This photo series, Beach Finds, is all from the beach of my hometown in Rocky Harbour, Newfoundland, from walks I took in the early spring of 2021. Particularly after storms, there would be an abundance of garbage washed ashore. Upregulated expression of the oncogenic splicing factor TRA2β occurs in human tumors partly through decreased inclusion of its autoregulatory non-coding poison exon (PE). Here, we reveal that low TRA2β-PE inclusion negatively impacts patient survival across several tumor types. We demonstrate the ability of splice-switching antisense oligonucleotides (ASOs) to promote TRA2β-PE inclusion and lower TRA2β protein levels in pre-clinical cancer models. TRA2β-PE-targeting ASOs induce anti-cancer phenotypes and widespread transcriptomic alterations with functional impact on RNA processing, mTOR, and p53 signaling pathways. Surprisingly, the effect of TRA2β-PE-targeting ASOs on cell viability are not phenocopied by TRA2β knockdown. Mechanistically, we find that the …

Domestic violence is a prevailing issue in the United States for both men and women, growing exponentially during COVID-19. In the law, there are two approaches to remedy this issue. The first being a criminal conviction of the abuser, which requires witness testimony and is held to a higher burden of proof than the other. The other approach is for the victim to obtain a protective order against the abuser. However, in many states, protective orders are only available to victims once the abuser physically attacks them, despite there being reliable signs of abuse before physical violence occurs. Managing the spread of introduced invasive species relies on educating recreationists that currently or potentially harbor such species. By partnering with a local brewery, we developed the Lagers for Lakes Draft Series to facilitate outreach about aquatic invasive species in Erie, Pennsylvania. To date, Lagers for Lakes drafts and associated launch events have reached over 230 individuals and increased awareness of invasive species by 86%. With the materials provided here and the lessons learned from these events, we hope similar outreach efforts can be developed elsewhere to educate and minimize the spread of invasive species.

The network of proteins at the interface between cell-cell adherens junctions and the actomyosin cytoskeleton provides robust yet dynamic connections that facilitate cell shape change and motility. While this was initially thought to be a simple linear connection via classic cadherins and their associated catenins, we now have come to appreciate that many more proteins are involved, providing robustness and mechanosensitivity. Defining the full set of proteins in this network remains a key objective in our field. Proximity proteomics provides a means to define these networks. Discrimination is a social adversity that is linked to several age-related outcomes. However, the molecular drivers of these observations are poorly understood. Social adverse factors are associated with proinflammatory and interferon gene expression, but little is known about whether additional genes are associated with discrimination among both African American and White adults. In this study, we examined how perceived discrimination in African American and White adults was associated with genome-wide transcriptome differences using RNA sequencing. Perceived discrimination was measured based on responses to self-reported lifetime discrimination and racial discrimination. A 47-year-old male with a history of immunosuppression and recent intensive care unit admission presented with progressive orbital swelling and pain. Blood cultures grew Trichosporon asahii and Candida guilliermondii, with matching positive cultures from peripheral and central venous samples. Given his fungemia and worsening orbital involvement, induction therapy with amphotericin B and isavuconazole was initiated. Within weeks, clinical improvement was noted, prompting a transition to long-term consolidation therapy with oral posaconazole and isavuconazole. At follow-up, the patient demonstrated sustained clinical stability with no recurrence of infection. Orbital involvement from fungemia due to T. asahii and C. guilliermondii is rare, and …

Offal Wildlife Watching is a participatory science project that engages deer hunters in wildlife research. The goal of the project is to better understand the ecology of scavenger species that visit deer gut piles across Minnesota. We asked hunters to deploy remote cameras at white-tailed deer gut piles that resulted from their freshly killed deer. Thus far 117 hunters have contributed images and recorded at least 49 different scavenger species. This project is a natural fit in Extension programs and can easily be adapted to other hunted species, biomes, and locations.

This article shares research outcomes and recommendations for replication of a Food, Agriculture and Marine Ecosystems (FAME) pilot project that successfully supported high school aged youth involved in a new 4-H special interest program. The goal of the program was to recruit and engage underserved youth in STEM learning through the production of food systems video stories in partnership with university plant scientists and science communication researchers and storytellers. This pilot offers an experiential method for underserved youth to engage in STEM learning through science storytelling that features youths' own bio-cultural food system knowledge.

Extension’s evolving role in urban food production will require intensive reflexivity and ongoing collaboration. Extension educators around the country have already made progress in engaging with both the social and horticultural sides of urban agriculture. Designed appropriately, urban food systems hold the potential for healthy food access, community and environmental resilience, and economic prosperity (Rangarajan & Riordan 2019). Moving forward, we offer recommendations for Extension staff to apply within their institutions and beyond. Specifically, we urge Extension to prioritize the following: 1) mediate the rural/urban dichotomy, 2) tackle structural and institutional power dynamics, and 3) intensify strategies for community resilience.

Background: Cyclin-dependent kinase 4/6 inhibitors (CDK4/6is) in combination with endocrine therapy are the standard treatment for patients with hormone receptor-positive, HER2-negative metastatic breast cancer (mBC). Despite the efficacy of CDK4/6is, intrinsic resistance occurs in approximately one-third of patients, highlighting the need for reliable predictive biomarkers.
Methods: Single-cell RNA sequencing analyzed metastatic tumors from HR+/HER2- mBC patients pre-CDK4/6i treatment at baseline (BL) and/or at disease progression. BL samples were from CDK4/6i responders (median progression-free survival [mPFS] = 25.5 months), while progressors were categorized as early-progressors (EP, mPFS = 3 months) and late-progressors (LP, mPFS = 11 months). Native mountain goats must cross U.S. Highway 2 in order to gain access to a natural salt lick along the southern boundary of Glacier National Park, Montana. Prior to 1981, goats approached through forest cover and then crossed over the highway. Reconstruction of the highway incorporated two goat underpasses which substantially altered the migration pattern. Large numbers of goats now utilize an open, south-facing slope, and cause extensive erosion. Repeat photography illustrates the extent of erosion over the period since 1981 , and provides estimates of the amount of sediment removed. Erosion rates are startlingly high, and are a direct …

Background: Endometriosis, a poorly studied gynecological condition, is characterized by the presence of ectopic endometrial lesions resulting in pelvic pain, inflammation, and infertility. These associated symptoms contribute to a significant burden, often exacerbated by delayed diagnosis. Current diagnostic methods involve invasive procedures, and existing treatments provide no cure.
Methods: Microbiome-metabolome signatures in stool samples from individuals with and without endometriosis were determined using unbiased metabolomics and 16S bacteria sequencing. Functional studies for selected microbiota-derived metabolites were conducted in vitro using patient-derived cells and in vivo by employing murine and human xenograft pre-clinical disease models.

Adaptive immunity is critical in eliminating tumors, but cancer-intrinsic factors can subvert this function. Melanoma antigen-A4 (MAGE-A4), a cancer-testis antigen, is expressed in solid tumors and correlates with poor survival, but its role in tumorigenesis and antitumor immunity remains unclear. We found that expression of MAGE-A4 was highly associated with the loss of PTEN, a tumor suppressor, in human non–small cell lung cancers (NSCLC). Here, we show that constitutive expression of human MAGE-A4 with Pten loss in mouse airway epithelia results in metastatic adenocarcinoma. In this paper, we present the implementation of the Cha-Cheon Identity-Based Signature (IBS) scheme to enhance secure data transfer in cloud computing environments. Cloud computingrely on traditional Public Key Infrastructure (PKI) systems, which is burdened by certificate management infrastructure. The primary focus of this research to simplify key and certificate management by leveraging identity-based elliptic curve cryptography (ECC) within the Cha-Cheon IBS framework. We show that the proposed IBS solution integrates seamlessly with Amazon Web Services (AWS), utilizing services like S3 for secure data storage and KMSfor key management. Background: Kidney proton- and sodium-dependent monocarboxylate transporters (MCT/SMCT) are involved in the renal reabsorption of substrates, and thus factors involved in their regulation may have pharmacokinetic implications. Previous studies have demonstrated sex hormone-dependent regulation of MCTs and SMCTs in tissues involved in drug disposition. The present study evaluates the impact of puberty on renal MCT/SMCT expression with ovariectomy and castration conducted before puberty, removing the initial exposure to sex hormones. Methods: Male and female rats were castrated or ovariectomized before puberty (4 weeks of age), and subsequently treated with testosterone, 17β-estradiol, progesterone, or both 17β-estradiol and progesterone for 21 days …

Despite media outcry and societal outcry, gun violence on college campuses in the United States continues to be an unresolved problem. Prior to on-campus shootings, college campuses were safe locations that were conducive to learning. Sadly, this is no longer the case. The purpose of this qualitative study was to identify the key characteristics of mass shootings on college campuses so that officials would be provided with information that would enable them to create safer campuses. Inflammation is a significant driver of ischemic stroke pathology in the brain. To identify potential regulators of inflammation, we performed single-cell RNA sequencing (scRNA-seq) of young and aged mouse brains following stroke and found that interferon alpha-inducible protein 27 like 2 A (Ifi27l2a) was significantly up-regulated, particularly in microglia of aged brain. Ifi27l2a is induced by interferons for viral host defense and has been linked with pro-inflammatory cellular mechanisms. However, its potential role in neurodegeneration is unknown. Using a combination of cell culture, experimental stroke models in mice, and human autopsy brain samples, we demonstrated that induction of Ifi27l2a occurs …

This study aimed to determine the pharmacokinetics of free and liposomal levamisole in sheep following single oral (O),intramuscular (IM), and subcutaneous (SC) administrations at a dose of 7.5 mg/kg. The study developed a levamisole liposome andinvestigated relevant parameters: particle size, zeta potential, polydispersity index, encapsulation efficiency, pH, and morphology. TheBangham method, a thin-film hydration technique, was employed to prepare the liposomal levamisole. Blood samples were collectedby jugular venipuncture in heparinized tubes immediately before each free and liposomal levamisole treatment and at 2.5, 5, 10, 15,30, 60, 90, 120, 240, 360, and 480 min and 12, 16, and 24 h after …

Aberrant Homeodomain-Dna Cooperative Dimerization Underlies Distinct Developmental Defects In Two Dominant Crx Retinopathy Models, Yiqiao Zheng, Gary D Stormo, Shiming Chen

Paired-class homeodomain (HD) transcription factors (TFs) play essential roles in vertebrate development, and their mutations are linked to human diseases. One unique feature of a paired-class HD is cooperative dimerization on specific palindrome DNA sequences. Yet, the functional significance of HD cooperative dimerization in animal development and its dysregulation in diseases remains elusive. Using the retinal TF cone-rod homeobox (CRX) as a model, we have studied how blindness-causing mutations in the paired HD, p.E80A and p.K88N, alter CRX's cooperative dimerization, leading to gene misexpression and photoreceptor developmental deficits in dominant manners. CRX
