Tumor necrosis factor (TNF) receptor-associated factor (TRAF)-interacting protein with forkhead-associated domain B (TIFAB), an inhibitor of NF-κB signaling, plays critical roles in hematopoiesis, myelodysplastic neoplasms, and leukemia. We previously demonstrated that Tifab enhances KMT2A::MLLT3-driven acute myeloid leukemia (AML) by either upregulating Hoxa9 or through ubiquitin-specific peptidase 15-mediated downregulation of p53 signaling. In this study, we show that Tifab deletion in KMT2A::MLLT3-induced AML impairs leukemia stem/progenitor cell (LSPC) engraftment, glucose uptake, and mitochondrial function. Gene set enrichment analysis reveals that Tifab deletion downregulates MYC, HOXA9/MEIS1, mTORC1 signaling, and genes involved in glycolysis and oxidative phosphorylation. Post-surgical spinal infection occurs in up to 20 % of patients, despite aggressive peri-operative antibiotic treatments. To improve prophylaxis, we have designed and evaluated an ultrasound-activated prophylactic antibiotic release system to combat post-surgical bacterial survival. Polylactic acid (PLA) clips (1 cm3) were 3D-printed with an interior reservoir (0.8 cm3) for carrying drug payload, specifically vancomycin (VAN). Under IACUC approval, clips were surgically implanted into the spines of sheep (n = 9) and swine (n = 2) by removing the spinous process at several levels of the lumbar spine. Metabolism plays a key role in the maintenance of sleep/wake states. Brain lactate fluctuations are a biomarker of sleep/wake transitions, where increased interstitial fluid (ISF) lactate levels are associated with wakefulness and decreased ISF lactate is required for sleep. ATP-sensitive potassium (KATP) channels couple glucose-lactate metabolism with excitability. Using mice lacking KATP channel activity (e.g., Kir6.2−/− mice), we explored how changes in glucose utilization affect cortical electroencephalography (EEG) activity and sleep/wake homeostasis. In the brain, Kir6.2−/− mice shunt glucose toward glycolysis, reducing neurotransmitter biosynthesis and dampening cortical EEG activity. Kir6.2−/− mice spent more time …

BACKGROUND: Although premature aging is a significant concern in adult survivors of childhood cancer, little is known about the role of diet in premature aging in this vulnerable population. Therefore, we examined whether dietary patterns specific to childhood cancer survivors are associated with premature aging.
METHODS: This cross-sectional study included 2904 adult survivors of childhood cancer (mean age = 31 years, SD = 8 years) in the St. Jude Lifetime Cohort. Diet was assessed using a food frequency. Better Generalization Through Distraction? Concurrent Load Reduces The Size Of The Inverse Base-Rate Effect, Lenard Dome, Andy J. Wills

The inverse base-rate effect (IBRE) is an irrational phenomenon in predictive learning. It occurs when people try to generalize what they have experienced to novel and ambiguous events. This irrational generalization manifests as a preference for rare, unlikely outcomes in the face of ambiguity. At least two formal mathematical models of this irrational preference (EXIT, NNRAS) lead to a counter-intuitive prediction: the effect reduces under concurrent load. We tested this prediction across two experiments (N1 = 72, Mage = 20.12; N2 = 160, Mage = 20.88). We confirm the prediction, but only when participants were under an obvious time constraint. …

Enteric pathogen rotavirus (RV) primarily infects mature enterocytes at the tips of the intestinal villi; however, the role of secretory Paneth and goblet cells in RV pathogenesis remains unappreciated. Atoh1 knockout mice (Atoh1cKO) were used to conditionally delete Paneth, goblet, and enteroendocrine cells in the epithelium to investigate the role of secretory cells in RV infection. Unexpectedly, the number of infected enterocytes and the amount of RV shedding in the stool were greatly decreased following secretory cell deletion. Resistance to RV infection persisted for 7 days after virus inoculation, and Atoh1 knockout mice co-housed with infected wild-type mice were uninfected, …

Background: Small nucleolar RNAs (snoRNAs) are non-coding RNAs that function in ribosome and spliceosome biogenesis, primarily by guiding modifying enzymes to specific sites on ribosomal RNA (rRNA) and spliceosomal RNA (snRNA). However, many orphan snoRNAs remain uncharacterized, with unidentified or unvalidated targets, and studies on additional snoRNA-associated proteins are limited.
Results: We adapted an enhanced chimeric eCLIP approach to comprehensively profile snoRNA-target RNA interactions using both core and accessory snoRNA-binding proteins as baits. Using core snoRNA-binding proteins, we confirmed most annotated snoRNA-rRNA and snoRNA-snRNA interactions in mouse and human cell lines and called novel, high-confidence interactions for orphan snoRNAs. The transcription factor carbohydrate response element binding protein (ChREBP) activates genes of glucose, fructose, and lipid metabolism in response to carbohydrate feeding. Integrated transcriptomic and metabolomic analyses in rats with GalNac-siRNA-mediated suppression of ChREBP expression in liver reveal other ChREBP functions. GalNac-siChREBP treatment reduces expression of genes involved in coenzyme A (CoA) biosynthesis, with lowering of CoA and short-chain acyl-CoA levels. Despite suppression of pyruvate kinase, pyruvate levels are maintained, possibly via increased expression of pyruvate and amino acid transporters. In addition, expression of multiple anaplerotic enzymes is decreased by GalNac-siChREBP treatment, affecting TCA cycle intermediates. Finally, GalNAc-siChREBP treatment suppresses …

Treatment with antibiotics is a major risk factor for Clostridioides difficile infection, likely due to depletion of the gastrointestinal microbiota. Two microbiota-mediated mechanisms thought to limit C. difficile colonization include the conversion of conjugated primary bile salts into secondary bile salts toxic to C. difficile growth and competition between the microbiota and C. difficile for limiting nutrients. Using a continuous flow model that simulates the nutrient conditions of the distal colon, we investigated how treatment with 6 clinically used antibiotics influenced susceptibility to C. difficile infection in 12 different microbial communities cultivated from healthy individuals. Mutation or deletion of the deubiquitinase USP7 causes Hao-Fountain syndrome (HAFOUS), which is characterized by speech delay, intellectual disability, and aggressive behavior and highlights important unknown roles of USP7 in the nervous system. Here, we conditionally delete USP7 in glutamatergic neurons in the mouse forebrain, triggering disease-relevant phenotypes, including sensorimotor deficits, impaired cognition, and aggressive behavior. Although USP7 deletion induces p53-dependent neuronal apoptosis, most behavioral abnormalities in USP7 conditional knockout mice persist following p53 loss. The COVID-19 pandemic of 2020 has had rippling effects on a plethora of industries, with outdoor tourism being one of the greatest examples. As United States citizens left their homes, they yearned for their natural resources, leading to a palpable uptake in visitation as public parks reopened. While there is municipal, county, state, and federal lands accessible to visitation, the following commentary is focused specifically on National Wildlife Refuges (Federal). Familial roles, household work distribution, and opinions concerning the operation of a household have been a popular topic of discussion within recent years. Many people push for fathers and mothers to take on equal amounts of housework and childcare duties, but previous research suggests that this often fails to happen. This paper uses 2022 survey data to investigate how demographic characteristics can predict who does the chores in the home, and how an individual believes a home should operate. This study investigates the airflow properties around the airfoil using numerical simulation, addressing the longstanding challenge of accurately predicting transition phenomena in turbulent flows. The importance of this research lies in its potential to improve the design and performance of airfoils in various engineering applications, such as wind turbines, and aircraft. A custom 2-dimensional airfoil model was created using Airfoil instruments, featuring a 6% maximum curvature, 40% curvature position, and 20% thickness. COMSOL Multiphysics software performed computations at a velocity of over 19 hours, with data analyzed at 60-minute intervals. The Transition turbulent model was employed to simulate the airflow …

Tom Bombadil, with his iconic singing―”Hey! Come merry doll! Derry doll and merry-o!―has captivated scholars worldwide, due to the enigmatic nature Tolkien intentionally left unexplained. This deliberate mystery has fuelled extensive debate, leading many to explore possible mythological and cultural frameworks that might decode Bombadil’s origins. Among these hypotheses, a Celtic interpretation has often been dismissed despite its potential relevance. Pengaturan kepemilikan dan penggunaan sumber daya alam di luar jurisdiksi nasional selalu menjadi perebutan dan perdebatan antara negara maju dan negara berkembang. Sampai saat ini sudah disahkan tiga rezim hukum internasional khusus yang berlaku untuk tiga wilayah sumber daya alam, yaitu Antartika, Laut Bebas khusus Dasar Laut Dalam, dan Antariksa khususnya Bulan. Ketiga rezim tersebut menggunakan konsepsi ‘common heritage of mankind/warisan bersama umat manusia’ sebagai prinsip utama pengaturannya.
