The biguanide metformin attenuates mitochondrial oxidation and is proposed as an anti-cancer therapy. However, recent clinical studies suggest increased proliferation and fatty acid β-oxidation (FAO) in a subgroup of patients with breast cancer (BC) after metformin therapy. Considering that FAO can activate Src kinase in aggressive triple-negative BC (TNBC), we postulate that low-dose biguanide-driven AMPK-ACC-FAO signaling may activate the Src pathway in TNBC. The low bioavailability of metformin in TNBC xenografts mimics metformin's in vitro low-dose effect. Pharmacological or genetic inhibition of FAO significantly enhances the anti-tumor properties of biguanides. Lower doses of biguanides induce and higher doses suppress Src …

Obesity is a multifaceted disease that poses a significant public health challenge. Recent discoveries in understanding the biological pathways that regulate satiety and metabolism have led to a shift in the treatment paradigm for obesity. Thus, the gap between pharmacological and surgical interventions has diminished. The latest approved anti-obesity medications are achieving weight loss comparable to surgery. These GLP-1 analog-based therapies not only cause substantial weight loss but also improve obesity-associated comorbidities. Background/aimOsteobiologics are increasingly used in orthopedics and spine surgery to facilitate bone healing and prevent non-union. Synthetic osteobiologics are artificial materials crafted in laboratories that aim to replicate the natural composition and functionality of bone. Notable examples, such as calcium phosphate and calcium sulfate, are engineered to mirror the mineral aspect of bone. They mimic human bone functionality, exhibiting osteoconductive, osteoinductive, and osteogenic properties. These characteristics promote cell attachment, migration, recruitment, and differentiation. Consequently, synthetic osteobiologics (osteoconductive grafts) have been introduced in bone fracture repair. Background/aim: The objective of this study is to evaluate the effect of empagliflozin on cyclophosphamide-induced neurotoxicity in rats.Materials and methods: A total of 32 male Wistar rats were separated into 4 groups as 8 rats each: control group, cyclophosphamide group, empagliflozin group and the cyclophosphamide plus empagliflozin group. At the end of the experiment rats' brains were removed for biochemical analysis of oxidative stress parameters (malondialdehyde, total sulfhydryl, total oxidant status and total antioxidant capacity) and histopathological analysis.Results: Total sulfhydryl levels were increased in group empagliflozin and group empagliflozin and cyclophosphamide compared to the cyclophosphamide group but did not improve …

Background/aimCarotid artery is the majority artery for brain feeding. Carotid Bodies (CBs) are located in carotid bifurcation and regulate body fluid pH. We aimed to evaluate the changes in CB-related pH levels as a consequence of degeneration induced by the ligation of both Common Carotid Arteries (BCCAL).Materials and methodsWe included 23 adult male New Zealand rabbits. A control group (n= 5); a sham group (0.5 mL saline injected n=5) and the main study group (subjected to the BCCAL procedure, n = 13). Background/aimLaparoscopic surgery in gynecology has advantages of reduced scarring, less postoperative pain, faster recovery and lower risk of complications. Despite these adavantages, shoulder pain still remains a major concern affecting upto 90% of patients undergoing gynecologic laparoscopic surgery. Background/aim: The aim of this study is to investigate the genotype-phenotype correlation in Sanfilippo type B patients in terms of bone formation/resorption parameters, and to determine the release/inhibition of biomarkers accompanying osteoporosis.Materials and Methods: Plasma levels of Osteoprotegerin, Matrix metalloproteinases (MMP2, MMP9), TIMP-1, TIMP-2, BALP, Cathepsin K, and TRACP-5b were examined by ELISA method in MPS IIIB patients. This study is the first to compare the protective effects of estradiol and ascorbic acid against malathion-induced nephrotoxicity through histopathological assessment and molecular docking analyses.

[Objective] A molecular -imprinted electrochemical sensor was developed to detect ceftiofur sodium in milk.[Methods] Firstly,a molecular -imprinted film was prepared on the surface of a gold electrode by electropolymerization with ceftiofur sodium as a template molecule and acrylamide as a functional monomer in phosphate buffer solution.The electrochemical performance of the sensor was characterized by differential pulse voltammetry and cyclic voltammetry,and the construction parameters and detection conditions were optimized for the sensor.Under the optimal conditions,the sensor was used to quantitatively determine the content of ceftiofur sodium in milk.[Results]] The sensor constructed under the optimal conditions exhibited a good linear detection range …

Allostatic load (AL), the cumulative physiological burden of chronic stress, is a significant health problem, and it has been associated with adverse health outcomes, including cognitive functioning. There is a paucity of literature examining associations between AL and incident dementia in the United States. This quantitative study used data from the United States Health and Retirement Study cohort (2016-2020) to examine the association between AL quartile and incident dementia among participants who were aged 56 and older at baseline. Multiple myeloma is a clonal plasma cell (PC) dyscrasia that arises from precursors and has been studied utilizing approaches focused on CD138+ cells. By combining single-cell RNA sequencing (scRNA-seq) with scB-cell receptor sequencing (scBCR-seq), we differentiate monoclonal/neoplastic from polyclonal/normal PCs and find more dysregulated genes, especially in precursor patients, than we would have by analyzing bulk PCs. To determine whether this approach can identify oncogenes that contribute to disease pathobiology, mitotic arrest deficient-2 like-1 (MAD2L1) and S-adenosylmethionine synthase isoform type-2 (MAT2A) are validated as targets with drug-like molecules that suppress myeloma growth in preclinical models. Lynch syndrome (LS), caused by inherited mutations in DNA mismatch repair genes, including MSH2, carries a 60% lifetime risk of developing endometrial cancer (EC). Beyond hypermutability, mechanisms driving LS-associated EC (LS-EC) remain unclear. We investigated MSH2 loss in EC pathogenesis using a mouse model (PR-Cre Msh2LoxP/LoxP, abbreviated Msh2KO), primary cell lines, human tissues, and human EC cells with isogenic MSH2 knockdown. By 8 months, 58% of Msh2KO mice developed endometrial atypical hyperplasia (AH), a precancerous lesion. Extracellular vesicles (EVs) are cell derived nanovesicles which are implicated in both physiological and pathological intercellular communication, including the initiation, progression, and metastasis of cancer. The exchange of biomolecules between stromal cells and cancer cells via EVs can provide a window to monitor cancer development in real time for better diagnostic and interventional strategies. In addition, the process of secretion and internalization of EVs by stromal and cancer cells in the tumor microenvironment (TME) can be exploited for delivering therapeutics. Gut mucosae are composed of stratified layers of microbes, a selectively permeable mucus, an epithelial lining, and connective tissue homing immune cells. Studying cellular and chemical interactions between the gut mucosal components has been limited without a good model system. We have engineered a three-dimensional (3D) multi-cellular co-culture system we coined “3D Flipwell system” using cell culture inserts stacked against each other. This system allows an assessment of the impact of a gut mucosal environmental change on interactions between gut bacteria, epithelia, and immune cells. This study explores the relationship between circadian rhythms and human performance across three key domains: cardiovascular endurance, vestibular function, and strength. Through the review of data obtained through performance testing at two different time points along with 24-hour physiological tracking, this study attempts to find correlations between circadian function and physical performance.

Integrated science, technology, engineering, and mathematics (STEM) education provides opportunities for interdisciplinary collaborations. To explore the nature of interdisciplinary collaborations, we investigated the disciplinary identity of 11 secondary preservice teachers (PSTs) enrolled in a semester-long secondary integrated STEM methods course. The pedagogical approach in the course aimed to provide secondary PSTs with the opportunity to experience interdisciplinary collaboration, where secondary PSTs from different disciplines collaborate on designing and implementing integrated STEM curricula. Through positioning theory, we analyzed the PSTs’ discussion during each team’s initial collaboration meeting. A new species Parabuthus starhai sp. n. from Somalia (Puntland) is described. Combination of two characters, pectine teeth number 56–59 in both sexes and pedipalp chela length/ width ratio 3.65 in male and 5.47 in female differ P. starhai sp. n. from all other species in the region. Included is also a map of distribution of Parabuthus species in the Horn of Africa.
