Inflammatory cytokines such as interleukin-6 (IL-6) and interleukin-12 (IL-12) are central regulators of immune signaling and key biomarkers of disease, yet existing assays for their detection remain slow, invasive, and lack multiplexing ability. This dissertation advances the development of single-walled carbon nanotube (SWCNT) optical nanosensors capable of real-time, multiplexed, and molecularly specific cytokine detection through innovative use of single-stranded DNA (ssDNA) interfaces.
First, an IL-6-specific DNA aptamer was employed as both a dispersing agent and recognition probe for SWCNT fluorescence sensing. Sequence modifications, including anchor domains, truncations, and thermally induced refolding, were systematically tested to optimize sensitivity and selectivity. The …

Evaluating the effectiveness of transcranial direct current stimulation (tDCS) is essential for guiding its integration into therapeutic and performance-enhancement applications. In our laboratory, we investigate the efficacy of tDCS across multiple experimental models, including both animal and human studies. I have contributed significantly to the execution and analysis of these experiments, which include studies in rats and healthy human participants aimed at evaluating whether electrical stimulation of the motor cortex can enhance motor learning. These studies assess improvements in fine motor performance resulting from tDCS. In stroke patients, I contribute to our investigation of tDCS as a rehabilitative intervention, particularly …

Non-invasive electrotherapies offer promising treatments for a wide range of conditions such as pain, headaches, depression, addiction, cognitive decline, wound healing, and drug delivery. However, patient compliance remains low due to the high initial cost, cumbersomeness and inconvenience of existing technology compared to pharmaceuticals. Current electrotherapy devices are often bulky and require users to connect disposable electrodes, initiate programs, and manage charging cycles, which hinders widespread adoption.

Navigating urban environments poses significant challenges for blind and low vision (BLV) individuals, particularly at street intersections where determining when it is safe to cross can be life-threatening. In New York City, where pedestrian fatalities are on the rise and only 2% of intersections are equipped with Accessible Pedestrian Signals (APS), alternative solutions are urgently needed. This thesis proposes an audio-based deep learning approach to support BLV individuals at crosswalks by detecting traffic movement direction and idling states using spatial sound. Adhesively bonded joints present significant potential for reducing structural weight and enhancing performance in modern and future engineering structures. By enabling the elimination or reduction of traditional mechanical fasteners and facilitating the integration of dissimilar materials, adhesives can contribute to more efficient, lightweight designs. However, to fully leverage these benefits—especially in high-performance applications—adhesive materials must exhibit robust behavior under both static and dynamic loading conditions, such as ballistic impact. The blood–brain barrier (BBB) is important in the normal function of the central nervous system (CNS). An altered BBB has been described in various neuropsychiatric disorders. The brain-specific microvascular endothelial cell (BMEC) is an essential structural component of the BBB. Antibiotics are helpful for limiting duration and complications of Salmonella andCampylobacter isolates in selected age groups and people who are immunosuppressed. However, antibiotic-resistant (AR) foodborne pathogens have been on the rise in the United States over the past 10 years and are often linked to international travel (IT). Long-term trends on AR have not been examined in Connecticut (CT). Thirty years have passed since a published comparison of natural language and terms and connectors searching in legal databases, and that analysis included only Westlaw. The current study compares natural language and terms and connectors searching in both Westlaw and Lexis for two hypothetical scenarios. Searchers ran four searches for each hypothetical: Lexis natural language, Westlaw natural language, Lexis terms and connectors, and Westlaw terms and connectors. The results showed no statistically significant difference between natural language and terms and connectors for either hypothetical. This thesis examines period poverty as a consequence of systemic neglect, disproportionately affecting those made vulnerable by intersecting structures of racial, gendered, and economic injustice. Situating menstruation as a public health crisis shaped by policy failure, social stigma, and institutional disregard, this research critiques dominant legal and policy frameworks that approach menstrual equity through a limited scope that overlooks deeper structural inequalities rooted in race and class. This thesis explores how marginalized populations experience legal and welfare systems as sites of surveillance and moral judgment in the context of menstrual equity. Leptospirosis, caused by the spirochetes of the genus Leptospira, is a zoonotic bacterialdisease endemic to tropical regions and lower middle income countries (LMICs). Japanese encephalitis (JE) is a mosquito-borne viral disease that poses a significant public health threat across Asia, including China. Climate change has been increasingly recognized as a key driver of JE transmission dynamics, influencing mosquito vector distribution and virus proliferation. This study aimed to assess how climatic variables affect the ecological suitability for Japanese encephalitis virus (JEV) transmission, using species distribution modeling with MaxEnt.

Sterile Insect Technique (SIT) has emerged as an important tool in the control of vector-borne diseases, providing an alternative to traditional chemical insecticides. Initially based on the irradiation of insects to induce sterility, SIT has proven effective in eradication campaigns, including the cases of the screwworm and tsetse fly. SIT’s primary strengths lie in its species specificity, environmental safety, and the avoidance of pesticide resistance. However, “classic” irradiating SIT is limited by high operational costs, reduced mating competitiveness of sterilized males, and difficulties in addressing high-density vector populations. Chronic hepatitis B (CHB) remains a significant global health burden despite advances in antiviral therapy. Current treatments, including nucleoside analogs (NAs) and pegylated interferon (Peg-IFN), effectively suppress viral replication but rarely achieve functional cure, defined as sustained hepatitis B surface antigen (HBsAg) loss. Emerging therapeutic strategies focus on antigen reduction and immune modulation to overcome the limitations of existing therapies.

1,4-Dioxane, a probable human carcinogen and widespread environmental contaminant, has been consistently associated with liver tumor development in rodents. However, limited research has addressed its role in epigenetic dysregulation. This study investigated epigenetic alterations in liver tissue following chronic exposure to 1,4-dioxane. Male mice of wild-type (WT) and glutamate-cysteine ligase modifier subunit knockout (Gclm-KO) genotypes were exposed to 0, 50, or 500 ppm of 1,4-dioxane in drinking water for six months. qPCR, Western blotting, and immunohistochemistry (IHC) were used to assess changes in DNA methylation-related enzyme gene expression, histone acetylation, and SAM (S-adenosylmethionine) distribution. qPCR results showed significant downregulation of …

Background: Shiga toxin-producing Escherichia coli (STEC) causes over 265,000 infections, 3,600 hospitalizations, and 30 deaths annually in the U.S.. While STEC O157 is the most common serotype, the increased use of culture-independent diagnostic tests (CIDT) led to a rise in non-O157 serotypes detected. Nationally, STEC incidence declined during the COVID-19 pandemic but has since surpassed pre-pandemic levels. Trypanosomiasis, caused by protozoan parasites of the Trypanosoma genus, remains a significant health and veterinary burden in sub-Saharan Africa, with Human African Trypanosomiasis (HAT) and Animal African Trypanosomiasis (AAT) posing ongoing threats to underserved populations. Despite advances in therapeutics, the lack of sensitive and scalable diagnostic tools continues to limit early detection, effective disease management, and evaluation of emerging vaccine candidates. Current diagnostic methods such as light microscopy and serological tests frequently fail to detect low levels of parasitemia during the early stages of infection, thereby delaying intervention and increasing the risk of disease progression and transmission.

Introduction: Advanced age is a primary risk factor for many chronic diseases and conditions; however, age-related immune dysregulation is not well understood. Animal models, particularly those that resemble human age-related physiological changes, are needed to better understand immunosenescence and to improve health outcomes. Here, we explore the utility of the olive baboon (Papio anubis) in studying age-related changes to the immune system and understanding mechanisms of immunosenescence.
Methods: We examined immune cell, inflammatory responses, cytokines, and cortisol levels using hematology and flow cytometry, mitogen stimulation, multiplex cytokine assay, and cortisol immunoassay.

This thesis examines the contemporary artist Patricia Piccinini and her posthuman hybrid creations within the context of a feminist ethic of care. Largely referring to scholarship by the political scientist Joan C. Tronto, I argue that Piccinini expands the definition of an ethic of care to include non-human entities as well as human ones. This thesis examines the role of spiritual beliefs in the literary mode now known as magical realism in Cuban Alejo Carpentier’s The Kingdom of This World (1949), Indian-Born British and American Salman Rushdie’s The Satanic Verses (1988), and a lesser-known novel, Household Saints (1981) by Francine Prose, an American writer. The Kingdom of This World, itself the originator of the mode of magical realism, has allowed me to conclude with Carpentier that magical realism arises from “privileged revelations,” as he notes in the preface to his seminal book. Organisms match their phenotypes to prevailing environmental conditions leading to habitat specific polymorphisms. Polymorphisms arise from genetic divergence, phenotypic plasticity, or a combination of both processes. I examined the presence of habitat-specific polymorphism in Physa acuta snails collected from three streams and their corresponding downstream reservoir. In lotic systems shells were rotund with larger apertures, while those from lentic systems were more elongate with smaller apertures. In a reciprocal transplant lab experiment that crossed flow regime and crayfish predator cues, I tested for the underlying mechanisms responsible for the observed phenotypic differences. Metabolic dysfunction-associated steatotic liver disease (MASLD) is a prevalent liver condition characterized by lipid accumulation and associated inflammation, driven by genetic, environmental, and metabolic factors. Much is still unknown about the mechanisms of the disease, and research on this is important for advancing effective treatments. This study investigated lipid accumulation and oxidative stress responses to varying concentrations of oleic acid (OA) and palmitic acid (PA) in induced pluripotent stem cell-derived hepatocytes (iPSC-Heps) from MASLD and healthy donors. BODIPY staining revealed that MASLD iPSC-Heps exhibit significantly higher lipid accumulation than controls, especially under prolonged OA and PA exposure. Additionally, PLIN2 expression …

The congenital, autosomal recessive disorder combined methylmalonic acidemia and homocystinuria - cblC type, is the most common inborn error of cobalamin (vitamin B12) metabolism. In its early onset form, cblC profoundly impacts fetal development of the central nervous system, hematopoietic system, and other tissues. Previously, mutations in the MMACHC gene, which encodes a protein required for the intracellular trafficking and enzymatic processing of free cobalamin into active coenzyme forms, were found to cause cblC. These coenzymes are required in two metabolic pathways which produce either succinyl-CoA in the mitochondria or methionine in the cytosol. However, due to a lack of …

Popular culture reveals much about the perceived role of lawyers in contemporary life. In this article, I draw lessons from the portrayal of lawyers in Aaron Sorkin's classic television series, The West Wing. As a drama centred around a Democratic presidential administration, Republicans often provide the foil. From time to time, however, the show lionizes what might be termed ‘the good Republican’. That ‘good Republican’ is most often a practicing lawyer whose desire to serve is grounded in duty or faith. In this essay, I use a trio of these characters to explore the role of lawyers in public service. …

Skin carotenoids can be measured non-invasively using spectroscopy methods to provide a biomarker of total dietary carotenoid and carotenoid-rich fruit and vegetable intake. However, the degree to which skin carotenoid biomarkers reflect intakes of specific carotenoids must be determined for specific devices. Previously, findings were mixed regarding the correlation between reflection spectroscopy (RS)-assessed skin carotenoids and individual plasma carotenoid concentrations. The current study expands on prior analyses to examine the cross-sectional associations between adult RS-assessed skin carotenoids and individual carotenoid species intakes and plasma concentrations, controlling for potential covariates. As the public’s awareness of the histories of theft and smuggling that brought many cultural artifacts from their communities of origin to American collections has grown, attitudes toward the ethics of retaining these artifacts has also shifted. This presentation will first consider challenges posed by existing legal remedies available for source countries who seek to reclaim their heritage and then discuss the emerging practice of voluntary repatriations, which occur when the current owner of an artifact returns it to a source country or community even though legal authorities would likely not compel its surrender.
