The Utah Soil Health Program (USHP) developed a five-year project to increase understanding of how best to implement soil health practices into Utah’s diverse farming systems. Fifteen producers were selected to participate in the USHP On-Farm Soil Health Demonstration Project. During 2022, the trial’s first year, the participating farmers and ranchers were interviewed. We begin by discussing participant demographics and operation characteristics. Next, we turn to challenges the producers faced in implementing soil health practices in the IMW and then turn to motivations for the producers to enroll in the USHP trial. This will help (1) document what is currently …

Perennial crops are thought as a solution for enhancing food security and providing ecosystem services under a changing climate, including forage-biomass production, reduced erosion and nutrient leaching, and soil C accumulation. However, the drivers of root decomposition and C allocation in perennial multifunctional forage-biomass buffer-strips as affected by fertility management are poorly elucidated. Thus, study objectives were to assess root decomposition and soil CO2 efflux on switchgrass (Panicum virgatum L.), eastern gamagrass (Tripsacum dactyloides L.), silphium (Silphium integrifolium Michx.), and intermediate wheatgrass Kernza (Thinopyrum intermedium [Host] Barkworth & D.R. Dewey) treated with poultry litter (PL) relative to the unfertilized control. …

Lipid nanoparticles (LNPs) for mRNA delivery have advanced significantly, but LNP-mediated DNA delivery still faces clinical challenges. This study compared various LNP formulations for delivering DNA-encoded biologics, assessing their expression efficacy and the protective immunity generated by LNP-encapsulated DNA in different models. The LNP formulation used in Moderna's Spikevax mRNA vaccine (LNP-M) demonstrated a stable nanoparticle structure, high expression efficiency, and low toxicity. Notably, a DNA vaccine encoding the spike protein, delivered via LNP-M, induced stronger antigen-specific antibody and T cell immune responses compared to electroporation. Single-cell RNA sequencing (scRNA-seq) analysis revealed that the LNP-M/pSpike vaccine enhanced CD80 activation signaling …

Glioblastoma is a highly aggressive primary brain tumor with glioblastoma stem cells (GSCs) enforcing the intra-tumoral hierarchy. Plasma cells (PCs) are critical effectors of the B-lineage immune system, but their roles in glioblastoma remain largely unexplored. Here, we leverage single-cell RNA and B cell receptor sequencing of tumor-infiltrating B-lineage cells and reveal that PCs are aberrantly enriched in the glioblastoma-infiltrating B-lineage population, experience low level of somatic hypermutation, and are associated with poor prognosis. PCs secrete immunoglobulin G (IgG), which stimulates GSC proliferation via the IgG-FcγRIIA-AKT-mTOR axis. Disruption of IgG-FcγRIIA paracrine communication inhibits GSC proliferation and self-renewal. Glioblastoma-infiltrating PCs are …

Soft ticks (Argasidae) of the subgenus Pavlovskyella Pospelova-Shtrom are worldwide distributed parasites of medical importance. However, the systematics of the subgenus are currently under debate because genetic data shows that the group is paraphyletic. Meanwhile, species of Pavlovskyella continue to be discovered. In this study a novel species of the subgenus is described from specimens collected on a fox in central Chile. The larva of this new species differentiates from other Pavlovskyella spp. by having the following combination of characters: subpyriform dorsal plate; 15 pairs of setae, 7 anterolateral, 3 central, and 5 posterolateral, and hypostome with denticles in the …

Glioblastoma (GBM) is the most common primary malignant brain tumor in adults with a poor prognosis despite aggressive therapy. Here, we hypothesized that daily host signaling regulates tumor growth and synchronizes circadian rhythms in GBM. We find daily glucocorticoids promote or suppress GBM growth through glucocorticoid receptor (GR) signaling depending on time of day and the clock genes, Bmal1 and Cry. Blocking circadian signals, like vasoactive intestinal peptide or glucocorticoids, dramatically slows GBM growth and disease progression. Analysis of human GBM samples from The Cancer Genome Atlas (TCGA) shows that high GR expression significantly increases hazard of mortality. Finally, mouse …

Purpose : Retinitis pigmentosa (RP) is the most common form of inherited retinal disease with ~1:5000 people affected worldwide. The substitution of Proline to Histidine (P23H) at position 23 in the rhodopsin protein is the most common form of adRP in North America. We have shown that treating a transgenic P23H human RHO pig model of adRP (TgP23H hRHO) with a gene-editing meganuclease Rho 1-2, recovers rod structure and restores rod function. We ran WT, Rho1-2 treated and untreated TgP23H hRHO (Tg) pigs through an obstacle course to quantify visually guided mobility under light- and dark-adapted conditions using motion analysis …

A Byzantine-Period Monastery And Farmhouse In Nahal Pehar (Giv‘Ot Bar), In The Northern Negev, Nir-Shimshon Paran

'Atiqot

A Byzantine-period monastery and farmhouse were uncovered on the southwestern bank of Naḥal Peḥar, within the modern settlement of Giv‘ot Bar, in the northern Negev. The monastery was well-preserved, attesting to the daily life of the monks in the Negev in the Byzantine period. It was paved with poorly executed mosaic floors, probably laid by the monks themselves. Most of the finds in the monastery were uncovered on the floors, attesting to its orderly abandonment in the late Byzantine or beginning of the Umayyad period (sixth–seventh centuries CE). The monastery seems to have belonged to the widespread ecclesiastical system documented …

Intermediate Bronze Age Fauna From A Suspected Cultic Area At Nahal RefaʼIm, Liora Kolska Horwitz Jan 2025

Intermediate Bronze Age Fauna From A Suspected Cultic Area At Nahal RefaʼIm, Liora Kolska Horwitz

'Atiqot

A small assemblage of faunal remains was recovered from four archaeological contexts excavated on the northern bank of Naḥal Refaʼim. These include Bronze Age rock-cut pits (Area 100), a structure (Area 300), a burial cave (Area 500) and an Ottoman-period shomera (Area 400). The remains represent almost exclusively domestic taxa—sheep, goat, cattle and donkey—some of which exhibit butchery marks, other modifications or burning. The faunal assemblage from the pits, together with other complementary data, may attest that the pits were used as favissae (cultic storage pits) associated with ritual activities at the site.

Intermediate Bronze Age, Middle Bronze Age Ii And Iron Age Ii Pottery From Nahal Refa’Im, Lilly Gershuny

'Atiqot

The pottery assemblage from the Naḥal Refa’im site originated in three different archaeological contexts: pits, structure(s) and a burial cave. These findings attest that the main period of occupation was during the Intermediate Bronze Age. One pit may have been reused for burial in Middle Bronze Age II, and the burial cave was visited in Iron Age II, possibly by the inhabitants of nearby Khirbat er-Ras.

Intermediate And Middle Bronze Age Settlement Remains, Domestic Worship, Pits And Burial Practices In Nahal RefaʼIm, Jerusalem, Shlomit Weksler-Bdolah, Alexander Onn

'Atiqot

Excavations on the northern bank of the Refa’im Valley uncovered four sites: a shaft tomb and building remains with a standing stone dating to the Intermediate Bronze Age; a concentration of eight pits dating to the Intermediate Bronze Age and the Middle Bronze Age II; and an Ottoman-period agricultural watchtower. The building remains and installations were probably part of the Intermediate Bronze Age settlement along Naḥal Refa’im. The standing stone’s prominent location, and the large concentration of pits and installations near it, may support its identification with domestic worship. The thin fill at the bottom of some of the pits …

Light-Microscopic And Spectroscopic Analyses Of The Skin Materials Used In The Tefillin And Mezuzah Slips From The Judean Desert, Yonatan Adler, Theresa Emmerich Kamper

'Atiqot

Among the Dead Sea Scrolls is an assemblage of ancient tefillin and mezuzah slips—ritual artifacts composed of thin animal skins inscribed with passages from the Pentateuch. This paper presents the results of light-microscopic and spectroscopic analyses conducted on 26 tefillin and 8 mezuzot from the Judean Desert caves. These analyses provide the first opportunity to identify the following characteristics on these artifacts: animal species; skin thickness; skin layers present; tannage; side/s of skin with writing. In time, these characteristics took on ritual significance, especially in later rabbinic halakhah. A significant finding is that the ancient tefillin analyzed here were all …

An Iron Age Iib–C Hebrew Stamp Seal With Apotropaic And Prophylactic Properties From The Southwestern Corner Of The Temple Mount, Jerusalem, Filip Vukosavović, Ronny Reich, Navot Rom, Yuval Baruch

'Atiqot

An Iron Age Hebrew iconic stamp seal was uncovered near the southwestern corner of the Temple Mount in Jerusalem. The seal, made of black stone, depicts a winged genie and a personal name, Yehoʽezer son of Hoshaʽyahu. This is the first depiction of the winged genie iconography in the region found in an archaeological excavation. Influenced by Neo-Assyrian art, the seal was created in Jerusalem by a local artist for Hoshaʽyahu, a member of the Judean elite, during the Assyrian Century. Later on, the seal was inherited by his son, Yehoʽezer. In addition to being used as a stamp, the …

Evidence Of Worship In The Rock-Cut Rooms On The Eastern Slope Of The City Of David, Jerusalem, Eli Shukron, Liora Freud, Helena Roth, Reli Avisar, Efrat Bocher

'Atiqot

This paper presents the finds retrieved from a unique series of rock-cut rooms found on the eastern slope of the City of David. These rock-cut rooms stand out in terms of their architectural features, as well as the artifacts discovered within them, all of which reflect on their function as a cultic complex, likely constructed in the Middle Bronze Age, with its final stage in Iron Age IIB. This complex offers profound evidence for the diversity of cultic practices in the capital of the Kingdom of Judah.

The Emergence Of Worship And Religion In The Neolithic Period: Direct And Indirect Evidence From Moẓa (Motza), Hamoudi Khalaily, Anna Eirikh-Rose, Ianir Milevski Jan 2025

The Emergence Of Worship And Religion In The Neolithic Period: Direct And Indirect Evidence From Moẓa (Motza), Hamoudi Khalaily, Anna Eirikh-Rose, Ianir Milevski

'Atiqot

Evidence of worship and religion in the form of architectural remains is rather scanty before the shift in human behavior, from hunter-gatherers to sedentary communities, and the emergence of permanent settlements. Communal and ritual structures first appeared in the Natufian culture (12,500–9400 BCE) and expanded during the Neolithic period (9400–5300 BCE), alongside the treatment of burials linked to ritual related to ancestor worship. The excavations at Motza’s Final PPNB (7100–6700 BCE) layer exposed many non-residential structures, some presumably used for worship and ritual. Some burials, including the one interpreted here as a shaman burial, included objects of symbolic importance. Cultic …

Chromosome 4 Duplication Associated With Strabismus Leads To Gene Expression Changes In Ipsc-Derived Cortical Neurons., Mayra Martinez-Sanchez, William C Skarnes, Ashish Jain, Sampath Vemula, Liang Sun, Shira Rockowitz, Mary C Whitman

Faculty Research 2025

BACKGROUND/OBJECTIVES: Strabismus is the most common ocular disorder of childhood. Three rare, recurrent genetic duplications have been associated with both esotropia and exotropia, but the mechanisms by which they contribute to strabismus are unknown. This work aims to investigate the mechanisms of the smallest of the three, a 23 kb duplication on chromosome 4 (hg38|4:25,554,985-25,578,843).

METHODS: Using CRISPR and bridging oligos, we introduced the duplication into the Kolf2.1J iPSC line. We differentiated the parent line and the line with the duplication into cortical neurons using a three-dimensional differentiation protocol, and performed bulk RNASeq on neural progenitors (day 14) and differentiated …

The field of immunology has traditionally focused on immune checkpoint modulation of adaptive immune cells. However, many malignancies such as glioblastoma are mostly devoid of T cells and rather are enriched with immunosuppressive myeloid cells of the innate immune system. While some immune checkpoint targets are shared between adaptive and innate immunity, myeloid-specific checkpoints could also serve as potential therapeutics. To better understand the impact of immune checkpoint blockade on myeloid cells, we systematically summarize the current literature focusing on the direct immunological effects of PD-L1/PD-1, CD24/Siglec-10, collagen/LAIR-1, CX3CL1/CX3CR1, and CXCL10/CXCR3. By synthesizing the molecular mechanisms and the translational implications, …

Changes In Vertical Stratification Of Neotropical Nymphalid Butterflies At Forest Edges Are Not Directly Caused By Light And Temperature Conditions, Brian K. Oye, Ryan I. Hill Jan 2025

Changes In Vertical Stratification Of Neotropical Nymphalid Butterflies At Forest Edges Are Not Directly Caused By Light And Temperature Conditions, Brian K. Oye, Ryan I. Hill

Pacific Faculty Work

Habitat fragmentation and land use changes threaten neotropical habitats and alter patterns of diversity at forest edges. Like other arthropod assemblages, neotropical fruit-feeding butterfly communities show strong vertical stratification within forests, with some recent work showing its potential role in speciation. At forest edges, species considered to be forest canopy specialists have been observed descending to the forest understory, with the similarity in light conditions between the canopy and understory strata at edges hypothesized to be responsible for this phenomenon. We conducted a study using standardized sampling to document and quantify this edge effect, characterize edge and forest strata, and …

Dealing with refugees, was the late Pope Francis’ legacy, during his pontificate, both as the leader of the Catholic Church and a global figure. This article reviews Francis’ pastoral approach of mercy, especially in his concern and advocacy for migrants and refugees, as expressed through his gestures, statements, thoughts, proposals, and policies. Along with issues of global inequality and ecological damage, Francis believed that forced migration can only be addressed through joint efforts based on recognizing human dignity, solidarity, the need for participation, as well as connectivity and inclusivity that encompasses the entire human family. This article then highlights the …

The human transmembrane protease, serine 2 (TMPRSS2), essential for SARS-CoV-2 entry, is a key antiviral target. Here, we computationally profiled the TMPRSS2-binding affinities of 15 antiviral compounds. Molecular dynamics (MD) simulations for the docked complexes revealed that three compounds exited the substrate-binding cavity (SBC), suggesting noncompetitive inhibition. Of the remaining compounds, five charged ones exhibited reduced binding stability due to competing electrostatic interactions and increased solvent exposure, while seven neutral compounds showed stronger binding affinity driven by van der Waals (vdW) interactions compensating for unfavorable electrostatic effects (including electrostatic interactions and desolvation penalties). Positive and negative hotspot residues were identified …
