amounts of Zn are required and dosing must be continued, at regular intervals, throughout the period when animals are exposed to the fungus. While practicable for dairy cattle, these requirements preclude the routine use of Zn for FB prophylaxis in sheep. For these animals, a …
